2Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. 3An imputed whole-genome sequence-based GWAS approach pinpoints causal mutations for complex traits in a specific swine population显示文摘Sequencing-based genome-wide association studies(GWAS) have facilitated the identification of causal associations between genetic variants and traits in diverse species. However, it is cost-prohibitive for the majority of research groups to sequence a large number of samples. Here, we carried out genotype imputation to increase the density of single nucleotide polymorphisms in a large-scale Swine F;population using a reference panel including 117 individuals, followed by a series of GWAS analyses. The imputation accuracies reached 0.89 and 0.86 for allelic concordance and correlation, respectively. A quantitative trait nucleotide(QTN) affecting the chest vertebrate was detected directly, while the investigation of another QTN affecting the residual glucose failed due to the presence of similar haplotypes carrying wild-type and mutant allelesin the reference panel used in this study. A high imputation accuracy was confirmed by Sanger sequencing technology for the most significant loci. Two candidate genes,CPNE5 and MYH3, affecting meat-related traits were proposed. 6Effects of gamma-aminobutyric acid A-receptor antagonist on sleep-wakefulness cycles following lesion to the ventrolateral preoptic area in rats显示文摘BACKGROUND: Neurons expressing gamma-aminobutyric acid (GABA) play an important role in the regulation of wakefulness to sleep, as well as the maintenance of sleep. However, the role of GABAergic neurons in the tuberomammillary nucleus (TMn), with regard to the sleep-wakefulness cycle, is poorly understood. OBJECTIVE: To investigate the effects of GABAergic neurons in the TMn on the sleep-wakefulness cycle. DESIGN, TIME AND SETTING: Randomized controlled study, performed at the Laboratory of Neurobiology, Department of Anatomy, Histology and Embryology, School of Basic Medical Sciences, Lanzhou University from July 2007 to February 2008. MATERIALS: Fifteen healthy, adult, male, Sprague Dawley rats were randomly divided into three groups(n = 5): control, ventrolateral preoptic area (VLPO) lesion, and VLPO lesion plus GABAA receptor antagonist-treated. Ibotenic acid and bicuculline were provided by Sigma (St. Louis, USA). METHODS: Four electroencephalogram screw electrodes were implanted into the skull at a frontal region (two) and parietal bones (two) on each side. Three flexible electromyogram wire electrodes were placed into the nuchal muscles. On day 8, a fine glass micropipette (10–20 mm tip diameter) containing ibotenic acid solution (10 nmol/L) was injected into the VLPO in both hemispheres following bone wax removal under anesthesia. One week after the second surgery, sleep-wakefulness states were recorded in rats from the VLPO lesion group. On day 10 after VLPO lesion, bicuculline (10 nmol/L), a GABAA-receptor antagonist, was microinjected into the TMn and sleep-wakefulness states were recorded for 24 hours. MAIN OUTCOME MEASURES: Duration of the sleep-wakefulness cycle in each group using a Data acquisition unit (Micro1 401 mk2) and Data collection software (Spike Ⅱ). RESULTS: VLPO lesion induced an increased duration of wakefulness (W, 13.17%) and light slow-wave sleep (SWS1, 28.9%), respectively. Deep slow-wave sleep (SWS2, 43.74%) and paradoxical sleep (PS, 44.07%) were respectively decreased for 24 hours at day 9 post-lesion, compared with pre-lesion (P < 0.01). Microinjection of bicuculline into the TMn following VLPO lesion at 10:00 am on the 10th day elicited a wake state for 40–55 minutes, with a latency of 15 minutes. However, 24-hour sleep-wake states demonstrated that the ratio of W and SWS1 were increased by 12.61% (P < 0.01) and 50.97% (P < 0.01), respectively. In addition, SWS2 and PS were decreased by 68.08% (P < 0.01) and 39.92% (P < 0.05), respectively, compared with prior to VLPO lesion. CONCLUSION: The evidence of decreased deep slow-wave sleep, which was induced by VLPO lesion, suggested that GABAergic neurons in the VLPO play an important role in maintaining sleep. Bicuculline microinjection into the TMn, following VLPO lesion, elicited wakefulness and sleep depression for 50 minutes, with contrary increased light slow-wave sleep for 24 hours, which suggested that GABAergic neurons in the TMn play a role in sleep drive (sleepiness) via local circuit to directly inhibit histaminergic neurons.Xin Zhang Yina Sun Peng Xie Xuguang Yang Yiping Hou 2009Neural Regeneration Research2009,4,1:1
