1Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. 7Growth and characterization of 0.8-μm gate length AlGaN/GaN HEMTs on sapphire substrates显示文摘AlGaN/GaN high electron mobility transistor (HEMT) structures were grown on 2 inch sapphire substrates by MOCVD, and 0.8-μm gate length devices were fabricated and measured. It is shown by resistance mapping that the HEMT structures have an average sheet resistance of approximately 380 ?/sq with a uniformity of more than 96%. The 1-mm gate width devices using the materials yielded a pulsed drain current of 784 mA/mm at Vgs=0.5 V and Vds=7 V with an extrinsic transconductance of 200 mS/mm. A 20-GHz unity current gain cutoff frequency (?T ) and a 28-GHz maximum oscillation frequency (?max) were obtained. 8Establishment of the model of vascular endothelial cell membrane chromatography and its preliminary application显示文摘A model of vascular endothelial cell membrane chromatography was established by using an ECV304 cell membrane stationary phase (ECV304 CMSP) prepared by immobilizing the ECV304 cell membrane onto the surface of silica carrier. The surface and chromatographic characteristics of ECV304 CMSP were studied. The active component from Caulophyllum robustum was screened by using the model of vascular endothelial cell membrane chromatography. The interaction between the active component and membrane receptor was determined by using a replace experiments. The effect of the active component was tested by using tube formation of ECV304 cell. The results indicated that the model of ECV304 cell membrane chromatograph (ECV304 CMC) can stimulate the interaction between drug and receptor in vitro and the retention characteristics of taspine as active component was similar to that of model molecule in the model of ECV304 CMC. And therefore, taspine acted on VEGFR2 and inhibited the tube formation of ECV304 cell induced by VEGF. 9Efficacy and advantages of modified Traditional Chinese Medicine treatments based on'kidney reinforcing'for chronic aplastic ane-mia:a randomized controlled clinical trial显示文摘OBJECTIVE:To compare the efficacy of modified treatments based on 'kidney reinforcing' in the management of chronic aplastic anemia(CAA),and explore their advantages and specialties.METHODS:One hundred and eleven patients with CAA were randomly divided into three groups:kidney reinforcing alone(KA),'kidney reinforcing and Qi tonifying'(KQ),and 'kidney reinforcing and blood circulation invigorating'(KC).Normal and positive control groups were also formed.All patients were treated for 6 months(two courses).Hemograms,Traditional Chinese Medicine(TCM) syndrome scores,and therapeutic effects were assessed,and changes in T-lymphocyte subsets,regulatory T cells and cytokines were detected.RESULTS:The KQ and KC groups had lower TCM syndrome scores than the positive control group after 6 months(P < 0.05).The KQ group had a higher overall efficacy than the positive control group after 3 months(P < 0.05),while platelet counts increased in the KC group after 6 months(P < 0.05).CD3+ T-lymphocyte ratios decreased only in the KQ group,while CD3 + CD4 + CD8-Tlymphocytes increased only in the KC group after 6 months(P <0.05).Levels of interferon-γ,tumor necrosis fac-tor-α,interleukin(IL)-2 and IL-6 decreased and levels of IL-4 and IL-10 increased in all treated groups after 6 months.Levels of IL-6 in the KQ and KC groups were lower than those in the positive control group(P < 0.05).CONCLUSION:Treatments based on kidney reinforcing have a rebalancing effect on cytotoxic and T helper cells,and regulate expression of interferon-γ,IL-2,IL-6 and IL-4.KQ may be more effective in treating CAA,and KC may have an advantage in platelet recovery.Wu Dijiong Shen Yiping Ye Baodong Fang Bingmu Lin Shengyun Chen Zhilu Jiang Huifang Feng Changwei He Lüyuan Gao Yanting Liu Yonglin Liu Yonghua Zhu Jiajia Wu Liqiang Shao Keding Zhou Yuhong 2016Journal of Traditional Chinese Medicine2016,36,4:6
