1Virus-induced gene silencing and its application in plant functional genomics显示文摘Virus-induced gene silencing is regarded as a powerful and efficient tool for the analysis of gene function in plants because it is simple, rapid and transformation-free. It has been used to perform both forward and reverse genetics to identify plant functional genes. Many viruses have been developed into virus-induced gene silencing vectors and gene functions involved in development, biotic and abiotic stresses, metabolism, and cellular signaling have been reported. 2Identification of a novel DNA molecule associated with Tobacco leaf curl virus显示文摘A circular DNA molecule, designated as DNAp, was identified in tobacco plants infected with Tobacco leaf curl virus (TLCV) isolates Y5 and Y8 by PCR using primers based on the conserved region of the two reported DNAp sequences of whitefly-transmitted geminiviruses (WTGs). The complete nucleotide sequences of DNAβ of Y5 and Y8 (TLCV DNAβ) were determined. Y5 DNAβ comprises 1333 nucleotides encoding 8 predicted ORFs with 4 ORFs in virion-sense DNA and 4 ORFs in complementary-sense DNA; Y8 DNAP consists of 1338 nucleotides encoding 7 predicted ORFs with 4 ORFs in virion-sense DNA and 3 ORFs in complementary-sense DNA. TLCV DNAβhas little sequence homology to DNA-A of TLCV, except that it shares conserved TAATATTAC loop sequence with TLCV DNA-A. Sequence comparison showed that Y5 DNAβ shared 85% sequence homology with Y8 DNAβ, and both Y5 DNAβ and Y8 DNAβ had relatively low sequence identity (51%-65%) with the reported DNAβ molecules associated with Agera-tum yellow vein virus and Cotton leafXIE Yan ZHOU Xueping LI Zhenghe ZHANG Zhongkai LI Guixin 2002Chinese Science Bulletin2002,47,15:6
3Physics issues in diffraction limited storage ring design显示文摘Diffraction limited electron storage ring is considered a promising candidate for future light sources,whose main characteristics are higher brilliance,better transverse coherence and better stability.The challenge of diffraction limited storage ring design is how to achieve the ultra low beam emittance with acceptable nonlinear performance.Effective linear and nonlinear parameter optimization methods based on Artificial Intelligence were developed for the storage ring physical design.As an example of application,partial physical design of HALS(Hefei Advanced Light Source),which is a diffraction limited VUV and soft X-ray light source,was introduced.Severe emittance growth due to the Intra Beam Scattering effect,which is the main obstacle to achieve ultra low emittance,was estimated quantitatively and possible cures were discussed.It is inspiring that better performance of diffraction limited storage ring can be achieved in principle with careful parameter optimization.FAN Wei BAI ZhengHe GAO WeiWei FENG GuangYao LI WeiMin WANG Lin HE DuoHui 2012Science China(Physics,Mechanics & Astronomy)2012,55,5:2
4Engineered biocontainable RNA virus vectors for non-transgenic genome editing across crop species and genotypes显示文摘CRISPR/Cas genome-editing tools provide unprecedented opportunities for basic plant biology research and crop breeding.However,the lack of robust delivery methods has limited the widespread adoption of these revolutionary technologies in plant science.Here,we report an efficient,non-transgenic CRISPR/Cas delivery platform based on the engineered tomato spotted wilt virus(TSWV),an RNA virus with a host range of over 1000 plant species.We eliminated viral elements essential for insect transmission to liberate genome space for accommodating large genetic cargoes without sacrificing the ability to infect plant hosts.The resulting non-insect-transmissible viral vectors enabled effective and stable in planta delivery of Cas12a and Cas9 nucleases as well as adenine and cytosine base editors.In systemically infected plant tissues,the deconstructed TSWV-derived vectors induced efficient somatic gene mutations and base conversions in multiple crop species with little genotype dependency.Plants with heritable,bi-allelic mutations could be readily regenerated by culturing the virus-infected tissues in vitro without antibiotic selection.Moreover,we showed that antiviral treatment with ribavirin during tissue culture cleared the viral vectors in 100%of regenerated plants and further augmented the recovery of heritable mutations.Because many plants are recalcitrant to stable transformation,the viral delivery system developed in this work provides a promising tool to overcome gene delivery bottlenecks for genome editing in various crop species and elite varieties.Qian Liu Chenglu Zhao Kai Sun Yinlu Deng Zhenghe Li 2023Molecular Plant2023,16,3:2

