3Delivery of triptolide with reduction-sensitive polymer nanoparticles for liver cancer therapy on patient-derived xenografts models显示文摘Hepatocellular carcinoma(HCC)has become the fourth predominant cause of cancer-related deaths worldwide,and HCC is still one of the worst prognoses for survival as it is poorly responsive to both chemotherapy and surgical treatment due to drug resista nce and great toxic effects.Triptolide(TP),a key ingredient from the traditional Chinese medical herb,has been utilized to treat inflammation and antitumor for centuries.However,investigations of this potent agent have been met with only limited success due to the severe systemic toxicities in patients and low water solubility as well as its high toxicity over the past two decades.Herein,we reported the development of a reduction-responsive drug delive ry system loaded with TP fo r glutathione(GSH)-trigge red drug release for cancer therapy.With the GSH-sensitive TP loaded nanoparticles,the remarkable increases in tumor accumulation and amelioration of drug toxicity in animals are demonstrated,which is likely due to sustained stepwise release of active TP within cancer cells.Moreover,in a patient-derived tumor xenograft model of liver cancer,administration of tritolide nanoparticles enhances the antitumor efficacy relative to administration of free TP.These findings indicate that GSH-sensitive release of TP may be a promising strategy for cancer treatment.Mengxue He Ling Yu Yuanyuan Yang Binhua Zou Wen Ma Meng Yu Jiandong Lu Guoliang Xiong Zhiqiang Yu Aimin Li 2020Chinese Chemical Letters2020,31,12:5
416S rRNA Gene Phylogenesis of Culturable Predominant Bacteria from Diseased Apostichopus japonicus(Holothuroidea,Echinodermata)显示文摘Cultured Apostichopus japonicus in China suffers from a kind of skin ulceration disease that has caused severe economic loss in recent years. The disease,pathogens of which are supposed to be bacteria by most researchers,is highly infectious and can often cause all individuals in the same culture pool to die in a very short time. The 16S rRNA gene phylogenesis of the culturable bacteria from the lesions of diseased individuals was conducted to study the biodiversity of the bacterial communities in the lesions and to identify probable pathogen(s) associated with this kind of disease. S. japonica samples were selected from a hatchery located in the eastern part of Qingdao,China. Bacterial universal primers GM5F and DS907R were used to amplify the 16S rRNA gene of bacteria colonies,and touchdown PCR was performed to amplify the target sequences. 5Addition Al and/or Ti Induced Modifi cations of Microstructures, Mechanical Properties, and Corrosion Properties in CoCrFeNi High-Entropy Alloy Coatings显示文摘The purpose of this work is to study the influences of Al and/or Ti addition on the microstructures, mechanical properties and corrosion properties of CoCrFeNi-(Al, Ti) high entropy alloy(HEA) coatings. Three coatings, AlCoCrFeNi(Ⅰ), CoCrFeNiTi_(0.5)(Ⅱ) and AlCoCrFeNiTi_(0.5)(Ⅲ), were fabricated by laser cladding successfully. The AlCoCrFeNi(Ⅰ) coating exhibited a simple body-centered cubic(BCC) solid-solution structure, whereas the CoCrFeNiTi_(0.5)(Ⅱ) alloy exhibited a face-centered cubic(FCC) solid-solution and a small amount of Laves phase. The BCC phases in AlCoCrFeNiTi _(0.5)(Ⅲ) coating were characterized as Fe–Cr rich disordered BCC phases(A2) and Al-Ni–Ti-rich ordered BCC phases(L2_1) separately. The AlCoCrFeNiTi_(0.5)(Ⅲ) coating with dual-phase BCC structure showed the optimal performance of both mechanical and corrosion properties, which was superior to BCC-based AlCoCrFeNi(Ⅰ) and FCC-based CoCrFeNiTi_(0.5)(Ⅱ) coatings. Nanoindentation tests and quantitative investigations on the strengthening mechanisms of AlCoCrFeNiTi_(0.5)(Ⅲ) coating were conducted, suggesting that the precipitation strengthening is the dominant strengthening mechanism. 6Influence of N doping and the functional groups of graphene on a RGO/TiO2 composite photocatalyst显示文摘Reduced graphene oxide(RGO)assisted TiO2 composite photocatalysts have attracted increased attention because of their high photocatalytic performance under both UV-and visible-light irradiation.However,the corresponding properties under the long wavelength irradiation is far from expected.In this study,a series of RGO samples with well-designed N impurity and optimized surface functional groups are prepared and employed during hybridization with TiO2(named N-RGO/TiO2).The coupling between the doped N atoms and surface functional groups is analyzed based on the resulting photocatalytic performances,and the specific mechanism is revealed by the EPR,IR and lifetime of the photoinduced electrons.After comparing the corresponding performances of the RGO/TiO2 and RGO/N-TiO2 specimens,the results show that the surface functional groups of the RGO serve as a precondition to achieveπ-d coupling between the graphene basal plane and TiO2 nanoparticles,while N atoms in the RGO enhance the resulting photoactivity under a long wavelength range(>510 nm).The findings provide a potential approach to promote the photocatalytic properties of graphene-based composite photocatalysts with the low-energy incident photons.TANG Bo HE YanFeng ZHANG ZhenYu WANG ZhengWei JI Li MA TingTing LI Sen DAI YuanZhe ZHANG GuoLiang 2020Science China(Technological Sciences)2020,63,6:3
