| 1 | Dual-targeting and microenvironment-responsive micelles as a gene delivery system to improve the sensitivity of glioma to radiotherapy显示文摘Dbait is a small double-stranded DNA molecule that has been utilized as a radiosensitizer to enhance the sensitivity of glioma to radiotherapy(RT). However, there is no effective drug delivery system to effectively overcome the blood–brain barrier(BBB). The aim of this study was to develop a gene delivery system by using the BBB and glioma dual-targeting and microenvironment-responsive micelles(ch-Kn(s-s)R8-An) to deliver Dbait into glioma for RT. Angiopep-2 can target the low-density lipoprotein receptor-related protein-1(LRP1) that is overexpressed on brain capillary endothelial cells(BCECs) and glioma cells. In particular, due to upregulated matrix metalloproteinase 2(MMP-2) in the tumor microenvironment, we utilized MMP-2-responsive peptides as the enzymatically degradable linkers to conjugate angiopep-2. The results showed that ch-Kn(s-s)R8-An micelles maintained a reasonable size(80–160 nm) with a moderate distribution and a decreased mean diameter from the cross-linking as well as exhibited low critical micelle concentration(CMC) with positive surface charge, ranging from 15 to40 mV. The ch-K5(s-s)R8-An/pEGFP showed high gene transfection efficiency in vitro, improved uptake in glioma cells and good biocompatibility in vitro and in vivo. In addition, the combination of ch-K5(s-s)R8-An/Dbait with RT significantly inhibited the growth of U251 cells in vitro. | 6 | Quantitative distinction of the relative actions of climate change and human activities on vegetation evolution in the Yellow River Basin of China during 1981-2019显示文摘Under the combined influence of climate change and human activities,vegetation ecosystem has undergone profound changes.It can be seen that there are obvious differences in the evolution patterns and driving mechanisms of vegetation ecosystem in different historical periods.Therefore,it is urgent to identify and reveal the dominant factors and their contribution rates in the vegetation change cycle.Based on the data of climate elements(sunshine hours,precipitation and temperature),human activities(population intensity and GDP intensity)and other natural factors(altitude,slope and aspect),this study explored the spatial and temporal evolution patterns of vegetation NDVI in the Yellow River Basin of China from 1989 to 2019 through a residual method,a trend analysis,and a gravity center model,and quantitatively distinguished the relative actions of climate change and human activities on vegetation evolution based on Geodetector model.The results showed that the spatial distribution of vegetation NDVI in the Yellow River Basin showed a decreasing trend from southeast to northwest.During 1981-2019,the temporal variation of vegetation NDVI showed an overall increasing trend.The gravity centers of average vegetation NDVI during the study period was distributed in Zhenyuan County,Gansu Province,and the center moved northeastwards from 1981 to 2019.During 1981-2000 and 2001-2019,the proportion of vegetation restoration areas promoted by the combined action of climate change and human activities was the largest.During the study period(1981-2019),the dominant factors influencing vegetation NDVI shifted from natural factors to human activities.These results could provide decision support for the protection and restoration of vegetation ecosystem in the Yellow River Basin. | LIU Yifeng GUO Bing LU Miao ZANG Wenqian YU Tao CHEN Donghua | 2023 | Journal of Arid Land2023,15,1: | 2 |
| 7 | Hormones induce the formation of luminal-derived basal cells in the mammary gland显示文摘In the mammary gland, it is widely believed that the luminal cells are unipotent after birth, contributing only to the luminal compartment in normal development. Here, by lineage tracing, we uncovered an unexpected potential of luminal cells that can give rise to basal cells during pregnancy. These luminal-derived basal cells (LdBCs) persisted through mammary regression and gen erated more proge ny in successive rounds of preg nan cies. LdBCs express basal markers as well as estroge n receptor a (ERa). I n ovariectomized (OVX) mice, stimulation with estrogen and progesterone promoted the formation of LdBCs. In serial transplantation assays, LdBCs were able to rec on stitute new mammary glands in a hormone-depende nt manner. Tran scriptome an alysis