10Review of Femtosecond-Laser-Inscribed Fiber Bragg Gratings:Fabrication Technologies and Sensing Applications显示文摘Fiber Bragg grating(FBG)is the most widely used optical fiber sensor due to its compact size,high sensitivity,and easiness for multiplexing.Conventional FBGs fabricated by using an ultraviolet(UV)laser phase-mask method require the sensitization of the optical fiber and could not be used at high temperatures.Recently,the fabrication of FBGs by using a femtosecond laser has attracted extensive interests due to its excellent flexibility in creating FBGs array or special FBGs with complex spectra.The femtosecond laser could also be used for inscribing various FBGs on almost all fiber types,even fibers without any photosensitivity.Such femtosecond-laser-induced FBGs exhibit excellent thermal stability,which is suitable for sensing in harsh environment.In this review,we present the historical developments and recent advances in the fabrication technologies and sensing applications of femtosecond-laser-inscribed FBGs.Firstly,the mechanism of femtosecond-laser-induced material modification is introduced.And then,three different fabrication tech no logies,i.e.,femtosecond laser phase mask technology,femtosecond laser holographic interferometry,and femtosecond laser direct writing technology,are discussed.Finally,the advances in high-temperature sensing applications and vector bending sensing applications of various femtosecond-laser-inscribed FBGs are summarized.Such femtosecond-laser-inscribed FBGs are promising in many industrial areas,such as aerospace vehicles,nuclear plants,oil and gas explorations,and advanced robotics in harsh environments.Jun HE Baijie XU Xizhen XU Changrui LIAO Yiping WANG 2021Photonic Sensors2021,11,2:6
11Xeno-repopulation of Fah^(-/-)Nod/Scid mice livers by human hepatocytes显示文摘Functional human hepatocytes xenografted into the liver of mice can be used as a model system to study pharmacokinetics,infection of hepatitis viruses,and the efficacy of hepatitis vaccines.Significant levels of liver xeno-repopulation have been reported in Fah-/-Rag2-/-Il2rg-/-mice.However,the high mortality and low breeding rate of this model may hinder its application.A new model,termed Fah-/-Nod/Scid mice,which combines the advantages of liver repopulation in Fah-/-mice with the ease of xenotransplantation in Nod/Scid mice was obtained by gradual cross-breeding.Fah-/-Nod/Scid mice were easily maintained in breeding colonies and in adult animal care facilities.FK506 treatment combined with gradual withdrawal of NTBC before cell transplantation ensured that Fah-/-Nod/Scid mice were susceptible to liver xeno-repopulation by human hepatocytes;the proportion of engrafted human hepatocytes reached 33.6%.The function of the expanded human hepatocytes within the chimeric liver was confirmed by weight curve analysis,the expression of characteristic proteins,and the biochemical analysis of liver function.These results show that Fah-/-Nod/Scid mice are an ideal humanized liver mouse model with many useful applications.SU BaoLiang LIU ChangCheng XIANG Dao ZHANG HaiBin YUAN SiMing WANG MinJun CHEN Fei ZHU HaiYing HE ZhiYing WANG Xin HU YiPing 2011Science China(Life Sciences)2011,54,3:5

14Single-mode helical Bragg grating waveguide created in a multimode coreless fiber by femtosecond laser direct writing显示文摘We demonstrate the fabrication of single-mode helical Bragg grating waveguides(HBGWs)in a multimode core-less fiber by using a femtosecond laser direct writing technique.This approach provides a single-step method for creating Bragg grating waveguides.Specifically,the unique helical structure in such an HBGW serves as a depressed cladding waveguide and also generates strong Bragg resonance due to its periodicity.Effects of pulse energy,helical diameter,and helical pitch used for fabricating HBGWs were studied,and a single-mode HBGW with a narrow bandwidth of 0.43 nm and a Bragg wavelength of 1546.50 nm was achieved by using appropriate parameters,including a diameter of 10μm and a helical pitch of 1.07μm.The measured cross-sectional refractive index profile indicates that a depressed cladding waveguide has been created in this