1Ferroptosis is essential for diabetic cardiomyopathy and is prevented by sulforaphane via AMPK/NRF2 pathways显示文摘Herein,we define the role of ferroptosis in the pathogenesis of diabetic cardiomyopathy(DCM)by examining the expression of key regulators of ferroptosis in mice with DCM and a new ex vivo DCM model.Advanced glycation end-products(AGEs),an important pathogenic factor of DCM,were found to induce ferroptosis in engineered cardiac tissues(ECTs),as reflected through increased levels of Ptgs2 and lipid peroxides and decreased ferritin and SLC7 A11 levels.Typical morphological changes of ferroptosis in cardiomyocytes were observed using transmission electron microscopy.Inhibition of ferroptosis with ferrostatin-1 and deferoxamine prevented AGE-induced ECT remodeling and dysfunction.Ferroptosis was also evidenced in the heart of type 2 diabetic mice with DCM.Inhibition of ferroptosis by liproxstatin-1 prevented the development of diastolic dysfunction at 3 months after the onset of diabetes.Nuclear factor erythroid 2-related factor 2(NRF2)activated by sulforaphane inhibited cardiac cell ferroptosis in both AGE-treated ECTs and hearts of DCM mice by upregulating ferritin and SLC7 A11 levels.The protective effect of sulforaphane on ferroptosis was AMP-activated protein kinase(AMPK)-dependent.These findings suggest that ferroptosis plays an essential role in the pathogenesis of DCM;sulforaphane prevents ferroptosis and associated pathogenesis via AMPK-mediated NRF2 activation.This suggests a feasible therapeutic approach with sulforaphane to clinically prevent ferroptosis and DCM.Xiang Wang Xinxin Chen Wenqian Zhou Hongbo Men Terigen Bao Yike Sun Quanwei Wang Yi Tan Bradley B.Keller Qian Tong Yang Zheng Lu Cai 2022Acta Pharmaceutica Sinica B2022,12,2:20
2Cotton GhPOX1 encoding plant class Ⅲ peroxidase may be responsible for the high level of reactive oxygen species production that is related to cotton fiber elongation显示文摘The accumulation of reactive oxygen species(ROS) is involved in plant cell development.In plant,class Ⅲ peroxidases are heme-containing enzymes encoded by a large multi-gene family participated in the release or consumption of ROS.The specific function of each member of the family is still elusive.Here,we showed that ROS was significantly generated during cotton fiber initiation and elongation,whereas,application of NADPH oxidase inhibitor diphenyleneiodonium(DPI) and peroxidase inhibitor salicylhydroxamic acid(SHAM) to the wild-type cotton ovule culture significantly suppressed fiber growth,respectively.Their inhibitory effects were caused by the reduction of superoxide radical(O2-).Ten GhPOX genes(cDNAs) encoding cotton class Ⅲ peroxidases were isolated,among them eight GhPOX genes were reported for the first time.Microarray analyses indicated that GhPOX1 was the mostly predomi-nantly expressed in fast-elongating cotton fiber cells.Real-time quantitative PCR analysis revealed the transcript level of GhPOX1 was over 400-fold higher in growing fiber cells than in ovules,flowers,roots,stems and leaves.To reveal the role of GhPOX1 in plant devel-opment,its Arabidopsis orthologue atpox13 mutant was demonstrated to be defective in branch root development.Taken together,the data suggest that GhPOX1 plays an important role during fiber cell elongation possibly by mediating production of reactive oxygen spe-cies.Wenqian Mei Yongmei Qin Wenqiang Song Jun Li Yuxian Zhu 2009Journal of Genetics and Genomics2009,36,3:19

4Dual-targeting and microenvironment-responsive micelles as a gene delivery system to improve the sensitivity of glioma to radiotherapy显示文摘Dbait is a small double-stranded DNA molecule that has been utilized as a radiosensitizer to enhance the sensitivity of glioma to radiotherapy(RT). However, there is no effective drug delivery system to effectively overcome the blood–brain barrier(BBB). The aim of this study was to develop a gene delivery system by using the BBB and glioma dual-targeting and microenvironment-responsive micelles(ch-Kn(s-s)R8-An) to deliver Dbait into glioma for RT. Angiopep-2 can target the low-density lipoprotein receptor-related protein-1(LRP1) that is overexpressed on brain capillary endothelial cells(BCECs) and glioma cells. In particular, due to upregulated matrix metalloproteinase 2(MMP-2) in the tumor microenvironment, we utilized MMP-2-responsive peptides as the enzymatically degradable linkers to conjugate angiopep-2. The results showed that ch-Kn(s-s)R8-An micelles maintained a reasonable size(80–160 nm) with a moderate distribution and a decreased mean diameter from the cross-linking as well as exhibited low critical micelle concentration(CMC) with positive surface charge, ranging from 15 to40 mV. The ch-K5(s-s)R8-An/pEGFP showed high gene transfection efficiency in vitro, improved uptake in glioma cells and good biocompatibility in vitro and in vivo. In addition, the combination of ch-K5(s-s)R8-An/Dbait with RT significantly inhibited the growth of U251 cells in vitro. 