| 2 | Resequencing of 145 Landmark Cultivars Reveals Asymmetric Sub-genome Selection and Strong Founder Genotype Effects on Wheat Breeding in China显示文摘Controlled pedigrees and the multi-decade timescale of national crop plant breeding programs offer a unique experimental context for examining how selection affects plant genomes.More than 3000 wheat cultivars have been registered,released,and documented since 1949 in China.In this study,a set of 145 elite cultivars selected from historical points of wheat breeding in China were re-sequenced.A total of 43.75 Tb of sequence data were generated with an average read depth of 17.94x for each cultivar,and more than 60.92 million SNPs and 2.54 million InDels were captured,based on the Chinese Spring RefSeq genome v1.0.Seventy years of breeder-driven selection led to dramatic changes in grain yield and related phenotypes,with distinct genomic regions and phenotypes tar-geted by different breeders across the decades.There are very clear instances illustrating how introduced Italian and otherforeign germplasm was integrated into Chinese wheat programs and reshaped the genomic landscape of local modern cultivars.Importantly,the resequencing data also highlighted significant asymmetric breeding selec-tion among the three sub-genomes:this was evident in both the collinear blocks for homeologous chromosomes and among sets of three homeologous genes.Accumulation of more newly assembled genes in newer cultivars implied the potential value of these genes in breeding.Conserved and extended sharing of linkage disequilibrium(LD)blocks was highlighted among pedigree-related cultivars,in which fewer haplotype differences were detected.Fixation or replacement of haplotypes from founder genotypes after generations of breeding was related to their breeding value.Based on the haplotype frequency changes in LD blocks of pedigree-related cultivars,we propose a strategy for evaluating the breeding value of any given line on the basis of the accumulation(pyramiding)of bene-ficial haplotypes.Collectively,our study demonstrates the influence of 'founder genotypes' on the output of breeding efforts over many decades and also suggests that founder genotype perspectives are in fact more dy-namic when applied in the context of modern genomics-informed breeding. | Chenyang Hao Chengzhi Jiao Jian Hou Tian Li Hongxia Liu Yuquan Wang Jun Zheng Hong Liu Zhihong Bi Fengfeng Xu Jing Zhao Lin Ma Yamei Wang Uzma Majeed Xu Liu Rudi Appels Marco Maccaferri Roberto Tuberosa Hongfeng Lu Xueyong Zhang | 2020 | Molecular Plant2020,13,12: | 9 |
| 3 | RESUSPENSION METHOD FOR ROAD SURFACE DUST COLLECTION AND AERODYNAMIC SIZE DISTRIBUTION CHARACTERIZATION显示文摘Traffic-generated fugitive dust is a source of urban atmospheric particulate pollution in Beijing. This paper introduces the resuspension method, recommended by the US EPA in AP-42 documents, for collecting Beijing road-surface dust. Analysis shows a single-peak distribution in the number size distribution and a double-peak mode for mass size distribution of the road surface dust. The median diameter of the mass concentration distribution of the road dust on a high-grade road was higher than that on a low-grade road. The ratio of PM2.5 to PM10 was consistent with that obtained in a similar study for Hong Kong. For the two selected road samples, the average relative deviation of the size distribution was 10.9% and 11.9%. | 4 | Devonian Sequence Stratigraphy and Relative Sea Level Changes in Longmenshan Area, Sichuan显示文摘The Longmenshan area is located at the western margin of Yangtze platform, where the Devonian is composed of clastic rocks, mixed clastic-carbonate rocks and carbonate rocks in ascending order, and was formed in the marine environment from nearshore to outer shelf. Based on a study of six sections the Devonian in Longemenshan area is divided into 18 sequences and 5 sequence sets. The maximum transgressive high in the Devonian of this area occurred in Early Frasnian, corresponding to asymmetricus zone. The boundaries among the sequence sets are roughly corresponding to the bottom boundaries of Ⅰa’,Ⅰc’,Ⅱa, and Ⅱd in the Devonian of West Europe and North America, respectively. | 5 | Less and shrunken pollen 1(LSP1) encodes a member of the ABC transporter family required for pollen wall development in rice(Oryza sativa L.)显示文摘Pollen fertility is an agronomic trait that strongly influences rice yield. Recent studies have revealed that the development of the pollen wall is required for pollen fertility and is regulated by several genes. However, the mechanisms underlying pollen and pollen wall development in rice remain largely unknown. In the present study, a point mutation in a gene on chromosome 1 was identified that resulted in the production of less and shrunken pollen(LSP) and led to defects in pollen wall formation. This gene was named LSP1 and was found to encode a member of the adenosine triphosphate-binding cassette(ABC)transporter G subfamily, OsABCG3. Two other loss-of-function mutants of LSP1/OsABCG3,generated using CRISPR/Cas9 technology, showed the same male sterile phenotype. The LSP1/OsABCG3 gene showed a spatio-temporal expression pattern in the developing anthers, and is an ortholog of the Arabidopsis genes At ABCG1 and At ABCG16, which play an important role in pollen wall development. Mutation of LSP1/OsABCG3 affected the expression of several genes involved in pollen and pollen wall formation. | 6 | Recent Climate Trends on the Northern Slopes of the Tianshan Mountains, Xinjiang, China显示文摘In arid regions, mountains fulfill important ecological and economic functions for the surrounding lowlands. In the scenario of global warming, mountain ecosystems change rapidly, especially in the arid region of northwestern China. This paper provides an assessment of the changes in temperature and precipitation in the historical records of climate on the northern slopes of the eastern Tianshan Mountains. A Mann-Kendall nonparametric trend and Sen's tests are employed to analyze the interannual changes and innerannual variability in temperature and precipitation in the regions of low to high altitude. The present study finds that the largest increases in annual temperature are observed at stations in the low altitude regions. The significant increasing trends in temperature tend to occur mainly in late winter and early spring at stations from middle to high altitude, but in summer and autumn at stations of low altitudes. The increasing trends in annual precipitation are found from the middle to high altitude areas, but decreasing trends are found in the low altitude areas. | 9 | Regulation of autophagic flux by CHIP显示文摘Autophagy is a major degradation system which processes substrates through the steps of autophagosome formation, autophagosome-lysosome fusion, and substrate degradation. Aberrant autophagic flux is present in many pathological conditions including neurodegeneration and tumors. CHIP/STUB1, an E3 ligase, plays an important role in neurodegeneration. In this study, we identified the regulation of autophagic flux by CHIP(carboxy-terminus of Hsc70-interacting protein). Knockdown of CHIP induced autophagosome formation through increasing the PTEN protein level and decreasing the AKT/m TOR activity as well as decreasing phosphorylation of ULK1 on Ser757. However, degradation of the autophagic substrate p62 was disturbed by knockdown of CHIP, suggesting an abnormality of autophagic flux. Furthermore, knockdown of CHIP increased the susceptibility of cells to autophagic cell death induced by bafilomycin A1. | 14 | Pulsar candidate selection using ensemble networks for FAST drift-scan survey显示文摘The Commensal Radio Astronomy Five-hundred-meter Aperture Spherical radio Telescope(FAST) Survey(CRAFTS) utilizes the novel drift-scan commensal survey mode of FAST and can generate billions of pulsar candidate signals. The human experts are not likely to thoroughly examine these signals, and various machine sorting methods are used to aid the classification of the FAST candidates. In this study, we propose a new ensemble classification system for pulsar candidates. This system denotes the further development of the pulsar image-based classification system(PICS), which was used in the Arecibo Telescope pulsar survey, and has been retrained and customized for the FAST drift-scan survey. In this study, we designed a residual network model comprising 15 layers to replace the convolutional neural networks(CNNs) in PICS. The results of this study demonstrate that the new model can sort >96% of real pulsars to belong the top 1% of all candidates and classify >1.6 million candidates per day using a dual-GPU and 24-core computer. This increased speed and efficiency can help to facilitate real-time or quasi-real-time processing of the pulsar-search data stream obtained from CRAFTS. | 15 | Shexian Bupleurum Alleviates Insomnia and Improves Symptoms of Anxiety and Depression:A Randomized Double-Blind Placebo-Controlled Clinical Trial显示文摘Objective:To observe the clinical efficacy of Shexian bupleurum on insomnia based on evaluation scales and polysomnogram(PSG).Methods:A total of 260 patients suffering from insomnia admitted to the outpatient department of Handan Central Hospital were included in a randomized,double blind,placebo-controlled trial.The patients were randomly divided into two groups:a control group(receiving placebo,n=150),and an intervention group(receiving Shexian bupleurum treatment,n=110).The clinical trial lasted for 4 weeks.The Pittsburgh Sleep Quality Index(PSQI),Insomnia Severity Index(ISI),Patient Health Questionnaire(PHQ)-9,Generalized Anxiety Disorder(GAD)-7,17 items of Hamilton Depression Scale(HAMD-17),and Hamilton Anxiety Scale(HAMA)were used to evaluate the patients at baseline as well as two weeks and four weeks after treatment;the Treatment Emergent Symptom Scale(TESS)was used to evaluate adverse reactions;polysomnography(PSG)was used to monitor and analyze their sleep characteristics at baseline and four weeks after treatment.Results:The PSQI,ISI,PHQ-9,HAMD-17,and HAMA scores of the intervention group significantly decreased compared to the control group,while the total sleep time,rapid eye movement sleep latency,stage 2 sleep,deep sleep,rapid eye movement sleep,and sleep efficiency of the intervention group significantly increased compared to the control group.The PHQ-9 score of the control group only decreased two weeks after treatment(p<0.05)compared to the intervention group.In addition,there were no obvious adverse events in both the intervention group and the control group.Conclusion:Shexian bupleurum not only improves sleep quality,but also relieves depression and anxiety in patients who suffer from insomnia. | Kai Zhang Xianling Zheng Hongfeng Zhang Xueqiang Zhang Yanmin Xu Shiying Jin Shuang Huo | 2022 | Journal of Clinical and Nursing Research2022,6,4: | 0 |