7A small scale expression screen identifies tissue specific markers in the Dugesia japonica strain Pek-1显示文摘Freshwater planaria has tremendous capacity to reform the missing part of the body and therefore is considered as one of the most important model organism for regeneration study. At present, Schmidtea mediterranea and Dugesia japonica are the two major species utilized for laboratory manipulations. Dugesia japonica flatworms are widely distributed in the Far East including Cherry Valley region in the northwest area of Beijing, China. We reported here the establishment of an asexual Dugesia japonica strain Pek1, as a suitable system for regeneration study. Using morphological, karyotypical as well as phylogenetic analyses, we confirmed that these flatworms indeed belonged to Dugesia japonica. We went on to show that the commonly used in situ probes and immunohistochemistry reagents and protocols were applicable to the Pek1 strain. Using this strain, we carried out small scale analysis on EST, RNAi and gene expression. We identified 193 unique EST sequences and 65 of them had not been reported in planarian. By RNAi analysis, we showed that 48 genes, when downregulated individually, had no effect on regeneration. Furthermore, we identified 3 groups of tissue specific expressing genes that were useful for cell lineage analysis. 9Explore the QCD phase transition phenomena from a multiphase transport model显示文摘We study the phase structure of QCD matter in the framework of a multiphase transport model by implementing a strong local parton density fluctuation scenario. Our calculations on the beam energy dependence of net-proton high moment show that local parton density fluctuation only has a small effect. But it becomes important when all baryons are included. We then study the effect on elliptic flow and find that an enhanced local parton density fluctuation leads to a significant effect on protons but a small effect on pions. 10The corrosion behaviors of Mg-7Gd-5Y-1Nd-0.5Zr alloy under(NH4)_(2)SO_(4),NaCl and Ca(NO_(3))_(2)salts spray condition显示文摘The corrosion behaviors of Mg–7Gd–5Y–1Nd–0.5Zr alloys after T5 treatment under(NH4)_(2)SO_(4),NaCl and Ca(NO_(3))_(2)salt spray condition were investigated by weight loss rates,residual mechanical properties,scanning electron microscope(SEM),X-ray Diffraction(XRD)and potentiodynamic polarization tests.The corrosion degree of Mg–7Gd–5Y–1Nd–0.5Zr alloys in Ca(NO_(3))_(2)salt spray was very shallow by corrosion morphology and the corrosion route was extended along the surface in texture-like shape,while the alloy in NaCl and(NH4)_(2)SO_(4)salt spray were major local corrosion and there were serious corrosion pits on the surface.The weight loss rates in(NH4)_(2)SO_(4),NaCl and Ca(NO_(3))_(2)salt spray was respectively 0.4147,0.1618 and 0.0725 mg/(cm2 d−1).The results of residual mechanical properties indicated that the corrosion order in salts spray of Mg–7Gd–5Y–1Nd–0.5Zr alloys is NH4SO4>NaCl>Ca(NO_(3))_(2),which was consistent with the results of potentiodynamic polarization tests.The type of the salts will play a vital role in the initiation of the corrosion of EW75 alloy when they are used in the atmosphere environments.Inorganic salts with the smaller PH value after dissolution will have a stronger impact on the corrosion of EW75 magnesium alloys.Quantong Jiang Kui Zhang Xinggang Li Yongjun Li Minglong Ma Guoliang Shi Jiawei Yuan 2013Journal of Magnesium and Alloys2013,1,3:2
11Antifouling mechanism of natural product-based coatings investigated by digital holographic microscopy显示文摘Using natural product-based antifouling coatings has proven to be an effective strategy to combat biofouling.However,their antifouling mechanisms are still unclear.In this study,the antifouling mechanism of natural product-based coatings consisting of bio-sourced poly(lactic acid)-based polyurethane and ecofriendly antifoulant(butenolide)derived from marine bacteria was revealed by observing 3D bacterial motions utilizing a 3D tracking technique-digital holographic microscopy(DHM).As butenolide content increases,the density of planktonic marine bacteria(Pseudomonas sp.)near the surface decreases and thus leads to a reduced adhesion,indicating that butenolide elicits the adaptive response of Pseudomonas sp.to escape from the surface.Meanwhile,among these remained cells,an increased percentage is found to undergo subdiffusive motions compared with the case of smaller dose of butenolide.Further experiments show that butenolide can accelerate their swimming velocity and reduce flick frequency.Antibacterial assay confirms that butenolide-based coating shows high efficacy of antifouling performance against Pseudomonas sp.but without killing them like 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one(DCOIT).Jiansen Pan Qingmei Peng Guoliang Zhang Qingyi Xie Xiangjun Gong Pei-Yuan Qian Chunfeng Ma Guangzhao Zhang 2021Journal of Materials Science & Technology2021,,25:2
