1Effect of nanoheat stimulation mediated by magnetic nanocomposite hydrogel on the osteogenic differentiation of mesenchymal stem cells显示文摘Hyperthermia has been considered as a promising healing treatment in bone regeneration. We designed a tissue engineering hydrogel based on magnetic nanoparticles to explore the characteristics of hyperthermia for osteogenic regeneration. This nanocomposite hydrogel was successfully fabricated by incorporating magnetic Fe_3O_4 nanoparticles into chitosan/polyethylene glycol(PEG) hydrogel, which showed excellent biocompatibility and were able to easily achieve increasing temperatures under an alternative magnetic field(AMF). With uniformly dispersed nanoparticles, the composite hydrogel resulted in high viability of mesenchymal stem cells(MSCs), and the elevated temperature contributed to the highest osteogenic differentiation ability compared with direct heat treatment applied under equal temperatures. 4Aligned fibrin/functionalized self-assembling peptide interpenetrating nanofiber hydrogel presenting multi-cues promotes peripheral nerve functional recovery显示文摘Nerve guidance conduits with hollow lumen fail to regenerate critical-sized peripheral nerve defects(15 mm in rats and 25 mm in humans),which can be improved by a beneficial intraluminal microenvironment.However,individual cues provided by intraluminal filling materials are inadequate to eliminate the functional gap between regenerated nerves and normal nerves.Herein,an aligned fibrin/functionalized self-assembling peptide(AFG/fSAP)interpenetrating nanofiber hydrogel that exerting synergistic topographical and biochemical cues for peripheral nerve regeneration is constructed via electrospinning and molecular self-assembly.The hydrogel possesses an aligned structure,high water content,appropriate mechanical properties and suitable biodegradation capabilities for nerve repair,which enhances the alignment and neurotrophin secretion of primary Schwann cells(SCs)in vitro,and successfully bridges a 15-mm sciatic nerve gap in rats in vivo.The rats transplanted with the AFG/fSAP hydrogel exhibit satisfactory morphological and functional recovery in myelinated nerve fibers and innervated muscles.The motor function recovery facilitated by the AFG/fSAP hydrogel is comparable with that of autografts.Moreover,the AFG/fSAP hydrogel upregulates the regeneration-associated gene expression and activates the PI3K/Akt and MAPK signaling pathways in the regenerated nerve.Altogether,the AFG/fSAP hydrogel represents a promising approach for peripheral nerve repair through an integration of structural guidance and biochemical stimulation.Shuhui Yang Jinjin Zhu Changfeng Lu Yi Chai Zheng Cao Jiaju Lu Zhe Zhang He Zhao Yin-Yuan Huang Shenglian Yao Xiangdong Kong Peixun Zhang Xiumei Wang 2022Bioactive Materials2022,7,2:3
5Dynamic model of computer viruses under the effect of removable media and external computers显示文摘This paper investigates the propagation of computer viruses and establishes a novel propagation model. In contrast to the existing models,this model can directly indicate the impact of removable media and external computers on the propagation of computer virus. The stability results of equilibrium point are derived by Hurwitz criterion and Bendixson Dulac criterion. The effectiveness of the proposed results is shown by numerical simulation. In order to show the superiority of the proposed model,some comparisons with the existing models are presented. 8White matter regeneration induced by aligned fibrin nanofiber hydrogel contributes to motor functional recovery in canine T12 spinal cord injury显示文摘A hierarchically aligned fibrin hydrogel(AFG)that possesses soft stiffness and aligned nanofiber structure has been successfully proven to facilitate neuroregeneration in vitro and in vivo.However,its potential in promoting nerve regeneration in large animal models that is critical for clinical translation has not been sufficiently specified.Here,the effects of AFG on directing neuroregeneration in canine hemisected T12 spinal cord injuries were explored.Histologically obvious white matter regeneration consisting of a large area of consecutive,compact and aligned nerve fibers is induced by AFG,leading to a significant motor functional restoration.The canines with AFG implantation start to