2T cell–associated immunoregulation and antiviral effect of oxymatrine in hydrodynamic injection HBV mouse model显示文摘Although oxymatrine(OMT) has been shown to directly inhibit the replication of hepatitis B virus(HBV) in vitro, limited research has been done with this drug in vivo. In the present study, the antiviral effect of OMT was investigated in an immunocompetent mouse model of chronic HBV infection.The infection was achieved by tail vein injection of a large volume of DNA solution. OMT(2.2, 6.7 and20 mg/kg) was administered by daily intraperitoneal injection for 6 weeks. The efficacy of OMT was evaluated by the levels of HBV DNA, hepatitis B surface antigen(HBs Ag), hepatitis B e antigen(HBe Ag)and hepatitis B core antigen(HBc Ag). The immunoregulatory activity of OMT was evaluated by serum ELISA and flow cytometry. Results shows that OMT at 20 mg/kg inhibited HBV replication, and it was more efficient than entecavir(ETV) in the elimination of serum HBs Ag and intrahepatic HBc Ag. Inaddition, OMT accelerated the production of interferon-γ(IFN-γ) in a dose-dependent manner in CD4^+T cells. Our findings demonstrate the beneficial effects of OMT on the enhancement of immunological function and in the control of HBV antigens. 4Oxidative stress, nutritional antioxidants and beyond显示文摘Free radical-induced oxidative stress contributes to the development of metabolic syndromes(Mets), including overweight,hyperglycemia, insulin resistance and pro-inflammatory state. Most free radicals are generated from the mitochondrial electron transport chain;under physiological conditions, their levels are maintained by efficient antioxidant systems. A variety of transcription factors have been identified and characterized that control gene expression in response to oxidative stress status.Natural antioxidant compounds have been largely studied for their strong antioxidant capacities. 5Study on stabilities and electrochemical behavior of V(V) electrolyte with acid additives for vanadium redox flow battery显示文摘Several acid compounds have been employed as additives of the V(V) electrolyte for vanadium redox flow battery(VRB) to improve its stability and electrochemical activity. Stability of the V(V) electrolyte with and without additives was investigated with ex-situ heating/cooling treatment at a wide temperature range of-5 ?C to 60 ?C. It was observed that methanesulfonic acid, boric acid, hydrochloric acid, trifluoroacetic acid,polyacrylic acid, oxalic acid, methacrylic acid and phosphotungstic acid could improve the stability of the V(V) electrolyte at a certain range of temperature. Their electrochemical behaviors in the V(V) electrolyte were further studied by cyclic voltammetry(CV), steady state polarization and electrochemical impedance spectroscopy(EIS). 8LncRNA Nron regulates osteoclastogenesis during orthodontic bone resorption显示文摘Activation of osteoclasts during orthodontic tooth treatment is a prerequisite for alveolar bone resorption and tooth movement.However,the key regulatory molecules involved in osteoclastogenesis during this process remain unclear.Long noncoding RNAs(lnc RNAs)are a newly identified class of functional RNAs that regulate cellular processes,such as gene expression and translation regulation.Recently,lnc RNAs have been reported to be involved in osteogenesis and bone formation.However,as the most abundant noncoding RNAs in vivo,the potential regulatory role of lnc RNAs in osteoclast formation and bone resorption urgently needs to be clarified.We recently found that the lnc RNA Nron(long noncoding RNA repressor of the nuclear factor of activated T cells)is highly expressed in osteoclast precursors.Nron is downregulated during osteoclastogenesis and bone ageing.To further determine whether Nron regulates osteoclast activity during orthodontic treatment,osteoclastic Nron transgenic(Nron c TG)and osteoclastic knockout(Nron CKO)mouse models were generated.When Nron was overexpressed,the orthodontic tooth movement rate was reduced.In addition,the number of osteoclasts decreased,and the activity of osteoclasts was inhibited.Mechanistically,Nron controlled the maturation of osteoclasts by regulating NFATc1 nuclear translocation.In contrast,by deleting Nron specifically in osteoclasts,tooth movement speed increased in Nron CKO mice.These results indicate that lnc RNAs could be potential targets to regulate osteoclastogenesis and orthodontic tooth movement speed in the clinic in the future.Ruilin Zhang Junhui Li Gongchen Li Fujun Jin Zuolin Wang Rui Yue Yibin Wang Xiaogang Wang Yao Sun 2020International Journal of Oral Science2020,12,2:9