single-mode HBGW.Moreover,five single-mode HBGWs with various Bragg wavelengths were successfully fabricated by controlling the helical pitch,and this technique could be used for achieving a wavelength-division-multiplexed HBGW array.Then,the temperature and strain responses of the fabricated single-mode HBGW were tested,exhibiting a temperature sensitivity of 11.65 pm/℃ and a strain sensitivity of 1.29 pm/με,respectively.In addition,the thermal stability of the single-mode HBGW was also studied by annealing at a high temperature of 700℃ for 15 h.The degeneration of the single-mode waveguide into a multimode waveguide was observed during the isothermal annealing process,and the peak reflection and the Bragg wavelength of the fundamental mode exhibited a decrease of∼7 dB and a“blue”shift of 0.36 nm.Hence,such a femtosecond laser directly written single-mode HBGW could be used in many applications,such as sapphire fiber sensors,photonic integrated circuits,and monolithic waveguide lasers.JUN HE JIA HE XIZHEN XU BIN DU BAIJIE XU CHANGRUI LIAO ZHIYONG BAI YIPING WANG 2021Photonics Research2021,9,10:4
15Debrominated and methoxylated polybrominated diphenyl ether metabolites in rainbow trout(Oncorhynchus mykiss) after exposure to decabromodiphenyl ether显示文摘Decabromodiphenyl ether(BDE209) is the primary component in a commonly used flame retardant.Previous studies had proved that BDE209 itself was not toxic,while its metabolites including debrominated diphenyl ethers(De-BDEs) and methoxylated brominated diphenyl ethers(MeO-BDEs) posed a potential threat to organisms.Many studies had indicated that BDE209 could metabolize quickly in mammals,but lacking in the basic data about the metabolism of BDE209 in fish.In the present study,two replicate treatment groups of rainbow trout(Oncorhynchus mykiss) were exposed to BDE209 via a single intraperitoneal injection approximately 100 and 500 ng/g,respectively.Muscle,liver and blood samples were collected to analyze the specific metabolites on day 1 and day 28 post injection.The highest concentration of BDE209 was detected in muscle tissues,from 796.1 ng/g wet weight(day 1) to 687.1 ng/g wet weight(day 28) in high dose group,suggesting that BDE209 could accumulate slightly in muscle tissues.However,BDE209 was not detected in the blood for all treatments.Most congeners of De-BDEs were found in muscle and liver tissues,with the highest concentration in the liver.The main De-BDEs were nona-,octa-,hepta-and penta-De-BDEs.A total of seven MeO-BDE metabolites were observed among different fish tissues.Blood had the highest contribution of the MeO-BDE metabolites.Each MeO-BDE congener increased over the 28 days.These results in contrast to other studies suggested possible species-specific differences in metabolic abilities.Chenglian Feng,Yiping Xu,Yue He,Qian Luo,Jinmiao Zha,Zijian Wang State Key Laboratory of Environmental Aquatic Chemistry,Research Center for Eco-Environmental Sciences,Chinese Academy of Sciences,Beijing 100085,China. 2010Journal of Environmental Sciences2010,22,9:4
16Woodland strawberry WRKY71 acts as a promoter of flowering via a transcriptional regulatory cascade显示文摘The WRKY proteins are a large family of transcription factors that play important roles in stress responses and plant development.However,the roles of most WRKYs in strawberry are not well known.In this study,FvWRKY71 was isolated from the woodland strawberry‘Ruegen’.FvWRKY71 was highly expressed in the shoot apex and red fruit.Subcellular localization analysis showed that FvWRKY71 was located in the nucleus.Transactivation analysis showed that FvWRKY71 presented transcriptional activation activity in yeast.Overexpression of FvWRKY71 in Arabidopsis and woodland strawberry revealed early flowering in the transgenic plants compared with the wild-type control.Gene expression analysis indicated that the transcript levels of the flowering time and development integrator genes AP1,LFY,FT,AGL42,FUL,FPF1,SEP1,SEP2,and SEP3 were increased in FvWRKY71-overexpressing Arabidopsis and strawberry plants compared with the wild-type controls,which may result in accelerated flowering in transgenic plants.Furthermore,FvWRKY71 was proven to directly bind to the W-boxes(TTGACT/C)of the FvFUL,FvSEP1,FvAGL42,FvLFY,and FvFPF1 promoters in vitro and in vivo.Taken together,our results reveal a transcriptional regulatory cascade of FvWRKY71 involved in promoting flowering in woodland strawberry.Yingying Lei Yiping Sun BaotianWang Shuang Yu Hongyan Dai He Li Zhihong Zhang Junxiang Zhang 2020Horticulture Research2020,7,1:2