12Omnidirectional Antenna Diversity System for High-Speed Onboard Communication显示文摘In this article,an omnidirectional dual-polarized antenna with synergetic electromagnetic and aerodynamic properties is propounded for high-speed diversity systems.The propounded antenna comprises a probe-fed cavity for horizontally polarized radiation and a microstrip-fed slot for vertical polarization.Double-layer metasurfaces are properly designed as artificial magnetic conductor boundaries with direct metal-mountable onboard installation and compact sizes.An attached wedge-shaped block is utilized for windage reduction in hydrodynamics.The propounded antenna is fabricated for design verification,and the experimental results agree well with the simulated ones.For vertical polarization,the operating bandwidth is in the range of 2.37–2.55 GHz,and the realized gain variation in the azimuthal radiation pattern is 3.67 decibels(dB).While an impedance bandwidth in the range of 2.45–2.47 GHz and a gain variation of 3.71 dB are also achieved for horizontal polarization.A port isolation more than 33 dB is obtained in a compact volume of 0.247λ_(0)×0.345λ_(0)×0.074λ_(0),whereλ_(0)represents the wavelength in vacuum at the center frequency,wherein the wedge-shaped block is included.The propounded diversity antenna has electromagnetic and aerodynamic merits,and exhibits an excellent potential for high-speed onboard communication.Yongjian Zhang Yue Li Weiquan Zhang Zhijun Zhang Zhenghe Feng 2022Engineering2022,8,4:0
13Analysis of black soil compaction with driver-agricultural machinery-soil system under corn sowing with high-power tractor in Northeast China显示文摘Soil compaction leads to crop yield reduction in Northeast of China.The interaction mechanism of driveragricultural machinery-black soil is not clear.A comprehensive field experiment of 4 hm2 of maize seeding was carried out in Baiquan County Cooperative.The results showed that the average increase rates of soil compaction before and after sowing were 118.82%and 71.02%.The SEM showed that waist fatigue had the greatest impact on soil compaction,and the unit fatigue of waist caused 1.51 and 1.27 unit compactions to the soil at the depths of 10 cm and 20 cm.The neck,waist,arm and leg fatigue of drivers increased the surface soil compaction by 1.83,1.76,1.78 and 1.55 units,and the deep soil compaction by 1.65,1.58,1.60 and 1.40 units.The results can provide a reference for the integration of human factor efficiency and conservation tillage.Xiao Yang Zhiqiang Zhai Weijie Guo Wenjie Li Minli Yang Zhenghe Song 2023International Journal of Agricultural and Biological Engineering2023,16,4:0

15Effect of Culture Parameters on the Growth of Sparassis crispa Mycelium显示文摘The effect of carbon and nitrogen sources, pH, temperature and substrate water content on the growth and vigor of Sparassis crispa mycelium was evaluated. Mycelial growth rates were highest when rice starch served as the carbon source and when peptone, yeast extract or soybean meal were adopted as the nitrogen source. The optimum pH and substrate water content values were 5.25 and 62.5% respectively. 16PD-L1 expression is regulated by ATP-binding of the ERBB3 pseudokinase domain显示文摘How PD-L1 expression is regulated in cancer is poorly understood.Here,we report that the ATP-binding activity of ERBB3 pseudokinase regulates PD-L1 gene expression in colorectal cancers(CRCs).ERBB3 is one of the four members of the EGF receptor family,all with protein tyrosine kinase domains.ERBB3 is a pseudokinase with a high binding affin-ity to ATP.We showed that ERBB3 ATP-binding inactivation mutant reduces tumorigenicity in genetically engineered mouse models and impairs xenograft tumor growth of CRC cell lines.The ERBB3 ATP-binding mutant cells dramatically reduce IFN-g-induced PD-L1 expres-sion.Mechanistically,ERBB3 regulates IFN-g-induced PD-L1 expression through the IRS1-PI3K-PDK1-RSK-CREB signaling axis.CREB is the transcription factor that regulates PD-L1 gene expression in CRC cells.Knockin of a tumor-derived ERBB3 mutation located in the ki-nase domain sensitizes mouse colon cancers to anti-PD1 antibody therapy,suggesting that ERBB3 mutations could be predictive biomarkers for tumors amenable to immune check-point therapy.Yamu Li Zhonghua Liu Yiqing Zhao Jie Yang Tsan Sam Xiao Ronald A.Conlon Zhenghe Wang 2023Genes & Diseases2023,10,4:0