7Utility of Triti-Map for bulk-segregated mapping of causal genes and regulatory elements in Triticeae显示文摘Triticeae species,including wheat,barley,and rye,are critical for global food security.Mapping agronomically important genes is crucial for elucidating molecular mechanisms and improving crops.However,Triticeae includes many wild relatives with desirable agronomic traits,and frequent introgressions occurred during Triticeae evolution and domestication.Thus,Triticeae genomes are generally large and complex,making the localization of genes or functional elements that control agronomic traits challenging.Here,we developed Triti-Map,which contains a suite of user-friendly computational packages specifically designed and optimized to overcome the obstacles of gene mapping in Triticeae,as well as a web interface integrating multi-omics data from Triticeae for the efficient mining of genes or functional elements that control particular traits.The Triti-Map pipeline accepts bothDNA and RNAbulk-segregated sequencing data as well as traditional QTL data as inputs for locating genes and elucidating their functions.We illustrate the usage of Triti-Map with a combination of bulk-segregated ChIP-seq data to detect a wheat disease-resistance gene with its promoter sequence that is absent from the reference genome and clarify its evolutionary process.We hope that Triti-Map will facilitate gene isolation and accelerate Triticeae breeding.Fei Zhao Shilong Tian Qiuhong Wu Zijuan Li Luhuan Ye Yili Zhuang Meiyue Wang Yilin Xie Shenghao Zou Wan Teng Yiping Tong Dingzhong Tang Ajay Kumar Mahato Moussa Benhamed Zhiyong Liu Yijing Zhang 2022Plant Communications2022,3,4:1

9Effects of the layer thickness ratio on the enhanced ductility of laminated aluminum显示文摘The layered structural parameters have been reported to be critical for tuning the tensile properties of laminated metals.Here,we investigated the effects of the thickness ratio(rc/f)of coarse-grained layers(CLs)to fine-grained layers(FLs)on the enhanced ductility of the laminated Al.The local strain evolution demonstrates that the strain delocalization ability of laminated Al is improved with the decrease of rc/f.The interfacial strain gradients,which can produce extra work hardening,gradually approach and cover the CLs with the rc/fdecreasing,explaining the trend of uniform elongation in laminated Al with various rc/f.The integrated fracture morphology characterization reveals that the increase of the rc/fleads to an improvement in the tolerance of the interfacial microcracks,which is corresponding to the variation of fracture elongation in the laminated Al.Moreover,there is an evident transition of transverse propagation path of interfacial microcracks from the CLs to FLs with increasing the rc/f.Based on a geometrical criterion of microcracks connectivity,the preferential transverse propagation path of interfacial microcracks in these laminated Al was rationalized.The calculation based on this criterion also predicted the critical rc/fcorresponding to the optimal combination of strength and fracture elongation.This work deepens the understanding of the role of structural parameters of laminated metals in achieving the strength and ductility synergy.Yiping Xia Hao Wu Kesong Miao Xuewen Li Chao Xu Lin Geng Honglan Xie Guohua Fan 2022Journal of Materials Science & Technology2022,,16:1

16Unclear but present danger:An asymptomatic SARS-CoV-2 carrier显示文摘Superspreaders are critical infectious resources in multiple infectious diseases.They can be asymptomatic or present mild symptoms but can transmit pathogens to susceptible populations,leading to severe symptoms,and even death.Early identification of this population is extremely important to inhibit the spread of infectious diseases.Right now,the whole global world is suffering from a devastating infectious disease caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).In this article,a superspreader cluster event in coronavirus disease 2019(COVID-19)was identified by tracking contacting histories of infected patients.This cluster was found to be originated from an asymptomatic SARS-CoV-2 carrier,which resulted in 13 secondary cases getting infected.All the secondary patients presented with non-typical symptoms of COVID-19,such as fever,dry cough,and myalgia,one of which died of respiratory failure at the end.From this cluster,we learn that people with older ages,low immunity,multiple underlying diseases,especially pulmonary diseases,can contribute to a poor prognosis.Thus,asymptomatic superspreaders of COVID-19 can be extremely dangerous and must be handled time-efficiently.Xuejing Yu Dongchuan Ran Jinhui Wang Yuan Qin Ruishan Liu Xueli Shi Yiping Wang Chang Xie Jia Jiang Jianzhong Zhou 2020Genes & Diseases2020,7,4:0

