3Gas Film Disturbance Characteristics Analysis of High-Speed and High-Pressure Dry Gas Seal显示文摘The dry gas seal(DGS) has been widely used in high parameters centrifugal compressor, but the intense vibrations of shafting,especially in high-speed condition, usually result in DGS's failure. So the DGS's ability of resisting outside interference has become a determining factor of the further development of centrifugal compressor. However, the systematic researches of which about gas film disturbance characteristics of high parameters DGS are very little. In order to study gas film disturbance characteristics of high-speed and high-pressure spiral groove dry gas seal(S-DGS) with a flexibly mounted stator, rotor axial runout and misalignment are taken into consideration, and the finite difference method and analytical method are used to analyze the influence of gas film thickness disturbance on sealing performance parameters, what's more, the effects of many key factors on gas film thickness disturbance are systematically investigated. The results show that, when sealed pressure is 10.1MPa and seal face average linear velocity is 107.3 m/s, gas film thickness disturbance has a significant effect on leakage rate, but has relatively litter effect on open force; Excessively large excitation amplitude or excessively high excitation frequency can lead to severe gas film thickness disturbance; And it is beneficial to assure a smaller gas film thickness disturbance when the stator material density is between 3.1 g/cm^3 to 8.4 g/cm^3; Ensuring sealing performance while minimizing support axial stiffness and support axial damping can help to improve dynamic tracking property of dry gas seal. 5Fast-Suppressor Screening for New Components in Protein Trafficking,Organelle Biogenesis and Silencing Pathway in Arabidopsis thaliana Using DEX-Inducible FREE1-RNAi Plants显示文摘Membrane trafficking is essential for plant growth and responses to external signals.The plant unique FYVE domain-containing protein FREE1 is a component of the ESCRT complex(endosomal sorting complex required for transport).FREE1 plays multiple roles in regulating protein trafficking and organelle biogenesis including the formation of intraluminal vesicles of multivesicular body(MVB),vacuolar protein transport and vacuole biogenesis,and autophagic degradation.FREE1 knockout plants show defective MVB formation,abnormal vacuolar transport,fragmented vacuoles,accumulated autophagosomes,and seedling lethality.To further uncover the underlying mechanisms of FREE1 function in plants,we performed a forward genetic screen for mutants that suppressed the seedling lethal phenotype of FREE1-RNAi transgenic plants.The obtained mutants are termed as suppressors of free1(sof).To date,229 putative sof mutants have been identified.Barely detecting of FREE1 protein with M3plants further identified 84 FREE1-related suppressors.Also145 mutants showing no reduction of FREE1 protein were termed as RNAi-related mutants.Through next-generation sequencing(NGS)of bulked DNA from F2mapping population of two RNAi-related sof mutants,FREE1-RNAi T-DNA inserted on chromosome 1 was identified and the causal mutation of putative sof mutant is being identified similarly.These FREE1-and RNAi-related sof mutants will be useful tools and resources for illustrating the underlying mechanisms of FREE1 function in intracellular trafficking and organelle biogenesis,as well as for uncovering the new components involved in the regulation of silencing pathways in plants.Qiong Zhao Caiji Gao PoShing Lee Lin Liu Shaofang Li Tangjin Hu Jinbo Shen Shuying Pan Hao Ye Yunru Chen Wenhan Cao Yong Cui Peng Zeng Sheng Yu Yangbin Gao Liang Chen Beixin Mo Xin Liu Shi Xiao Yunde Zhao Silin Zhong Xuemei Chen Liwen Jiang 2015Journal of Genetics and Genomics2015,42,6:4

17Ameliorative Effect of Ginsenoside RT-5 on CDDP-Induced Nephrotoxicity显示文摘The aim of this study is to explore whether RT-5, one novel active ginsenosides from Ginseng, has protective effects against cisplatin(CDDP)-induced nephrotoxicity in vivo. One model of acute renal failure induced by CDDP in rats and one model of xenograft tumors of human cervical cancer in nude mice are established. Four groups are assigned: control, CDDP, RT-5, CDDP plus RT-5, in which the RT-5 is administered via oral gavage 24 h before CDDP intraperitoneal injection. The significant increase in blood urea nitrogen and creatinine are induced by CDDP treatment at 6 mg/kg, which are attenuated by pre-treated with RT-5 at 10 mg/kg. RT-5 could ameliorate CDDP-induced morphological damages in kidney by PAS staining, and reduce tubular apoptosis evaluated by TUNEL staining. Pretreatment with RT-5 notably inhibits CDDP-induced oxidative stress in kidney tissues. Interestingly, RT-5 does not interfere with the in vivo anti-cancer effects of CDDP against the growth of xenograft tumors in nude mice.