12Microstructure evolution,mechanical properties and creep mechanisms of Mg-12Gd-1MM-0.6Zr(wt%)magnesium alloy显示文摘In this research,the microstructure evolution,mechanical properties,and creep mechanisms of Mg-12 Gd-1 MM-0.6 Zr(wt%)alloy under different conditions were systematically studied using scanning electron microscopy(SEM),transmission electron microscopy(TEM),X-ray diffraction(XRD),and tensile creep tests.Regarding the microstructure of the as-cast sample,the average grain size is about 42μm,and the eutectic compounds were determined to be Mg_(5)(Gd_(0.8)MM_(0.2)).During homogenization,these eutectic compounds gradually dissolve,and Mg_(12)MM particles are precipitated.During hot extrusion,complete dynamic recrystallization(DRX)occurs,resulting in equiaxial grains with an average grain size of about 12μm and the formation of streamlines consisting of Mg_(12)MM particles along the extrusion direction(ED).After T5 treatment(225℃for 7 h),a large number ofβ'(Mg_(7)Gd)phases are precipitated on the{11-20}αhabit plane and are interconnected,forming an interlaced network structure.The ultimate tensile strength(R_(m)=405 MPa)and yield strength(R_(P0.2)=288 MPa)of the T5 sample are significantly higher than those of the as-extruded sample(R_(m)=289 MPa,R_(P0.2)=185 MPa),but the elongation(A=4%)was remarkably lower than that of the as-extruded sample(A=18%).When crept at225℃under 100 MPa,the steady-state creep rates of the as-cast,as-extruded,and T5 samples are1.59×10^(-8),1.08×10^(-8),and 1.40×10^(-8)s^(-1),respectively,and their total strains within 100 h are respectively breaking,0.81%,and 0.92%,indicating that the as-extruded alloy exhibits the best creep resistance.TEM analysis reveals that,during the creep process of the T5 sample,theβ'particles coarsen and the precipitate-free zones(PFZs)widen,which increase the steady-state creep rate and the total strain within 100 h as compared with the as-extruded sample.Bibo Li Kui Zhang Guoliang Shi Kaikun Wang Yongjun Li Xinggang Li Minglong Ma Jiawei Yuan 2021Journal of Rare Earths2021,39,5:2
13Dynamic precipitation during multi-axial forging of an Mg-7Gd-5Y-1Nd-0.5Zr alloy显示文摘The dynamic precipitation behavior during multi-axial forging in an Mg-7Gd-5Y-1Nd-0.5Zr alloy has been investigated and compared with that in static precipitation treatment. The results indicated that dynamic precipitation does occur during multi-axial forging. The dynamic precipitate can be deduced as β phase with face-centered cubic crystal structure (a = 2.22 nm). Most of the β precipitates locate at the dynamic recrystallization grain boundaries. The morphology and orientation relationship is different from that of the β phase formed in the static precipitation treated alloys, although the crystal structure is the same. 20Brocaeloid D,a novel compound isolated from a wheat pathogenic fungus, Microdochium majus 99049显示文摘Microbes serve as the most important resource for drug discovery.During our screening for bioactive compounds from our natural products library,a pathogenic fungus,Microdochium majus strain 99049,from wheat was selected for further investigation.A new alkaloid named brocaeloid D(1),together with six previously characterized compounds(2–7)were identified.Compound 1 belongs to 4-oxoquinoline with C-2 reversed prenylation and a succinimide substructure.All the structures of these newly isolated compounds were determined by different means in spectroscopic experiments.The absolute configurations of 1 was further deduced from comparison of its CD spectrum with that of known compound 2.The bioactivities of these identified compounds were evaluated against several pathogenic microorganisms and cancer cell lines.Compounds 1–5 showed activity against HUH-7 human hepatoma cells with IC50 values of 80μg/mL.Compound 6 showed mild activity against HeLa cells(IC50=51.9μg/mL),weak anti-MTB activity(MIC=80μg/mL),and moderate anti-MRSA activity(MIC=25μg/mL),and compound 7 showed weak anti-MRSA activity(MIC=100μg/mL).Jingyu Zhang Zhenzhen Wang Zhijun Song Loganathan Karthik Chengjian Hou Guoliang Zhu Lan Jiang Jianying Han Rong Ma Li Li Lixin Zhang Xueting Liu Tom Hsiang 2019Synthetic and Systems Biotechnology2019,4,4:1
