2De novo characterization of the root transcriptome of a traditional Chinese medicinal plant Polygonum cuspidatum显示文摘Various active components have been extracted from the root of Polygonum cuspidatum.However,the genetic basis for their activity is virtually unknown.In this study,25600002 short reads (2.3 Gb) of P.cuspidatum root transcriptome were obtained via Illumina HiSeq 2000 sequencing.A total of 86418 unigenes were assembled de novo and annotated.Twelve,18,60 and 54 unigenes were respectively mapped to the mevalonic acid (MVA),methyl-D-erythritol 4-phosphate (MEP),shikimate and resveratrol biosynthesis pathways,suggesting that they are involved in the biosynthesis of pharmaceutically important anthraquinone and resveratrol.Eighteen potential UDP-glycosyltransferase unigenes were identified as the candidates most likely to be involved in the biosynthesis of glycosides of secondary metabolites.Identification of relevant genes could be important in eventually increasing the yields of the medicinally useful constituents of the P.cuspidatum root.From the previously published transcriptome data of 19 non-model plant taxa,1127 shared orthologs were identified and characterized.This information will be very useful for future functional,phylogenetic and evolutionary studies of these plants.HAO DaCheng MA Pei MU Jun CHEN ShiLin XIAO PeiGen PENG Yong HUO Li XU LiJia SUN Chao 2012Science China(Life Sciences)2012,55,5:36

4Osteoimmunomodulatory effects of biomaterial modification strategies on macrophage polarization and bone regeneration显示文摘Biomaterials as bone substitutes are always considered as foreign bodies that can trigger host immune responses.Traditional designing principles have been always aimed at minimizing the immune reactions by fabricating inert biomaterials.However,clinical evidence revealed that those methods still have limitations and many of which were only feasible in the laboratory.Currently,osteoimmunology,the very pioneering concept is drawing more and more attention-it does not simply regard the immune response as an obstacle during bone healing but emphasizes the intimate relationship of the immune and skeletal system,which includes diverse cells,cytokines,and signaling pathways.Properties of biomaterials like topography,wettability,surface charge,the release of cytokines,mediators,ions and other bioactive molecules can impose effects on immune responses to interfere with the skeletal system.Based on the bone formation mechanisms,the designing methods of the biomaterials change from immune evasive to immune reprogramming.Here,we discuss the osteoimmunomodulatory effects of the new modification strategies—adjusting properties of bone biomaterials to induce a favorable osteoimmune environment.Such strategies showed potential to benefit the development of bone materials and lay a solid foundation for the future clinical application.Yajuan Xie Cheng Hu Yi Feng Danfeng Li Tingting Ai Yulei Huang Xiaodan Chen Lijia Huang Jiali Tan 2020Regenerative Biomaterials2020,7,3:22
5A new ophiolitic mélange containing boninitic blocks in Alxa region:Implications for Permian subduction events in southern CAOB显示文摘The Alxa region, located in the southernmost part of Central Asian Orogenic Belt, is a key region for understanding the tectonic processes associated with the closure of the Paleo-Asian Ocean. Issues of late Paleozoic tectonic settings and tectonic unit divisions of the Alxa region still remain controversial. In this study, we report a new ophiolitic mélange named the Tepai ophiolitic mélange in the northern Alxa region, northwest of Alxa Youqi. The tectonic blocks in the Tepai ophiolitic mélange are mainly composed of serpentinized peridotites, serpentinites, mylonitized gabbros, gabbros, basalts, and quartzites, with a matrix comprising highly deformed clastic rocks. A gabbro exhibits a zircon LA-ICP-MS Ue Pb age of278.4 ± 3.3 Ma. Gabbros exhibit high Mg O and compatible element contents, but extremely low TiO_2,totally rare earth element and high field strength element contents. These rocks exhibit light rare earth element depleted patterns, and display enriched in large-ion lithophile elements and depleted in high field strength elements. Boninite-like geochemical data show that they were formed in a subductionrelated environment, and derived from an extremely depleted mantle source infiltrated by subduction-derived fluids and/or melts. The Tepai ophiolitic mélange exhibits similar zircon U-Pb-O isotopic compositions and whole-rock geochemical characteristics to those of the Quagan Qulu ophiolite.Therefore, we propose that the Tepai ophiolitic mélange may have been the western continuation of the Quagan Qulu ophiolite. 