| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 5 | Bearing behavior and failure mechanism of squeezed branch piles显示文摘The current practice for the design of squeezed branch piles is mainly based on the calculated bearing capacity of circular piles. Insufficient considerations of the load-transfer mechanism, branch effect and failure mechanism, as well as overreliance on pile load tests, have led to conservative designs and limited application. This study performs full-scale field load tests on instrumented squeezed branch piles and shows that the shaft force curves have obvious drop steps at the branch position, indicating that the branches can effectively share the pile top load. The effects of branch position, spacing, number and diameter on the pile bearing capacity are analyzed numerically. The numerical results indicate that the squeezed branch piles have two types of failure mechanisms, i.e. individual branch failure mechanism and cylindrical failure mechanism. | 6 | Three-dimensional dissipative stress space considering yield behavior in deviatoric plane显示文摘Naturally deposited soils are always found in the complex three-dimensional stress state.Constitutive models developed for modeling the three-dimensional mechanical behavior of soils should obey the basic laws of thermo-mechanical principles.Based on the incremental dissipation function,a new deviatoric shift stress is derived and then introduced into the existing constitutive models to describe the yield behavior in the deviatoric plane for geomaterials.By adopting the proposed shift stress,the relationship between dissipative stress tensors and true stress tensors can be established.Therefore,the threedimensional plastic strain can be calculated reasonably through the associated flow rule in the three-dimensional dissipative stress space.At the same time,three methods that are conventionally adopted for generalizing constitutive models to model the three-dimensional stress-strain relationships are examined under the thermo-mechanical framework.The TS(transformed stress)method is shown to obey the thermo-mechanical rules and the TS space adopted in TS method is actually a translational three-dimensional dissipative stress space.However,it is illustrated that the other two approaches,the method of using failure criterion directly and the method of using g()function,violate the basic rules of thermo-mechanical theories although they may bring convenience and simplicity to numerical analysis for geotechnical engineering.Comparison between model predictions and experimental data confirms the validity of the proposed three-dimensional dissipative stress space. | YAO YangPing CUI WenJie WANG NaiDong | 2013 | Science China(Technological Sciences)2013,56,8: | 4 |
| 7 | Optimal power allocation for cognitive relay networks: amplify-and-forward versus selection relay显示文摘We consider a cognitive relay network which is defined by a source,a destination,and cognitive relay nodes and primary user nodes.In this network,a source is assisted by cognitive relay nodes which allow coexisting with primary user nodes by imposing severe constraints on the transmission power so that they operate below the noise floor of primary user nodes.In this paper,we mainly study the power allocation strategies of this system to minimize the outage probability subject to total and individual power constraints for cognitive relay nodes and subject to interference constraints for primary user nodes.A relay transmission scheme,namely amplify-and-forward (AF),is considered.We first present an optimal power allocation (OPA) scheme to minimize the system outage probability,based on instantaneous channel state information among the source,destination and relay nodes and mean channel gains between primary user nodes and cognitive relays.Next,we propose a selection AF scheme (S-AF) where the single best relay is chosen to assist in the transmission and study power allocation strategy for S-AF to minimize outage probability.The analytical power allocation strategies have been validated through numerical simulations.The results indicate that the AF with OPA and S-AF with OPA have significantly better outage behavior and average throughput than AF and S-AF schemes respectively.We also find that S-AF with OPA achieves a higher throughput,and hence lower outage probability than AF with OPA. | YUE WenJing ZHENG BaoYu MENG QingMin YUE WenJie CUI JingWu | 2011 | Science China(Information Sciences)2011,54,4: | 3 |
