3Optimization of Extraction of Total Flavonoids from HERBA BLUMEAE RIPARIAE Using Response Surface Methodology显示文摘[Objectives]This study aimed to optimize the ultrasonic-assisted extraction process of total flavonoids from a Zhuang medicine HERBA BLUMEAE RIPARIAE using response surface methodology.[Methods]Using rutin as a reference substance,a standard curve was drawn at a wavelength of 292 nm.Content determination was performed through absorbance under different conditions.The effects of ultrasonic time,ethanol volume fraction and liquid-to-solid ratio on the yield of total flavonoids from HERBA BLUMEAE RIPARIAE were investigated by single-factor tests.By using Box-Behnken test,the ultrasonic-assisted extraction process of total flavonoids from HERBA BLUMEAE RIPARIAE was optimized.[Results]The optimal extraction process of total flavonoids from HERBA BLUMEAE RIPARIAE was as follows:ultrasonic time of 30 min,ethanol volume fraction of 50%and liquid-to-solid ratio of 40∶1(mL/g).Under the optimal extraction conditions,the average yield of total flavonoids from HERBA BLUMEAE RIPARIAE was 85.47 mg/g.[Conclusions]The optimized extraction process was convenient and feasible.This study will provide a reference for the extraction of total flavonoids from HERBA BLUMEAE RIPARIAE,and provide certain guidance and basis for further development and utilization of the plan resource of Blumea riparia(Bl.)DC.Hongna SU Yining ZHANG Xuexue LI Xuelian ZHAO Yisong LI Wenbing LI Yuan LIU 2020Medicinal Plant2020,11,2:5
4DNA barcoding and rapid identification of the precious herb Herba Anoectochili显示文摘Herba Anoectochili is a commonly used medicinal material. However, its adulteration is a serious concern. Due to the similar morphological characteristics of Herba Anoectochili and its adulterants, traditional identification techniques often fail to distinguish between them accurately, which is not conducive to the circulation management and safety of the medicinal materials. To improve the distinction between Herba Anoectochili and its adulterants accurately, this study identified 41 Herba Anoectochili and its adulterant samples based on the ITS2 sequence. Sequence characteristics, Basic Local Alignment Search Tool(BLAST) application, genetic distance, construction of phylogenetic tree, secondary structure prediction, and other methods showed the ITS2 sequence to accurately identify Herba Anoectochili from its adulterants. Furthermore, in this study, we designed a specific primer, based on the ITS2 sequence, and established a real-time PCR detection system for the rapid, sensitive, and specific identification of the original plant of Herba Anoectochili. 6Pharmacognostic Identification of Yi Medicine Herba Boenninghauseniae Albiflorae显示文摘[Objectives] To identify original plant and traits of Yi medicine Herba Boenninghauseniae Albiflorae,and provide experimental data for its identification and application. [Methods] The original plant,traits,and microscopic identification were used. [Results] Essential identification characteristics of Yi medicine Herba Boenninghauseniae Albiflorae: small leaf was 1-2. 5 cm long,while the gynophore was 4-8mm long when bearing fruit; traits mainly included characteristic odor,strong and bitter taste,and spicy flavor; microscopic exclusive identifying characteristics: rich aleurone grain in the root; leaf had lysigenous secretory cavity,and non-glandular hairs and oil droplets were common. [Conclusions] Yi medicine Herba Boenninghauseniae Albiflorae( Ebazema) has distinctive identifying characteristics in original plant,traits,and microscopic tissues. 