9Recent developments in electrocatalysts and future prospects for oxygen reduction reaction in polymer electrolyte membrane fuel cells显示文摘The main difficulty in the extensive commercial use of polymer electrolyte membrane fuel cells(PEMFCs)is the use of noble metals such as Pt-based electrocatalyst at the cathode, which is essential to ease the oxygen reduction reaction(ORR) in fuel cells(FCs). To eliminate the high loading of Pt-based electrocatalysts to minimize the cost, extensive study has been carried out over the previous decades on the non-noble metal catalysts. Development in enhancing the ORR performance of FCs is mainly due to the doped carbon materials, Fe and Co-based electrocatalysts, these materials could be considered as probable substitutes for Pt-based catalysts. But the stability of these non-noble metal electrocatalysts is low and the durability of these metals remains unclear. The three basic reasons of instability are:(i) oxidative occurrence by H_2O_2,(ii) leakage of the metal site and(iii) protonation by probable anion adsorption of the active site. Whereas leakage of the metal site has been almost solved, more work is required to understand and avoid losses from oxidative attack and protonation. The ORR performance such as stability tests are usually run at low current densities and the lifetime is much shorter than desired need. Therefore,improvement in the ORR activity and stability are the key issues of the non-noble metal electrocatalyst.Based on the consequences obtained in this area, numerous future research directions are projected and discussed in this paper. 11Comparative genomic and transcriptomic analyses uncover the molecular basis of high nitrogen-use efficiency in the wheat cultivar Kenong 9204显示文摘Studying the regulatory mechanisms that drive nitrogen-use efficiency(NUE)in crops is important for sustainable agriculture and environmental protection.In this study,we generated a high-quality genome assembly for the high-NUE wheat cultivar Kenong 9204 and systematically analyzed genes related to nitrogen uptake and metabolism.By comparative analyses,we found that the high-affinity nitrate transporter gene family had expanded in Triticeae.Further studies showed that subsequent functional differentiation endowed the expanded family members with saline inducibility,providing a genetic basis for improving the adaptability of wheat to nitrogen deficiency in various habitats.To explore the genetic and molecular mechanisms of high NUE,we compared genomic and transcriptomic data from the high-NUE cultivar Kenong 9204(KN9204)and the low-NUE cultivar Jing 411 and quantified their nitrogen accumulation under high-and low-nitrogen conditions.Compared with Jing 411,KN9204 absorbed significantly more nitrogen at the reproductive stage after shooting and accumulated it in the shoots and seeds.Transcriptome data analysis revealed that nitrogen deficiency clearly suppressed the expression of genes related to cell division in the young spike of Jing 411,whereas this suppression of gene expression was much lower in KN9204.In addition,KN9204 maintained relatively high expression of NPF genes for a longer time than Jing 411 during seed maturity.Physiological and transcriptome data revealed that KN9204 was more tolerant of nitrogen deficiency than Jing 411,especially at the reproductive stage.The high NUE of KN9204 is an integrated effect controlled at different levels.Taken together,our data provide new insights into the molecular mechanisms of NUE and important gene resources for improving wheat cultivars with a higher NUE trait.Xiaoli Shi Fa Cui Xinyin Han Yilin He Long Zhao Na Zhang Hao Zhang Haidong Zhu Zhexin Liu Bin Ma Shusong Zheng Wei Zhang Jiajia Liu Xiaoli Fan Yaoqi Si Shuiquan Tian Jianqing Niu Huilan Wu Xuemei Liu Zhuo Chen Deyuan Meng Xiaoyan Wang Liqiang Song Lijing Sun Jie Han Hui Zhao Jun Ji Zhiguo Wang Xiaoyu He Ruilin Li Xuebin Chi Chengzhi Liang Beifang Niu Jun Xiao Junming Li Hong-Qing Ling 2022Molecular Plant2022,15,9:6

14Clinical characteristics of 96 patients with drug-induced acute liver failure:a comparison between Traditional Chinese and Western Medicine显示文摘OBJECTIVE: To compare the causes, clinical symptoms, laboratory test results, and prognosis in patients with acute liver failure(ALF) induced by traditional Chinese medicines(TCM) and by Western Medicines(WM).METHODS: The medical histories of patients who were diagnosed with drug-induced ALF(DALF)(n =96) after hospitalization in the 302 Military Hospital between January 2010 and December 2015 were retrospectively examined.RESULTS: Fifty-eight of the 96 DALF patients(60.4%) had a hepatocellular pattern of DALF, 16 patients(16.7%) had a cholestatic pattern, and 22 patients(22.9%) had a mixed pattern. DALF resolved in 24 patients(25.0%). Twenty-five patients(26.0%)developed chronic liver injury, 43 patients(44.8%)died, and 4 patients(4.2%) underwent liver transplantation. There were 42 ALF patients(43.8%) who received WM, and 32 ALF patients(33.3%) who received TCM. TCM-induced ALF patients had a higher average age [42.4 ± 18.4) vs(33.5 ± 17.9) years,P = 0.04] and higher creatinine and urine nitrogen levels [(155.2 ± 108.8) vs(97.5 ± 130.4) mmol/L, P =0.047;(9.1 ± 7.7) vs(4.3 ± 5.0) mmol/L, P = 0.002, respectively]. Patients with TCM-induced ALF exhibited an increased risk of renal injury [odds ratio(OR),3.75; 95% confidence interval(CI), 1.330-10.577].The 14 patients with TCM-induced ALF who died exhibited higher creatinine levels than the 18 patients with TCM-induced ALF patients who survived[(218.7 ± 111.6) vs(105.8 ± 78.4) mmol/L, P =0.002]. 