and genetic experiments suggest that Wnt/p-catenin signaling is essential for the formation and maintenance of LdBCs. Our data uncover an unexpected bi-potency of luminal cells in a physiological context. | 8 | SHANK2 is a frequently amplified oncogene with evolutionarily conserved roles in regulating Hippo signaling显示文摘Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth.However,for a significant portion of human cancer,how Hippo signaling is perturbed remains unknown.To answer this question,we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome.In our screen,Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth.Interestingly,a mammalian homolog of Prosap,SHANK2,is the most frequently amplified gene on 11 q13,a major tumor amplicon in human cancer.Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification.More importantly,across the human cancer genome,SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon.Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator,and as a potential oncogene,SHANK2 promotes cellular transformation and tumor formation in vivo.In cancer cell lines with deregulated Hippo pathway,depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation.Taken together,these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator,commonly amplified in human cancer and potently promotes cancer.Our study for the first time illustrated oncogenic function of SHANK2,one of the most frequently amplified gene in human cancer.Furthermore,given that in normal adult tissues,SHANK2 s expression is largely restricted to the nervous system,SHANK2 may represent an interesting target for anticancer therapy. | Liang Xu Peixue Li Xue Hao Yi Lu Mingxian Liu Wenqian Song Lin Shan Jiao Yu Hongyu Ding Shishuang Chen Ailing Yang Yi Arial Zeng Lei Zhang Hai Jiang | 2021 | Protein & Cell2021,12,3: | 1 |

| 10 | Characterization of three Arabidopsis AP2/EREBP family transcription factors involved in ABA sensitivity, freeze and salt tolerance显示文摘AP2/EREBP transcription factors (TFs) play very important roles in plant development, hormonal regulation and stress response. Upon genome-wide cDNA cloning, phylogenetic and expression pattern analyses of this plant specific TF family, we found that three of the members including At1g71450, At1g50680 and At5g13910, were likely involved in responses to ABA, cold and salt. Complementary DNAs containing putative full-length ORFs of these three TFs were obtained and fused individually to the GAL4 DNA-binding domains. All the 3 genes functioned effectively as trans-activators using yeast one-hybrid assays. RT-PCR experiments showed that the At1g71450 gene was induced by ABA and low temperature; the At1g50680 gene was responsive to quite a few stress conditions, but especially to freezing temperature; and the At5g13910 gene was induced by high salt treatment, drought and ethyl-ene. By searching the ABRC T-DNA insertion mutant stocks, we obtained knockout lines for these TFs. Homozygous ko1 (At1g71450) plants showed a hypersensitive response to ABA during seed germina-tion and also in stomata movement. Homozygous ko2 (At1g50680) plants showed a significant reduc-tion in plant freezing tolerance compared to the wild type after chilling treatment. Homozygous ko3 (At5g13910) were less tolerant to high salinity than wild type plants. | 11 | Serum from asthmatic rat treated with acupuncture inhibits acetyl-choline-induced contractile responses of airway smooth muscle cells显示文摘OBJECTIVE: To investigate the influence of serum from asthmatic rat treated with acupuncture(acupuncture serum) on the contractile responses of airway smooth muscle cells(ASMCs).METHODS: OVA-induced asthmatic rats were subject to acupuncture treatment at the points of Dazhui(GV 14), Fengmen(BL 12), and Feishu(BL 13).The resulting serum was collected, as well as serum from normal animals. Contraction of ASMCs was induced with 10 μM acetylcholine(Ach), and acupuncture serum or normal serum was supplemented 15 min later. The effects of the sera on the changes in cell length, width, and surface areawere monitored in real time via a live cell imaging system.RESULTS: The contraction rates of ASMCs 15 min and 30 min after Ach stimulation reached 38.2% ±3.8% and 49.7% ± 4.6%, both of which were significantly higher than unstimulated control cells(P <0.01). Acupuncture serum at the 1∶100 dosage reduced the contraction rate of ASMCs from 40.1% ±5.1% to 17.6% ± 6.5%(P < 