4Simultaneous Determination of Plant Growth Regulators in Fruits Using a Modified Qu ECh ERS Procedure and UPLC–MS/MS显示文摘Six plant growth regulators frequently used in fruit were detected using UPLC–MS/MS including chlormequat chloride, GA3, forchlorfenuron,paclobutrazol, thidiazuron and 2,4-D. A modified Qu ECh ERS method was used in pretreatment: each compound was extracted with acetonitrile containing 1% acetic acid and cleaned with a mixture of ostade-cylsilane(ODS) and Mg SO_4. The detection was performed using electrospray ionization(ESI) and multiple reaction monitoring mode(MRM) and quantified using the external standard method. Results indicated that the calibration curves of six plant growth regulators were linear within the range of 1–100 μg·kg^(-1) with correlation coefficients over 0.999. The recovery at four spiked concentrations, 1, 20, 50, and 100 μg·kg^(-1), ranged from 60.77% to 119.43% and relative standard deviation(RSD) were between 0.18% and 32.94%. The limits of detection(LOD) and limits of quantitation(LOQ) were 0.01–0.14 μg·kg^(-1) and 0.02–0.46 μg·kg^(-1), respectively, for apples, 0.01–0.18 μg·kg^(-1) and 0.03–0.59 μg·kg^(-1) for grapes, 0.01–0.08 μg·kg^(-1) and 0.04–0.28 μg·kg^(-1) for kiwis, 0.003–0.110 μg·kg^(-1) and 0.01–0.38 μg·kg^(-1) for peaches, and 0.01–0.08 μg·kg^(-1) and 0.03–0.25 μg·kg^(-1) for oranges.YAN Zhen NIE Jiyun XU Guofeng LI Haifei LI Jing LI Zhixia WU Yonglong KUANG Lixue 2016Horticultural Plant Journal2016,2,4:8
5CNN Based No-Reference HDR Image Quality Assessment显示文摘Motivated by the problems of non-universality and over-reliance on the original reference image in High dynamic range(HDR)Image quality assessment(IQA),a convolutional neural network-based algorithm for no-reference HDR image quality assessment is proposed.The Salience detection by self-resemblance(SDSR)algorithm which extracts the salient regions of the HDR image,is used to simulate the human visual attention mechanism.Then a visual quality perception network for training quality prediction models is designed according to the visual characteristics of luminance and contrast sensitivity.And this network consists of an Error estimation network(Error-net),a Perceptual resistance network(PR-net)and a mixing function.The experimental results indicate that the method proposed has high consistency with subjective perception,and the value of assessment metrics Spearman rank-order correlation coefficient(SROCC),Pearson product-moment correlation coefficient(PLCC)and Root mean square error(RMSE)correspondingly reaches 0.941,0.910 and 8.176 as well.It is comparable with classic full-reference HDR IQA methods.FAN Kefeng LIANG Jiyun LI Fei QIU Puye 2021Chinese Journal of Electronics2021,30,2:4
6Parallel ADR Detection Based on Spark and BCPNN显示文摘Adverse Drug Reaction(ADR) is one of the major challenges to the evaluation of drug safety in the medical field. The Bayesian Confidence Propagation Neural Network(BCPNN) algorithm is the main algorithm used by the World Health Organization to monitor ADRs. Currently, ADR reports are collected through the spontaneous reporting system. However, with the continuous increase in ADR reports and possible use scenarios, the efficiency of the stand-alone ADR detection algorithm will encounter considerable challenges. Meanwhile, the BCPNN algorithm requires a certain number of disk I/O, which leads to considerable time consumption. In this study,we propose a Spark-based parallel BCPNN algorithm, which speeds up data processing and reduces the number of disk I/O in BCPNN, and two optimization strategies. Then, the ADR data collected from the FDA Adverse Event Reporting System are used to verify the performance of the proposed algorithm and its optimization strategies.Experiments show that the parallel BCPNN can significantly accelerate data processing and the optimized algorithm has a high acceleration rate and can effectively prevent memory overflow. Finally, we apply the proposed algorithm to a dataset provided by a real medical consortium. 7Global analysis of lysine succinylation in patchouli plant leaves显示文摘Lysine succinylation is a novel,naturally occurring posttranslational modification(PTM)in living organisms.Global lysine succinylation identification has been performed at the proteomic level in various species;however,the study of lysine succinylation in plant species is relatively limited.Patchouli plant(P.cablin(Blanco)Benth.,Lamiaceae)is a globally important industrial plant and medicinal herb.In the present study,lysine succinylome analysis was carried out in patchouli plants to determine the potential regulatory role of lysine succinylation in patchouli growth,development,and physiology.The global succinylation sites and proteins in patchouli plants were screened with an immunoprecipitation affinity enrichment technique and advanced mass spectrometry-based proteomics.Several bioinformatic analyses,such as function classification and enrichment,subcellular location predication,metabolic pathway enrichment and protein−protein interaction networking,were