18Breeding and Cultivation Techniques of a New Table Use Purple-fleshed Sweetpotato Cultivar,Xuzishu 2显示文摘Xuzishu 2 is a new high-quality table use purple-fleshed sweetpotato cultivar bred from hybrid progenies of AIS35-2 × Xushu 22-5 by Jiangsu Xuzhou Sweetpotato Research Center. In the sweetpotato variety regional trial of Jiangsu Province during 2007- 2008,average fresh storage root yield was 30 185. 3kg / hm2; average dry matter content was 29. 26%; average dry storage root yield was 8 868. 7 kg/hm2. In the sweetpotato variety production test of Jiangsu Province,average fresh storage root yield was 30 683. 9 kg / hm2; average dry storage root yield was 9 399. 0 kg/hm2. Multi-location sampling analysis in the regional trial indicated that the anthocyanin content in fresh storage roots was 104. 6 μg / g. Xuzishu 2 was approved by Crop Variety Approval Committee of Jiangsu Province in March 2010,which exhibited high fresh storage root yield,sweet taste,high anthocyanin content,good commodity,root rot resistance,and was tolerant to moisture and easy to store. 19Neddylation is a novel therapeutic target for lupus by regulating double negative T cell homeostasis显示文摘Systemic lupus erythematosus(SLE),a severe autoimmune disorder,is characterized by systemic inflammatory response,autoantibody accumulation and damage to organs.The dysregulation of double-negative(DN)T cells is considered as a crucial commander during SLE.Neddylation,a significant type of protein post-translational modification(PTM),has been well-proved to regulate T cell-mediated immune response.However,the function of neddylation in SLE is still unknown.Here,we reported that neddylation inactivation with MLN4924,a specific inhibitor of NEDD8-activating enzyme E1(NAE1),or genetic abrogation of Ube2m in T cells decreased DN T cell accumulation and attenuated murine lupus development.Further investigations revealed that inactivation of neddylation blocked Bim ubiquitination degradation and maintained Bim level in DN T cells,contributing to the apoptosis of the accumulated DN T cells in lupus mice.Then double knockout(KO)lupus-prone mice(Ube2m-/-Bim-/-lpr)were generated and results showed that loss of Bim reduced Ube2m deficiency-induced apoptosis in DN T cells and reversed the alleviated lupus progression.Our findings identified that neddylation inactivation promoted Bim-mediated DN T cell apoptosis and attenuated lupus progression.Clinically,we also found that in SLE patients,the proportion of DN T cells was raised and their apoptosis was reduced.Moreover,compared to healthy groups,SLE patients exhibited decreased Bim levels and elevated Cullin1 neddylation levels.Meantime,the inhibition of neddylation induced Bim-dependent apoptosis of DN T cells isolated from SLE patients.Altogether,our findings provide the direct evidence about the function of neddylation during lupus,suggesting a promising therapeutic approach for this disease.Yun Zhang Lijun Du Chenxi Wang Zhangsheng Jiang Qingchi Duan Yiping Li Zhijun Xie Zhixing He Yi Sun Lin Huang Liwei Lu Chengping Wen 2024Signal Transduction and Targeted Therapy2024,9,2:0
20Population genetics and forensic efficiency of 30 InDel markers in four Chinese ethnic groups residing in Sichuan显示文摘Sichuan Province is located at the transitional junction regions of the Qinghai-Tibet Plateau and the low-altitude plains.It also serves as the corridor of Sino Tibetan-speaking population migration and expansion since neolithic expansion of Proto-Tibeto-Burman populations from Middle/Upper Yellow River during Majiayao period(3300-2000 BC).However,the population structure and the corresponding genetic diversity of forensic-related markers in this region remain unclear.Thus,we genotyped 30 insertion-deletion(InDel)markers in 444 samples from four ethnic groups(Han,Tibetan,Hui and Yi)from Sichuan Province using the Investigator■ DIPplex kit to explore the characteristics of population genetics and forensic genetic focuses.All the loci were found to be in Hardy-Weinberg Equlibrium(HWE)after applying a Bonferroni correction and no pairwise loci showed prominent linkage disequilibrium.The combined matching probability(CMP)and the combined power of discrimination(CPD)are larger than 1.8089×10^(11)and 0.9999999995,respectively.Principal component analysis,multi-dimen-sional scaling plots and Neighbour-Joining tree among 65 worldwide populations indicated that Sichuan Hui and Han are genetically close to Hmong-Mien and Tai-Kadai-speaking popula-tions,and Sichuan Tibetan and Yi bear a strong genetic affinity with Tibeto-Burman-speaking populations.The model-based genetic structure further supports the genetic affinity between the studied populations and linguistically close populations.Fei Wang Guanglin He Zheng Wang Mengge Wang Jing Liu Xing Zou Shouyu Wang Mengyuan Song Ziwei Ye Mingkun Xie Yiping Hou 2022Forensic Sciences Research2022,7,3:0
