Individualized prediction of perineural invasion in colorectal cancer: development and validation of a radiomics prediction model显示文摘Objective: To develop and validate a radiomics prediction model for individualized prediction of perineural invasion(PNI) in colorectal cancer(CRC).Methods: After computed tomography(CT) radiomics features extraction, a radiomics signature was constructed in derivation cohort(346 CRC patients). A prediction model was developed to integrate the radiomics signature and clinical candidate predictors [age, sex, tumor location, and carcinoembryonic antigen(CEA) level]. Apparent prediction performance was assessed. After internal validation, independent temporal validation(separate from the cohort used to build the model) was then conducted in 217 CRC patients. The final model was converted to an easy-to-use nomogram. Results: The developed radiomics nomogram that integrated the radiomics signature and CEA level showed good calibration and discrimination performance [Harrell's concordance index(c-index): 0.817; 95% confidence interval(95% CI): 0.811–0.823]. Application of the nomogram in validation cohort gave a comparable calibration and discrimination(c-index: 0.803; 95% CI: 0.794–0.812).Conclusions: Integrating the radiomics signature and CEA level into a radiomics prediction model enables easy and effective risk assessment of PNI in CRC. This stratification of patients according to their PNI status may provide a basis for individualized auxiliary treatment. | Yanqi Huang Lan He Di Dong Caiyun Yang Cuishan Liang Xin Chen Zelan Ma Xiaomei Huang Su Yao Changhong Liang Jie Tian Zaiyi Liu | 2018 | Chinese Journal of Cancer Research2018,30,1: | 23
Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations, respectively, and may benefit from immunotherapy or targeted therapies. We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment. | Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show | 2020 | Cell Research2020,30,10: | 19
Schisandra sphenanthera extract(Wuzhi Tablet)protects against chronic-binge and acute alcohol-induced liver injury by regulating the NRF2-ARE pathway in mice显示文摘Alcohol abuse leads to alcoholic liver disease and no effective therapy is currently available.Wuzhi Tablet(WZ), a preparation of extract from Schisandra sphenanthera that is a traditional hepatoprotective herb, exerted a significant protective effect against acetaminophen-induced liver injury in our recent studies, but whether WZ can alleviate alcohol-induced toxicity remains unclear. This study aimed to investigate the contribution of WZ to alcohol-induced liver injury by using chronic-binge and acute models of alcohol feeding. The activities of ALT and AST in serum were assessed as well as the level of GSH and the activity of SOD in the liver. The expression of CYP2E1 and proteins in the NRF2-ARE signaling pathway including NRF2, GCLC, GCLM, HO-1 were measured, and the effect of WZ on NRF2 transcriptional activity was determined. We found that both models resulted in liver steatosis accompanied by increased transaminase activities, but that liver injury was significantly attenuated by WZ. WZ administration also inhibited CYP2E1 expression induced by alcohol, and elevated the level of GSH and the activity of SOD in the liver. Moreover, the NRF2-ARE signaling pathway was activated by WZ andthe target genes were all upregulated. Furthermore, WZ significantly activated NRF2 transcriptional activity. Collectively, our study demonstrates that WZ protected against alcohol-induced liver injury by reducing oxidative stress and improving antioxidant defense, possibly by activating the NRF2-ARE pathway. | Xuezhen Zeng Xi Li Chenshu Xu Fulin Jiang Yufei Mo Xiaomei Fan Yaoting Li Yiming Jiang Dongshun Li Min Huang Huichang Bi | 2017 | Acta Pharmaceutica Sinica B2017,7,5: | 13
Field emission characteristics of ZnO nanotetrapods and the effect of thermal annealing in hydrogen显示文摘Large-scale tetrapod-like ZnO nanostructures have been synthesized using a thermal chemical vapor deposition method on a silicon substrate. The high-purity nanotetrapods show sharp tips geometry with a wurtzite structure. The field emission properties of the uniform ZnO nanostructural material are investigated at different anode-cathode distances. The turn-on field for the ZnO nanotetrapods is found to be about 3.7 V/μm at a current density of 1 μA/cm2. The field emission behavior obeys Fowler-Nordheim relationship. More importantly, the field emission properties are improved after an-nealing in hydrogen, and therefore high emission current and low turn-on field are obtained. These results indicate that tetrapod-like ZnO nanostructures are a promising candidate for cold cathode emitters. | CHEN HongSheng QI JunJie ZHANG Yue LIAO QingLiang ZHANG XiaoMei HUANG YunHua | 2007 | Chinese Science Bulletin2007,52,9: | 5
Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude. However, the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood. Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss, polycythemia, and pathological cardiac hypertrophy in hypoxic rats, accompanied by a large compositional shift in the gut microbiota, which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae, and Streptococcaceae. The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes, and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces, suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly, the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota, partially inhibited the increased abundance of the Bacteroides and Alistipes genera, restored the decrease of plasma propionate, and moderately ameliorated cardiac hypertrophy in hypoxic rats. Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats. | Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang | 2022 | Science China(Life Sciences)2022,65,10: | 5
Progress in the development of techniques based on light scattering for single nanoparticle detection显示文摘Nanoparticles have recently attracted extensive attention in view of their great potential in biomedicine and bioanalytical ap- plications. Single particle detection via light scattering offers a simple and efficient approach for the size, size distribution, and concentration analysis of nanoparticles. In particular, intrinsic heterogeneity or rare events masked by ensemble averaging can be revealed. However, the sixth power dependence of Rayleigh scattering on particle size makes it very challenging to detect individual nanoparticles of small sizes. This article is intended to provide an overview of recent progress in the development of techniques based on light scattering for the detection of single nanoparticles. | ZHU ShaoBin, WANG Shuo, YANG LingLing, HUANG TianXun & YAN XiaoMei Department of Chemical Biology, College of Chemistry and Chemical Engineering Key Laboratory for Chemical Biology of Fujian Province Key Laboratory of Analytical Science, Xiamen University, Xiamen 361005, China | 2011 | Science China Chemistry2011,54,8: | 5
Evaluation of human epidermal growth factor receptor 2 status of breast cancer using preoperative multidetector computed tomography with deep learning and handcrafted radiomics features显示文摘Objective:To evaluate the human epidermal growth factor receptor 2(HER2)status in patients with breast cancer using multidetector computed tomography(MDCT)-based handcrafted and deep radiomics features. Methods:This retrospective study enrolled 339 female patients(primary cohort,n=177;validation cohort,n=162)with pathologically confirmed invasive breast cancer.Handcrafted and deep radiomics features were extracted from the MDCT images during the arterial phase. After the feature selection procedures, handcrafted and deep radiomics signatures and the combined model were built using multivariate logistic regression analysis.Performance was assessed by measures of discrimination, calibration, and clinical usefulness in the primary cohort and validated in the validation cohort. Results:The handcrafted radiomics signature had a discriminative ability with a C-index of 0.739[95%confidence interval(95%CI):0.661-0.818]in the primary cohort and 0.695(95%CI:0.609-0.781)in the validation cohort. The deep radiomics signature also had a discriminative ability with a C-index of 0.760(95%CI:0.690-0.831)in the primary cohort and 0.777(95%CI:0.696-0.857)in the validation cohort. The combined model, which incorporated both the handcrafted and deep radiomics signatures, showed good discriminative ability with a C-index of 0.829(95%CI:0.767-0.890)in the primary cohort and 0.809(95%CI:0.740-0.879)in the validation cohort. Conclusions:Handcrafted and deep radiomics features from MDCT images were associated with HER2 status in patients with breast cancer. Thus, these features could provide complementary aid for the radiological evaluation of HER2 status in breast cancer. | Xiaojun Yang Lei Wu Ke Zhao Weitao Ye Weixiao Liu Yingyi Wang Jiao Li Hanxiao Li Xiaomei Huang Wen Zhang Yanqi Huang Xin Chen Su Yao Zaiyi Liu Changhong Liang | 2020 | Chinese Journal of Cancer Research2020,32,2: | 4
Anti-cavitation performance of a splitter-bladed inducer under different flow rates and different inlet pressures显示文摘The anti-cavitation performance of a high-speed centrifugal pump with a splitter-bladed inducer is investigated under different flow rates and different inlet pressures. Simulations and external characteristics experiments are carried out. Static pressure and the vapor volume fraction distributions on the inducer and the impeller of the pump under various operation conditions are obtained. The results show that the cavitation developments on the impeller and on the inducer with the flow rates are reverse, while the development of the inlet pressure on the inducer and the impeller is the same. Cavitation on the impeller increases with the increase of flow rates, and it extends to the near passages with rotating, while cavitation on the inducer is more complex than that on the impeller. Cavitation at the inlet of the inducer decreases with the increase of flow rates, while cavitation at the outlet of the inducer is opposite. The results also show that cavitation development on the impeller and on the inducer with the inlet pressure is the same. Cavitation both decreases with the increase of the inlet pressure at the same flow rate. Furthermore, asymmetric cavitation on the impeller and on the inducer is both observed. And the asymmetric degree of cavitation on the impeller is higher than that on the inducer. | GUO XiaoMei ZHU ZuChao CUI BaoLing HUANG Yong | 2015 | Science China(Technological Sciences)2015,58,12: | 4
