1Regulation of HMGB1 release by infl ammasomes显示文摘High mobility group box 1(HMGB1)is an evolutionarily conserved non-histone chromatin-binding protein.During infection or injury,activated immune cells and damaged cells release HMGB1 into the extracellular space,where HMGB1 functions as a proinflammatory mediator and contributes importantly to the pathogenesis of inflammatory diseases.Recent studies reveal that infl ammasomes,intracellular protein complexes,critically regulate HMGB1 release from activated immune cells in response to a variety of exogenous and endogenous danger signals.Double stranded RNA dependent kinase(PKR),an intracellular danger-sensing molecule,physically interacts with inflammasome components and is important for infl ammasome activation and HMGB1 release.Together,these studies not only unravel novel mechanisms of HMGB1 release during infl ammation,but also provide potential therapeutic targets to treat HMGB1-related infl ammatory diseases.Ben Lu Haichao Wang Ulf Andersson Kevin J.Tracey 2013Protein & Cell2013,4,3:21
2High Performance In Vivo Near-IR (>1 μm) Imaging and Photothermal Cancer Therapy with Carbon Nanotubes显示文摘Short single-walled carbon nanotubes(SWNTs)functionalized by PEGylated phospholipids are biologically non-toxic and long-circulating nanomaterials with intrinsic near infrared photoluminescence(NIR PL),characteristic Raman spectra,and strong optical absorbance in the near infrared(NIR).This work demonstrates the first dual application of intravenously injected SWNTs as photoluminescent agents for in vivo tumor imaging in the 1.0-1.4μm emission region and as NIR absorbers and heaters at 808 nm for photothermal tumor elimination at the lowest injected dose(70μg of SWNT/mouse,equivalent to 3.6 mg/kg)and laser irradiation power(0.6 W/cm2)reported to date.Ex vivo resonance Raman imaging revealed the SWNT distribution within tumors at a high spatial resolution.Complete tumor elimination was achieved for large numbers of photothermally treated mice without any toxic side effects after more than six months post-treatment.Further,side-by-side experiments were carried out to compare the performance of SWNTs and gold nanorods(AuNRs)at an injected dose of 700μg of AuNR/mouse(equivalent to 35 mg/kg)in NIR photothermal ablation of tumors in vivo.Highly effective tumor elimination with SWNTs was achieved at 10 times lower injected doses and lower irradiation powers than for AuNRs.These results suggest there are significant benefits of utilizing the intrinsic properties of biocompatible SWNTs for combined cancer imaging and therapy.Joshua T.Robinson Kevin Welsher Scott M.Tabakman Sarah P.Sherlock Hailiang Wang Richard Luong Hongjie Dai 2010Nano Research2010,3,11:20
3Food safety regulatory systems in Europe and China:A study of how co-regulation can improve regulatory effectiveness显示文摘Food safety has received a great deal of attention in both developed and developing countries in recent years. In China,the numerous food scandals and scares that have struck over the past decade have spurred significant food safety regulatory reform,which has been increasingly oriented towards the public-private partnership model adopted by the Europe Union's(EU) food safety regulatory system. This paper analyzes the development of both the EU's and China's food safety regulatory systems,identifies the current challenges for China and additionally considers the role of public-private partnership. The success of co-regulation in the food regulatory system would bring significant benefits and opportunities for China. 5TREM-1 multimerization is essential for its activation on monocytes and neutrophils显示文摘The triggering receptor expressed on myeloid cells-1(TREM-1)is a receptor expressed on innate immune cells.By promoting the amplification of inflammatory signals that are initially triggered by Toll-like receptors(TLRs),TREM-1 has been characterized as a major player in the pathophysiology of acute and chronic inflammatory diseases,such as septic shock,myocardial infarction,atherosclerosis,and inflammatory bowel diseases.However,the molecular events leading to the activation of TREM-1 in innate immune cells remain unknown.Here,we show that TREM-1 is activated by multimerization and that the levels of intracellular Ca 2+release,reactive oxygen species,and cytokine production correlate with the degree of TREM-1 aggregation.TREM-1 activation on primary human monocytes by LPS required a two-step process consisting of upregulation followed by clustering of TREM-1 at the cell surface,in contrast to primary human neutrophils,where LPS induced a rapid cell membrane reorganization of TREM-1,which confirmed that TREM-1 is regulated differently in primary human neutrophils and monocytes.In addition,we show that the ectodomain of TREM-1 is able to homooligomerize in a concentration-dependent manner,which suggests that the clustering of TREM-1 on the membrane promotes its oligomerization.We further show that the adapter protein DAP12 stabilizes TREM-1 surface expression and multimerization.TREM-1 multimerization at the cell surface is also mediated by its endogenous ligand,a conclusion supported by the ability of the TREM-1 inhibitor LR12 to limit TREM-1 multimerization.These results provide evidence for ligand-induced,receptor-mediated dimerization of TREM-1.Collectively,our findings uncover the mechanisms necessary for TREM-1 activation in monocytes and neutrophils.Kevin Carrasco Amir Boufenzer Lucie Jolly Helene Le Cordier Guanbo Wang Albert JR Heck Adelheid Cerwenka Emilie Vinolo Alexis Nazabal Alexandre Kriznik Pierre Launay Sebastien Gibot Marc Derive 2019Cellular & Molecular Immunology2019,16,5:13