6OsSIDP366,a DUF1644 gene,positively regulates responses to drought and salt stresses in rice显示文摘Domain of unknown function 1644(DUF1644) is a Rhighly conserved amino acid sequence motif present only in plants. Analysis of expression data of the family of DUF1644-containing genes indicated that they may regulate responses to abiotic stress in rice. Here we present our discovery of the role of Os SIDP366, a member of the DUF1644 gene family, in response to drought and salinity stresses in rice. Transgenic rice plants overexpressing Os SIDP366 showed enhanced drought and salinity tolerance and reduced water loss as compared to that in the control, whereas plants with downregulated Os SIDP366 expression levels using RNA interference(RNAi) were more sensitive to salinity and drought treatments. The sensitivity to abscisic acid(ABA)treatment was not changed in Os SIDP366-overexpressing plants, and Os SIDP366 expression was not affected in ABAdeficient mutants. Subcellular localization analysis revealed that Os SIDP366 is presented in the cytoplasmic foci that colocalized with protein markers for both processing bodies(PBs) and stress granules(SGs) in rice protoplasts. Digital gene expression(DGE) profile analysis indicated that stress-related genes such as SNAC1, Os HAK5 and PRs were upregulated in Os SIDP366-overexpressing plants. 8Methylation of phytohormones by the SABATH methyltransferases显示文摘In plants,one of the most common modifications of secondary metabolites is methylation catalyzed by various methyltransferases. Recently,a new class of methyltransferases,the SABATH family of methyltransferases,was found to modify phytohormones and other small molecules.The SABATH methyltransferases share little sequence similarity with other well characterized methyltransferases.Arabidopsis has 24 members of the SABATH methyltransferase genes,and a subset of them has been shown to catalyze the formation of methyl esters with phytohormones and other small molecules.Physiological and genetic analyses show that methylation of phytohormones plays important roles in regulating various biological processes in plants,including stress responses,leaf development,and seed maturation/germination.In this review,we focus on phytohormone methylation by the SABATH family methyltransferases and the implication of these modifications in plant development.QU LiJia LI Shuang XING ShuFan 2010Chinese Science Bulletin2010,55,21:6
9Downregulation of caveolin-1 contributes to the synaptic plasticity deficit in the hippocampus of aged rats显示文摘Caveolin-1 is involved in the regulation of synaptic plasticity,but the relationship between its expression and cognitive function during aging remains controversial.To explore the relationship between synaptic plasticity in the aging process and changes in learning and memory,we examined caveolin-1 expression in the hippocampus,cortex and cerebellum of rats at different ages.We also examined the relationship between the expression of caveolin-1 and synaptophysin,a marker of synaptic plasticity.Hippocampal caveolin-1 and synaptophysin expression in aged(22–24 month old)rats was significantly lower than that in young(1 month old)and adult(4 months old)rats.Expression levels of both proteins were significantly greater in the cortex of aged rats than in that of young or adult rats,and levels were similar between the three age groups in the cerebellum.Linear regression analysis revealed that hippocampal expression of synaptophysin was associated with memory and learning abilities.Moreover,synaptophysin expression correlated positively with caveolin-1 expression in the hippocampus,cortex and cerebellum.These results confirm that caveolin-1 has a regulatory effect on synaptic plasticity,and suggest that the downregulation of hippocampal caveolin-1 expression causes a decrease in synaptic plasticity during physiological aging.Yang Liu Zhanhua Liang Jing Liu Wei Zou Xiaoyan Li Yachen Wang Lijia An 2013Neural Regeneration Research2013,8,29:6
