| 1 | Viral and Bacterial Etiology of Acute Febrile Respiratory Syndrome among Patients in Qinghai, China显示文摘Objective This study was conducted to investigate the viral and bacterial etiology and epidemiology of patients with acute febrile respiratory syndrome(AFRS) in Qinghai using a commercial routine multiplex-ligation-nucleic acid amplification test(NAT)-based assay. Methods A total of 445 nasopharyngeal swabs specimens from patients with AFRS were analyzed using the RespiFinderSmart22 kit(PathoFinder BV, Netherlands) and the LightCycler 480 real-time PCR system. Results Among the 225(225/445, 51%) positive specimens, 329 positive pathogens were detected, including 298(90.58%) viruses and 31(9%) bacteria. The most commonly detected pathogens were influenza virus(IFV;37.39%;123/329), adenovirus(AdV;17.02%;56/329), human coronaviruses(HCoVs;10.94%;36/329), rhinovirus/enterovirus(RV/EV;10.03%;33/329), parainfluenza viruses(PIVs;8.51%;28/329), and Mycoplasma pneumoniae(M. pneu;8.51%;28/329), respectively. Among the co-infected cases(17.53%;78/445), IFV/AdV and IFV/M. pneu were the most common co-infections. Most of the respiratory viruses were detected in summer and fall. Conclusion In our study, IFV-A was the most common respiratory pathogen among 22 detected pathogens, followed by AdV, HCoV, RV/EV, PIV, and M. pneu. Bacteria appeared less frequently than viruses, and co-infection was the most common phenomenon among viral pathogens. Pathogens were distributed among different age groups and respiratory viruses were generally active in July, September, and November. | 4 | Effect of traditional Chinese medicine compounds on rumen fermentation, methanogenesis and microbial flora in vitro显示文摘This study was conducted to investigate the effects of traditional Chinese medicine compounds(TCMC)on rumen fermentation, methane emission and populations of ruminal microbes using an in vitro gas production technique. Cablin patchouli herb(CPH), Atractylodes rhizome(AR), Amur Cork-tree(AC) and Cypsum were mixed with the weight ratios of 1:1:1:0.5 and 1:1:1:1 to make up TCMC1 and TCMC2,respectively. Both TCMC were added at level of 25 g/kg of substrate dry matter. In vitro gas production was recorded and methane concentration was determined at 12 and 24 h of incubation. After 24 h, the incubation was terminated and the inoculants were measured for pH, ammonia nitrogen, volatile fatty acids(VFA). Total deoxyribonucleic acid of ruminal microbes was extracted from the inocula, and populations were determined by a real-time quantitative polymerase chain reaction. Populations of total rumen methanogens, protozoa, total fungi, Ruminococcus albus, Fibrobacter succinogenes and Ruminococcus flavefaciens were expressed as a proportion of total rumen bacterial 16 S ribosomal deoxyribonucleic acid. Compared with the control, the 2 TCMC decreased(P < 0.05) total VFA concentration,acetate molar proportion, acetate to propionate ratio, gas and methane productions at 12 and 24 h,hydrogen(H) produced and consumed, and methanogens and total fungi populations, while the 2 TCMC increased(P < 0.05) propionate molar proportion. Traditional Chinese medicine compound 1 also decreased(P ≤ 0.05) R. flavefaciens population. From the present study, it is inferred that there is an effect of the TCMC in suppressing methanogenesis, probably mediated via indirect mode by channeling H2 utilized for methanogenesis to synthesis of propionate and direct action against the rumen microbes involved in methane formation. | 6 | A parameterization scheme for upward lightning in the cloud model and a discussion of the initial favorable environmental characteristics in the cloud显示文摘The upward lightning(UL) initiated from the top of tall buildings(at least above 100 m) is a type of atmospheric discharge. Currently, we understand the nature of the UL from ground observations, but the corresponding theoretical research is lacking. Based on an existing bidirectional leader stochastic model, a stochastic parameterization scheme for the UL has been built and embedded in an existing two-dimensional thundercloud charge/discharge model. The ULs simulated from the experiments with two-dimensional high resolution agree generally with the observation results. By analyzing the charge structure of thunderstorm clouds, we determined the in-cloud environmental characteristics that favor the initiation of conventional cloud-to-ground(CG) flashes and analyzed the differences and similarities of some characteristics of the positive and the negative UL. Simulation results indicate that the positive ULs are typically other-lightning-triggered ULs(OLTUL) and are usually a discharge phenomenon between the ground and the lower positive charge region appearing below the main middle negative charge region. The effect of the previous in-cloud lightning(IC) process of space electrical field provides favorable conditions for the initiation of a positive UL. Its entire discharge process is limited, and the branches