| 1 | ALK,the Key Gene for Gelatinization Temperature,is a Modifier Gene for Gel Consistency in Rice显示文摘Gelatinization temperature(GT) is an important parameter in evaluating the cooking and eating quality of rice.Indeed,the phenotype,biochemistry and inheritance of GT have been widely studied in recent times.Previous map-based cloning revealed that GT was controlled by ALK gene,which encodes a putative soluble starch synthase II-3.Complementation vector and RNAi vector were constructed and transformed into Nipponbare mediated by Agrobacterium.Phenotypic and molecular analyses of transgenic lines provided direct evidence for ALK as a key gene for GT.Meanwhile,amylose content,gel consistency and pasting properties were also affected in transgenic lines.Two of four nonsynonymous single nucleotide polymorphisms in coding sequence of ALK were identified as essential for GT.Based on the single nucleotide polymorphisms(SNPs),two new sets of SNP markers combined with one cleaved amplified polymorphic sequence marker were developed for application in rice quality breeding. | Zhenyu Gao Dali Zeng Fangmin Cheng Zhixi Tian Longbiao Guo Yan Su Meixian Yan Hua Jiang Guojun Dong Yuchen Huang Bin Han Jiayang Li Qian Qian | 2011 | Journal of Integrative Plant Biology2011,53,9: | 13 |

| 3 | Optimizing the CRISPR/Cas9 system for genome editing in grape by using grape promoters显示文摘The efficacy of the CRISPR/Cas9 system in grapevine(Vitis vinifera L.)has been documented,but the optimization of this system,as well as CRISPR/Cas9-mediated multiplex genome editing,has not been explored in this species.Herein,we identified four VvU3 and VvU6 promoters and two ubiquitin(UBQ)promoters in grapevine and demonstrated that the use of the identified VvU3/U6 and UBQ2 promoters could significantly increase the editing efficiency in grape by improving the expression of sgRNA and Cas9,respectively.Furthermore,we conducted multiplex genome editing using the optimized CRISPR/Cas9 vector that contained the conventional multiple sgRNA expression cassettes or the polycistronic tRNA-sgRNA cassette(PTG)by targeting the sugar-related tonoplastic monosaccharide transporter(TMT)family members TMT1 and TMT2,and the overall editing efficiencies were higher than 10%.The simultaneous editing of TMT1 and TMT2 resulted in reduced sugar levels,which indicated the role of these two genes in sugar accumulation in grapes.Moreover,the activities of the VvU3,VvU6,and UBQ2 promoters in tobacco genome editing were demonstrated by editing the phytoene desaturase(PDS)gene in Nicotiana benthamiana leaves.Our study provides materials for the optimization of the CRISPR/Cas9 system.To our knowledge,our simultaneous editing of the grape TMT family genes TMT1 and TMT2 constitutes the first example of multiplex genome editing in grape.The multiplex editing systems described in this manuscript expand the toolbox of grape genome editing,which would facilitate basic research and molecular breeding in grapevine. | Chong Ren Yanfei Liu Yuchen Guo Wei Duan Peige Fan Shaohua Li Zhenchang Liang | 2021 | Horticulture Research2021,8,1: | 6 |

| 6 | A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. | 8 | Evaluation of the weakening behavior of gas on the coal strength and its quantitative influence on the coal deformation显示文摘The coal strength and deformation properties are key factors affecting safe coal mining and highefficiency coalbed methane(CBM)development.In this paper,reconstituted coal samples are chosen to investigate the weakening behavior of gas on coal strength,meanwhile,its effects on coal deformation are quantitatively evaluated.The results indicate that the weakening degree of gas on coal strength is closely related to the confining stress and gas pressure.Compared with non-gas-saturated coals,the maximum weakening ratios of adsorbed gas to coal strength are 10.58%,18.12%,8.55%and 14.65%under the conditions of confining stress CS=3 MPa and gas pressure GP=1 MPa,CS=3 MPa and GP=2 MPa,CS=4 MPa and GP=1 MPa,and CS=4 MPa and GP=2 MPa,respectively.Furthermore,the maximum weakening ratios of free gas to coal strength are 18.27%,36.54%,14.79%and 29.58%,respectively,under above four conditions.The maximum coal bulk strain decreases as particle sizes of coal powders increase,and it has a maximum value of 0.0227 and a minimum value of 0.0191 in particle size ranges of 0.01–0.041 and 0.5–1 mm.Under the same conditions,the coal bulk strain increases with increasing gas pressure,revealing that coal deformation properties can be enhanced by gas. | Haijun Guo Kai Wang Yuchen Wu Hanlu Tang Jianguo Wu Lianhe Guan Chenyang Chang Chao Xu | 2021 | International Journal of Mining Science and Technology2021,31,3: | 2 |