0.01). Thus, the treatment significantly inhibited the Ach-induced contractile responses of ASMCs and yielded a relaxation efficiency of 58%, which was greater than the effects obtained with a 1∶250 dosage. | 14 | Single-atom Fe Embedded Co_(3)S_(4) for Efficient Electrocatalytic Oxygen Evolution Reaction显示文摘Constructing atomically dispersed active sites with densely exposed and dispersed double metal-Sx catalytic sites for favorable OER catalytic activity remains rare and challenging.Herein,we design and construct a Fe_(1)S_(x)@Co_(3)S_(4) electrocatalyst with Fe single atoms epitaxially confined in Co_(3)S_(4) nanosheets for catalyzing the sluggish alkaline oxygen evolution reaction(OER).Consequently,in ultralow concentration alkaline solutions(0.1 mol/L KOH),such a catalyst is highly active and robust for OER with low overpotentials of 300 and 333 mV at current densities of 10 and 30 mA/cm^(2),respectively,accompanying long-term stability without significant degradation even for 350 h.In addition,Fe_(1)S_(x)@Co_(3)S_(4) shows a turnover frequency(TOF)value of 0.18 s−1,nearly three times that of Co_(3)S_(4)(0.07 s−1),suggesting the higher atomic utilization of Fe single atoms.Mössbauer and in-situ Raman spectra confirm that the OER activity of Fe_(1)S_(x)@Co_(3)S_(4) origins from a thin catalytic layer of Co(Fe)OOH that interacts with trace-level Fe species in the electrolyte,creating dynamically stable active sites.Combined with experimental characterizations,it suggests that the most active S-coordinated dual-metal site configurations are 2S-bridged(Fe-Co)S4,in which Co-S and Fe-S moieties are shared with two S atoms,which can strongly regulate the adsorption energy of reaction intermediates,accelerating the OER reaction kinetics. | QI Yuxue LI Tingting HU Yajie XIANG Jiahong SHAO Wenqian CHEN Wenhua MU Xueqin LIU Suli CHEN Changyun YU Min MU Shichun | 2022 | Chemical Research in Chinese Universities2022,38,5: | 0 |

| 18 | A new media content trusted dissemination architecture based on AV-blockchain and ChinaDRM显示文摘The diffusion of all-media content plays a vital role in guiding public opinion and ideology.However,at present,most of the media content exists on all kinds of mainstream media platforms,which poses great challenges to the effective supervision of relevant departments and society.This has led to arbitrary charges,chaotic media content,difficulties in supervision and evidence collection,and infringements of the rights and interests of original content creators.To address these problems,this paper constructs a trustworthy propagation architecture that supports multi-platform media content sharing.This architecture collaboratively builds an audio-visual blockchain through public and consortium blockchains,coupled with an improved ChinaDRM to provide digital rights management and content encryption.Simultaneously,we employ an enhanced Diffie-Hellman key agreement protocol to offer distributed encryption and decryption for media content.Within this model,various media platforms and national regulatory authorities are responsible for content storage and distribution as consortium nodes and public blockchain nodes,respectively.At the same time,users,as light nodes of public chain or service consumers of consortium blockchain,can consume and comment on content.Analysis shows that the trusted communication framework of media content based on the audio-visual blockchain has certain expansibility and practicability.It can facilitate the supervision of mainstream media platforms by national authorities and society through inter-blockchain technology,offering a novel solution for multi-platform trustworthy cooperative information sharing. | Wenqian Shang Zaifu Yu | 2023 | Intelligent and Converged Networks2023,4,2: | 0 |

| 20 | Concise synthesis of NaTi_2(PO_4)_3 nanocrystals with size and morphology control显示文摘NaTi_2 (PO_4)_3 (NTP) nanocrystals with high room-temperature ionic conductivity of 1.1×10~3 S/cm were prepared by a concise solvothermal method at 140 ℃ for 3 h, and the aspect ratios of all the NTP nanocrystals are the closest to 0.7. It implies a moderate size-distribution of NTP nanocrystals obtained at 140 ℃ for 3 h is helpful for increasing packing density, and the packing density is the larger, so its conductivity is the higher. The controllability over size and morphology of the NTP nanocrystals via solvothermal temperature and time were investigated.
