| 1 | Long-term follow-up of the effects of fecal microbiota transplantation in combination with soluble dietary fiber as a therapeutic regimen in slow transit constipation显示文摘As some studies have reported that strategies targeting the gut microbiota such as fecal microbiota transplantation(FMT) with or without other microecological therapy might have efficacy in treating slow transit constipation(STC), we conducted a singlecenter, open-label trial to study the long-term effect of FMT combined with soluble dietary fiber(pectin) on STC. Thirty-one adult patients with STC were enrolled into the trial. Patients received 6-day FMT procedures repeatedly for the first 3 months and soluble dietary fiber(pectin) daily during the follow-up. The rate of clinical remission and improvement, stool consistency, the Wexner constipation scale, and assessment of constipation-related symptoms were evaluated at week 4 and 1 year later. The clinical remission and improvement rates at week 4 were 69.0%(20/29) and 75.9%(22/29), respectively. At the end of the study,48.3%(14/29) of patients continued to have at least three complete spontaneous bowel movements per week and 58.6%(17/29)of patients showed clinical improvements. Stool consistency, the Wexner constipation scale, and constipation symptoms improved both at short-term and long-term follow-up. | 2 | Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. | 3 | Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. | 10 | Significance of ultrasound evaluation of carotid atherosclerotic plaque for diagnosing ischemic cerebrovascular disease显示文摘BACKGROUND: Carotid artery is the main source for craniocerebral blood supply. Its intimal plaque formation and arterial stenosis degree both are the risk factors for ischemic cerebrovascular disease. Therefore, the close relationship of carotid atherosclerotic plaque and ischemic cerebrovascular disease, and ultrasound evaluation of carotid atherosclerotic plaque have become the hot spot in studying ischemic cerebrovascular disease. OBJECTIVE: This study was to detect the degree of carotid atherosclerosis of ischemic cerebrovascular disease patients by ultrasonography, and to analyze the situation of carotid atherosclerosis and its relationship with clinic. DESIGN: Clinical randomized concurrent control experiment. SETTING: Lintong Convalescent Hospital of Lanzhou Military Area Command of Chinese PLA. PARTICIPANTS: Totally 60 outpatients and inpatients with ischemic cerebrovascular disease, 42 males and 18 females, admitted to Lintong Convalescent Hospital of Lanzhou Military Area Command of Chinese PLA between January 2006 and December 2006 were involved in the patient group. They met the diagnosis criteria of ischemic cerebrovascular disease constituted by the 4th Cerebrovascular Disease Conference in 1996, and were confirmed to suffer from ischemic cerebrovascular disease by skull CT and MRI. Another 20 subjects who received healthy examination concurrently in the same hospital, 12 males and 8 females, were involved in the control group. Informed consents of detected items were obtained from involved subjects. METHODS: The plaque thickness of mid portion, distal end and crotch of common carotid artery (CCA), internal carotid artery (ICA), external carotid artery (ECA) and vertebral artery (VA) of involved subjects, who received health examination was separately detected with color Doppler ultrasonograph (HDI-5000). Then, total integral of plaque was calculated. The intima-media thickness (IMT) was measured with two-dimensional ultrasonography. The inner diameter stenosis degree of subjects who had plaque was measured. Blood flow parameters were recorded, and stenosis degree and plaque area were calculated. Blood flow volume of bilateral carotid artery and VA was separately measured with ultrasound equipment software, and brain blood flow volume was calculated. MAIN OUTCOME MEASURES: Atherosclerotic degree and blood flow volume of patients of two groups. RESULTS: Sixty patients with ischemic cerebrovascular disease and twenty subjects who received health examination participated in the final analysis. ①The IMT thickness, total plaque score, and total plaque area of patient group was significantly superior to that of control group, respectively(t=5.216–10.158,P < 0.05). ② There were significant differences in the stenosis degree of CCA, ICA and VA between patient group and control group (t=6.720–12.816,P < 0.05). ③ The blood flow volume of CCA, ICA, VA and brain of patient group was significantly lower than that of control group, respectively (t=2.872–10.860, P < 0.05). | 11 | Cross-cultural adaption and psychometric properties of the Chinese version of the Diabetes Behavior Rating Scale: a pilot study显示文摘Self-care behavior plays a major role in diabetic management. However, in China, a satisfactory instrument has not yet been developed to evaluate the compliance of self-care behavior for young patients with type 1 diabetes mellitus(T1 DM). The Diabetes Behavior Rating Scale(DBRS) has a potential to be the first mature instrument. The purpose of this study is to cross-culturally adapt the DBRS, and preliminarily evaluate its psychometric properties. The instrument translation included translation, back