5Spatial and temporal change patterns of net primary productivity and its response to climate change in the Qinghai–Tibet Plateau of China from 2000 to 2015显示文摘The vegetation ecosystem of the Qinghai–Tibet Plateau in China,considered to be the′′natural laboratory′′of climate change in the world,has undergone profound changes under the stress of global change.Herein,we analyzed and discussed the spatial-temporal change patterns and the driving mechanisms of net primary productivity(NPP)in the Qinghai–Tibet Plateau from 2000 to 2015 based on the gravity center and correlation coefficient models.Subsequently,we quantitatively distinguished the relative effects of climate change(such as precipitation,temperature and evapotranspiration)and human activities(such as grazing and ecological construction)on the NPP changes using scenario analysis and Miami model based on the MOD17A3 and meteorological data.The average annual NPP in the Qinghai–Tibet Plateau showed a decreasing trend from the southeast to the northwest during 2000–2015.With respect to the inter-annual changes,the average annual NPP exhibited a fluctuating upward trend from 2000 to 2015,with a steep increase observed in 2005 and a high fluctuation observed from 2005 to 2015.In the Qinghai–Tibet Plateau,the regions with the increase in NPP(change rate higher than 10%)were mainly concentrated in the Three-River Source Region,the northern Hengduan Mountains,the middle and lower reaches of the Yarlung Zangbo River,and the eastern parts of the North Tibet Plateau,whereas the regions with the decrease in NPP(change rate lower than–10%)were mainly concentrated in the upper reaches of the Yarlung Zangbo River and the Ali Plateau.The gravity center of NPP in the Qinghai–Tibet Plateau has moved southwestward during 2000–2015,indicating that the increment and growth rate of NPP in the southwestern part is greater than those of NPP in the northeastern part.Further,a significant correlation was observed between NPP and climate factors in the Qinghai–Tibet Plateau.The regions exhibiting a significant correlation between NPP and precipitation were mainly located in the central and eastern Qinghai–Tibet Plateau,and the regions exhibiting a significant correlation between NPP and temperature were mainly located in the southern and eastern Qinghai–Tibet Plateau.Furthermore,the relative effects of climate change and human activities on the NPP changes in the Qinghai–Tibet Plateau exhibited significant spatial differences in three types of zones,i.e.,the climate change-dominant zone,the human activity-dominant zone,and the climate change and human activity interaction zone.These research results can provide theoretical and methodological supports to reveal the driving mechanisms of the regional ecosystems to the global change in the Qinghai–Tibet Plateau.GUO Bing ZANG Wenqian YANG Fei HAN Baomin CHEN Shuting LIU Yue YANG Xiao HE Tianli CHEN Xi LIU Chunting GONG Rui 2020Journal of Arid Land2020,12,1:12

8Permian integrative stratigraphy and timescale of China显示文摘A series of global major geological and biological events occurred during the Permian Period. Establishing a highresolution stratigraphic and temporal framework is essential to understand their cause-effect relationship. The official International timescale of the Permian System consists of three series(i.e., Cisuralian, Guadalupian and Lopingian in ascending order) and nine stages. In China, the Permian System is composed of three series(Chuanshanian, Yansingian and Lopingian) and eight stages, of which the subdivisions and definitions of the Chuanshanian and Yangsingian series are very different from the Cisuralian and Guadalupian series. The Permian Period spanned ~47 Myr. Its base is defined by the First Appearance Datum(FAD) of the conodont Streptognathodus isolatus at Aidaralash, Kazakhstan with an interpolated absolute age 298.9±0.15 Ma at Usolka, southern Urals, Russia. Its top equals the base of the Triassic System and is defined by the FAD of the conodont Hindeodus parvus at Meishan D section, southeast China with an interpolated absolute age 251.902±0.024 Ma. Thirty-five conodont, 23 fusulinid, 17 radiolarian and 20 ammonoid zones are established for the Permian in China, of which the Guadalupian and Lopingian conodont zones have been served as the standard for international correlation. The Permian δ13 Ccarbtrend indicates that it is characterized by a rapid negative shift of 3–5‰ at the end of the Changhsingian, which can be used for global correlation of the end-Permian mass extinction interval, but δ13 Ccarbrecords from all other intervals may have more or less suffered subsequent diagenetic alteration or represented regional or local signatures only. Permian δ18 Oapatitestudies suggest that an icehouse stage dominated the time interval from the late Carboniferous to Kungurian(late Cisuralian). However, paleoclimate began to ameriolate during the late Kungurian and gradually shifted into a greenhouse-dominated stage during the Guadalupian.The Changhsingian was a relatively cool stage, followed by a globally-recognizable rapid temperature rise of 8–10°C at the very end of the Changhsingian. The87 Sr/86 Sr ratio trend shows that their values at the beginning of the Permian were between 0.70800,then gradually decreased to the late Capitanian minimum 0.70680–0.70690, followed by a persistent increase until the end of the Permian with the value 0.70708. Magenetostratigraphy suggests two distinct stages separated by the Illawarra Reversal