2First dark matter search results from the PandaX-Ⅰ experiment显示文摘We report on the first dark-matter(DM)search results from PandaX-I,a low threshold dual-phase xenon experiment operating at the China JinPing Underground Laboratory.In the 37-kg liquid xenon target with 17.4 live-days of exposure,no DM particle candidate event was found.This result sets a stringent limit for low-mass DM particles and disfavors the interpretation of previously-reported positive experimental results.The minimum upper limit,3.7×10-44cm2,for the spin-independent isoscalar DM-particle-nucleon scattering cross section is obtained at a DM-particle mass of 49 GeV/c2at 90%confidence level.XIAO MengJiao XIAO Xiang ZHAO Li CAO XiGuang CHEN Xun CHEN YunHua CUI XiangYi FANG DeQing FU ChangBo GIBONI Karl L. 4Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 5Super-assembled core-shell mesoporous silica-metal-phenolic network nanoparticles for combinatorial photothermal therapy and chemotherapy显示文摘Multimodal combinatorial therapy merges different modes of therapies in one platform,which can overcome several clinical challenges such as premature drug loss during blood circulation and significantly improve treatment efficiency.Here we report a combinatorial therapy nanoplatform that enables dual photothermal therapy and pH-stimulus-responsive chemotherapy.By super-assembly of mesoporous silica nanoparticles(MSN)with metal-phenolic networks(MPN),anti-cancer drugs can be loaded in the MSN matrix,while the outer MPN coating allows dual photothermal and pH-responsive properties.Upon near-infrared light irradiation,the MSN@MPN nanoplatform exhibits excellent photothermal effect,and demonstrates outstanding pH-triggered drug release property.In vitro cell experiments suggest the MSN@MPN system exhibits superior biocompatibility and can effectively kill tumor cells after loading anti-cancer drugs.Consequently,the MSN@MPN system shows promising prospects in clinical application for tumor therapy.Bo Yang Shan Zhou Jie Zeng Liping Zhang Runhao Zhang Kang Liang Lei Xie Bing Shao Shaoli Song Gang Huang Dongyuan Zhao Pu Chen Biao Kong 2020Nano Research2020,13,4:3

9DNA demethylases remodel DNA methylation in rice gametes and zygote and are required for reproduction显示文摘Fertilization constitutes a critical step in the plant life cycle during which the gamete genomes undergo chromatin dynamics in preparation for embryogenesis. In mammals, parental chromatin is extensively reprogrammed through the global erasure of DNA methylation. However, in flowering plants it remains unclear whether and how DNA methylation is remodeled in gametes and after fertilization in the zygote. In this study, we characterize DNA methylation patterns and investigate the function of DNA glycosylases in rice eggs, sperm, and unicellular zygotes and during embryogenesis. We found that DNA methylation is locally reconfigured after fertilization and is intensified during embryogenesis. Genetic, epigenomic, and transcriptomic analysis revealed that three rice DNA glycosylases, DNG702, DNG701, and DNG704, demethylate DNA at distinct genomic regions in the gametes and the zygote, and are required for zygotic gene expression and development. 12Evolutionarily conservative and non-conservative regulatory networks during primate interneuron development revealed by single-cell RNA and ATAC sequencing显示文摘The differences in size and function between primate and rodent brains,and the association of disturbed excitatory/inhibitory balance with many neurodevelopmental disorders highlight the importance to study primate ganglionic eminences(GEs)development.Here we used single-cell RNA and ATAC sequencing to characterize the emergence of cell diversity in monkey and human GEs where most striatal and cortical interneurons are generated.We identified regional and temporal diversity among progenitor cells which give rise to a variety of interneurons.These cells are specified within the primate GEs by well conserved gene regulatory networks,similar to those identified in mice.However,we detected,in human,several novel regulatory pathways or factors involved in the specification and migration of interneurons.Importantly,comparison of progenitors between our human and published mouse GE datasets led to the discovery and confirmation of outer radial glial cells in GEs in human cortex.Our findings reveal both evolutionarily conservative and nonconservative regulatory networks in primate GEs,which may contribute to their larger brain sizes and more complex neural networks compared with mouse.Ziqi Zhao Dan Zhang Fuqiang Yang Mingrui Xu Shaoli Zhao Taotao Pan Chuanyu Liu Yongjie Liu Qingfeng Wu Qiang Tu Ping Zhou Rong Li Jia Kang Lan Zhu Fei Gao Yaqing Wang Zhiheng Xu 2022Cell Research2022,32,5:1

