| 1 | CD146三十年研究的回顾与展望显示文摘CD146分子发现于1987年,最初被认为是黑色素瘤标志分子,随后30余年的研究,逐渐揭示了该分子在早期发育、免疫应答、代谢等生理过程,以及在肿瘤、炎症、自身免疫病中的重要作用及机制.目前认为,CD146不仅是一个黏附分子,还是新生血管、T细胞活化和多种肿瘤的标志分子,更是一种细胞膜受体,接受细胞外信号并调控细胞内多种重要信号途径.最近,越来越多的临床研究报道,无论是细胞膜表面的CD146,还是血清和其他体液中的可溶性CD146(soluble CD146,sCD146),都已成为多种疾病诊断和预后评价的重要参考,将成为疾病治疗的新靶点.迄今,全球已有40余株CD146抗体,有些治疗性抗体已进入临床试验阶段.本文全面回顾了CD146的研究历史,分析了当前亟待解决的关键科学问题,展望了其未来临床应用的潜能,以期为全面认识CD146,充分发挥其在疾病诊疗中的价值提供更多借鉴. | 段红霞 熊朝亮 景林 徐庆吉 刘晶玉 马欣然 王大吉 向建全 何志恒 冯静 阎锡蕴 | 2020 | 中国科学:生命科学2020,50,12: | 6 |
| 2 | 血清sCD40L和CD146与急性缺血性脑卒中后局部血流灌注患者预后的相关性显示文摘目的探究血清可溶性CD40配体(sCD40L)、CD146与急性缺血性脑卒中(CIS)后局部血流灌注患者预后的相关性。方法选取2017年1月~2019年12月我院收治的102例CIS患者依据影像学检查结果分为低灌注组(局部血流低灌注)、对照组(局部血流无灌注),依据随访结果将低灌注组患者分为预后良好组与预后不良亚组。对比低灌注组和对照组一般资料、实验室指标、血清sCD40L、mRS评分、CD146水平。单因素和二元Logistic回归分析影响CIS后局部血流灌注患者预后的高危因素,Spearman相关分析血清sCD40L、CD146水平与CIS后局部血流灌注患者预后的关系。结果影像学检查发现102例患者中56例存在局部血流灌注(54.90%),其中低灌注组sCD40L、mRS评分均高于对照组,CD146水平明显低于对照组(P<0.05)。56例CIS后局部血流灌注患者随访结果显示,预后良好者9例,预后不良者47例,预后不良者年龄≥60岁、LDLC、sCD40L较预后良好者明显高,TC、HDLC、CD146较预后良好组明显低(P<0.05)。二元Logistic回归分析显示,sCD40L、CD146水平是CIS后局部血流灌注患者预后的高危因素(P<0.05)。Spearman相关分析显示,CIS后局部血流灌注患者血清sCD40L水平与预后呈负相关,而CD146水平与预后呈正相关(P<0.05)。结论血清sCD40L、CD146水平是影响CIS后局部血流灌注患者预后的高危因素,其检测结果可作为CIS后局部血流灌注患者预后的有效预测指标。 | 赖琼 赵晓玲 李立 周咏梅 刘艳 | 2020 | 西部医学2020,32,11: | 3 |

| 4 | 3种不同来源CD146阳性间充质干细胞亚群的生物学特性比较显示文摘目的比较脐带间充质干细胞(UCMSCs)、脂肪间充质干细胞(AMSCs)以及骨髓间充质干细胞(BM-MSCs)中CD146^+细胞亚群的生物学特性。方法用磁珠分选法分选不同组织来源的间充质干细胞,获得高纯度CD146^+亚群;用流式细胞计量术分析表型;透射电子显微镜观察细胞结构;成脂诱导分化后油红O染色;RT-qPCR检测成脂相关基因LPL、C/EBPα和PPARγ表达;成骨诱导分化后ALP染色,检测成骨相关基因ALP、OPN和RUNX2表达;检测细胞干性基因及血管生成相关基因表达;检测细胞体外成管能力。结果3种组织来源CD146^+MSCs具有相似的形态,表达除CD106外相似的细胞表面标志分子。与其他两种细胞比较,CD146^+AMSCs表达更高的干性基因OCT-4、SOX2和NANOG。在相同的诱导时间,CD146^+UCMSCs成脂和成骨能力较其他两种来源的CD146^+MSCs弱。3种组织来源的CD146^+MSCs均可表达血管内皮相关刺激因子BFGF、VEGF、Ang-1和EGF,但CD146^+BM-MSCs具有更强的体外成管能力。结论3种不同来源的CD146^+MSC具有不同的生物学特性,为进一步研究不同组织来源的间充质干细胞的特定应用提供了基础。 | 王巧玲 张晓彤 徐衍 张乃月 朱榕嘉 赵春华 | 2020 | 基础医学与临床2020,40,1: | 2 |

| 7 | CD146阳性亚群有作为软骨组织工程种子细胞的应用潜力显示文摘背景:再生医学的发展、组织工程技术的出现为软骨缺损重建提供了新的解决思路。在组织工程中,间充质干细胞是应用广泛的种子细胞,然而干细胞作为一个异质性的细胞群体其不同亚群发挥着不同的功能。因此,应用间充质干细胞关键功能亚群进行软骨修复具有广泛应用前景。目的:从人脂肪间充质干细胞中分离出CD146阳性亚群细胞,验证其生物学特性及其作为软骨组织工程种子细胞的潜力。方法:人脂肪间充质干细胞由浙江金时代生物技术有限公司提供,通过流式细胞术对人脂肪间充质干细胞表面标志物进行鉴定,应用免疫磁珠分选方法从人脂肪间充质干细胞中分选出CD146阳性表达的细胞亚群。通过基因芯片检测技术及生物信息学分析技术揭示2种细胞的分子特性;体外诱导2种细胞成软骨分化并进行鉴定;冻存复苏前后检测2种细胞的细胞活性及凋亡情况。结果与结论:①人脂肪间充质干细胞表达高水平干细胞相关标志物CD73、CD90,不表达造血干细胞相关标志物CD34、CD45、HLA-DR;②生物信息分析结果表明CD146阳性亚群细胞与脂肪间充质干细胞相比在炎症通路及骨骼肌肉系统疾病有不同功能;③CD146阳性亚群细胞能够成球软骨分化,并且其成软骨分化能力要优于人脂肪间充质干细胞;④CD146阳性亚群细胞复苏后凋亡情况和活性均要优于人脂肪间充质干细胞;⑤结果表明,CD146阳性亚群细胞具有良好的成软骨分化潜力,是一种具有前景的软骨组织工程种子细胞。 | 眭翔 田广招 杨振 李旭 刘舒云 郭全义 | 2020 | 中国组织工程研究2020,24,19: | 0 |