stand well with their defective legs from 3 to 4 weeks postoperatively and even effortlessly climb the steps from 7 to 8 weeks.Moreover,high-resolution multi-shot diffusion tensor imaging illustrates the spatiotemporal dynamics of nerve regeneration rapidly crossing the lesion within 4 weeks in the AFG group.Our findings indicate that AFG could be a potential therapeutic vehicle for spinal cord injury by inducing rapid white matter regeneration and restoring locomotion,pointing out its promising prospect in clinic practice.Zheng Cao Weitao Man Yuhui Xiong Yi Guo Shuhui Yang Dongkang Liu He Zhao Yongdong Yang Shenglian Yao Chuzhong Li Lingyun Zhao Xiaodan Sun Hua Guo Guihuai Wang Xiumei Wang 2022Regenerative Biomaterials2022,9,1:0
9Stem cell properties and neural differentiation of sheep amniotic epithelial cells显示文摘This study was designed to verify the stem cell properties of sheep amniotic epithelial cells and their capacity for neural differentiation. Immunofluorescence microscopy and reverse transcription-PCR revealed that the sheep amniotic epithelial cells were positive for the embryonic stem cell marker proteins SSEA-1, SSEA-3, SSEA-4, TRA-1-60 and TRA-1-81, and the totipotency-associated genes Oct-4, Sox-2 and Rex-1, but negative for Nanog. Amniotic epithelial cells expressed β-III-tubulin, glial fibrillary acidic protein, nestin and microtubule-associated protein-2 at 28 days after induction with serum-free neurobasal-A medium containing B-27. 10The effect of laser acupuncture on hypoxia tolerance and inflammation reaction in mice with optical fiber acupuncture needle intra body显示文摘Hypoxia is a disorder in which the tissues are not oxygenated adequately.The lack of oxygen may cause irreversible damage to the major organs such as the brain,heart and lungs.In severe cases,coma,seizures,and death may happen.In addition to the routine medication,acupuncture.treatment has been applied as a complementary treatment to counter hypoxia.In this paper,the optical fiber acupuncture needle was fabricated using the optical fiber imbedding into the metal capillary tube with needle tip,therefore made it easy to insert into the body for the laser treatment.The laser optical fiber needle treatment to the ST36 acupoint conducted with the laser.irradiation intra body.The normobaric hypoxia tolerance test results in mice show that the optical fber needle treatment to the ST36 acupoint with laser acupuncture appears to improve the tolerance to hypoxia.The mice treated with laser acupuncture expressed high level of IL-1βin serum.Our results suggest that laser optical fiber needle acupuncture may serve as a potential treatment for hypoxia.Shupeng Liu Minmin Xu Xiumei Cao Junyi Hou Na Chen Fufei Pang Zhenyi Chen Xiangfei Yang Tingyun Wang Jianshe Yan 2017Journal of Innovative Optical Health Sciences2017,,2:0
11ISS:Efficient Search Scheme Based on Immune Method in Modern Unstructured Peer-to-Peer Networks显示文摘Flooding is the most famous technique for locating contents in unstructured P2P networks. Recently traditional flood-ing has been replaced by more efficient dynamic query(DQ) and different variants of such algorithms. Dynamic query is a new flooding technique which could estimate a proper time-to-live(TTL) value for a query flooding by estimating the popularity of the searched files,and retrieve sufficient results under controlled flooding range for reducing network traffic. However,all DQ-like search algorithms are 'blind' so that a large amount of redundant messages are caused. In this paper,we proposed a new search scheme,called Immune Search Scheme(ISS) ,to cope with this problem. In ISS,an immune systems inspired concept of similar-ity-governed clone proliferation and mutation for query message movement is applied. 