| 1 | CapOX as neoadjuvant chemotherapy for locally advanced operable colon cancer patients: a prospective single-arm phase Ⅱ trial显示文摘Objective: The aim of this prospective, single-arm phase II trial was to confirm the safety and efficacy of neoadjuvant chemotherapy(NAC) using oxaliplatin plus capecitabine(Cap OX) for patients with operable locally advanced colon cancer(CC).Methods: Patients with computed tomography-defined T4 or lymph node-positive CCs were enrolled. After radiological staging, patients were treated with at least 2 cycles of NAC consisting of 130 mg/m2 oxaliplatin on d 1,plus 1,000 mg/m2 capecitabine twice daily for 14 d every 3 weeks, followed by surgery, and then with the rest cycles of adjuvant chemotherapy. Radiological response was evaluated after 2 cycles of NAC. Tumor response, treatment toxicity, and surgical complications were recorded. The pathological response to therapy was evaluated according to the tumor regression grade(TRG) score. The primary endpoint was pathologic tumor response. This trial is registered in Clinical Trials.gov(No: NCT02415829).Results: Forty-seven patients were enrolled in the study. Forty-two patients completed the planned treatments.The total radiological response rate was 68%(32/47), including complete and partial response rates of 2%(1/47)and 66%(31/47), respectively. Stable disease was observed in 32%(15/47) and progressive disease was observed in none. Complete pathologic response, major regression, and at least moderate regression were achieved in 1(2%), 2(4%), and 29(62%) patients, respectively. Four patients developed grade 3 treatment toxicities. One patient with wound infection occurred after operation(1/47, 2%). | 4 | Arabidopsis calcium-dependent protein kinase CPK28 is potentially involved in the response to osmotic stress显示文摘As calcium sensors, plant calcium-dependent protein kinases(CDPKs) play important roles in plants' responses to various abiotic stresses. Here, we report the functional characterization of CPK28, a member of the CDPK family in Arabidopsis, in response to osmotic stress.The cpk28 mutant, a loss-of-function mutant, exhibited an NaCl- and mannitol-sensitive phenotype in green cotyledons, while CPK28-overexpressing plants displayed stronger tolerance to NaCl and mannitol stresses than wildtype plants. Reverse transcriptase-polymerase chain reaction and beta-glucuronidase staining assays showed that NaCl and mannitol stresses induced CPK28. CPK28-overexpressing lines accumulated significantly more proline relative to wild-type plants and mutant plants under NaCl and mannitol stresses. Transient expression of CPK28-GFP in mesophyll cell protoplasts, as well as stable transgenic lines expressing CPK28-GFP, showed that CPK28 was localized in the plasma membrane. Expression levels of known stress-responsive genes were not significantly altered in the null mutant and overexpression lines,suggesting that CPK28 possibly mediated the stress response via the regulation of target proteins rather than via regulation at the level of transcription. Meanwhile, CPK28could autophosphorylate. | 5 | Phytophthora Effectors Modulate Genome-wide Alternative Splicing of Host mRNAs to Reprogram Plant Immunity显示文摘Alternative splicing(AS)of pre-mRNAs increases transcriptome and proteome diversity,regulates gene expression through multiple mechanisms,and plays important roles in plant development and stress responses.However,the prevalence of genome-wide plant AS changes during infection and the mechanisms by which pathogens modulate AS remain poorly understood.Here,we examined the global AS changes in tomato leaves infected with Phytophthora infestans,the infamous Irish famine pathogen.We show that more than 2000 genes exhibiting significant changes in AS are not differentially expressed,indicating that AS is a distinct layer of transcriptome reprogramming during plant-pathogen interactions.Furthermore,our results show that P.infestans subverts host immunity by repressing the AS of positive regulators of plant immunity and promoting the AS of susceptibility factors.To study the underlying mechanism,we established a luminescence-based AS reporter system in Nicotiana benthamiana to screen pathogen effectors modulating plant AS.We identified nine splicing regulatory effectors(SREs)from 87 P.infestans effectors.Further studies revealed that SRE3 physically binds U1-70K to manipulate the plant AS machinery and subsequently modulates AS-mediated plant immunity.Our study not only unveils genome-wide plant AS reprogramming during infection but also establishes a novel AS screening tool to identify SREs from a wide range of plant pathogens,providing opportunities to understand the splicing regulatory mechanisms through which pathogens subvert plant immunity. | Jie Huang Xinyu Lu Hongwei Wu Yuchen Xie Qian Peng Lianfeng Gu Juyou Wu Yuanchao Wang Anireddy SNReddy Suomeng Dong | 2020 | Molecular Plant2020,13,10: | 4 |