translation and culture adaptation. Psychometric properties were assessed in a sample of 116 young patients with T1 DM adapting insulin injection therapy. The Chinese version of the DBRS was divided to four subscales. Cronbach's α for the total scale was 0.92. The mean inter-item and item-total correlations were 0.35 and 0.54 respectively. Test-retest reliability showed good temporal stability(r=0.81, P=0.001). Negative correlations were found between DBRS scores with the Diabetes Distress Scale scores(r=.0.32, P=0.003) and hemoglobin A1 c(HbA1 c) levels(r=.0.36, P=0.002). Higher DBRS scores correlated with better glycemic control. The Chinese insulin injection therapy version of the DBRS is well translated and culturally adapted. A thin homogeneous elastic interphase with different shear and bulk moduli, located between the reinforced particle and the host matrix, was introduced to model the interfacial bonding state. The effects of such an interphase on the coherent plane waves were studied numerically. Numerical simulations were carried out for SiC-Al composites with four typical cases of interphase. It was found that the property of interphase has significant influences on the effective propagation constants of coherent waves and the dynamic effective elastic moduli of the composites. The influences on the coherent longitudinal wave and the coherent shear waves were different and dependent upon the frequency range. Moreover, several imperfect interface models, i.e., the spring model, mass model, and spring-mass model, were studied numerically and compared with the interphase model. | 17 | Effects of Snow Cover on Ground Thermal Regime: A Case Study in Heilongjiang Province of China显示文摘The important effects of snow cover to ground thermal regime has received much attention of scholars during the past few decades. In the most of previous research, the effects were usually evaluated through the numerical models and many important results are found. However, less examples and insufficient data based on field measurements are available to show natural cases. In the present work, a typical case study in Mohe and Beijicun meteorological stations, which both are located in the most northern tip of China, is given to show the effects of snow cover on the ground thermal regime. The spatial(the ground profile) and time series analysis in the extremely snowy winter of 2012–2013 in Heilongjiang Province are also performed by contrast with those in the winter of 2011–2012 based on the measured data collected by 63 meteorological stations. Our results illustrate the positive(warmer) effect of snow cover on the ground temperature(GT) on the daily basis, the highest difference between GT and daily mean air temperature(DGAT) is as high as 32.35℃. Moreover, by the lag time analysis method it is found that the response time of GT from 0 cm to 20 cm ground depth to the alternate change of snow depth has 10 days lag, while at 40 cm depth the response of DGAT is not significant. | 20 | Effects of different forms of nitrogen addition on microbial extracellular enzyme activity in temperate grassland soil显示文摘Background:Nitrogen(N)deposition alters litter decomposition and soil carbon(C)sequestration by influencing the microbial community and its enzyme activity.Natural atmospheric N deposition comprises of inorganic N(IN)and organic N(ON)compounds.However,most studies have focused on IN and its effect on soil C cycling,whereas the effect of ON on microbial enzyme activity is poorly understood.Here we studied the effects of different forms of externally supplied N on soil enzyme activities related to decomposition in a temperate steppe.Ammonium nitrate was chosen as IN source,whereas urea and glycine were chosen as ON sources.Different ratios of IN to ON(Control,10:0,7:3,5:5,3:7,and 0:10)were mixed with equal total amounts of N and then used to fertilize the grassland soils for 6 years.Results:Our results show that IN deposition inhibited lignin-degrading enzyme activity,such as phenol oxidase(POX)and peroxidase(PER),which may restrain decomposition and thus induce accumulation of recalcitrant organic C in grassland soils.By contrast,deposition of ON and mixed ON and IN enhanced most of the C-degrading enzyme activities,which may promote the organic matter decomposition in grassland soils.In addition,theβ-N-acetyl-glucosaminidase(NAG)activity was remarkably stimulated by fertilization with both IN and ON,maybe because of the elevated N availability and the lack of N limitation after long-term N fertilization at the grassland site.Meanwhile,differences in soil pH,soil dissolved organic carbon(DOC),and microbial biomass partially explained the differential effects on soil enzyme activity under different forms of N treatments.Conclusions:Our results emphasize the importance of organic N deposition in controlling soil processes,which are regulated by microbial enzyme activities,and may consequently change the ecological effect of N deposition.Thus,more ON deposition may promote the decomposition of soil organic matter thus converting C sequestration in grassland soils into a C source. | Lili Dong Björn Berg Weiping Gu Zhengwen Wang Tao Sun | 2022 | Ecological Processes2022,11,1: | 1 |