conducted to characterize the functions of the identified sites and proteins.In total,1097 succinylation sites in 493 proteins were detected in patchouli plants,among which 466 succinylation sites in 241 proteins were repeatedly identified within three independent experiments.The functional characterization of these proteins indicated that the tricarboxylic acid(TCA)cycle,oxidative phosphorylation,photosynthesis processes,and amino acid biosynthesis may be regulated by lysine succinylation.In addition,these succinylated proteins showed a wide subcellular location distribution,although the chloroplast and cytoplasm were the top two preferred cellular components.Our study suggested the important role of lysine succinylation in patchouli plant physiology and biology and could serve as a useful reference for succinylation studies in other medicinal plants.Xiaobing Wang Xiuzhen Chen Junren Li Xuanxuan Zhou Yanting Liu Liting Zhong Yun Tang Hai Zheng Jiyun Liu Ruoting Zhan Likai Chen 2019Horticulture Research2019,6,1:2
8Characterization of NDM-1-producing carbapenemase in Acinetobacter spp.and E.coli isolates from diseased pigs显示文摘In recent years,the mobile metallo-β-lactamase(MBL)genes have been found to correspond to one of the most important resistance characters identified in Gramnegative bacteria,severely affecting clinical chemotherapy and threatening public health.The prevalence of mobile MBL genes and their flanking regions in Gram-negative bacteria from diseased pigs in China was investigated.A total of 334 lung samples from diseased pigs were screened for Gram-negative bacteria classified as non-susceptible to meropenem(MIC≥4mg·L^(–1)).Six isolates,including three Escherichia coli,two Acinetobacter baumanii and one A.calcoaeticus,exhibitedMBL production and carried the blaNDM-1 gene.S1-PFGE and Southern blot analysis showed that the blaNDM-1 gene was located on the chromosome of one A.baumanii isolate and on plasmids of various sizes in the other five isolates.MIC testing using broth microdilution revealed that all blaNDM-1-carrying isolates and some of their transconjugants exhibited resistance to almost allβ-lactams tested.Whole genome sequencing revealed that the flanking region of the blaNDM-1 gene from all porcine isolates had high levels of similarity with the corresponding regions in human isolates.One porcine E.coli isolate carrying blaNDM-1 was typed as ST48,a common sequence type in human E.coli isolates.These results suggest the possibility of human-tofood animal transfer of blaNDM-1-producing E.coli,highlighting the need for surveillance of carbapenemase producers among bacteria from food animals.In addition,the prudent use of antimicrobial agents to decrease the opportunities for co-selection of carbapenemase genes in food animals is also urgently needed.Rongmin ZHANG Yang WANG Zhihai LIU Jiyun LI Wenjuan YIN Lei LEI Congming WU Jianzhong SHEN 2015Frontiers of Agricultural Science and Engineering2015,2,3:2
9Molecular Epidemiology of Klebsiella pneumoniae from Clinical Bovine Mastitis in Northern Area of China,2018–2019显示文摘Klebsiella pneumoniae(K.pneumonia,KpⅠ)is a predominate inducement of bovine mastitis,which is associated with high mortality and milk yield reduction.However,data is lacking on the molecular characteristics of bovine K.pneumoniae,limiting the risk assessment of its transmission through the food chain.Herein,we investigated the prevalence of K.pneumoniae in 6301 clinical mastitis(CM)milk samples from dairy cattle in northern area of China.In total,183 K.pneumoniae isolates were recovered,with detection rates of 3.0% and 2.8% in 2018 and 2019,respectively.Like human clinical K.pneumoniae,all CM K.pneumoniae isolates belonged to one of three phylogroups:KpⅠ(n=143),Klebsiella.quasipneumoniae subsp.similipneumoniae(KpⅡ-B)(n=37),and Klebsiella variicola(KpⅢ)(n=3).We detected the extendedspectrum β-lactamase-encoding genes bla_(SHV-2a),blac_(CTX-M-14),and bla_(CTX-M-15),as well as clpC,lpfA,lacI,lacZ,lacY,and the fecABDEIR operon in the KpⅠ isolates,which may contribute to their pathogenicity and host adaptability in cows.The high prevalence of KpⅠ in dairy farms may be problematic,as it showed relatively higher rates of antibiotic resistance and virulence gene carriage than the KpⅡ-B and KpⅢ isolates.Furthermore,we observed distinct differences in population structure between CM-and human infection-associated KpⅠ isolates,with the genes associated with invasive infection in humans rarely being observed in bovine isolates,indicating that few CM-associated K.pneumoniae isolates pose a threat to human health.Nevertheless,bovine KpⅡ-B isolates shared a high level of nucleotide sequence identity with isolates from human infections and frequently carried the nitrogen-fixation gene nif,suggesting an association between KpⅡ-B isolates from cattle and humans,and plant-derived bacteria.Shikai Song Wenjuan He Dawei Yang Manar Benmouffok Yao Wang Jiyun Li Chengtao Sun Xiangbin Song Shizhen Ma Chang Cai Shuangyang Ding Congming Wu Zhangqi Shen Yang Wang 2022Engineering2022,8,3:1