| 9 | A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. | 10 | Shape and position of Earth's bow shock near-lunar orbit based on ARTEMIS data显示文摘Earth's bow shock is the result of interaction between the supersonic solar wind and Earth's magnetopause. However, data limitations mean the model of the shape and position of the bow shock are based largely on near-Earth satellite data. The model of the bow shock in the distant magnetotail and other factors that affect the bow shock, such as the interplanetary magnetic field(IMF) B_y, remain unclear. Here, based on the bow shock crossings of ARTEMIS from January 2011 to January 2015, new coefficients of the tail-flaring angle a of the Chao model(one of the most accurate models currently available) were obtained by fitting data from the middle-distance magnetotail(near-lunar orbit, geocentric distance -20R_E>X>-50R_E). In addition, the effects of the IMF B_y on the flaring angle a were analyzed. Our results showed that:(1) the new fitting coefficients of the Chao model in the middle-distance magnetotail are more consistent with the observed results;(2) the tail-flaring angle a of the bow shock increases as the absolute value of the IMF B_y increases. Moreover, positive IMF B_y has a greater effect than negative IMF B_y on flaring angle. | 12 | Review of flash sintering with strong electric field显示文摘Flash sintering is a novel field‐assisted sintering technology for ceramics that allows a dramatic reduction in processing time and temperature.Since 2010,when flash sintering was first reported,it has been a focus of research interest in the field of materials science.Recent study results have confirmed that the sintering temperature decreases with the strength of the applied electric field;for some ceramics,the sintering temperature can be lowered even to room temperature.This represents an innovative breakthrough for ceramic sintering at ultra‐low temperatures.However,once the electric field strength in flash sintering is increased,new questions and challenges arise,such as whether or not partial discharge occurs in the green body if under electro‐thermal coupling stress,how to quantitatively analyse the impact of discharge on sintering,and,moreover,whether the sintering mechanism under a strong electric field is consistent with that under a weak electric field.These research questions require knowledge of partial discharge detection,dielectric theory,and other topics that are beyond the scope of materials science.To address this need,this review summarises the work carried out in flash sintering with a strong electric field from the perspective of high voltage and insulation technology.First,the flash sintering process and mechanism are briefly introduced.Then,the published literature on flash sintering with a strong electric field for various ceramic materials is summarised in depth.In addition,experimental phenomena that are observed in flash sintering with a strong electric field,such as flash and blackening,are discussed.Finally,some suggestions for future work are presented.It is anticipated that the knowledge gap between different areas of study can be filled by this review. | Hongyang Zhou Xiang Li Yuchen Zhu Jieming Liu Angxuan Wu Guoming Ma Xilin Wang Zhidong Jia Liming Wang | 2022 | High Voltage2022,7,1: | 2 |

In this work, we prepare several CuCo_(2)O_(4)@Ni-Co-S composite electrodes by a controllable hydrothermal and electrodeposition route. Onedimensional nanowires can shorten the ions transport path, while two-dimensional nanosheets expose many active sites. This enables three-dimensional structured composite with high electrochemical activity. The as-prepared heterostructured materials show a specific of 1048 C/g at 1 A/g. It still maintains 75.6% of initial capacity after 20000 cycles at 10 A/g. The device delivers an energy density of 79.2 Wh/kg when the power density reaches to 2280 W/kg. | 19 | Taurocholic acid inhibits the response to interferon-αtherapy in patients with HBeAg-positive chronic hepatitis B by impairing CD8^(+)T and NK cell function显示文摘Pegylated interferon-alpha (PegIFNα) therapy has limited effectiveness in hepatitis B e-antigen (HBeAg)-positive chronic hepatitis B (CHB) patients. However, the mechanism underlying this failure is poorly understood. We aimed to investigate the influence of bile acids (BAs), especially taurocholic acid (TCA), on the response to PegIFNα therapy in CHB patients. Here, we used mass spectrometry to determine serum BA profiles in 110 patients with chronic HBV infection and 20 healthy controls (HCs). We found that serum BAs, especially TCA, were significantly elevated in HBeAg-positive CHB patients compared with those in HCs and patients in other phases of chronic HBV infection. Moreover, serum BAs, particularly TCA, inhibited the response to PegIFNα therapy in HBeAg-positive CHB patients. Mechanistically, the expression levels of IFN-γ, TNF-α, granzyme B, and perforin were measured using flow cytometry to assess the effector functions of immune cells in patients with low or high BA levels. We found that BAs reduced the number and proportion and impaired the effector functions of CD3^(+)CD8^(+) T cells and natural killer (NK) cells in HBeAg-positive CHB patients. TCA in particular reduced the frequency and impaired the effector functions of CD3^(+)CD8^(+) T and NK cells in vitro and in vivo and inhibited the immunoregulatory activity of IFN-α in vitro. Thus, our results show that BAs, especially TCA, inhibit the response to PegIFNα therapy by impairing the effector functions of CD3^(+)CD8^(+) T and NK cells in HBeAg-positive CHB patients.