| 8 | 显示恶性孤立肺结节分叶征的理想CT层厚(英文)显示文摘Objective: The aim of this study was to determine an optimal slice thickness that was efficient in revealing lobulation of malignant solitary pulmonary nodules (SPNs) on multi-slice spiral computed tomography (MSCT) images preliminarily. Methods: Fifty patients with malignant SPNs (diameter ≤ 3 cm) underwent multidetector-row computed tomography of the chest in a single-breath-hold technique. The raw data were acquired with a collimation of 0.625 mm. Three sets of contiguous images were reconstructed with 1-, 2-, and 5-mm slice thickness, respectively. The lobulation sign of SPNs on the computed tomography (CT) images presented in 1-, 2-, and 5-mm slice thickness was compared. Using the 1-mm sections as the gold standard, an optimal slice thickness in revealing lobulation sign of SPNs was determined. Results: The 1-mm-thick images CT revealed 98 lobulations (25 with chord distance < 1 mm; 30 with chord distance 1-2 mm; 43 with chord distance > 2 mm) of 45 malignant SPNs. 18 lobulations with chord distance < 1 mm presented in 2-mm-thick sections were as same as those in 1-mm-thick sections. Statistically significant difference in lobulations number was found between that revealed in 2-mm-thick images and that in 1-mm-thick images (P = 0.023 < 0.05). 16 lobulations with chord distance < 1 mm presented in 5-mm-thick sections were as same as that in 1-mm-thick sections. There was statistically significant difference in lobulations number between that revealed in 5-mm-thick images and that in 1-mm-thick images (P = 0.004 < 0.05). The 24 lobulations with chord distance 1-2 mm presented in 2-mm-thick sections were as same as that in 1-mm-thick sections. No statistically significant difference in lobulations number were found between that revealed in 2-mm-thick images and that in 1-mm-thick images (P = 0.261 > 0.05). 13 lobulations with chord distance 1-2 mm presented in 5-mm-thick sections were as same as that in 1-mm-thick sections. There was statistically significant difference in lobulations number between that revealed in 5-mm-thick images and that in 1-mm-thick images (P = 0.003 < 0.05). 40 lobulations with chord distance ﹥2 mm presented in 2-mm-thick sections were as same as that in 1-mm-thick sections. No statistically significant difference in lobulations number was found between that revealed in 2-mm-thick images and that in 1-mm-thick images (P = 0.631 > 0.05). 36 lobulations with chord distance ﹥2 mm presented in 5-mm-thick sections were as same as that in 1-mm-thick sections. | 9 | Homeobox B8 Targets Sterile Alpha Motif Domain-Containing Protein 9 and Drives Glioma Progression显示文摘Gliomas are the most commonly occurring tumors of the central nervous system. Glioblastoma multiforme (GBM) is the most malignant and aggressive brain cancer in adults. Further understanding of the mechanisms underlying the aggressive nature of GBM is urgently needed. Here we identified homeobox B8(HOXB8), a member of the homeobox family, as a crucial contributor to the aggressiveness of GBM. Data mining of publicly accessible RNA sequence datasets and our patient cohorts confirmed a higher expression of HOXB8 in the tumor tissue of GBM patients, and a strong positive correlation between the expression level and pathological grading of tumors and a negative correlation between the expression level and the overall survival rate. We next showed that HOXB8 promotes the proliferation and migration of glioblastoma cells and is crucial for the activation of the PI3K/AKT pathway and expression of epithelial–mesenchymal transition-related genes, possibly through direct binding to the promoter of SAMD9 (Sterile Alpha Motif Domain-Containing Protein 9) and activating its transcription. | 14 | Complete Genome Sequences of the SARS-CoV: the BJ Group (Isolates BJ01-BJ04)显示文摘Beijing has been one of the epicenters attacked most severely by the SARS-CoV (severe acute respiratory syndrome-associated coronavirus) since the first patient was diagnosed in one of the city's hospitals. We now report complete genome sequences of the