10Acoustic emission generated during the gas sorption-desorption process in coal显示文摘An experimental system for monitoring the acoustic signals generated in coal during gas sorption and/or desorption was designed and the acoustic signals were observed under different gas pressures. The experimental results show that signals generated by the coal during gas adsorption are attenuated over time. Also, the signals are not continuous but are impulsive. The intensity of the signals generated during gas desorption is far smaller than that observed during adsorption. The signal seen during desorption remains essentially stable. Cycles of sorption and desorption cause acoustic emission signals that exhibit a memory effect, which depends upon the maximum gas pressure the sample was exposed to in earlier cycles. Lower pressures in subsequent cycles, compared to the maximum adsorption pressure in previous cycles, cause both the energy and impulse frequency to be lower than previously. 11Construction of chimeric inducible promoters by elicitors of rice fungal blast pathogen and their expression in transgenic rice显示文摘The promoter fragments of wheat GstA1 and potato Gst1 have been amplified by PCR, cloned and fused respectively to the minimal promoter sequence of rice actin gene (Act1)) and its 5’ untranslated leader sequence together with GUS. The constructs with 2 chimeric promoters (WGA and PGA) have been transferred into rice in order to analyze their inducibility patterns in transgenic rice plants. 12Adsorption and mechanistic study of the invasive plant-derived biochar functionalized with CaAl-LDH for Eu(Ⅲ) in water显示文摘Herein,we developed the invasive plant-derived biochar (IPB) functionalized with CaAl-LDH at five mass ratios using a physical mixture method,assessed their adsorption perform for Eu(Ⅲ),and explored the relative mechanisms.Results show that the IPB successfully loaded CaAl-LDH in five composites and their Eu(Ⅲ) sorption affinities were strongly affected by solution p H,contact time,temperature,and the mass ratio of LDH and IPB.All the sorpiton process for Eu(Ⅲ) occurred on the heterogeneous surface of five composites and the boundary layer diffusion limited the chemical sorption rate.Interestingly,the CaAl-LDH/IPB composite with high ratio of IPB had higher sorption capacity than the one with high ratio of LDH due to larger porosity of the former.Three mechanisms containing ion exchange between Al and Eu ions,surface complexation with carboxyl-and oxygen-containing functional groups,and precipitation were involved in the Eu(Ⅲ) sorption,but the dominant sorption mechanism for each CaAl-LDH/IPB composite differed with different mass ratio of CaAl-LDH and IPB.In composite with more IPB (e.g.,CaAl-LDH/IPB-13),both ion exchange and surface complexes dominated the sorption process and the intensity of Eu^3+ was identified with the one of Eu2O3.Whereas in composites with high LDH,ion exchange dominated the sorption and the intensity of Eu^3+ was obviously higher than the one of Eu_2O_3.This research will provide a new perspective for the application of the LDH/biochar materials.Shengbo Li Lijia Dong Zuofu Wei Guodong Sheng Kui Du Baowei Hu 2020Journal of Environmental Sciences2020,32,10:5
13Determination of copy number for 5S rDNA and centromeric sequence RCS2 in rice by Fiber-FISH显示文摘The copy number of 5S rDNA and centromerie sequence RCS2 was determined by extended DNA fiber based fluorescence in situ hybridization (Fiber-FISH) in rice (Oryza sativa ssp. indica cv. Guangluai No. 4) genome. In order to determine the copy number, it is necessary to know the basepair number that a given length DNA fiber contains under a microscope. Therefore, the length of two DNA frag-ments, in which the basepair number had been already known, was measured. The insert sequence of the tested BAC 38D17 was 136 kb and its extended DNA was 56.4 μm long, 2.41 kb/μm on average, while that of the tested BAC 44B4 was 144.5 kb in total and 55.7 μm long, 2.60 kb/μm on average under the microscope. They were very close to the theoretical value of B-DNA in the Watson-Crick DNA model, which is 2.97 kb/μm. 16Prussian blue nanoparticle-loaded microbubbles for photothermally enhanced gene delivery through ultrasound-targeted microbubble destruction显示文摘By adsorbing chitosan(CS)-functionalized Prussian blue(PB) nanoparticles(CS/PB NPs) complexing DNA onto the surface of gas encapsulated microbubbles(MBs), a multifunctional gene delivery system of MBs@CS/PB/DNA was fabricated for photothermally enhanced gene transfection through ultrasound-targeted microbubble destruction. CS/PB NPs of(2.69 ± 0.49) nm could complex DNA effectively when the mass ratio was2:1. It was found that MBs@CS/PB/DNA could enhance ultrasound imaging greatly both in vitro and in vivo. In addition, MBs@CS/PB/DNA could be disrupted by applying a higher-intensity ultrasound irradiation to release CS/PB/DNA, which could effectively transform the nearinfrared(NIR) light into heat to assist the uptake of CS/PB/DNA by cells. With the aid of ultrasound irradiation and NIR light irradiation, the gene transfection efficiency was significantly enhanced to(43.08 ± 1.13) %, much higher than polyethylenimine. 