1Tobacco curly shoot virus isolated in Yunnan is a distinct species of Begomovirus显示文摘Virus isolate Y1 was obtained from tobacco showing curly shoot symptoms in Baoshan, Yunnan Province. Whitefly transmission test and virion morphology observa-tion showed that it is a begomovirus. In reactions with 14 monoclonal antibodies raised against begomoviruses, Y1 was readily differentiated from begomoviruses reported in China, Pakistan and India. The complete nucleotide sequence of DNA-A was determined, it contains 2746 nucleotides, with two ORFs in virion-sense DNA and four ORFs in comple-mentary-sense DNA. Comparisons with total DNA-A, inter-genie region and deduced amino acid sequences of individual ORFs showed that Y1 is a distinct Begomovirus species, for which the name Tobacco curly shoot virus (TCSV) is pro-posed. The total DNA-A of TCSV is most closely related to that of Tomato leaf curl virus from India (85% sequence identity). 2Sulfonated polyaniline/vanadate composite as anode material and its electrochemical property in microbial fuel cells on ocean floor显示文摘A unique sulfonated polyaniline/vanadate composite was synthesized and utilized as a composite anode in microbial fuel cells on ocean floor (BMFCs). X-ray diffraction (XRD) and thermogravimetric analysis (TGA) were employed to characterize its chemical composition and morphology. Wettability of the composite anodes decreases due to the addition of polytetrafluoroethylene (PTFE). The electrochemical behavior of the composite anodes was investigated by means of linear sweep voltammetry and Tafel plot measurements. Compared with the plain graphite anode,the composite anode significantly improves the power density,5.5-fold higher,reaching 187.1 mW/m2 and gives a 27-fold higher exchange current density and a higher kinetic activity. A novel synergistic mechanism between sulfonated polyaniline and vanadate is proposed to explain the excellent electrochemical performance. 3Purification and Characterization of a New Ribosome Inactivating Protein from Cinchonaglycoside C-treated Tobacco Leaves显示文摘A new ribosome-inactivating protein(RIP)with a molecular weight of 31 kDa induced by Cinchonaglycoside C(1)designatedCLP31,was isolated from tobacco leaves.Analysis of this protein sequence indicated that it belongs to the RIP family and itwas distinct from the other plant RIPs reported previously at its N-terminal amino acid sequence.CIP31 can directly impairsynthesis of coat protein(CP)of tobacco mosaic virus(TMV),which resulted in inhibition of TMV long distance movementand multiplication in tobacco plants at concentrations of ng/mL.Furthermore,no toxicity was shown to the growth andfertility of the plants.CIP31 was synthesized only in the presence of Cinchonaglycoside C(1)and was independent of thesalicylic acid(SA)signal pathway.We provided evidence for the SA-independent biological induction of resistance.Yanmei Li Yantao Jia Zhongkai Zhang Xiaoying Chen Hongping He Rongxiang Fang Xiaojiang Hao 2007Journal of Integrative Plant Biology2007,49,9:8
4Identification of a novel DNA molecule associated with Tobacco leaf curl virus显示文摘A circular DNA molecule, designated as DNAp, was identified in tobacco plants infected with Tobacco leaf curl virus (TLCV) isolates Y5 and Y8 by PCR using primers based on the conserved region of the two reported DNAp sequences of whitefly-transmitted geminiviruses (WTGs). The complete nucleotide sequences of DNAβ of Y5 and Y8 (TLCV DNAβ) were determined. Y5 DNAβ comprises 1333 nucleotides encoding 8 predicted ORFs with 4 ORFs in virion-sense DNA and 4 ORFs in complementary-sense DNA; Y8 DNAP consists of 1338 nucleotides encoding 7 predicted ORFs with 4 ORFs in virion-sense DNA and 3 ORFs in complementary-sense DNA. TLCV DNAβhas little sequence homology to DNA-A of TLCV, except that it shares conserved TAATATTAC loop sequence with TLCV DNA-A. 5Fragility assessment of tunnels in soft soils using artificial neural networks显示文摘Recent earthquakes have shown that tunnels are prone to damage,posing a major threat to safety and having major cascading and socioeconomic impacts.Therefore,reliable models are needed for the seismic fragility assessment of underground structures and the quantitative evaluation of expected losses.Based on previous researches,this paper presented a probabilistic framework based on an artificial neural network(ANN),aiming at the development of fragility curves for circular tunnels in soft soils.Initially,a two-dimensional incremental dynamic analysis of the nonlinear soil-tunnel system was performed to estimate the response of the tunnel under ground shaking.The effects of soil-structure-interaction and the ground motion characteristics on the seismic response and the fragility of tunnels were adequately considered within the proposed framework.An ANN was employed to develop a probabilistic seismic demand model,and its results were compared with the traditional linear regression models.Fragility curves were generated for various damage states,accounting for the associated uncertainties.The results indicate that the proposed ANN-based probabilistic framework can results in reliable fragility models,having similar capabilities as the traditional approaches,and a lower computational cost is required.The proposed fragility models can be adopted for the risk analysis of typical circular tunnel in soft soils subjected to seismic loading,and they are expected to facilitate decision-making and risk management toward more resilient transport infrastructure.Zhongkai Huang Sotirios A.Argyroudis Kyriazis Pitilakis Dongmei Zhang Grigorios Tsinidis 2022Underground Space2022,7,2:4