18Statistical Analysis of Regularity of Pesticide Residues in Vegetables Produced in Inner Mongoli显示文摘[Objective]The paper was to study regularity of pesticide residues in vegetables produced in Inner Mongolia.[Method]Mathematical statistic analysis was carried out on 6 800 samples of veggies,fruits,leguminous vegetables,nuts and seeds produced in Inner Mongolia.[Result]The dominated vegetables in Inner Mongolia were green leafy vegetables,solanaceous vegetables and melon vegetables,and their yields accounted for 70%of the total vegetables production.Since 2003,with the rapid increase of detected vegetable numbers,the status of vegetable quality safety had entered into a new stage of sustainable steady after a rapid decline or periodic fluctuation,and the differences in safety levels were obvious,of which the range of exceeding standard rate of pesticide residues in solanaceous vegetables,melon vegetables and leguminous vegetables(about 55%of the total vegetables)was under 2%,with average values of 1.1%,1.6%and 3.1%,respectively.They belonged to stable type.The exceeding standard rate of pesticide residues in green leafy vegetables and Chinese cabbage group(about 30%of total vegetables)presented a decreasing trend year by year,wondering in the range of 7%-10%.They belonged to main risk type.The time period of exceeding standard of pesticide residues in root vegetables and cole vegetables was under predictable,and its exceeding standard rate in some years was over 5%(amplitude variation is over 15percentage points).They belonged to random risk type.The kinds of pesticides,which exceeding standard rate in vegetables were relatively intensive,presented variance in different vegetable species.70%of the pesticides belonged to intermittent over-limits and the probability was below 5%.About 20%tradition pesticides often exceeded standard and their probabilities were over 30%.The exceeding standard of organophosphorus and carbamates pesticides in vegetables presented a decreasing tend,while the risk of some new pesticides containing nitrogen element and high effective pyrethroids showed an increasing tendency.[Conclusion]The paper provided decision making services for agricultural management departments.Fujin ZHANG Dekun HOU Jiang HE Tianyun GAO Hong LUO Songyan LANG Xinxin ZHANG Yiping YAO 2013Agricultural Science & Technology2013,14,10:2
19Shikonin alleviates collagen-induced arthritis mice by inhibiting M1 macrophage polarization显示文摘OBJECTIVE: To investigate the effects of shikonin(SKN) on M1 and M2 polarization of macrophages both in vitro and in vivo. METHODS: Collagen-induced arthritis(CIA) in male DBA/1 mice were treated with a dose of 4 mg/kg/day of SKN for 23 d(n = 6/group). The histopathology of inflamed joints in CIA mice was evaluated to test the antiarthritic effect of SKN. M1/M2 polarization of macrophages induced by lipopolysaccharide(LPS) and interferon(IFN)-γ or interleukin(IL)-4 and IL-13, were used to assess the effect of SKN(0.05, 0.1, and 0.2 μM). The effect of SKN on the protein expression of nitric oxide synthase, arginase, CD68, and CD206 was evaluated using western blot analysis. RESULTS: The results of this study revealed that SKN delayed the arthritis feet symptom score, reduced the incidence rate of arthritis, and relieved the inflammation of joints in CIA mice. SKN inhibited M1 macrophage polarization but did not affect M2 macrophage polarization in the joints of CIA mice. Moreover, SKN inhibited M1 polarization induced by LPS and IFN-γ, but did not affect M2 polarization induced by IL-4 and IL-13.