in the middle Wordian, of which the lower is the reverse polarity Kiaman Superchron and the upper is the mixed-polarity Illawarra Superchron. The end-Guadalupian(or pre-Lopingian) biological crisis occurred during the late Capitanian, when faunal changeovers of different fossil groups had different paces, but generally experienced a relatively long time from the Jinogondolella altudensis Zone until the earliest Wuchiapingian. The end-Permian mass extinction was a catastrophic event that is best constrained at the Meishan section, which occurred at 251.941±0.037 Ma and persisted no more than 61±48 kyr. After the major pulse at Bed 25, the extinction patterns are displayed differently in different sections. The global end-Guadalupian regression is manifested between the conodont Jinogondolella xuanhanensis and Clarkina dukouensis zones and the endChanghsingian transgression began in the Hindeodus changxingensis-Clarkina zhejiangensis Zone. 9Molecular biomarkers for gestational diabetes mellitus and postpartum diabetes显示文摘Gestational diabetes mellitus(GDM)is a growing public health problem worldwide that threatens both maternal and fetal health.Identifying individuals at high risk for GDM and diabetes after GDM is particularly useful for early intervention and prevention of disease progression.In the last decades,a number of studies have used metabolomics,genomics,and proteomic approaches to investigate associations between biomolecules and GDM progression.These studies clearly demonstrate that various biomarkers reflect pathological changes in GDM.The established markers have potential use as screening and diagnostic tools in GDM and in postpartum diabetes research.In the present review,we summarize recent studies of metabolites,single-nucleotide polymorphisms,microRNAs,and proteins associated with GDM and its transition to postpartum diabetes,with a focus on their predictive value in screeningand diagnosis.Wenqian Lu Cheng Hu 2022Chinese Medical Journal2022,135,16:9

11Challenges of evaluating immunotherapy efficacy in solid tumors显示文摘Immunotherapy is one of the most promising treatments for multiple tumor types.The significant clinical benefits and durable responses of immunotherapy have led to the emergence of various immune-related clinical response patterns that extend beyond those achieved with cytotoxic agents.Various studies investigated the efficacy of immunotherapy,including the effect on tumor size,long-term survival benefits,and the ability to overcome the particularly challenging survival curves tailing phenomenon.The current immune-related methods guidelines,such as immune-related Response Criteria(irRC),immune-related Response Evaluation Criteria in Solid Tumors(irRECIST),immune Response Evaluation Criteria in Solid Tumors(iRECIST),and immune-modified Response Evaluation Criteria in Solid Tumors(imRECIST),could be well-adapted to identify the heterogeneity of responses that appear in patients receiving immunotherapy,such as pseudoprogression(PsPD)and hyperprogressive disease(HPD),and to some extent to overcome the limitation of evaluating the efficacy of immunotherapy on tumor size by imaging.Additionally,a second type of evaluation method was proposed based on survival,which includes milestone analysis and restricted mean survival time.Currently,milestone analysis is a complementary tool to summarize and interpret trial results along with more conventional measures of survival and other less established metrics.A golden standard evaluation method to distinguish the efficacy of immunotherapy may improve the process of imaging and aid survival-based efficacy evaluation in patients with solid tumors.Rilan Bai Wenqian Li Nawen Du Jiuwei Cui 2019Chinese Journal of Cancer Research2019,31,6:8

14Soil phosphorus availability and rice phosphorus uptake in paddy fields under various agronomic practices显示文摘Agronomic practices affect soil phosphorus(P) availability, P uptake by plants, and subsequently the efficiency of P use. A field experiment was carried out to investigate the effects of various agronomic practices(straw incorporation, paddy water management, nitrogen(N) fertilizer dose, manure application,and biochar addition) on soil P availability(e.g., soil total P(STP), soil available P(SAP), soil microbial biomass P(SMBP), and rice P uptake as well as P use efficiency(PUE)) over four cropping seasons in a rice-rice cropping system, in subtropical central China. Compared to the non-straw treatment(control,using full dose of chemical N fertilizer), straw incorporation increased SAP and SMBP by 9.3%–18.5% and 15.5%–35.4%, respectively;substituting half the chemical N fertilizer dose with pig manure and the biochar application increased STP, SAP, and SMBP by 10.5%–48.3%, 30.2%–236.0%, and 19.8%–72.4%,respectively, mainly owing to increased soil P and organic carbon inputs;adding a half dose of N and no N input(reduced N treatments) increased STP and SAP by 2.6%–7.5% and 19.8%–33.7%, respectively, due to decreased soil P outputs. Thus, soil P availability was greatly affected by soil P input and use. The continuous flooding water regime without straw addition significantly decreased SMBP by 11.4% compared to corresponding treatments under a mid-season drainage water regime. Total P uptake by rice grains and straws at the harvest stage increased under straw incorporation and under pig manure application, but decreased under the reduced N treatments and under biochar application at a rate of 48 t ha-1, compared to the control. Rice P uptake was significantly positively correlated with rice biomass, and both were positively correlated with N fertilizer application rates, SAP, SMBP, and STP. Phosphorus use efficiency generally increased under straw incorporation but decreased under the reduced N treatments and under the manure application(with excessive P input), compared to the control. These results showed that straw incorporation can be used to increase soil P availability and PUE while decreasing the use of chemical P fertilizers. 