| 2 | Impact of dissolved organic matter on the photolysis of the ionizable antibiotic norfloxacin显示文摘Norfloxacin(NOR), an ionizable antibiotic frequently used in the aquaculture industry, has aroused public concern due to its persistence, bacterial resistance, and environmental ubiquity.Therefore, we investigated the photolysis of different species of NOR and the impact of a ubiquitous component of natural water — dissolved organic matter(DOM), which has a special photochemical activity and normally acts as a sensitizer or inhibiter in the photolysis of diverse organics; furthermore, scavenging experiments combined with electron paramagnetic resonance(EPR) were performed to evaluate the transformation of NOR in water. The results demonstated that NOR underwent direct photolysis and self-sensitized photolysis via hydroxyl radical(U OH) and singlet oxygen(1O2) based on the scavenging experiments. In addition, DOM was found to influence the photolysis of different NOR species, and its impact was related to the concentration of DOM and type of NOR species. Photolysis of cationic NOR was photosensitized by DOM at low concentration, while zwitterionic and anionic NOR were photoinhibited by DOM, where quenching of U OH predominated according to EPR experiments, accompanied by possible participation of excited triplet-state NOR and1O2. Photo-intermediate identification of different NOR species in solutions with/without DOM indicated that NOR underwent different photodegradation pathways including dechlorination, cleavage of the piperazine side chain and photooxidation, and DOM had little impact on the distribution but influenced the concentration evolution of photolysis intermediates. | 4 | IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. | 6 | mTOR signaling promotes stem cell activation via counterbalancing BMP-mediated suppression during hair regeneration显示文摘Hair follicles(HFs)undergo cycles of degeneration(catagen),rest(telogen),and regeneration(anagen)phases.Anagen begins when the hair follicle stem cells(HFSCs)obtain sufficient activation cues to overcome suppressive signals,mainly the BMP pathway,from their niche cells.Here,we unveil that mTOR complex 1(mTORC1)signaling is activated in HFSCs,which coincides with the HFSC activation at the telogen-to-anagen transition.By using both an inducible conditional gene targeting strategy and a pharmacological inhibition method to ablate or inhibit mTOR signaling in adult skin epithelium before anagen initiation,we demonstrate that HFs that cannot respond to mTOR signaling display significantly delayed HFSC activation and extended telogen.Unexpectedly,BMP signaling activity is dramatically prolonged in mTOR signaling-deficient HFs.Through both gain-and loss-of-function studies in vitro,we show that mTORC1 signaling negatively affects BMP signaling,which serves as a main mechanism whereby mTORC1 signaling facilitates HFSC activation.Indeed,in vivo suppression of BMP by its antagonist Noggin rescues the HFSC activation defect in mTORC1-null skin.Our findings reveal a critical role for mTOR signaling in regulating stem cell activation through counterbalancing BMP-mediated repression during hair regeneration. | Zhili Deng Xiaohua Lei Xudong Zhang Huishan Zhang Shuang Liu Qi Chen Huimin Hu Xinyue Wang Lina Ning Yujing Cao Tongbiao Zhao Jiaxi Zhou Ting Chen Enkui Duan | 2015 | Journal of Molecular Cell Biology2015,7,1: | 3 |

| 10 | mTOR regulates TLR-induced c-fos and Th1 responses to HBV and HCV vaccines显示文摘Although IL-12 plays a critical role in priming Th1 and cytotoxic T lymphocyte(CTL) responses, Toll-like receptor(TLR) signaling only induces low amounts of IL-12 in dendritic cells and macrophages, implying the existence of stringent regulatory mechanisms. In this study, we sought to uncover the mechanisms underlying TLR-induced IL-12 expression and the Th1 response. By systemic screening, we identified a number of protein kinases involved in the regulation of TLRinduced IL-12 expression. In particular, PI3 K, ERK, and m TOR play critical roles in the TLR-induced Th1 response by regulating IL-12 and IL-10 production in innate immune cells. Moreover, we identified c-fos as a key molecule that mediates m TOR-regulated IL-12 and IL-10 expression in TLR signaling. Mechanistically, m TOR plays a crucial role in c-fos expression, thereby modulating NFκB binding to promoters of IL-12 and IL-10. By controlling the expression of a special innate gene program, m TOR can specifically regulate the TLR-induced T cell response in vivo. Furthermore, blockade of m TOR by rapamycin efficiently boosted TLR-induced antigen-specific T and B cell responses to HBV and HCV vaccines. | 11 | Electron dynamics and wave activities associated with mirror mode structures in the near-Earth magnetotail显示文摘We report the observation of mirror mode structures