| 1 | Effects of freeze-thaw on soil erosion processes and sediment selectivity under simulated rainfall显示文摘The freeze-thaw(FT) processes affect an area of 46.3% in China. It is essential for soil and water conservation and ecological construction to elucidate the mechanisms of the FT processes and its associated soil erosion processes. In this research, we designed the control simulation experiments to promote the understanding of FT-water combined erosion processes. The results showed that the runoff of freeze-thaw slope(FTS) decreased by 8% compared to the control slope(CS), and the total sediment yield of the FTS was 1.10 times that of the CS. The sediment yield rate from the FTS was significantly greater than that from the CS after 9 min of runoff(P<0.01). Both in FTS and CS treatments, the relationships between cumulative runoff and sediment yield can be fitted well with power functions(R2>0.98, P<0.01). Significant differences in the mean weight diameter(MWD) values of particles were observed for washed particles and splashed particles between the CS and the FTS treatments in the erosion process(P<0.05). The mean MWD values under CS were smaller than those under FTS for both washed and splashed particles. The ratio of the absolute value of a regression coefficient between the CS and the FTS was 1.15, being roughly correspondent with the ratio of K between the two treatments. Therefore, the parameter a of the power function between cumulative runoff and sediment yield could be an acceptable indicator for expressing the soil erodibility. | 2 | Lx2-32c,a novel semi-synthetic taxane,exerts antitumor activity against prostate cancer cells in vitro and in vivo显示文摘Tubulin has been shown to be an effective target for the development of cytotoxic agents against prostate cancer. Previously, we reported that Lx2-32 c is an anti-tubulin agent with high binding affinity to tubulin. In this study, we investigated the potential of Lx2-32 c to act as an effective cytotoxic agent in the treatment of prostate cancer. MTT assays showed that Lx2-32 c was cytotoxic to all tested prostate cancer cell lines. The Lx2-32c-treated cells typically exhibited a rounded morphology associated with the onset of apoptosis, as evidenced by immunocytochemical staining. Human prostate cancer cell lines treated with Lx2-32 c arrest in the G2/M phase of the cell cycle and the treatment is associated with an increased ratio of cells in the sub-G0/G1 phase as determined by flow cytometry. Furthermore, expression of the cleaved form of poly(ADP-ribose) polymerase in prostate cancer cell lines treated with Lx2-32 c was shown by Western blotting assay. Xenograft implants of LNCa P and PC3-derived tumors in nude mice showed that Lx2-32 c treatment significant inhibited tumor growth with effects equivalent to those of docetaxel. | 3 | Microstructure,tensile properties and thermal stability of AlMgSiScZr alloy printed by laser powder bed fusion显示文摘The new generation of Sc and Zr modified Al alloys has been attracted wide concerns in aerospace industry,owing to the excellent mechanical performances and superior thermal stability than other normal Al alloys.By microalloying with Sc and Zr,the Al3(Sc,Zr)particle forms as the grain refiner during the solidification,which is extremely beneficial for the laser powder bed fusion(LPBF)processed Al alloys.In this study,a new type Al-14.1 Mg-0.47 Si-0.31 Sc-0.17 Zr alloy was additively manufactured by LPBF,and the microstructure,tensile properties and thermal stability were studied in detail.By using a single melt-67°scanning strategy,the LPBF-processed specimen with a relative density of 99.4%and tensile strength of 487.7 MPa was obtained at 160 W-200 mm/s.And this AlMgSiScZr alloy can still exhibit an excellent tensile strength of 393.9 MPa at a moderate temperature of 473 K.After the aging treatment,the tensile properties further increased due to the precipitate hardening of Mg2Si and Al3(Sc,Zr),and the maximum value(580 MPa)was reached at an aging time of 10 h.The average crystal size was almost unchanged after aging treated at 325℃and 24 h,indicating this AlMgSiScZr alloy has an improved thermal stability.The AlMgSiScZr alloy is recommended to substitute some particular titanium alloys in aerospace field afterwards. | Jiang Bi Zhenglong Lei Yanbin Chen Xi Chen Ze Tian Nannan Lu Xikun Qin Jingwei Liang | 2021 | Journal of Materials Science & Technology2021,,10: | 4 |