17Overcoming chemotherapy resistance via simultaneous drug-efflux circumvention and mitochondrial targeting显示文摘Multidrug resistance(MDR) has been considered as a huge challenge to the effective chemotherapy. Therefore, it is necessary to develop new strategies to effectively overcome MDR. Here,based on the previous research of N-(2-hydroxypropyl)methacrylamide(HPMA) polymer–drug conjugates, we designed an effective system that combined drug-efflux circumvention and mitochondria targeting of anticancer drug doxorubicin(Dox). Briefly, Dox was modified with mitochondrial membrane penetrating peptide(MPP) and then attached to(HPMA) copolymers(P-M-Dox). Our study showed that macromolecular HPMA copolymers successfully bypassed drug efflux pumps and escorted Dox into resistant MCF-7/ADR cells via endocytic pathway. Subsequently, the mitochondria accumulation of drugs was significantly enhanced with 11.6-fold increase by MPP modification. The excellent mitochondria targeting then resulted in significant enhancement of reactive oxygen species(ROS) as well as reduction of adenosine triphosphate(ATP)production, which could further inhibit drug efflux and resistant cancer cell growth. By reversing Dox resistance, P-M-Dox achieved much better suppression in the growth of 3D MCF-7/ADR tumor spheroids compared with free Dox. 19Toward calibration-free Mach-Zehnder switches for next-generation silicon photonics显示文摘Silicon photonic Mach–Zehnder switches(MZSs)have been extensively investigated as a promising candidate for optical systems.However,conventional 2×2 MZSs are usually prone to the size variations of the arm waveguides due to imperfect fabrication,resulting in considerable random phase imbalance between the two arms,thereby imposing significant challenges for further developing next-generation N×N MZSs.Here we propose a novel design toward calibration-free 2×2 and N×N MZSs,employing optimally widened arm waveguides,enabled by novel compact tapered Euler S-bends with incorporated mode filters.With standard 180 nm CMOS foundry processes,more than thirty 2×2 MZSs and one 4×4 Benes MZS with the new design are fabricated and characterized.Compared with their conventional counterparts with 0.45-μm-wide arm waveguides,the present 2×2 MZSs exhibit significant reduction in the random phase imbalance.The measured extinction ratios of the present 2×2 and 4×4 MZSs operating in the all-cross state are 27-49 dB and∼20dB across the wavelength range of∼60nm,respectively,even without any calibrations.This work paves the way toward calibration-free large-scale N×N MZSs for next-generation silicon photonics.Lijia Song Tangnan Chen Weixi Liu Hongxuan Liu Yingying Peng Zejie Yu Huan Li Yaocheng Shi Daoxin Dai 2022Photonics Research2022,10,3:2
20Ecosystem service valuation of bays in East China Sea and its response to sea reclamation activities显示文摘Ecosystem service values(ESVs)of bays and their response to sea reclamation are of great practical importance for forming bay eco-compensation policy and extension of blue economic space.Based on land use information of bays collected during the period of 1990–2015,the spatiotemporal evolution of ESVs of 12 main bays in East China Sea and their response to sea reclamation activities over the past 25 years were quantitatively analyzed.The analysis results indicate that ESVs of bays in East China Sea showed a continuous downward trend and the whole ecosystem was continuously degraded,in which the degradation degree of ESV in the southern bays was higher than that in the northern bays.Spatial zoning of ESVs of bays in East China Sea was remarkable,showing a continuous downward trend from low-value to high-value zone.Spatial variation of ESVs of each bay was also significant,expanding from a city and from inland to the coast,which suggests that human activities,mainly reclamation,have become main agents for ESV evolution of bays in East China Sea.ESVs of bays have a significant response to sea reclamation,manifested as a significant negative correlation between ESV and reclamation intensity.The correlation in the southern bays was stronger than that in the northern bays,which was caused by different effects of sea reclamation modes on ESV evolution of muddy and bedrock bays.A negative effect of sea reclamation activities on bay ecosystem was hysteretic.Therefore,an attention should be paid to dynamic monitoring and early warning of development status in offshore areas,ecosystem-level reclamation control policy,and coastal wetland reserves planning.Moreover,the spatial coupling mechanism study between bay ecosystem service demand and its service supply capacity should be strengthened to realize systematic regulation of bay ecological security pattern.LIU Ruiqing XU Hao LI Jialin PU Ruiliang SUN Chao CAO Luodan JIANG Yimei TIAN Peng WANG Lijia GONG Hongbo 2020Journal of Geographical Sciences2020,30,7:2