8EGTA reduces the inflorescence stem mechanical strength of herbaceous peony by modifying secondary wall biosynthesis显示文摘The mechanical strength of inflorescence stems is an important trait in cut flowers.Calcium ions(Ca^(2+))play a pivotal role in maintaining stem strength,but little is known about the underlying molecular mechanisms.In this study,we treated herbaceous peony(Paeonia lactiflora Pall.)with ethyl glycol tetraacetic acid(EGTA),an effective Ca^(2+)chelator,and used morphology indicators,spectroscopic analysis,histochemical staining,electron microscopy,and proteomic techniques to investigate the role of Ca^(2+)in inflorescence stem mechanical strength.The EGTA treatment reduced the mechanical strength of inflorescence stems,triggered the loss of Ca^(2+)from cell walls,and reduced lignin in thickened secondary walls in xylem cells as determined by spectroscopic analysis and histochemical staining.Electron microscopy showed that the EGTA treatment also resulted in significantly fewer xylem cell layers with thickened secondary walls as well as in reducing the thickness of these secondary walls.The proteomic analysis showed 1065 differentially expressed proteins(DEPs)at the full-flowering stage(S4).By overlapping the Kyoto encyclopedia of genes and genomes(KEGG)and gene ontology(GO)analysis results,we identified 43 DEPs involved in signal transduction,transport,energy metabolism,carbohydrate metabolism,and secondary metabolite biosynthesis.Using quantitative real-time polymerase chain reaction(qRT-PCR)analysis,we showed that EGTA treatment inhibited Ca^(2+)sensors and secondary wall biosynthesis-related genes.Our findings revealed that EGTA treatment reduced the inflorescence stem mechanical strength by reducing lignin deposition in xylem cells through altering the expression of genes involved in Ca^(2+)binding and secondary wall biosynthesis.Yuhan Tang Daqiu Zhao Jiasong Meng Jun Tao 2019Horticulture Research2019,6,1:2
9Morphological and chemical studies of artificial Andrographis paniculata polyploids显示文摘Andrographis paniculata(Burm. f.) Nees(AP) is commonly used for the treatment of many infectious diseases and has been cultivated widely in Asian countries, and has been included in United States Pharmacopoeia as a dietary supplement, but the cultivars of A. paniculata are not abundant due to its self-pollinated. With the aims to enrich AP resources and provide materials for after breeding we explored the polyploidy induction. Different explants, colchicine concentration, and treatment time were tested. After identification by flow cytometry, eleven polyploid plants with different morphologic traits were obtained. The agronomic traits and andrographolide concentration of the polyploids were improved greatly. One of the polyploids(serial 3-7) was chosen for further study. The traits of the second and third generation polyploids(serial 3-7) were stable. Compared with the normal plants, the seeds(2 nd generation) weight increased by 31%, and the andrographolide concentration of the leaves increased by 14%(2 nd) and 28%(3 rd). 10Extraction Process of Volatile Oil of Artemisiae Scopariae Herba显示文摘[Objectives]To obtain the optimal extraction process parameters of volatile oil of Artemisiae Scopariae Herba.[Methods]The controlled variable method was used to take the extraction amount of volatile oil as the evaluation indicator,and the effects of single factor solid-to-liquid ratio,soaking time and extraction time on the yield rate of volatile oil were investigated.[Results]When the solid-to-liquid ratio reached 1∶12,the soaking time reached 3 h,and the extraction time reached 2 h,the yield of volatile oil was the highest.[Conclusions]The conditions of this process are stable and feasible,and it is expected to provide a certain reference for extracting volatile oil from Artemisiae Scopariae Herba.Zhaofeng LI 2020Medicinal Plant2020,11,2:1
11Extraction of Herba Lycopi Polysaccharide and Study of Its Antioxidant Activity by Cellulase Method显示文摘[Objectives] To optimize the extraction process of Herba Lycopi polysaccharide by cellulase,and study its antioxidant activity.[Methods]The single factor test and orthogonal test were conducted to study the effect of p H,enzyme dosage,enzymolysis temperature and extraction time on the extraction of Herba Lycopi polysaccharide. And the test was done about its scavenging effect on DPPH radicals and superoxide anions,and the antioxidant activity was evaluated. [Results] The optimum extraction process by cellulase method was as follows: p H value of 7; enzyme dosage of 4 ml; enzymolysis temperature of 40 ℃; extraction time of 2. 5 h. The average extraction rate of Herba Lycopi polysaccharide was 10. 70%,and RSD was 0. 