Testosterone attenuates pulmonary epithelial inflammation in male rats of COPD model through preventing NRF1-derived NF-κB signaling显示文摘Testosterone deficiency is common in male patients with chronic obstructive pulmonary disease (COPD) and may correlate with the deterioration of COPD. Clinical research suggests that testosterone replacement therapy may slow the COPD progression, but the specific biological pathway remains unclear. In this study, we explored the effect of testosterone on pulmonary inflammation in male COPD rats. The animals were co-treated with lipopolysaccharide (LPS) and cigarette to induce COPD. In COPD rats, nuclear respiratory factor 1 (NRF1) and NF-κB p65 were upregulated. In cigarette smoke extract (CSE)-, LPS-, or the combination of CSE and LPS-treated L132 cells, NRF1 and p65 were also upregulated. Silencing NRF1 resulted in the downregulation of p65. ChIP‒seq, ChIP‒qPCR, and luciferase results showed that NRF1 transcriptionally regulated p65. Both male and female COPD rats showed an upregulated NRF1 level and similar pulmonary morphology. But NRF1 was further upregulated in male castrated rats. Further supplementing testosterone in castrated male rats significantly reduced NRF1, pulmonary lesions, and inflammation. Supplementation of testosterone also reduced the phosphorylation of p65 and IKKβ induced by LPS or CSE in L132 cells. Our results suggest that testosterone plays a protective role in pulmonary epithelial inflammation of COPD through inhibition of NRF1-derived NF-κB signaling and the phosphorylation of p65. | Xueting Wang Linlin Huang Shan Jiang Kang Cheng Dan Wang Qianqian Luo Xiaomei Wu Li Zhu | 2021 | Journal of Molecular Cell Biology2021,13,2: | 4
Star-shaped conjugated oligoelectrolyte for bioimaging in living cells显示文摘A new star-shaped oligoelectrolyte (TEFCOONa) with triphenylamine as the core, acetylene as linkage and anionic fluorenes as arms was obtained and used for direct imaging in living PANC-1 cells. Because of the hydrophobic conjugated groups of the oli-goelectrolyte, TEFCOONa can form nanospheres with an average diameter of^75 nm in 10 mmol/L PBS. These nanospheres possess a relatively high absolute quantum yield (16.5% in PBS), low cytotoxicity and can penetrate into the nucleus through the cytoplasm, which is essential for living cellular imaging. Collectively, these results validate our rational design of conjugated oligoelectrolyte and even hyper branched polymers-copolyelectrolyte as effective nanovectors for bioimaging and other clinical applications. | SONG WenLi JIANG RongCui YUAN Yan LU XiaoMei HU WenBo FAN QuLi HUANG Wei | 2013 | Chinese Science Bulletin2013,58,21: | 4
An in situ ATR-FTIR study of mixed collectors BHA/DDA adsorption in ilmenite-titanaugite flotation system显示文摘This paper researched the enhanced flotation separation performance of ilmenite and titanaugite using the mixed collector benzhydroxamic acid/dodecylamine(BHA/DDA).The interface assembly mechanism was mainly investigated through in situ attenuated total reflectance Fourier transform infrared(ATRFTIR)spectroscopy combined with the two-dimensional correlation spectroscopy(2D-COS)and X-ray photoelectron spectroscopy(XPS).It has been found that BHA/DDA mixed collectors successfully separate ilmenite from titanaugite at a molar ratio of 8:1.Zeta potential experiments suggested that,in the presence of mixed collector system, the BHA-DDA complex adsorbed on the ilmenite surface via the chemically adsorbed BHA and the electrostatically adsorbed DDA,however, the complex adsorbed on the surface of titanaugite unstably.According to in situ ATR-FTIR combined with 2D-COS and XPS results, the interface assembly mechanism of BHA/DDA is summarized as:the function group of BHA molecules first binds to the metal sites on minerals to form bidentate ligand, then DDA co-adsorbed with BHA on the surface of minerals through hydrogen bonding.DDA may change the adsorption modes of some BHA on the ilmenite surface from four-membered ring to five-membered ring, while the modes on the titanaugite surface is true opposite. Finally, recommended adsorption configurations of the BHA/DDA complex on the two mineral surfaces are proposed. | Liping Luo Houqin Wu Longhua Xu Jinping Meng Jiahui Lu Huan Zhou Xiaomei Huo Lingyun Huang | 2021 | International Journal of Mining Science and Technology2021,31,4: | 4

