1Deubiquitinase JOSD2 stabilizes YAP/TAZ to promote cholangiocarcinoma progression显示文摘Cholangiocarcinoma(CCA)has emerged as an intractable cancer with scanty therapeutic regimens.The aberrant activation of Yes-associated protein(YAP)and transcriptional co-activator with PDZ-binding motif(TAZ)are reported to be common in CCA patients.However,the underpinning mechanism remains poorly understood.Deubiquitinase(DUB)is regarded as a main orchestrator in maintaining protein homeostasis.Here,we identified Josephin domain-containing protein 2(JOSD2)as an essential DUB of YAP/TAZ that sustained the protein level through cleavage of polyubiquitin chains in a deubiquitinase activity-dependent manner.The depletion of JOSD2 promoted YAP/TAZ proteasomal degradation and significantly impeded CCA proliferation in vitro and in vivo.Further analysis has highlighted the positive correlation between JOSD2 and YAP abundance in CCA patient samples.Collectively,this study uncovers the regulatory effects of JOSD2 on YAP/TAZ protein stabilities and profiles its contribution in CCA malignant progression,which may provide a potential intervention target for YAP/TAZ-related CCA patients.Meijia Qian Fangjie Yan Weihua Wang Jiamin Du Tao Yuan Ruilin Wu Chenxi Zhao Jiao Wang Jiabin Lu Bo Zhang Nengming Lin Xin Dong Xiaoyang Dai Xiaowu Dong Bo Yang Hong Zhu Qiaojun He 2021Acta Pharmaceutica Sinica B2021,11,12:5
2Temperature-sensitive polymeric nanogels encapsulating withβ-cyclodextrin and ICG complex for high-resolution deep-tissue ultrasound-switchable fluorescence imaging显示文摘One of the thorny problems currently impeding the applications of the fluorescence imaging technique is the poor spatial resolution in deep tissue.Ultrasound-switchable fluorescence(USF)imaging is a novel imaging tool that has recently been explored to possibly surmount the above-mentioned bottleneck.Herein,αβ-cyclodextrin/indocyanine green(ICG)complex-encapsulated poly(N-isopropylacrylamide)(PNIPAM)nanogel was synthesized and studied for ex vivo/in vivo deep tissue/high-resolution near infrared USF(NIR-USF)imaging.To be specific,our results revealed that the average diameter of the as-prepared nanogels was significantly decreased to-32 nm from-335 nm compared to the reported ICG-PNIPAM nanoparticles.Additionally,the excitation/emission characteristics of the ICG itself in present nanogels were almost completely retained,and the resultant nanogel exhibited high physiological stability and positive biocompatibility.In particular,the signal-to-noise ratio of the USF image for the PNIPAM/P-cyclodextrin/ICG nanogel(33.01±2.42 dB)was prominently higher than that of the ICG-PNIPAM nanoparticles(18.73±0.33 dB)in 1.5-cm-thick chicken breast tissues.The NIR-USF imaging in 3.5-cm-thick chicken breast tissues was achieved using this new probe.The e x v iv o NIR-USF imaging of the mouse liver was also successfully obtained.Animal experiments showed that the present nanogels were able to be effectively accumulated into U87 tumor-bearing mice via enhanced permeability and retention effects,and the high-resolution NIR-USF imaging of in v ivo tumor was efficiently acquired.The metabolism and in vivo biodistribution of the nanogels were evaluated.Overall,the results suggest that the current nanogel is a highly promising NIR-USF probe for deep tissue and high-resolution USF imaging.Ruilin Liu Tingfeng Yao Yang Liu Shuai Yu Liqin Ren Yi Hong Kytai T.Nguyen Baohong Yuan 2020Nano Research2020,13,4:1

6Surface etching induced ultrathin sandwich structure realizing enhanced photocatalytic activity显示文摘Photocatalytic conversion efficiency is limited by serious charge carrier recombination. Efficient carrier separation is usually achieved by elegantly-designed multi-component structures connected by directional electric field. Herein, we developed a twodimensional(2 D) sandwich structure, as a new photocatalytic system, to realize high-efficiency carrier separation. This strategy integrated multifunction into a single structure for the first time, which successfully introduces a stable built-in electric field,realizing high-effective carrier separation. Besides, the carrier concentration is dramatically increased due to dimensional confinement. Benefiting from above synergic advantages, 2 D sandwich photocatalyst achieves the highest nitrogen fixation rate(435 μmol g^(-1) h^(-1)) in inorganic solid photocatalysts under visible light irradiation. 7Interfacial ion regulation on 2D layered double hydroxide nanosheets for enhanced thermal insulation显示文摘Exploring new strategies to broaden the upper/lower limit of thermal conductivity is of great interest to develop thermal management materials that can adapt to extreme environments.In this work,we employ an interfacial ion regulation to enhance the thermal insulation performance of 2D layered double hydroxide nanosheets.The introduction of interfacial ion enlarges the interplanar spacing of Co(OH)_(2) nanosheets from 4.64 to 8.05 ?,which reduces phonon scattering length perpendicular to the two-dimensional plane and leads to enhanced interlayer thermal insulation.The interfacial ion-regulated Co(OH)_(2)(named as Co(OH)_(2)-M^(x-)) exhibits 3-fold enhancement of thermal insulation through decreasing the thermal conductivity to as low as 0.15 W m^(-1) K^(-1),which is among the top values in 2D solid materials.We anticipate that interfacial ion regulation for 2D nanosheets paves a new avenue to break through the thermal insulation limit.Chun Wang Hengyu Xu Han Cheng Hao Yu Si Liu Wenjie Wang Ruilin Yuan Hongfei Liu Tianpei Zhou Wangsheng Chu HengAn Wu Yi Xie Changzheng Wu 2022Science China Chemistry2022,65,5:0