of the leader are fewer in number as its discharge is not sufficient. A negative UL is generally a discharge phenomenon of the dipole charge structure between the ground and the main negative charge region. The lower temperature stratification and the sinking of the hydrometeors typically initiate a negative UL. Negative ULs develop strongly and have more branches. Methods A two-tube MCMRT-PCR assay was performed on 7900 HT Fast Real-Time PCR System(Applied Biosystems, USA). Amplification by PCR was optimized to obtain high efficiency. The sensitivity and specificity of assays were investigated. Results The detection limit of optimized MCMRT-PCR assay was 3.9×102 CFU/mL for S. aureus, 4.4×102 CFU/mL for L. monocytogenes, 3.0×102 CFU/mL for Salmonella, 2.5×102 CFU/mL for Shigella, 2.1×102 CFU/mL for V. parahaemolyticus, and 1.2×102 CFU/mL for E. coli. The feasibility of MCMRT-PCR was further evaluated using artificially contaminated milk, the sensitivity was at the level of 105 CFU/mL. Conclusion A two-tube MCMRT-PCR assay using six primer sets was developed for detection of multiple pathogens. | 10 | Measurement of the neutron total cross section of carbon at the Back-n white neutron beam of CSNS显示文摘To verify the performance of the neutron total cross-sectional spectrometer, the neutron total cross section of carbon is initially measured in the energy range of 1 eV to 20 MeV using the time-of-flight method. The measurement is performed at the Back-n white neutron source with a 76-m time-of-flight path using the China Spallation Neutron Source. A multilayer fast fission chamber with 235U and 238U is employed as the neutron detector. The diameter and thickness of the natural graphite sample are 70 mm and 40 mm, respectively. Signal waveforms are collected using a data acquisition system. Off-line data processing was used to obtain the neutron time-of-flight spectra and transmissions. The uncertainty of the counting statistics is generally approximately 3% for each bin in the energy range of 1–20 MeV. It is determined that the results for the neutron total cross section of carbon obtained using ^235U cells are in good agreement with the results obtained using 238U cells within limits of statistical uncertainty. With decreasing crystallite size, the magnetic transition becomes increasingly slow, accompanied by broadening of the negative thermal expansion (NTE) operation-temperature window. The NTE operation-temperature window for the 12-nm crystallite sample reaches at 140 K, which is about 75% larger than that of the 74-nm crystallite sample. The magnetic properties and NTE operation-temperature window can be tuned by varying the crystallite size. | 14 | Dominant changes in the breast muscle lipid profiles of broiler chickens with wooden breast syndrome revealed by lipidomics analyses显示文摘Background: Chicken is the most consumed meat worldwide and the industry has been facing challenging myopathies. Wooden breast(WB), which is often accompanied by white striping(WS), is a serious myopathy adversely affecting meat quality of breast muscles. The underlying lipid metabolic mechanism of WB affected broilers is not fully understood.Results: A total of 150 chickens of a white-feathered, fast-growing pure line were raised and used for the selection of WB, WB + WS and control chickens. The lipids of the breast muscle, liver, and serum from different chickens were extracted and measured using ultra performance liquid chromatography(UPLC) plus Q-Exactive Orbitrap tandem mass spectrometry. In the breast, 560 lipid molecules were identified. Compared to controls, 225/225 of 560 lipid molecules(40.2%) were identified with differential abundance(DA), including 92/100 significantly increased neutral lipids and 107/98 decreased phospholipids in the WB/WB + WS groups, respectively. The content of monounsaturated fatty acids(MUFA) was significantly higher, and the polyunsaturated fatty acids(PUFA) and saturated fatty acids(SFA) were significantly lower in the affected breasts. In the liver, 434 lipid molecules were identified, and 39/61 DA lipid molecules(6.7%/14.1%) were detected in the WB and WB + WS groups, respectively. In the serum, a total of 529 lipid molecules were identified and 4/44 DA lipid molecules(0.8%/8.3%) were detected in WB and WB + WS group, respectively. Compared to controls, the content of MUFAs in the serum and breast of the WB + WS group were both significantly increased, and the content of SFAs in two tissues were both significantly decreased. Only five lipid molecules were consistently increased in both liver and serum in WB + WS group.Conclusions: We have found for the first time that the dominant lipid profile alterations occurred in the affected breast muscle. The relative abundance of 40.2% of lipid molecules were changed and is characteristic of increased neutral lipids and decreased phospholipids in the affected breasts. Minor changes of lipid profiles in the liver and serum of the affected groups were founded.