Development and validation of a CT-based radiomics nomogram for preoperative prediction of tumor histologic grade in gastric adenocarcinoma显示文摘Objectives:To develop and validate a radiomics nomogram for preoperative prediction of tumor histologic grade in gastric adenocarcinoma(GA).Methods:This retrospective study enrolled 592 patients with clinicopathologically confirmed GA(low-grade:n=154;high-grade:n=438)from January 2008 to March 2018 who were divided into training(n=450)and validation(n=142)sets according to the time of computed tomography(CT)examination.Radiomic features were extracted from the portal venous phase CT images. The Mann-Whitney U test and the least absolute shrinkage and selection operator(LASSO)regression model were used for feature selection, data dimension reduction and radiomics signature construction.Multivariable logistic regression analysis was applied to develop the prediction model. The radiomics signature and independent clinicopathologic risk factors were incorporated and presented as a radiomics nomogram. The performance of the nomogram was assessed with respect to its calibration and discrimination. Results:A radiomics signature containing 12 selected features was significantly associated with the histologic grade of GA(P<0.001 for both training and validation sets).A nomogram including the radiomics signature and tumor location as predictors was developed. The model showed both good calibration and good discrimination,in which C-index in the training set,0.752[95%confidence interval(95%CI):0.701-0.803];C-index in the validation set,0.793(95%CI:0.711-0.874).Conclusions:This study developed a radiomics nomogram that incorporates tumor location and radiomics signatures, which can be useful in facilitating preoperative individualized prediction of histologic grade of GA. | Jia Huang Huasheng Yao Yexing Li Mengyi Dong Chu Han Lan He Xiaomei Huang Ting Xia Zongjian Yi Huihui Wang Yuan Zhang Jian He Changhong Liang Zaiyi Liu | 2021 | Chinese Journal of Cancer Research2021,33,1: | 3
Water-soluble light-emitting nanoparticles prepared by non-covalent bond self-assembly of a hydroxyl group functionalized oligo(p-phenyleneethynylene) with different water-soluble polymers显示文摘Water-soluble light-emitting nanoparticles were prepared from hydroxyl group functionalized oligos(p-phenyleneethynylene) (OHOPEL) and water-soluble polymers(PEG,PAA,and PG) by non-covalent bond self-assembly.Their structure and optoelectronic properties were investigated through dynamic light scattering(DLS) ,UV and PL spectroscopy. The optical properties of OHOPEL-based water-soluble nanoparticles exhibited the same properties as that found in OHOPEL films, indicating the existence of interchain-aggregation of OHOPELs in the nanoparticles.OHOPEL-based nanoparticles prepared from conjugated oligomers show smaller size and lower dispersity than nanoparticles from conjugated polymers, which means that the structures of water-soluble nanoparticles are linked to the conjugated length.Furthermore, the OHOPEL/PG and OHOPEL/PAA systems produced smaller particles and lower polydispersity than the OHOPEL/PEG system, indicating that there may exist influence of the strength of non-covalent bonds on the size and degree of dispersity of the nanoparticles. | Lu XiaoMei Fan QuLi Zhang GuangWei Pu KanYi Huang Wei | 2010 | Science China Chemistry2010,53,5: | 3
BioTSA: Annotating Token Semantic Association to Support Biomedical Text Mining显示文摘Corpus is a kind of important resource for knowledge acquisition in the natural language processing(NLP). However, up to now, in the biomedical domain comparatively fewer corpus focus on semantic association among all tokens in a sentence. We proposed an annotation scheme based on feature structure theory for enriching biomedical domain corpora with token semantic association(TSA). There are 227 documents of the Bio NLP GE ST training data annotated to form TSA corpus in which each annotated item shows a token semantic association that appears as a triple. The annotation of token semantic association has the potential to significantly advance biomedical text mining by providing rich token semantic information for NLP systems especially for the sophisticated IE systems, such as bio-event extraction. | WEI Xiaomei HUANG Sixing CHEN Bo JI Donghong | 2015 | Wuhan University Journal of Natural Sciences2015,20,2: | 2

Dynamics and manipulation of ferroelectric domain walls in bismuth ferrite thin films显示文摘Ferroelectric domain walls differ from domains not only in their crystalline and discrete symmetry, but also in their electronic, magnetic, and mechanical properties.Although domain walls provide a degree of freedom to regulate the physical properties at the nanoscale;the relatively lower controllability prevents their practical applications in nano-devices.In this work;with the advantages of 3D domain configuration detection based on piezoresponse force microscopy;we find that the mobility of three types of domain walls(tail-to-tail,head-to-tail,head-to-head)in(001)BiFeO3 films varies with the applied electrical field.Under low voltages;head-to-tail domain walls are more mobile than other domain walls;while, under high voltages,tail-to-tail domain walls become rather active and possess relatively long average lengths. This is due to the high nucleation energy and relatively low growth energy for charged domain walls. Finally;we demonstrate the manipulation of domain walls through successive electric writings, resulting in well-aligned conduction paths as designed;paving the way for their application in advanced spintronic;memory and communication nano-devices. | Shuyu Xiao Yaming Jin Xiaomei Lu Sang-Wook Cheong Jiangyu Li Yang Li Fengzhen Huang Jinsong Zhu | 2020 | National Science Review2020,7,2: | 2