BJ Group, including four isolates (Isolates B J01, B J02, B J03,and B J04) of the SARS-CoV. It is remarkable that all members of the BJ Group share a common haplotype, consisting of seven loci that differentiate the group from other isolates published to date. Among 42 substitutions uniquely identifled from the BJ group, 32 are non-synonymous changes at the amino acid level.Rooted phylogenetic trees, proposed on the basis of haplotypes and other sequence variations of SARS-CoV isolates from Canada, USA, Singapore, and China, gave rise to different paradigms but positioned the BJ Group, together with the newly discovered GD01 (GD-Ins29) in the same clade, followed by the H-U Group (from Hong Kong to USA) and the H-T Group (from Hong Kong to Toronto), leaving the SP Group (Singapore) more distant. | 15 | The transfection of embryonic stem cells with Tet-on system and its responsiveness to doxycycline显示文摘We transiently transfected pTet-on and pTRE2hyg-luciferase into the mouse em-bryonic stem cells (ESCs) using lipofectamine, and analyzed its inductive effect by adding serial concentrations of doxycycline (DOX). The results showed that in the transfected group, the luciferase activity of the cells was gradually increased along with the increasing concentration of DOX. While in the non-transfected group, the luciferase activity was not detectable even with DOX treatment. | 20 | Effects of Diet with'Lianhuang'Additive of Chinese Herb Medicine on Growth Performance and Metabolic Rates of Dietary Nutrients in Donglan Black Chicken显示文摘[Objectives]The study was conducted to evaluate the effects of diet with'Lianhuang'additive of Chinese herbal medicine(LACHM)on growth performance and metabolic rates of dietary nutrients in Donglan black chickens(DBCs).[Methods]80 DBCs aged 1 d were blocked into 4 groups randomly and assigned to 6 replicates in each group with 20 chicks in each replicate.Group A as control group fed basal diet.Groups B,C and D as treatment groups consumed the basal diet added with 0.25%,0.50%and 1.00%of LACHM,respectively during the test period of 70 d.In the end of feeding test,24 DBCs with similar body weight from each group were chosen out and used for a metabolism test to determine the metabolic rates of dietary nutrients from each treatment.The metabolism test was conducted as the same design as the feeding test and the DBCs from each group were fed the same diet as previous test.[Results]The ADG of chickens in the test increased by 5.00%-9.15%(P<0.05)except the stage of 50-70 d,and the individual average body weight(IABW)at 70 d in group B also increased by 4.86%(P<0.05).Compared with group A,the ADG of group C in the stages of 36-49,1-49 and 1-70 d and the IABW in the end of 70 d increased by 5.90%,2.66%,2.68%and 2.57%(P>0.05),respectively.The ADFI in the stage of 36-49 d from group A was 6.04%more than that from group B(P<0.01),and 5.12%and 4.13%more than those from groups C and D(P<0.05),respectively,and the ADFI of group A in the stage of 1-49 d was still 2.86%more than that from group C(P<0.05).However,no differences in ADFI were found in other stages in the test among the 4 groups(P>0.05).The F/G in the stages of 22-35 and 1-70 d from group B was 6.32%and 3.41%lower than those from group A(P<0.05),respectively,and even 11.43%and 7.31%lower in the stages of 36-49 and 1-49 d from group B than those from group A(P<0.01),respectively.The F/G of group C in the stage of 36-49 d was also 10.15%lower than that of group A(P<0.05),and still 5.29%lower than that from group A in 1-49 d(P<0.01),but no differences were found in other stages between the two groups(P>0.05).The metabolic rates of MD,CP,CF,NFE,CA,TE,Ca and P in the 3 treatment diets for group B,C and D all increased to some extent.Among them,the increasing extents of diets with 0.25%and 0.50%of LACHM for groups B and C were much greater.Therefore,the diet with 0.25%of LACHM could improve the growth performance and metabolic rates of dietary nutrients in DBCs significantly.[Conclusions]This study provides a theoretical reference for the effects of traditional Chinese medicine additives on the growth and metabolic rates of DBCs. | Jing YAN Lei QIU Min CHEN Fulin WANG Wenjie LU Menglei LI Cui YANG Jiahuang YANG | 2022 | Agricultural Biotechnology2022,11,3: | 1 |