6Calcium signaling and T-type calcium channels in cancer cell cycling显示文摘Regulation of intracellular calcium is an important signaling mechanism for cell proliferation in both normal and cancerous cells. In normal epithelial cells, free calcium concentration is essential for cells to enter and accomplish the S phase and the M phase of the cell cycle. In contrast, cancerous cells can pass these phases of the cell cycle with much lower cytoplasmic free calcium concentrations, indicating an alternative mechanism has developed for fulfilling the intracellular calcium requirement for an increased rate of DNA synthesis and mitosis of fast replicating cancerous cells. The detailed mechanism underlying the altered calcium loading pathway remains unclear; however, there is a growing body of evidence that suggests the T-type Ca2+ channel is abnormally expressed in cancerous cells and that blockade of these channels may reduce cell proliferation in addition to inducing apoptosis. Recent studies also show that the expression of T-type Ca2+ channels in breast cancer cells is proliferation state dependent, i.e. the channels are expressed at higher levels during the fast-replication period, and once the cells are in a non-proliferation state, expression of this channel isminimal. Therefore, selectively blocking calcium entry into cancerous cells may be a valuable approach for preventing tumor growth. 12A 60-year sedimentary record of natural and anthropogenic impacts on Lake Chenghai,China显示文摘Recent sediments from Lake Chenghai,China,were investigated at high temporal resolution to trace both natural and anthropogenic effects on the lake using total organic carbon(TOC),total nitrogen(TN),total phosphorus(TP),organic phosphorus(P o),inorganic phosphorus(P i) and organic carbon and nitrogen stable isotopes(δ 13 C org and δ 15 N) in a 137 Cs-dated sediment core.The results indicated that the sedimentary record covers the last 60 years,during which the lake had undergone apparent changes in nutrient sources and productivity in response to nutrient loading.Prior to the late 1980s,the nutrient contents in sediments mainly originated from algae and lake productivity was relatively stable.Since the late 1980s,increasing TOC,TN and TP concentrations together with the change of δ 13 C org and δ 15 N suggested anthropogenic perturbations in nutrient loading and lake productivity.Endogenic nutrients derived from algae and anthropogenic inputs were two important sources of sedimentary nutrients.The anthropogenic nutrients mainly originated from the discharge of industrial wastewater and artificial cultivation of Spirulina after the middle 1980s,and domestic wastewater discharged from Yongsheng County since 1993.Fengyu Zan Shouliang Huo Beidou Xi Jingtian Zhang Haiqing Liao Yue Wang Kevin M.Yeager 2012Journal of Environmental Sciences2012,24,4:8
13An Innovative Methodology of Product Design from Nature显示文摘Nature is an information sourcebook of behaviour, function, colour and shape which can inspire visual design and inven-tion. Studying the form and functional characteristics of a natural object can provide inspiration for product design and help to improve the marketability of manufactured products. The inspiration can be triggered either by direct observation or captured by three-dimensional (3D) digitising techniques to obtain superficial information (geometry and colour). An art designer often creates a concept in the form of a two-dimensional (2D) sketch while engineering methods lead to a point cloud in 3D. Each has its limitations in that the art designer commonly lacks the knowledge to build a final product from a 2D sketch and the engi-neering designer’s 3D point clouds may not be very beautiful. We propose a method for Product Design from Nature (PDN), coupling aesthetic intent and geometrical characteristics, exploring the interactions between designers and nature’s systems in PDN. 17Inferring the Tree of Life of the order Cypriniformes, the earth's most diverse clade of freshwater fishes: Implications of varied taxon and character sampling显示文摘The phylogenetic relationships of species are fundamental to any biological investigation, including all evolutionary studies. Accurate inferences of sister group relationships provide the researcher with an historical framework within which the attributes or geographic origin of species (or supraspecific groups) evolved. Taken out of this phylogenetic context, interpretations of evolutionary processes or origins, geographic distributions, or speciation rates and mechanisms, are subject to nothing less than a biological experiment without controls. Cypriniformes is the most diverse clade of freshwater fishes with estimates of diversity of nearly 3,500 species. These fishes display an amazing array of morphological, ecological, behavioral, and geographic diversity and offer a tremendous opportunity to enhance our understanding of the biotic and abiotic factors associated with diversification and adaptation to environments. Given the nearly global