12Novel Functions of Interleukin-1 Beta in Spotted Halibut(Verasper variegatus) at Its Embryonic and Metamorphic Stages显示文摘Interleukin-1 beta(IL-1β) is a cytokine involved in the control of inflammatory responses.The functions of IL-1β have been extensively studied in many vertebrates.However,few studies focused on the functions of IL-1β in the spotted halibut(Verasper variegatus),an endangered aquatic fish that easily suffers from bacterial and viral infections.We first cloned and sequenced the IL-1β gene from V.variegatus(Vv IL-1β).Its conservative interleukin-1 propeptide and interleukin-1/18 domains corresponding regions,characteristics of the IL-1 protein family and TA-rich motifs relating to inflammatory responses were similar to those of the IL-1β genes from other teleost and mammals.The phylogenetic analysis showed that Vv IL-1β was homologous to the IL-1β gene of other vertebrates.Treatment with pathogen-associated molecular patterns(PAMPs) induced a significant and rapid up-regulation of Vv IL-1β expression in the peripheral blood leukocytes(PBLs),revealing that Vv IL-1β is actively involved in the host immune responses against bacterial and viral pathogens.The expression level of Vv IL-1β was relatively high in the PBLs,gill,and intestine,indicating the immunological function of Vv IL-1β.Vv IL-1β also had interesting expression patterns at specific developmental stages,implying that the IL-1β of flatfishes may have some novel functions during embryonic development and metamorphosis.The 3 D structure prediction also provided information about how Vv IL-1β interacts with its receptor protein.These results suggested that Vv IL-1β may have multiple functions in V.variegatus and provided us valuable information for understanding the novel functions of IL-1β.LI Zan LIU Xiumei CAO DANDan LIU Yuezhong WANG Xubo LIU Jinxiang WANG Zhigang ZHANG Quanqi YU Haiyang 2018Journal of Ocean University of China2018,17,5:0
13Effects of electroacupuncture combined with hydrogel on the formation and changes in the glial scar in rats with spinal cord injury显示文摘Objective: To observe the effect of electroacupuncture(EA) combined with oriented conductive bioprotein hydrogel(OCBH) on the recovery of nerve function in rats with complete spinal cord injury(SCI)and to explore its effect and mechanism on the formation and changes of glial scars.Methods: A total of 72 female Sprague-Dawley rats were randomly divided into groups according to the treatment received. A rat model of complete SCI was constructed using a spinal cord transection.Behavioral assessments, hematoxylin-eosin(H&E) staining, immunofluorescence staining, and Western blotting were performed at a fixed period after the operation.Results: The material group and the material + EA group obtained better results in the behavioral assessments(all P <.05) and the H&E staining. In the immunofluorescence staining and Western blotting,the GFAP protein was expressed more and denser in the material group and the material + EA group than in the model group, and the density of the GFAP expression in the material + EA group was lower at week 12 than in the material group(all P <.05). The expression of complement C3 in the model, material,and material + EA groups decreased in turn. Some inflammatory factors and the NF-κB signaling pathway showed similar results in the Western blotting(all P <.05). 153D bio-printed living nerve-like fibers refine the ecological niche for long-distance spinal cord injury regeneration显示文摘3D bioprinting holds great promise toward fabricating biomimetic living constructs in a bottom-up assembly manner.To date,various emergences of living constructs have been bioprinted for in vitro applications,while the conspicuous potential serving for in vivo implantable therapies in spinal cord injury(SCI)has been relatively overlooked.Herein,living nerve-like fibers are prepared via extrusion-based 3D bioprinting for SCI therapy.The living nerve-like fibers are comprised of neural stem cells(NSCs)embedded within a designed hydrogel that mimics the extracellular matrix(ECM),assembled into a highly spatial ordered architecture,similar to densely arranged bundles of the nerve fibers.The pro-neurogenesis ability of these living nerve-like fibers is tested in a 4 mm-long complete transected SCI rat model.Evidence shows that living nerve-like fibers refine the ecological niche of the defect site by immune modulation,angiogenesis,neurogenesis,neural relay formations,and neural circuit remodeling,leading to outstanding functional reconstruction,revealing an evolution process of this living construct after implantation.This effective strategy,based on biomimetic living constructs,opens a new perspective on SCI therapies.Jia Yang Kaiyuan Yang Weitao Man Jingchuan Zheng Zheng Cao Chun-Yi Yang Kunkoo Kim Shuhui Yang Zhaohui Hou Guihuai Wang Xiumei Wang 2023Bioactive Materials2023,,7:0