17%. The antioxidant activity test showed that Herba Lycopi polysaccharide had a significant scavenging effect on radicals. Several herbs belonging to the genus Epimedium have been used in many variations of preparations and formulas for treating a wide range of diseases especially reinforcing Yang and toning the kidney by Chinese physicians for centuries[1,2]. More than 130 chemical constituents have been discovered and classified into many different chemical subgroups. Nearly half of these constituents belong to flavonoid glycosides and have been considered as the major active components in the Epimedium species. Icariin is thought to be the marker constituent of the genus because every Epimedium species contains it. The pharmaceutical effects of Epimedium species have been tested in various biological systems. It has been shown that the single constituent has less activity than that of decoctions, composite preparations or extracts of Epimedium. 14Identification and functional verification of PlFT gene associated with flowering in herbaceous peony based on transcriptome analysis显示文摘The herbaceous peony(Paeonia lactiflora Pall.)is considered to be a highly valued cut flower plant.It has large flower with rich colors.However,there has been little or no research into the genes related to its flower development.In this study,we used the Illumina HiSeq platform to analyze the RNA-Seq comparative transcriptome of the P.lactiflora'Dafugui'in three different flowering periods.Nine cDNA libraries were established,from which 92.53 Gb data with 81,788 unigenes were obtained.We screened the genes related to P.lactiflora flowering,isolated and cloned the PlFT gene related to flowering.The total length of the PlFT gene was 592 bp,which had a complete open reading frame of 522 bp and encoded 173 amino acids.The accession number of the PlFT gene is MT249229.To test the role of PlFT,we constructed an expression vector for genetic transformation.Its expression in Arabidposis mutant indicated that PlFT was involved in the flowering of P.lactiflora.This is the first transcriptome analysis of flower development in P.lactiflora.Our results provide some fundamental information for further analyzing the molecular mechanism underlying flower development of P.lactiflora.Jing Sun Tian Chen Yan Wu Jun Tao 2021Ornamental Plant Research2021,1,1:0
15Effects of Herba Picriae Extract on Growth Performance and Immune Organ Index of Guangxi Partridge Chickens显示文摘[Objectives]The purpose of this experiment was to study the effects of dietary supplementation of Herba Picriae extract on the growth performance and immune organ index of Guangxi partridge chickens.[Methods]A total of 500 one-day-old Guangxi partridge chickens were selected and randomly divided into five groups,five replicates for each group and 20 chickens for each replicate.Groups A,B and C were treatment groups,in which 0.70%,0.35%and 0.175%of Herba Picriae extract was added to the basal diet,respectively.Group D was a positive drug control group,in which 0.01%colistin sulfate premix was supplemented.Group E was a blank control group,in which the chickens were fed the basal diet.The experimental period was 105 d.During the experiment,five chickens were randomly selected from each group on days 21,35 and 49,respectively for the measurement of immune organ index.[Results]When the chickens were fed basal diet supplemented with 0.175%of Herba Picriae extract,their daily body weight gain increased significantly from day 22 to day 35 and from day 71 to day 105 and tended to increase from day 1 to day 21,from day 36 to day 49,and from day 50 to day 70,with a range of 7.37%-8.72%.Dietary supplementation of 0.70%and 0.175%of Herba Picriae extract significantly promoted the growth and development of Fabricius bursae in the experimental chickens.[Conclusions]Dietary supplementation of 0.175%of Herba Picriae extract improved the growth performance and feed intake,as well as 21-d-old bursal index of the experimental chickens.Cui YANG Xiaoguang LIAO Siqian WU Huofu WAN Jing SHI Huihui CAO Linling ZHAO Dawei LAI Fanquan ZENG Fulin WANG Jiahuang YANG Zhuyue WU 2020Asian Agricultural Research2020,12,4:0