distribution of these fishes, they serve as an important model group for a plethora of biological investigations, including indicator species for future cli-matic changes. The occurrence of the zebrafish, Danio rerio, in this order makes this clade a critical component in understanding and predicting the relationship between mutagenesis and phenotypic expressions in vertebrates, including humans. With the tremendous diversity in Cypriniformes, our understanding of their phylogenetic relationships has not proceeded at an acceptable rate, despite a plethora of morphological and more recent mo-lecular studies. Most studies are pre-Hennigian in origin or include relatively small numbers of taxa. Given that analyses of small numbers of taxa for molecular characters can be compromised by peculiarities of long-branch attraction and nodal-density effect, it is critical that significant progress in our understanding of the relationships of these important fishes occurs with increasing sampling of species to mitigate these potential problems. The recent Cypriniformes Tree of Life initiative is an effort to achieve this goal with morphological and molecular (mitochondrial and nuclear) data. In this early synthesis of our understanding of the phylogenetic relationships of these fishes, all types of data have contributed historically to improving our understanding, but not all analyses are complementary in taxon sampling, thus precluding direct understanding of the impact of taxon sampling on achieving accurate phylogenetic inferences. However, recent molecular studies do provide some insight and in some instances taxon sampling can be implicated as a variable that can influence sister group relationships. Other instances may also exist but without inclusion of more taxa for both mitochondrial and nuclear genes, one cannot distinguish between inferences being dictated by taxon sampling or the origins of the molecular data.Richard L. 18Role of autophagy in differential sensitivity of hepatocarcinoma cells to sorafenib显示文摘AIM: To investigate the role of sorafenib(SFN) in autophagy of hepatocellular carcinoma(HCC). We evaluated how SFN affects autophagy signaling pathway in human HCC cell lines. METHODS: Two different human HCC cell lines, Hep3 B and Huh7, were subjected to different concentrations of SFN. Cell viability and onset of apoptosis were determined with colorimetric assay and immunoblotting analysis, respectively. The changes in autophagy-related proteins, including LC3, ULK1, AMPK, and LKB, were determined with immunoblotting analysis in the presence or absence of SFN. To assess autophagic dynamics, autophagic flux was measured with chloroquine, a lysosomal inhibitor. The autophagic responsiveness between different HCC cell lines was compared under the autophagy enhancing conditions.RESULTS: Hep3 B cells were significantly more resistant to SFN than Huh7 cells. Immunoblotting analysis revealed a marked increase in SFN-mediated autophagy flux in Huh7 cells, which was, however, absent in Hep3 B cells. While both starvation and rapamycin enhanced autophagy in Huh7 cells, only rapamycin increased autophagy in Hep3 B cells. Immunoblotting analysis of autophagy initiation proteins showed that SFN substantially increased phosphorylation of AMPK and consequently autophagy in Huh7, but not in Hep3 B cells.CONCLUSION: The autophagic responsiveness to SFN is distinct between Hep3 B and Huh7 cells. 20Phyllosticta species associated with citrus diseases in China显示文摘Phyllosticta species associated with diseases of four commercial Citrus species grown in China are reported.Totally,496 Phyllosticta strains were isolated from mandarins(Citrus reticulata),pomeloes(C.maxima),oranges(C.sinensis)and lemons(C.limon)in the main citrus producing regions across China,and 74 strains were selected for phylogenetic analysis.Analyses inferred from the sequences of internal transcribed spacer region(ITS1,5.8S nrDNA and ITS2),partial translation elongation factor 1-alpha(TEF1)and partial actin gene(ACT),showed these representative Phyllosticta isolates clustered in four distinct clades corresponding to three known,and one undescribed species.The newly resolved taxon,Phyllosticta citrichinaensis was isolated from leaves and fruits of all four Citrus species and is introduced in this paper.This taxon caused minor damage,showing irregular spots or freckles.Phyllosticta citriasiana,associated with tan spot of pomeloes,was isolated only from pomeloes,and never from lemons,mandarins and oranges.Phyllosticta citricarpa,the citrus black spot pathogen,which is presently subjected to phytosanitary legislation in the EU and United States,was isolated from lemons,mandarins and oranges,but never from pomeloes.The isolates of P.citricarpa clustered in two subclades,one from mandarins,the other from oranges and lemons.P.capitalensis was isolated from all four Citrus species as an endophyte,causing false melanose,or together with P.citricarpa or P.citriasiana.Morphological,cultural and biochemical characters were consistent with the results of phylogenetic analysis.In addition,a specific primer pair Pca8/ITS4 was designed and selected,and its corresponding PCR procedure was developed for the detection of P.citriasiana in this study.Xinghong Wang Guoqing Chen Feng Huang Jingze Zhang Kevin D.Hyde Hongye Li 2012Fungal Diversity2012,,1:5