15Three-Dimensional Tidal Model and Its Application to Numerical Simulation of Water Quality in Coastal Waters显示文摘- The turbulence mechanism plays an important part in the mixing process and momentum transfer of turbulence. A three-dimensional Prandtl mixing length tidal model has been developed to simulate tidal flows and water quality. The eddy viscosities and diffusivities are computed from the Prandtl mixing length model. In order to model the water quality of an estuary or coastal area many interdependent processes need to be simulated. These may be conveniently separated into three main groups: transport and mixing processes, biochemical interaction of water quality variables and the utilization and re-cycling of nutrients by living matter. The model simulates full oxygen and nutrient balance, primary productivity and the transport, reaction mechanism and fate of pollutants over tidal time-scales. The model is applied to numerical simulation of tidal flows and water quality in Dalian Bay. 17Assessing the benthic quality status of the Bohai Bay(China)with proposed modifications of M-AMBI显示文摘Multivariate AZTI's Marine Biotic Index(M-AMBI) was designed to indicate the ecological status of European coastal areas.Based upon samples collected from 2009 to 2012 in the Bohai Bay,we have tested the response of variations of M-AMBI,using biomass(M-BAMBI) in the calculations,with different transformations of the raw data.The results showed that the ecological quality of most areas in the study indicated by M-AMBI was from moderate to bad status with the worse status in the coastal areas,especially around the estuaries,harbors and outfalls,and better status in the offshore areas except the area close to oil platforms or disposal sites.Despite large variations in nature of the input data,all variations of M-AMBI gave similar spatial and temporal distribution patterns of the ecological status within the bay,and showed high correlation between them.The agreement of new ecological status obtained from all M-AMBI variations,which were calculated according to linear regression,was almost perfect.The benthic quality,assessed using different input data,could be related to human pressures in the bay,such as water discharges,land reclamation,dredged sediment and drilling cuts disposal sites.It seems that M-BAMBI were more effective than M-NABMI(M-AMBI calculated using abundance data) in indicating human pressures of the Bay.Finally,indices calculated with more severe transformations,such as presence/absence data,could not indicate the higher density of human pressures in the coastal areas of the north part of our study area,but those calculated using mild transformation(i.e.,square root) did.CAI Wenqian BORJA Angel LIN Kuixuan ZHU Yanzhong ZHOU Juan LIU Lusan 2015Acta Oceanologica Sinica2015,34,10:5

20Effective control allocation using hierarchical multi-objective optimization for multi-phase flight显示文摘For different flight phases in an overall flight mission,different control and allocation preferences should be pursued considering lift,drag or maneuverability characteristics.The multi-objective flight control allocation problem for a multi-phase flight mission is studied.For an overall flight mission,different flight phases namely climbing,cruise,maneuver and gliding phases are defined.Firstly,a multi-objective control allocation problem considering drag,lift or control energy preference is constructed.Secondly,considering different control preferences at different flight phases,the analytic hierarchical process method is used to construct a comprehensive performance index from different objectives such as lift or drag preferences.The active set based dynamic programming optimization method is used to solve the real-time optimization problem.For the validation,the Innovative Control Effector(ICE)tailless aircraft nonlinear model and the angular acceleration measurements based adaptive Incremental Backstepping(IBKS)are used to construct the validation platform.Finally,an overall flight mission is simulated to demonstrate the efficiency of the proposed multi-phase and multi-objective flight control allocation method.The results show that the comprehensive performance index for different phases,which are determined from the Analytic Hierarchy Process(AHP)method,can suitably satisfy the preference requirements for different flight phases.Liguo SUN Qing ZHOU Baoxu JIA Wenqian TAN Hangxu LI 2020Chinese Journal of Aeronautics2020,33,7:4