by Cluster spacecraft at around X^-16 RE in the Earth’s magnetotail.The wavelength of the mirror structure is larger than 7000 km,corresponding to tens of ion gyroradii.Features of the mirror structures are similar to those detected in the magnetosheath:the anti-correlation between the magnetic field strength and plasma density,zero phase velocity in the plasma rest frame and linear polarization.The structures were observed in a region bounded by two dipolarizations during a substorm intensification.Thus,the dipolarization process may provide a plasma condition facilitating the growth of the mirror mode structures.Another interesting feature is the electron dynamics within the mirror structures.Thermal electron energy flux has an enhancement at 0°and 180°pitch angles inside the magnetic dips of the first three mirror structures and an enhancement at 90°pitch angle inside the magnetic dip of the last structure.The different electron distribution inside the mirror structures might be a result of different evolution stages of the mirror wave.The last structure may be in the nonlinear stage of the mirror instability,whereas the three others with quasi-sinusoidal waveforms may be in the linear stage.In addition,we found that intense whistler waves were confined within the magnetic dips.We conjecture that whistler waves observed in the first three dips were generated in a remote region,then they were trapped in the mirror mode troughs and transported toward the spacecraft;while the whistler wave detected in the last dip was excited locally by the electron anisotropy instability. | LI HuiMin ZHOU Meng DENG XiaoHua YUAN ZhiGang HUANG ShiYong | 2014 | Science China(Technological Sciences)2014,57,8: | 2 |

The directions of the core fields are the same as the ambient cross-tail magnetic field component(By) just outside of flux ropes. Therefore, we suggest that the core field of flux ropes is formed by compression of the local preexisting Byand that the directional change of core field is due to the change of local preexisting By. Recent kinetic simulations have found that DFs may break up into small-scale structures after they are produced by reconnection. Motivated by this simulation, we revisited the scale size of DFs in the dawn-dusk direction by using Cluster observations during the years when the inter-distance among Cluster spacecraft was between 1000 and 2000 km. We selected the DFs that were detected by more than one spacecraft and estimated the radii of these DFs by a simple geometrical analysis, which is based on comparison of DF normals observed by different spacecraft. We found a few DFs that were only a few ion inertial lengths in the dawn-dusk direction. | 18 | Mesenchymal stromal cells alleviate acute respiratory distress syndrome through the cholinergic anti-inflammatory pathway显示文摘Mesenchymal stromal cells(MSCs)have been considered a promising alternative for treatment of acute respiratory distress syndrome(ARDS).However,there is significant heterogeneity in their therapeutic efficacy,largely owing to the incomplete understanding of the mechanisms underlying the therapeutic activities of MSCs.Here,we hypothesize that the cholinergic antiinflammatory pathway(CAP),which is recognized as a neuroimmunological pathway,may be involved in the therapeutic mechanisms by which MSCs mitigate ARDS.Using lipopolysaccharide(LPS)and bacterial lung inflammation models,we found that inflammatory cell infiltration and Evans blue leakage were reduced and that the expression levels of choline acetyltransferase(ChAT)and vesicular acetylcholine transporter(VAChT)in lung tissue were significantly increased 6 hours after MSC infusion.When the vagus nerve was blocked orα7 nicotinic acetylcholine(ACh)receptor(α7nAChR)-knockout mice were used,the therapeutic effects of MSCs were significantly reduced,suggesting that the CAP may play an important role in the effects of MSCs in ARDS treatment.Our results further showed that MSC-derived prostaglandin E2(PGE2)likely promoted ACh synthesis and release.Additionally,based on the efficacy of nAChR andα7nAChR agonists,we found that lobeline,the nicotinic cholinergic receptor excitation stimulant,may attenuate pulmonary inflammation and alleviate respiratory symptoms of ARDS patients in a clinical study(ChiCTR2100047403).In summary,we reveal a previously unrecognized MSC-mediated mechanism of CAP activation as the means by which MSCs alleviate ARDS-like syndrome,providing insight into the clinical translation of MSCs or CAP-related strategies for the treatment of patients with ARDS. | Xiaoran Zhang Xuxia Wei Yiwen Deng Xiaofeng Yuan Jiahao Shi Weijun Huang Jing Huang Xiaoyong Chen Shuwei Zheng Jieying Chen Keyu Chen Ruiming Xu Hongmiao Wang Weiqiang Li Shiyue Li Huimin Yi Andy Peng Xiang | 2022 | Signal Transduction and Targeted Therapy2022,7,10: | 1 |