15Solvent effects on Pt-Ru/C catalyst for methanol electro-oxidation显示文摘Alloying degree,particle size and the level of dispersion are the key structural parameters of Pt-Ru/C catalyst in fuel cells.Solvent(s) used in the preparation process can affect the particle size and alloying degree of the object substance,which lead to a great positive impact on its properties.In this work,three types of solvents and their mixtures were used in preparation of the Pt-Ru/C catalysts by chemical reduction of metal precursors with sodium borohydride at room temperature.The structure of the catalysts was characterized by X-ray diffraction(XRD) and Transmission electron microscopy(TEM).The catalytic activity and stability for methanol electro-oxidation were studied by Cyclic Voltammetry(CV) and Chronoamperometry(CA).Pt-Ru/C catalyst prepared in H2O or binary solvents of H2O and isopropanol had large particle size and low alloying degree leading to low catalytic activity and less stability in methanol electro-oxidation.When tetrahydrofuran was added to the above solvent systems,Pt-Ru/C catalyst prepared had smaller particle size and higher alloying degree which resulted in better catalytic activity,lower onset and peak potentials,compared with the above catalysts.Moreover,the catalyst prepared in ternary solvents of isopropanol,water and tetrahydrofuran had the smallest particle size,and the high alloying degree and the dispersion kept unchanged.Therefore,this kind of catalyst showed the highest catalytic activity and good stability for methanol electro-oxidation.Jinwei Chen Chunping Jiang Hui Lu Lan Feng Xin Yang Liangqiong Li Ruilin Wang 2009Journal of Natural Gas Chemistry2009,18,3:2
16Multiplicity of solutions to geophysical inversion reflected by rupture slip distribution of the 2015 Nepal earthquake显示文摘The equivalence of geophysical fields,the finiteness of measurements and the measurement errors make the result of geophysical inversion non-unique.For example,the measurements and inversion method used,the priori rupture model determined and the slip distribution smoothing factor selected will have significant influences on the earthquake rupture slip distribution.Using different data and methods,different authors have given different rupture slip distribution models of the 2015 Mw7.9 Nepal earthquake,with the maximum slip ranging from 3.0 m to 6.8 m.In this paper,geometry parameters of the single rectangular fault model in elastic half-space were inferred constraining with the Global Positioning System(GPS) and Interferometric Synthetic Aperture Radar(InSAR) coseismic deformations and bounding the slip with approximate average value;and then,the single rectangular fault was divided into multiple sub-faults,and the final slip smoothing factor,the final slip distribution and the maximum slip were determined with the misfit-roughness tradeoff curve,the cross-validation sum of squares(CVSS) and the third-party observation data or indexes being comprehensively taken into account.The results show that,the rupture of the Nepal earthquake extended by over 100 km east by south.The maximum slip of the earthquake was about 6.5-6.7 m,and most of the slip is confined at depths of 8-20 km,consistent with the depth distribution of aftershocks.The method for reducing the multiplicity of solutions to rupture slip distribution in this paper was ever used in inversion of rupture slip distribution for the 2008 Wenchuan and 2013 Lushan earthquakes,and the third-party measurement-surface dislocation has very large effect on reducing the multiplicity of solutions to inversion of the Wenchuan earthquake.Other priori information or indicators,such as fault strike,dip,earthquake magnitude,seismic activity,Coulomb stress,and seismic period,can be used for beneficial validation of and comparison with inversion results.Kai Tan Caihong Zhang Bin Zhao Qi Wang Ruilin Du Rui Zhang Xuejun Qiao Yong Huang 2017Geodesy and Geodynamics2017,8,1:2

18Glycosylation of dentin matrix protein 1 is critical for fracture healing via promoting chondrogenesis显示文摘Fractures are frequently occurring diseases that endanger human health. Crucial to fracture healing is cartilage formation, which provides a bone-regeneration environment. Cartilage consists of both chondrocytes and extracellular matrix (ECM). The ECM of cartilage includes collagens and various types of proteoglycans (PGs), which play important roles in maintaining primary stability in fracture healing. The PG form of dentin matrix protein 1 (DMP1-PG) is involved in maintaining the health of articular cartilage and bone. Our previous data have shown that DMP1-PG is richly expressed in the cartilaginous calluses of fracture sites. However, the possible significant role of DMP1-PG in chondrogenesis and fracture healing is unknown. To further detect the potential role of DMP1-PG in fracture repair, we established a mouse fracture model by using a glycosylation site mutant DMP1 mouse (S89G-DMP1 mouse). Upon inspection, fewer cartilaginous calluses and down-regulated expression levels of chondrogenesis genes were observed in the fracture sites of S89G-DMP1 mice. Given the deficiency of DMP1-PG, the impaired IL-6/JAK/STAT signaling pathway was observed to affect the chondrogenesis of fracture healing.