16Treatment of Chronic Dysfunction of Transplantation Kidney in Rats——By Tanshinone, Lysimachiae Combined with Mycophenolate Mofetil or Cyclosporine Alone显示文摘WT5HZ]Objective:[WT5BZ] To evaluate the relationship between chronic kidney dysfunction after transplantation and chronic vascular rejection (CVR), and to evaluate the efficacy and safety of Tanshinone (Tan) and Herba Lysimachiae (Lys) combined with Mycophenolate Mofetil (MMF) to fight against CVR, and to reduce the incidence of chronic dysfunction in rat renal transplantation model. [WT5HZ]Methods:[WT5BZ] Sixty five male SD rats as donors and sixty five male Wistar rats as recipients were used. The recipients were divided into five Groups, including Group A: Lys+Cyclosporine A (CsA), Group B: Tan+CsA, Group C: MMF+CsA, Group D: CsA, and Group E: normal saline. Kidney function and morphological changes were assessed at 2, 4, and 6 weeks after transplantation. All sections of renal grafts were stained with monoclonal antibodies (McAB), including major histocompability complex class II (OX 6), lymphocyte function antigen 1 (CD11b/CD18), intercellular adhesive molecular 1 (IA 29 ), CD + 8 (OX 8), and proliferation cell nuclear antigen (PCNA) (5A10, IgG1k) were used. [WT5HZ]Results:[WT5BZ]The two control groups developed typical chronic rejection episodes, and the histologic feature indication of kidney chronic rejection includes loss of renal units and presence of an obliteration arteriopathy involving large renal arteries. These were associated with serum levels of BUN and SCr increased and different degrees of glomerulosclerosis. Their mean survival time was lower than that of other groups. By contrast, serum levels of cytokine in control groups was significantly increased when compared with group B and C ( P <0.05). Both group B and C had minimum changes in glomeruli and arteries, and expression levels of PCNA on the glomeruli and tubular cells were higher than those of other groups ( P <0.05). However, there was no significant difference ( P >0.05) between group B and C. [WT5HZ]Conclusions:[WT5BZ] CVR may activate the risk of the factor responsible for the development of graft chronic dysfunction that causes slow, progressive destruction of the transplanted kidney. Tanshinone was extracted from Salvia miltiorrhiza and purified for use in medicine. Especially when Tanshinone combined with a low dose of CsA, it may fight against the CVR by inhibiting cell infiltration, and improving microcirculation of the graft, and thus the incidence of CVR is reduced. It is suggested that Tanshinone can be applied to treat patients with chronic renal dysfunction when used in combination with a low dose Cyclosporine. 17Protective Effects and Action Mechanism of Total Flavonoids of POLYGONI PERFOLIATI HERBA on Rats with Nonalcoholic Steatohepatitis显示文摘[Objectives]To study the protective effects and mechanism of POLYGONI PERFOLIATI HERBA on rats with nonalcoholic steatohepatitis( NASH). [Methods] Sixty rats were randomly divided into normal group,model group,metformin group( 0. 5 mg/kg),and high dose,medium dose,and low dose groups of total flavonoids of POLYGONI PERFOLIATI HERBA( 600,300,150 mg/kg). The standard feed was given to the normal group,and the model group and the total flavonoids groups were fed with high-fat diet for 13 weeks to establish the NASH rat model. At the 8 th week,the metformin group and the POLYGONI PERFOLIATI HERBA total flavonoids groups were given with the corresponding drug treatment for 6 weeks,blood was taken from the eyeball to collect liver tissue. Biochemical method was used to determine ALT,AST,TC,TG,HDL-c,LDL-c,SOD,MDA,GSH-Px activity or content in serum and HOMA-IR,and ELISA was used to measure the TNF-α,IL-1β and IL-6 content in liver tissue; Western blotting was used to detect the expression levels of AMPK,p-AMPK and ACC in liver tissue. [Results] Total flavonoids of POLYGONI PERFOLIATI HERBA could significantly decrease the activity or content of ALT,AST and MDA in serum of NASH rats( P < 0. 05,P < 0. 01),and enhance the activity of SOD and GSH-Px( P < 0. 05,P < 0. 01),reduce serum TC,TG,LDL-c levels and insulin resistance index,increase HDL-c levels( P < 0. 05,P < 0. 01); down-regulate liver IL-1β,IL-6,TNF-α,ACC levels and up-regulate p-AMPK expression( P < 0. 05,P < 0. 01).
