| 1 | Osteoimmunomodulatory effects of biomaterial modification strategies on macrophage polarization and bone regeneration显示文摘Biomaterials as bone substitutes are always considered as foreign bodies that can trigger host immune responses.Traditional designing principles have been always aimed at minimizing the immune reactions by fabricating inert biomaterials.However,clinical evidence revealed that those methods still have limitations and many of which were only feasible in the laboratory.Currently,osteoimmunology,the very pioneering concept is drawing more and more attention-it does not simply regard the immune response as an obstacle during bone healing but emphasizes the intimate relationship of the immune and skeletal system,which includes diverse cells,cytokines,and signaling pathways.Properties of biomaterials like topography,wettability,surface charge,the release of cytokines,mediators,ions and other bioactive molecules can impose effects on immune responses to interfere with the skeletal system.Based on the bone formation mechanisms,the designing methods of the biomaterials change from immune evasive to immune reprogramming.Here,we discuss the osteoimmunomodulatory effects of the new modification strategies—adjusting properties of bone biomaterials to induce a favorable osteoimmune environment.Such strategies showed potential to benefit the development of bone materials and lay a solid foundation for the future clinical application. | Yajuan Xie Cheng Hu Yi Feng Danfeng Li Tingting Ai Yulei Huang Xiaodan Chen Lijia Huang Jiali Tan | 2020 | Regenerative Biomaterials2020,7,3: | 22 |
| 2 | Decreased osteogenesis of adult mesenchymal stem cel s by reactive oxygen species under cyclic stretch: a possible mechanism of age related osteoporosis显示文摘Age related defect of the osteogenic differentiation of mesenchymal stem cells(MSCs) plays a key role in osteoporosis. Mechanical loading is one of the most important physical stimuli for osteoblast differentiation.Here, we compared the osteogenic potential of MSCs from young and adult rats under three rounds of 2 h of cyclic stretch of 2.5% elongation at 1 Hz on 3 consecutive days. Cyclic stretch induced a significant osteogenic differentiation of MSCs from young rats, while a compromised osteogenesis in MSCs from the adult rats.Accordingly, there were much more reactive oxygen species(ROS) production in adult MSCs under cyclic stretch compared to young MSCs. Moreover, ROS scavenger N-acetylcysteine rescued the osteogenic differentiation of adult MSCs under cyclic stretch. Gene expression analysis revealed that superoxide dismutase 1(SOD1) was significantly downregulated in those MSCs from adult rats. | 3 | Dysregulation of the miR-146a-Smad4 axis impairs osteogenesis of bone mesenchymal stem cells under inflammation显示文摘Osteoporosis is a common disease that affects patient quality of life, especially among the elderly population.Although inflammation contributes significantly to osteoporosis, the underlying mechanism is unclear. In this study, we found that tumor necrosis factor(TNF)-α, an inflammatory environment mimic, inhibits osteogenesis of bone mesenchymal stem cells(BMSCs), induces mi R-146 a and decreases Smad4. Moreover,overexpression of mi R-146 a inhibited the osteogenic ability of BMSCs, whereas blocking mi R-146 a partially rescued the osteogenesis deficiency under TNF-α treatment. Molecularly, mi R-146 a decreased Smad4 expression at the protein level by binding to an element located in the Smad4 30-untranslated region, and restoration of Smad4 reversed the inhibitory effects of mi R-146 a on osteogenesis. | 4 | Essential sequence of the N-terminal cytoplasmic localization-related domain of huntingtin and its effect on huntingtin aggregates显示文摘Huntington's disease(HD) is caused by abnormal CAG repeat expansion in the 5′-end of the Huntingtin(HTT) gene.In addition to the canonical C-terminal full-length huntingtin(htt) nuclear export signal,a cytoplasmic localization-related domain(CLRD) in the N-terminus of htt has recently been reported.Here,we analyzed this domain by introducing deletion and substitution mutations in a truncated N-terminal htt protein and subsequently monitored htt expression,aggregation and subcellular localization by immunocytochemistry and Western blot analysis.We demonstrated that Htt4-17 was the essential sequence for htt cytoplasmic localization.We also found that the subcellular distribution of htt was altered when Htt1-17 was mutated to contain amino acids of different charges,suggesting a structural requirement of Htt1-17 for the cytoplasmic localization of htt. | 5 | Biodegradable magnesium pins enhanced the healing of transverse patellar fracture in rabbits显示文摘Displaced fractures of patella often require open reduction surgery and internal fixation to restore the extensor continuity and articular congruity.Fracture fixation with biodegradable magnesium(Mg)pins enhanced fracture healing.We hypothesized that fixation with Mg pins and their degradation over time would enhance healing of patellar fracture radiologically,mechanically,and histologically.Transverse patellar fracture surgery was performed on