| 4 | Essential sequence of the N-terminal cytoplasmic localization-related domain of huntingtin and its effect on huntingtin aggregates显示文摘Huntington's disease(HD) is caused by abnormal CAG repeat expansion in the 5′-end of the Huntingtin(HTT) gene.In addition to the canonical C-terminal full-length huntingtin(htt) nuclear export signal,a cytoplasmic localization-related domain(CLRD) in the N-terminus of htt has recently been reported.Here,we analyzed this domain by introducing deletion and substitution mutations in a truncated N-terminal htt protein and subsequently monitored htt expression,aggregation and subcellular localization by immunocytochemistry and Western blot analysis.We demonstrated that Htt4-17 was the essential sequence for htt cytoplasmic localization.We also found that the subcellular distribution of htt was altered when Htt1-17 was mutated to contain amino acids of different charges,suggesting a structural requirement of Htt1-17 for the cytoplasmic localization of htt. | 5 | Risk stratification in symptomatic intracranial atherosclerotic disease with conventional vascular risk factors and cerebral haemodynamics显示文摘Background and purpose Symptomatic intracranial atherosclerotic stenosis(sICAS)is associated with a considerable risk of recurrent stroke despite contemporarily optimal medical treatment.Severity of luminal stenosis in sICAS and its haemodynamic significance quantified with computational fluid dynamics(CFD)models were associated with the risk of stroke recurrence.We aimed to develop and compare stroke risk prediction nomograms in sICAS,based on vascular risk factors and these metrics.Methods Patients with 50%-99%sICAS confirmed in CT angiography(CTA)were enrolled.Conventional vascular risk factors were collected.Severity of luminal stenosis in sICAS was dichotomised as moderate(50%-69%)and severe(70%-99%).Translesional pressure ratio(PR)and wall shear stress ratio(WSSR)were quantified via CTA-based CFD modelling;the haemodynamic status of sICAS was classified as normal(normal PR&WSSR),intermediate(otherwise)and abnormal(abnormal PR&WSSR).All patients received guideline-recommended medical treatment.We developed and compared performance of nomograms composed of these variables and independent predictors identified in multivariate logistic regression,in predicting the primary outcome,recurrent ischaemic stroke in the same territory(SIT)within 1 year.Results Among 245 sICAS patients,20(8.2%)had SIT.The D2H2A nomogram,incorporating diabetes,dyslipidaemia,haemodynamic status of sICAS,hypertension and age≥50 years,showed good calibration(P for Hosmer-Lemeshow test=0.560)and discrimination(C-statistic 0.73,95%CI 0.60 to 0.85).It also had better performance in risk reclassification and provided larger net benefits in decision curve analysis,compared with nomograms composed of conventional vascular risk factors only,and plus the severity of luminal stenosis in sICAS.Sensitivity analysis in patients with anterior-circulation sICAS showed similar results.Conclusions The D2H2A nomogram,incorporating conventional vascular risk factors and the haemodynamic significance of sICAS as assessed in CFD models,could be a useful tool to stratify sICAS patients for the risk of recurrent stroke under contemporarily optimal medical treatment. | Xuan Tian Hui Fang Linfang Lan Hing Lung Ip Jill Abrigo Haipeng Liu Lina Zheng Florence S Y Fan Sze Ho Ma Bonaventure Ip Bo Song Yuming Xu Jingwei Li Bing Zhang Yun Xu Yannie O Y Soo Vincent Mok Ka Sing Wong Thomas W Leung Xinyi Leng | 2023 | Stroke & Vascular Neurology2023,8,1: | 2 |

| 18 | Morphine Re-arranges Chromatin Spatial Architecture of Primate Cortical Neurons显示文摘The expression of linear DNA sequence is precisely regulated by the three-dimensional(3D)architecture of chromatin.Morphine-induced aberrant gene networks of neurons have been extensively investigated;however,how morphine impacts the 3D genomic architecture of neurons is still unknown.Here,we applied digestion-ligation-only high-throughput chromosome conformation capture(DLO Hi-C)technology to investigate the effects of morphine on the 3D chromatin architecture of primate cortical neurons.After receiving continuous morphine administration for 90 days on rhesus monkeys,we discovered that morphine re-arranged chromosome territories,with a total of 391 segmented compartments being switched.Morphine altered over half of the detected topologically associated domains(TADs),most of which exhibited a variety of shifts,followed by separating and fusing types.Analysis of the looping events at kilobase-scale resolution revealed that morphine increased not only the number but also the length of differential loops.Moreover,all identified differentially expressed genes from the RNA sequencing data were mapped to the specific TAD boundaries or differential loops,and were further validated for changed expression.Collectively,an altered 3D genomic architecture of cortical neurons may regulate the gene networks associated with morphine effects.Our finding provides critical hubs connecting chromosome spatial organization and gene networks associated with the morphine effects in humans. | Liang Wang Xiaojie Wang Chunqi Liu Wei Xu Weihong Kuang Qian Bu Hongchun Li Ying Zhao Linhong Jiang Yaxing Chen Feng Qin Shu Li Qinfan Wei Xiaocong Liu Bin Liu Yuanyuan Chen Yanping Dai Hongbo Wang Jingwei Tian Gang Cao Yinglan Zhao Xiaobo Cen | 2023 | Genomics, Proteomics & Bioinformatics2023,21,3: | 0 |