6Distribution Features of the Nanhua-Sinian Rifts and their Significance to Hydrocarbon Accumulation in the Tarim Basin显示文摘On the basis of reprocessing 34 new two-dimensional spliced long sections(20,191 km) in the Tarim Basin, the deep structure features of the Tarim Basin were analyzed through interpreting 30,451 km of two-dimensional seismic data and compiling basic maps. Seismic interpretation and geological analysis conclude that the Nanhua-Sinian strata are a set of rift-depression depositional systems according to their tectonic and depositional features. The rift valley formed in the Nanhua Period, and the transformation became weaker during the late Sinian Period, which eventually turned into depression. From bottom to top, the deposited strata include mafic igneous, tillite, mudstone, and dolomite. Three major depocenters developed inside this basin during the rift stage and are distributed in the eastern Tarim Basin, the Awati area, and the southwestern Tarim Basin. Among them, the rift in the eastern Tarim Basin strikes in the near east-west direction on the plane and coincides with the aeromagnetic anomaly belt. This represents a strong magnetic zone formed by upwelling basic volcanic rock along high, steep normal faults of the Nanhua Period. Controlled by the tectonic background, two types of sedimentary systems were developed in the rift stage and depression stage, showing two types of sequence features in the Sinian depositional stage. The Nanhua System appears as a wedge-shaped formation, with its bottom in unconformable contact with the base. The rifting event has a strong influence on the current tectonic units in the Tarim Basin, and affects the distribution of source rock in the Yuertus Formation and reservoir beds in the Xiaoerbulake Formation in Lower Cambrian, as well as the gypseous cap rock in Middle Cambrian. The distribution features of the rifts have important and realistic significance for determining the direction of oil and gas exploration in the deep strata of the Tarim Basin. 10Flexible temperature sensor with high sensitivity ranging from liquid nitrogen temperature to 1200℃显示文摘Flexible temperature sensors have been extensively investigated due to their prospect of wide application in various flexible electronic products.However,most of the current flexible temperature sensors only work well in a narrow temperature range,with their application at high or low temperatures still being a big challenge.This work proposes a flexible thermocouple temperature sensor based on aerogel blanket substrate,the temperature-sensitive layer of which uses the screen-printing technology to prepare indium oxide and indium tin oxide.It has good temperature sensitivity,with the test sensitivity reaching 226.7μV℃^(−1).Most importantly,it can work in a wide temperature range,from extremely low temperatures down to liquid nitrogen temperature to high temperatures up to 1200℃,which is difficult to be achieved by other existing flexible temperature sensors.This temperature sensor has huge application potential in biomedicine,aerospace and other fields.Zhaojun Liu Bian Tian Zhuangde Jiang Shuimin Li Jiaming Lei Zhongkai Zhang Jiangjiang Liu Peng Shi Qijing Lin 2023International Journal of Extreme Manufacturing2023,5,1:1
11Functional analysis of the promoter of the dmrt1 gene in Chinese tongue sole,Cynoglossus semilaevis显示文摘The dmrt1 (doublesex and mab-3 related transcription factor 1) gene is considered to play an essential role in testis differentiation and development among metazoan species. As an economically important marine fish in China, Chinese tongue sole (Cynoglossus semilaevis) possess a ZW/ZZ sex-determining system, and the females usually grow two to four times bigger than males. Previous studies have confirmed that Z-linked dmrt1 gene is the male determining gene in C. semilaevis and TALENs-mediated knocking-out individuals showed increased growth phenotype. In order to investigate the function of dmrt1 regulatory regions and its potential application, we have cloned the promoter of C. semilaevis dmrt1. The acquired sequence consisted of a 5'-upstream sequence and a partial exon1;functional analysis showed that there were two positive regulatory regions in the promoter, as well as one negative region and one neutral region. These positive regions were then combined to obtain several newly constructed promoters. Both in vitro and in vivo analysis confirmed one of them (dmrt1-Δ3+Δ4) showed elevated promoting activity compared to the full-length promoter and could drive exogenous gene expression in tongue sole gonads. 14Auto machine learning-based modelling and prediction of excavationinduced tunnel displacement显示文摘The influence of a deep excavation on existing shield tunnels nearby is a vital issue in tunnelling engineering.Whereas,there lacks robust methods to predict excavation-induced tunnel displacements.In this study,an auto machine learning(AutoML)-based approach is proposed to precisely solve the issue.Seven input parameters are considered in the database covering two physical aspects,namely soil property,and spatial characteristics of the deep excavation.The 10-fold cross-validation method is employed to overcome the scarcity of data,and promote model’s robustness.Six genetic algorithm(GA)-ML models are established as well for comparison.The results indicated that the proposed AutoML model is a comprehensive model that integrates efficiency and robustness.Importance analysis reveals that the ratio of the average shear strength to the vertical effective stress E_(ur)/σ′_(v),the excavation depth H,and the excavation width B are the most influential variables for the displacements.Finally,the AutoML model is further validated by practical engineering.The prediction results are in a good agreement with monitoring data,signifying that our model can be applied in real projects.Dongmei Zhang Yiming Shen Zhongkai Huang Xiaochuang Xie 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,4:1