thirty-two 18-weeks old female New Zealand White Rabbits.The fracture was fixed with a pin made of stainless steel or pure Mg,and a figure-of-eight stainless steel band wire.Samples were harvested at week 8 or 12,and assessed with microCT,tensile testing,microindentation,and histology.Microarchitectural analysis showed that Mg group showed 12%higher in the ratio of bone volume to tissue volume at week 8,and 38.4%higher of bone volume at week 12.Tensile testing showed that the failure load and stiffness of Mg group were 66.9%and 104%higher than the control group at week 8,respectively.At week 12,Mg group was 60.8%higher in ultimate strength than the control group.Microindentation showed that,compared to the Control group,Mg group showed 49.9%higher Vickers hardness and 31%higher elastic modulus at week 8 and 12,respectively.At week 12,the new bone of Mg group remodelled to laminar bone,but those of the control group remained woven bone-like.Fixation of transverse patellar fracture with Mg pins and its degradation enhanced new bone formation and mechanical properties of the repaired patella compared to the Control group. | Dick Ho Kiu Chow Jiali Wang Peng Wan Lizhen Zheng Michael Tim Yun Ong Le Huang Wenxue Tong Lili Tan Ke Yang Ling Qin | 2021 | Bioactive Materials2021,6,11: | 3 |

| 7 | Hewei Jiangni granule alleviates visceral hypersensitivity in a rat model of non-erosive reflux disease via transient receptor potential channel signaling显示文摘Objective:To uncover the underlying mechanism of Hewei Jiangni granule(HWJNG)on non-erosive reflux disease(NERD)treatment by examining histological changes,gastrointestinal neurochemicals release and visceral hypersensitivity-related receptor expression in NERD model rats.Methods:A NERD rat model was established via a combination of basal sensitization and acid perfusion.HWJNG treatments at different doses were then administered.Pathological changes to tissues,mast cell(MC)activation,serum levels of esophageal visceral hypersensitivity-related neurochemicals,and transient receptor potential(TRP)receptor mRNA and protein levels were investigated.Results:Compared with the control group,the expression of tryptase in MCs,the changes of intercellular space,and the serum levels of substance P(SP),calcitonin gene-related peptides(CGRP)and proteinaseactivated receptor 2(PAR2)increased in the model group(all P<.05).The expression of TRP vanilloid 1(Trpv1)mRNA decreased in esophagus and dorsal root ganglia(DRG)of the model group(P=.030&P=.013),and the expression of TRP melastatin channel subfamily member 8(Trpm8)mRNA decreased in the esophagus of model group(P<.01).The level of esophageal TRPV1 protein increased in the model group(P<.01)and the level of TRPM8 protein decreased in esophagus and DRG of the model group(both P<.05).Compared with the model group,the serum levels of SP,CGRP,and PAR2 in the mediumdose HWJNG group showed significant decreases(all P<.05).The expression of Trpv1 mRNA in esophagus and DRG of the HWJNG groups and the Omeprazole group remarkably decreased(all P<.05),as was the expression of Trpm8 mRNA in esophagus of the HWJNG groups(all P<.05).Conclusion:HWJNG alleviated visceral hypersensitivity in NERD model rats by regulating TRP-mediated signaling.Our results indicate that HWJNG has potential as a therapeutic agent for NERD. | Jiali Liu Fushun Kou Xiang Tan Yi Dai Chune Xie Xiaohong Li Lei Shi Tangyou Mao Xiaojun Shi Junxiang Li | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,2: | 1 |

| 14 | Two New Species of Free-Living Marine Nematodes of the Desmodoridae from Mangrove Wetlands of Xiamen Bay, China显示文摘Mangroves are unique in their biodiversity,but studies on their meiobenthic biodiversity in China are scarce.Despite the importance of mangroves,little work has been done on the classification of nematodes in mangrove wetlands.Fujian Province is the most northern point of China’s natural mangrove distribution,and it is also one of the provinces with the earliest constructed mangrove forest.In this paper,two new free-living marine nematode species of Desmodoridae from the Xiamen mangrove wetlands in China are described.Metachromadora xiamenensis sp.nov.is characterized by a cylindrical body,and smooth head capsule set off from the rest of the body.The cuticle is finely annulated and thickened at the midbody.Lateral ridges run from the posterior end of the pharynx to the middle of the tail.The amphid foveae is loop shaped and opens at the top with a double contour amphidial,pharynx with bipartite cuticularized internal cavity.There are 18 tubular precloacal supplements and tail with three small protuberances.Molgolaimus euryformis sp.nov.is characterized by a relatively short and plump body with finely annulated cuticle,which is particularly obvious in the tail.The head is small and wide with intensive striates.The inner and outer labial sensilla are indistinct with short spicules and ventral apophysis,a gubernaculum with a block-shaped hook,a swollen conical-cylindrical tail and an absence of precloacal supplements. | ZHOU Xiping ZENG Jiali CAI Lizhe FU Sujing TAN Wenjuan | 2020 | Journal of Ocean University of China2020,19,1: | 0 |