14Clinical Epidemiology,Illness Profiles,and the Implication of COVID-19 Before and After the Nationwide Omicron Outbreak During the Winter of 2022显示文摘China.In this study,we aimed to investigate the clinical characteristics and outcomes of patients with coronavirus disease 2019(COVID-19)in the Beijing region.Methods:In this retrospective study,we enrolled inpatients admitted for COVID-19 in the Fifth Medical Center of Chinese PLA General Hospital in Beijing between November 10,2022,and January 30,2023.Demographic and clinical features and treatment outcomeswere comprehensively analyzed.We used logistic regression and linear regression analyses to explore the risk factors associated with disease severity and time of nucleic acid conversion,respectively.Results:A total of 1010 hospitalized patients with COVID-19 were enrolled.The median age was 43.0 years(interquartile range,28.0–63.0),and patients aged<60 years and≥60 years comprised 71.2%and 28.8%of total included patients,respectively.The clinical classification of mild(74.6%,753/1010),moderate(21.0%,212/1010),severe(2.7%,27/1010),and unidentified(1.8%,18/1010)was separately recorded;1005 patients were discharged,and 5 patients died in the hospital.The outbreak of the emerging epidemic witnessed an evident increase in the proportion of moderate(42.9%vs.16.4%)and severe(10.3%vs.1.1%)cases after December 7,2022.Patients with a moderate/severe classification had higher levels of procalcitonin,IL-6,serum ferritin,C-reactive protein,lactic dehydrogenase,serum urea nitrogen,and D-dimer and lower counts of CD4+T,CD8+T,and B cells(all P<0.001).Multivariable regression analysis revealed that increased odds of disease severity were associated with the following factors:age≥60 years,IL-6>7 pg/mL,lactic dehydrogenase level>245 U/L,cough,and fever at admission.Age≥80 years and chronic lung diseasewere independent risk factors in the nonmild group in elderly patients.In addition,the duration for nucleic acid to turn negativewas approximately 5.0 d(interquartile range,3.0–7.0).Prolonged time of nucleic acid conversion was associated with age≥60 years,serum urea nitrogen level>8.2 mmol/L,neutrophil count>7×10^(9)/L,and the presence of a chronic lung disease or carcinoma.Finally,unvaccinated patients accounted for 37.3%of enrolled patients;children and the elder people accounted for approximately half of that.The univariable analysis found that booster doses reduced disease severity and shortened the time of nucleic acid conversion in elderly patients.Conclusions:The outbreak ofOmicron rapidly increased the number of patientswith COVID-19 in Beijing.In elderly patients,booster doses may reduce disease severity and shorten the time of nucleic acid conversion.Healthcare systems should be optimized before an emerging epidemic outbreak.Yuming Guo Zhe Xu Wen-Xin Wang Cheng Zhen Jinhua Hu Jinsong Mu Chengcheng Ji Xin Yuan Ruonan Xu Lei Huang Lei Shi Fanping Meng Junliang Fu Shuangnan Zhou Siyu Wang Fengyi Li Bo Tu Dawei Zhang Huihuang Huang Yufeng Mao Wen Xu Chao Zhang Xiuying Mu Jun Zhao Bo Jin Haibin Su Yinying Lu Yongqian Cheng Dong Ji Shaoli You Jinghui Dong Changchun Liu Mengmeng Zhang Yuanyuan Li Tianjun Jiang Yonggang Li Fu-Sheng Wang 2023Infectious Diseases & Immunity2023,3,4:0
15Morphology modulation of artificial muscles by thermodynamic-twist coupling显示文摘Human muscles can grow and change their length with body development;therefore,artificial muscles that modulate their morphology according to changing needs are needed.In this paper,we report a strategy to transform an artificial muscle into a new muscle with a different morphology by thermodynamic-twist coupling,and illustrate its structural evolution during actuation.The muscle length can be continuously modulated over a large temperature range,and actuation occurs by continuously changing the temperature.This strategy is applicable to different actuation modes,including tensile elongation,tensile contraction and torsional rotation.This is realized by twist insertion into a fibre to produce torsional stress.Fibre anne aling causes partial thermodynamic relaxation of the spiral molecular chains,which serves as internal tethering and inhibits fibre twist release,thus producing a self-supporting artificial muscle that actuates under heating.At a sufficiently high temperature,further relaxation of the spiral molecular chains occurs,resulting in a new mus cle with a different length.A structural study provides an understanding of the thermo dynamic-twist coupling.This work provides a new design strategy for intelligent materials.Xiaoyu Hu Jiatian Li Sitong Li Guanghao Zhang Run Wang Zhongsheng Liu Mengmeng Chen Wenqian He Kaiqing Yu Wenzhong Zhai Weiqiang Zhao Abdul Qadeer Khan Shaoli Fang Ray HBaughman Xiang Zhou Zunfeng Liu 2023National Science Review2023,10,1:0
