1High efficiency production of ginsenoside compound K by catalyzing ginsenoside Rb1 using snailase显示文摘The rare ginsenoside Compound K(C-K) is attracting more attention because of its good physiological activity and urgent need. There are many pathways to obtain ginsenoside C-K, including chemical and biological methods. Among these, the conversion of PPD-type ginsenosides by enzymatic hydrolysis is a trend due to its high efficiency and mild conditions. For effectively extracting from the other panaxadiol saponins, the conversion process for ginsenoside C-K was investigated using snailases in this study. The univariate experimental design and response surface methodology were used to determine the optimal hydrolysis conditions for the conversion of ginsenoside Rb1 into ginsenoside C-K by snailases. The optimum conditions were as follows: p H 5.12, temperature 51 °C, ratio of snailase/substrate 0.21, and reaction time 48 h. On the basis of these parameters, the addition of 1.0 mmol·L^(-1) ferric ion was found to significantly improve the enzymolysis of snailases for the first time. With the above conditions, the maximum conversion rate reached 89.7%, suggesting that the process can obviously increase the yield of ginsenoside C-K. The bioassay tests indicated that the ginsenoside C-K showed anti-tumor activity in a series of tumor cell lines. 2Fine genetic mapping of the chromosome 11q23.3 region in a Han Chinese population:insights into the apolipoprotein genes underlying the blood lipid-lipoprotein variances显示文摘The unusual chromosome 11q23.3 harboring the apolipoprotein(APO)gene cluster has been well documented for its essential roles in plasma lipid-related traits and atherosclerotic cardiovascular diseases.However,its genetic architecture and the potential biological mechanisms underlying complex phenotypes have not been well assessed.We conducted a study for this target region in a Han Chinese population through a stepwise forward framework based on massive parallel sequencing,association analyses,genetic fine mapping,and functional interpretation.The present study identified new meaningful genetic associations that were not simply determined by statistical significance.In addition to the APOA5 gene,we found robust evidence of the genetic commitments of APOC3 and APOA1 to blood lipids.Several variants with high confidence were prioritized along with the potential biological mechanism interpretations in the wake of adaptive fine-mapping analyses.rs2849174 in the APOC3 enhancer was discovered with an unrivaled posterior probability of causality for triglyceride levels and could mediate APOC3 expression through enhancer activity modulated by a combination of histone modifications and transcription factor accessibility.Similarly,multiple lines of evidence converged in favor of rs3741297 as a causal variant influencing high-density lipoprotein cholesterol.Our findings provided novel insights into this genomic locus in the Chinese population.Weihua Shou Chenhui Zhang Jinxiu Shi Hong Wu Wei Huan 2020Journal of Genetics and Genomics2020,47,12:1
3Collision Attacks against AEZ-PRF for Authenticated Encryption AEZ显示文摘AEZ is an AES-based authenticated encryption submitted to the ongoing CAESAR competition and was presented at Eurocrypt2015 with AEZ v3. There are three models for AEZ, AEZ-core, AEZ-tiny and AEZ-prf. In this paper, we consider the security of AEZprf for AEZ v4.2, the latest version of AEZ.Our major finding is a collision of any 256-bit associated data for AES-prf. Then we launch collision attacks in a quantum setting and a classical setting respectively under different assumptions. In the quantum setting, by Simon's quantum algorithm, we amount a forgery with O(n) quantum superposition queries and an overwhelming probability close to 1.In the classical setting, one with the key of AEZ-prf can also construct the forgeries. Our results show that the AEZ-prf models of AEZ v4.2 is not secure in both the quantum setting and classical world. Furthermore, our results can also be applied to AEZ v3, which has been published on Eurocrypt 2015. 6A monocular visual SLAM system augmented by lightweight deep local feature extractor using in-house and low-cost LIDAR-camera integrated device显示文摘Simultaneous Localization and Mapping(SLAM)has been widely used in emergency response,self-driving and city-scale 3D mapping and navigation.Recent deep-learning based feature point extractors have demonstrated superior performance in dealing with the complex environmental challenges(e.g.extreme lighting)while the traditional extractors are struggling.In this paper,we have successfully improved the robustness and accuracy of a monocular visual SLAM system under various complex scenes by adding a deep learning based visual localization thread as an augmentation to the visual SLAM framework.In this thread,our feature extractor with an efficient lightweight deep neural network is used for absolute pose and scale estimation in real time using the highly accurate georeferenced prior map database at 20cm geometric accuracy created by our in-house and low-cost LiDAR and camera integrated device.The closed-loop error provided by our SLAM system with and without this enhancement is 1.03m and 18.28m respectively.The scale estimation of the monocular visual SLAM is also significantly improved(0.01 versus 0.98).In addition,a novel camera-LiDAR calibration workflow is also provided for large-scale 3D mapping.This paper demonstrates the application and research potential of deep-learning based vision SLAM with image and LiDAR sensors.Jing Li Chenhui Shi Jun Chen Ruisheng Wang Zhiyuan Yang Fan Zhang Jianhua Gong 2022International Journal of Digital Earth2022,15,1:0

11Bone morphogenetic protein-7 induced bone marrow stromal cells differentiate into neuron-like cells显示文摘Bone morphogenetic protein-7 is widely accepted as an inducer for bone marrow stem cells differ-entiating into osteoblasts and chondrocytes. Whether bone marrow stromal cells differentiate into neuron-like cells remains unclear. The current study examined the presence of positive cells for in-termediate filament protein and microtubule associated protein-2 in the cytoplasm of bone marrow stromal cells induced by bone morphogenetic protein-7 under an inverted microscope, while no expression of glial fibrillary acidic protein was found. Reverse transcription PCR electrophoresis also revealed a positive target band for intermediate filament protein and microtubule-associated protein 2 mRNA. 12A ɑ-KA fluorescent probe for discrimination of blood cancer serum显示文摘a-Ketoglutaric acid(a-KA) is an important metabolic intermediate in tricarboxylic acid circle in our body.The mutations of isocitrate dehydrogenase-1(IDH1) and isocitrate dehydrogenase-2(IDH2), however,will lead to the transformation of a-KA into 2-hydroxyglutarate(2-HG), which is confirmed to closely related to actue myeloid leukemia(AML). Therefore it is of great significance to detect a-KA level changes in serum. In this paper, a fluorescent 'off-on' probe CH for a-KA was designed based on naphthalimide fluorophore by introducing a hydrazine group for a-KA recognition and a long alkyl amino chain to enhance PET efficiency and water solubility. Cetyltrimethyl ammonium bromide(CTAB) was added toform self-assembly micelles for accelerating the recognition process. CH shows a 28-fold fluorescence enhancement((I - I_0)/I_0 at 550 nm) over other biological species by optimizing the chemical recognition process of CH with a-KA. Significantly, CH was successfully applied for thefluorescence discrimination of all kinds of blood cancer serum samples.