18High-Q collective Mie resonances in monocrystalline silicon nanoantenna arrays for the visible light显示文摘Dielectric optical antennas have emerged as a promising nanophotonic architecture for manipulating the propagation and localization of light.However,the optically induced Mie resonances in an isolated nanoantenna are normally with broad spectra and poor��-factors,limiting their performances in sensing,lasing,and nonlinear optics.Here,we dramatically enhance the��-factors of Mie resonances in silicon(Si)nanoparticles across the optical band by arranging the nanoparticles in a periodic lattice.We select monocrystalline Si with negligible material losses and develop a unique method to fabricate nanoparticle arrays on a quartz substrate.By extinction dispersion measurements and electromagnetic analysis,we can identify three types of collective Mie resonances with��-factors∼500 in the same nanocylinder arrays,including surface lattice resonances,bound states in the continuum,and quasi-guided modes.Our work paves the way for fundamental research in strong light-matter interactions and the design of highly efficient light-emitting metasurfaces.Zhenghe Zhang Pengbo Liu Wanli Lu Ping Bai Bingchang Zhang Zefeng Chen Stefan A.Maier Jaime Gómez Rivas Shaojun Wang Xiaofeng Li 2023Fundamental Research2023,3,5:0
19Design and experiment of a high-clearance self-propelled sprayer chassis显示文摘A large high-clearance self-propelled sprayer chassis was designed according to the requirements of the sprayer’s driving operation and the overall goal.The structural characteristics and working principle of the chassis drive system,suspension system,track adjustment system,and frame were analyzed.A finite element analysis of the frame was carried out under four extreme conditions:full load bending,full load torsion,full load emergency braking and full load emergency turning.Under these four conditions,the maximum stress values of the frame were 149.45 MPa,219 MPa,151.44 MPa,and 123.27 MPa respectively,and the maximum strain values were 1.12 mm,2.22 mm,0.95 mm,and 1.16 mm respectively.A theoretical analysis of the stability,steering ability,and obstacle navigation of the sprayer chassis was conducted.The results showed that the upper angleθlim,lower angleθ׳lim and lateral limit tumbling angleφlim of the sprayer chassis were 50.1°,30.0°and 35.3°respectively.The minimum turning radius of the chassis was 5816 mm,and the minimum turning width was 4113 mm.The maximum obstacle crossing heights of the front and rear wheels were 466 mm and 457 mm.The theoretical analysis showed that the designed chassis met the field management operations of large plots.A field experiment of the high-clearance self-propelled sprayer chassis at full load was conducted.The experimental results were consistent with the theoretical analysis results.The sprayer chassis not only good handling stability but also good ride comfort.The results of this study provide references for the design of high-clearance self-propelled sprayer chassis.Yu Chen Enrong Mao Wei Li Shuo Zhang Zhenghe Song Shanju Yang Jun Chen 2020International Journal of Agricultural and Biological Engineering2020,13,2:0
20Dispersion coding of ENZ media via multiple photonic dopants显示文摘Epsilon-near-zero (ENZ) media are opening up exciting opportunities to observe exotic wave phenomena. In this work, we demonstrate that the ENZ medium comprising multiple dielectric photonic dopants would yield a comb-like dispersion of the effective permeability, with each magnetic resonance dominated by one specific dopant. Furthermore, at multiple frequencies of interest, the resonant supercouplings appearing or not can be controlled discretely via whether corresponding dopants are assigned or not. Importantly, the multiple dopants in the ENZ host at their magnetic resonances are demonstrated to be independent. Based on this platform, the concept of dispersion coding is proposed, where photonic dopants serve as “bits” to program the spectral response of the whole composite medium. As a proof of concept, a compact multi-doped ENZ cavity is fabricated and experimentally characterized, whose transmission spectrum is manifested as a multi-bit reconfigurable frequency comb.