8Polymorphism Analysis of RXFP2 in Different Sheep Breeds from Qinghai-Tibet Plateau of China显示文摘Relaxin/insulin-like family peptide receptor 2(RXFP2) is a robust candidate gene related to horn types in sheep.A series of independent genome-wide association studies have reported that RXFP2 underlies the existence and lack of horns.In this study,High-Resolution Melting(HRM) analysis and DNA sequencing were employed to detect the polymorphism of RXFP2 gene in three sheep breeds from Qinghai-Tibet Plateau of China(Tibetan sheep,Qinghai fine wool sheep and Alpine Merino sheep) and to determine the impacts of genotypes of RXFP2 on expression of horn phenotypes.The results showed that one single nucleotide polymorphism(SNP) was identified as RXFP 2SNP c.29389966 A > G.The frequency of genotype AA in Alpine Merino ram(polled) was significantly higher than that in Tibetan ram(horned) and Qinghai fine wool ram(horned) [x^2(1,N = 421) = 72.25,P < 0.001;x^2(1,N = 402) = 4.28,P < 0.005) ];the frequency of genotype AA in Qinghai fine wool ewe(polled) was also much higher than that in Qinghai fine wool ewe(horned) and Tibetan ewe(horned) [x^2(1,N = 196) = 42.04,P < 0.001;x^2(1,N = 192) = 24.69,P < 0.005) ].This mutation could potentially be exploited in marker-assisted selection(MAS) programs within sheep industry to breed horned or polled animals.Megersa Ashenafi GETACHEW Tingting GUO Chao YUAN Jianbin LIU Jian GUO Ruilin FENG Chun'e NIU Xiaoping SUN Yaojing YUE Bohui YANG 2016Agricultural Biotechnology2016,5,2:0
9Transmission electron microscopy of synaptic ultrastructural white matter damage in neonatal rats显示文摘BACKGROUND: To date,animal models of white matter damage remain controversial.Mild grey matter damage should be the basis for animal models to investigate white matter disease.OBJECTIVE: To establish white matter damage in neonatal rats and evaluate feasibility of the established model by observing myelination and synaptic ultrastructure.DESIGN,TIME AND SETTING: A randomized,controlled,animal experiment was performed at the Laboratory of Histology and Embryology of Guangzhou Medical College from December 2008 to May 2009.MATERIALS: H600 transmission electron microscopy was provided by Hitachi,Japan.METHODS: A total of 39 neonatal,Sprague Dawley rats were randomly assigned to normal control (n = 12),sham-surgery (n = 12),and white matter damage (n = 15) groups.White matter damage rats were subjected to right common carotid artery ligation,followed by inhalation of nitrogen oxygen gas mixture (6% oxygen) for 4 hours.MAIN OUTCOME MEASURES: Myelin sheath and synaptic ultrastructure in the injured (right) hippocampal CA1 region in 1-month-old rats were observed through the use of transmission electron microscopy,and pathological changes in the cerebral cortex and corpus callosum of the right hemisphere were detected by hematoxylin-eosin staining.RESULTS: Obvious tissue loss was observed in the corpus callosum of the injured (right) hemisphere.Injured oligodendrocytes and disrupted myelination were observed in the white matter damage group.However,synaptic length in the active zones,width of synaptic cleft,thickness of postsynaptic density,and curvature of the synaptic interface remained unchanged following injury,compared with the control and sham-surgery groups (P > 0.05).CONCLUSION: The established white matter damage model resulted in changes in myelination and slightly altered synaptic ultrastructures.The model could function as an ideal model for white matter damage in neonatal rats.Baoli Yuan Sujuan Deng Zhengyu Zhang Ruilin Li 2010Neural Regeneration Research2010,5,5:0
10Fractal Characteristics and Main Controlling Factors of High-Quality Tight Sandstone Reservoirs in the Southeastern Ordos Basin显示文摘Due to the complex conditions and strong heterogeneity of tight sandstone reservoirs,the reservoirs should be classified and the controlling factors of physical properties should be studied.Cast thin section observations,cathodoluminescence,scanning electron microscopy(SEM),X-ray diffraction(XRD),and high-pressure mercury injection(HPMI)were used to classify and optimize the reservoir.The Brooks-Corey model and stepwise regression were used to study the fractal dimension and main controlling factors of the physical properties of the high-quality reservoir.The results show that the reservoirs in the study area can be divided into four types,and the high-quality reservoir has the best physical properties and pore-throat characteristics.In the high-quality reservoir,the homogeneity of transitional pores was the best,followed by that of micropores,and the worst was mesopores.The porosity was controlled by depth and kaolinite.The model with standardized coefficients is y=12.454−0.778×(Depth)+0.395×(Kaolinite).The permeability was controlled by depth,illite/montmorillonite,and siliceous cement,and the model with standardized coefficients is y=1.689−0.683×(Depth)−0.395×(Illite/Montmorillonite)−0.337×(Siliceous Cement).The pore-throat evolutionary model shows that the early-middle diagenetic period was when the reservoir physical properties were at their best,and the kaolinite intercrystalline pores and residual intergranular pores were the most important.Ruilin Hao Wenhui Huang Jiu Bo Li Yuan 2024Journal of Earth Science2024,35,2:0
