1Surface integrity and fatigue behavior when turning γ-TiAl alloy with optimized PVD-coated carbide inserts显示文摘This paper presents a comprehensive investigation on the effects of tool and turning parameters on surface integrity and fatigue behavior in turning c-Ti Al alloy. The wear of inserts surface, cutting forces, and surface roughness were studied to optimize PVD-coated carbide inserts.Surface topography, residual stresses, microhardness, and microstructure were analyzed to characterize the surfaces layer under different turning parameters. Surface integrity and fatigue life tests of c-Ti Al alloy were conducted under turning and turning-polishing processes. The results show that compared to CNMG120412-MF4, CNMG120408-SM is more suitable because it obtained low cutting force, surface roughness, and tool wear. With increasing the cutting speed and depth, the depths of the compressive residual stress layer, hardening layer, and plastic deformation layer increased. For turning and turning-polishing specimens, the compressive residual stress was relaxed by less than 20%–30% after 10~7 cycles. 2An Ensemble Approach for Emotion Cause Detection with Event Extraction and Multi-Kernel SVMs显示文摘In this paper, we present a new challenging task for emotion analysis, namely emotion cause extraction.In this task, we focus on the detection of emotion cause a.k.a the reason or the stimulant of an emotion, rather than the regular emotion classification or emotion component extraction. Since there is no open dataset for this task available, we first designed and annotated an emotion cause dataset which follows the scheme of W3 C Emotion Markup Language. We then present an emotion cause detection method by using event extraction framework,where a tree structure-based representation method is used to represent the events. Since the distribution of events is imbalanced in the training data, we propose an under-sampling-based bagging algorithm to solve this problem. Even with a limited training set, the proposed approach may still extract sufficient features for analysis by a bagging of multi-kernel based SVMs method. 4Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. 9A Novel Human Antibody,HF,against HER2/erb-B2 Obtained by a Computer-Aided Antibody Design Method显示文摘Fully human antibodies have minimal immunogenicity and safety profiles.At present,most potential antibody drugs in clinical trials are humanized or fully human.Human antibodies are mostly generated using the phage display method(in vitro)or by transgenic mice(in vivo);other methods include B lymphocyte immortalization,human–human hybridoma,and single-cell polymerase chain reaction.Here,we describe a structure-based computer-aided de novo design technology for human antibody generation.Based on the complex structure of human epidermal growth factor receptor 2(HER2)/Herceptin,we first designed six short peptides targeting the potential epitope of HER2 recognized by Herceptin.Next,these peptides were set as complementarity determining regions in a suitable immunoglobulin frame,giving birth to a novel anti-HER2 antibody named 'HF,'which possessed higher affinity and more effective anti-tumor activity than Herceptin.Our work offers a useful tool for the quick design and selection of novel human antibodies for basic mechanical research as well as for imaging and clinical applications in immune-related diseases,such as cancer and infectious diseases.Chunxia Qiao Ming Lv Xinying Li Xiaoling Lang Shouqin Lv Mian Long Yan Li Shusheng Geng Zhou Lin Beifen Shen Jiannan Feng 2021Engineering2021,7,11:0

11Multi-omics Analyses Provide Insight into the Biosynthesis Pathways of Fucoxanthin in Isochrysis galbana显示文摘Isochrysis galbana is considered an ideal bait for functional foods and nutraceuticals of humans because of its high fucoxanthin(Fx)content.However,multi-omics analysis of the regulatory networks for Fx biosynthesis in I.galbana has not been reported.In this study,we report a high-quality genome assembly of I.galbana LG007,which has a genome size of 92.73 Mb,with a contig N50 of 6.99 Mb and 14,900 protein-coding genes.Phylogenetic analysis confirmed the monophyly of Haptophyta,with I.galbana sister to Emiliania huxleyi and Chrysochromulina tobinii.Evolutionary analysis revealed an estimated divergence time between I.galbana and E.huxleyi of~133 million years ago.Gene family analysis indicated that lipid metabolism-related genes exhibited significant expansion,including IgPLMT,IgOAR1,and IgDEGS1.Metabolome analysis showed that the content of carotenoids in I.galbana cultured under green light for 7 days was higher than that under white light,andβ-carotene was the main carotenoid,accounting for 79.09%of the total carotenoids.Comprehensive multi-omics analysis revealed that the content ofβ-carotene,antheraxanthin,zeaxanthin,and Fx was increased by green light induction,which was significantly correlated with the expression of IgMYB98,IgZDS,IgPDS,IgLHCX2,IgZEP,IgLCYb,and IgNSY.These findings contribute to the understanding of Fx biosynthesis and its regulation,providing a valuable reference for food and pharmaceutical applications.Duo Chen Xue Yuan Xuehai Zheng Jingping Fang Gang Lin Rongmao Li Jiannan Chen Wenjin He Zhen Huang Wenfang Fan Limin Liang Chentao Lin Jinmao Zhu Youqiang Chen Ting Xue 2022Genomics, Proteomics & Bioinformatics2022,20,6:0
