1Identification of Nonstructural Protein 8 as the N-Terminus of the RNA-Dependent RNA Polymerase of Porcine Reproductive and Respiratory Syndrome Virus显示文摘Porcine reproductive and respiratory syndrome virus (PRRSV) is a member within the family Arteriviridae of the order Nidovirales. Replication of this positive-stranded RNA virus within the host cell involves expression of viral replicase proteins encoded by two ORFs, namely ORF1 a and ORF1 b. In particular, translation of ORF1 b depends on a-1-ribosomal frameshift strategy. Thus, nonstructural protein 9 (nsp9), the first protein within ORF1 b that specifies the function of the viral RNA-dependent RNA polymerase, is expressed as the C-terminal extension of nsp8, a small nsp that is encoded by ORF1 a. However, it has remained unclear whether the mature form of nsp9 in virus-infected cells still retains nsp8,addressing which is clearly critical to understand the biological function of nsp9. By taking advantage of specific antibodies to both nsp8 and nsp9, we report the following findings. (1) In infected cells, PRRSV nsp9 was identified as a major product with a size between 72 and 95 k Da (72–95 KDa form), which exhibited the similar mobility on the gel to the in vitro expressed nsp8–9 ORF1 b, but not the ORF1 b-coded portion (nsp9 ORF1 b). (2) The antibodies to nsp8, but not to nsp7 or nsp10, could detect a major product that had the similar mobility to the 72–95 KDa form of nsp9. Moreover, nsp9 could be co-immunoprecipitated by antibodies to nsp8, and vice versa. (3) Neither nsp4 nor nsp2 PLP2 was able to cleave nsp8–nsp9 in vitro. 2Tunnel vision optimization method for VR flood scenes based on Gaussian blur显示文摘The visualization of flood disasters in virtual reality(VR)scenes is useful for the representation and sharing of disaster knowledge and can effectively improve users’cognitive efficiency in comprehending disaster information.However,the existing VR methods of visualizing flood disaster scenes have some shortcomings,such as low rendering efficiency and poor user experience.In this paper,a tunnel vision optimization method for VR flood scenes based on Gaussian blur is proposed.The key techniques are studied,such as region of interest(ROI)calculation and tunnel vision optimization considering the characteristics of the human visual system.A prototype system has been developed and used to carry out an experimental case analysis.The experimental results show that the number of triangles drawn in a flood VR scene is reduced by approximately 30%–40%using this method and that the average frame rate is stable at approximately 90 frames per second(fps),significantly improving the efficiency of scene rendering and reducing motion sickness.Lin Fu Jun Zhu Weilian Li Qing Zhu Bingli Xu Yakun Xie Yunhao Zhang Ya Hu Jingtao Lu Pei Dang Jigang You 2021International Journal of Digital Earth2021,14,7:1
3Interoceptive regulation of skeletal tissue homeostasis and repair显示文摘Recent studies have determined that the nervous system can sense and respond to signals from skeletal tissue,a process known as skeletal interoception,which is crucial for maintaining bone homeostasis.The hypothalamus,located in the central nervous system(CNS),plays a key role in processing interoceptive signals and regulating bone homeostasis through the autonomic nervous system,neuropeptide release,and neuroendocrine mechanisms.These mechanisms control the differentiation of mesenchymal stem cells into osteoblasts(OBs),the activation of osteoclasts(OCs),and the functional activities of bone cells.Sensory nerves extensively innervate skeletal tissues,facilitating the transmission of interoceptive signals to the CNS.This review provides a comprehensive overview of current research on the generation and coordination of skeletal interoceptive signals by the CNS to maintain bone homeostasis and their potential role in pathological conditions.The findings expand our understanding of intersystem communication in bone biology and may have implications for developing novel therapeutic strategies for bone diseases.Yao Xiao Changhao Han Yunhao Wang Xinshu Zhang Rong Bao Yuange Li Huajiang Chen Bo Hu Shen Liu 2023Bone Research2023,11,3:1
4Theta Oscillations Support Prefrontal-hippocampal Interactions in Sequential Working Memory显示文摘The prefrontal cortex and hippocampus may support sequential working memory beyond episodic memory and spatial navigation.This stereoelectroencephalography(SEEG)study investigated how the dorsolateral prefrontal cortex(DLPFC)interacts with the hippocampus in the online processing of sequential information.Twenty patients with epilepsy(eight women,age 27.6±8.2 years)completed a line ordering task with SEEG recordings over the DLPFC and the hippocampus.Participants showed longer thinking times and more recall errors when asked to arrange random lines clockwise(random trials)than to maintain ordered lines(ordered trials)before recalling the orientation of a particular line.First,the ordering-related increase in thinking time and recall error was associated with a transient theta power increase in the hippocampus and a sustained theta power increase in the DLPFC(3–10 Hz).In particular,the hippocampal theta power increase correlated with the memory precision of line orientation.Second,theta phase coherences between the DLPFC and hippocampus were enhanced for ordering,especially for more precisely memorized lines.Third,the theta band DLPFC→hippocampus influence was selectively enhanced for ordering,especially for more precisely memorized lines.This study suggests that theta oscillations may support DLPFC-hippocampal interactions in the online processing of sequential information.Minghong Su Kejia Hu Wei Liu Yunhao Wu Tao Wang Chunyan Cao Bomin Sun Shikun Zhan Zheng Ye 2024Neuroscience Bulletin2024,40,2:0
5A New Method for Converting T_(2) Spectrum into Pore Radius显示文摘In this paper,a new method for converting the T_(2)(relaxation time) of NMR(nuclear magnetic resonance) into the pore radius is proposed.Combined with NMR and centrifugation experiments,the relationship between pore radius and T_(2) of the sample was established.The results show that the new method is more reasonable than the traditional method.When the sample was denser and the mercury saturation was lower,the pore distribution curve was obtained by traditional method had a worse agreement with mercury injection experiment,while pore distribution curve of the new method had a better agreement with the mercury injection curve,which reflected the greater advantage of the new method as the reservoir becomes denser.The new method can obtain all the pore information in the sample.The results show that the pores in tight sandstone are mainly consisted with mesopore and macropore,and the connectivity of macropore is better than that of mesopore.The new method can effectively characterize the full pore distribution and the seepage characteristics in different pores interval of tight reservoirs,which had a great significance to evaluate the recoverable resources of tight reservoir.Fan Zhang Zhenxue Jiang Yuanhao Zhang Bin Hu Zaiquan Yang Yuhua Yang Xianglu Tang Hanmin Xiao Lin Zhu Yunhao Han 2023Journal of Earth Science2023,34,4:0
