| 4 | Landscape of DILI-related adverse drug reaction in China Mainland显示文摘Drug-induced liver injury(DILI)is a type of bizarre adverse drug reaction(ADR)damaging liver(L-ADR)which may lead to substantial hospitalizations and mortality.Due to the general low incidence,detection of L-ADR remains an unsolved public health challenge.Therefore,we used the data of 6.673 million of ADR reports from January 1st,2012 to December 31st,2016 in China National ADR Monitoring System to establish a new database of L-ADR reports for future investigation.Results showed that totally 114,357 ADR reports were retrieved by keywords searching of liver-related injuries from the original heterogeneous system.By cleaning and standardizing the data fields by the dictionary of synonyms and English translation,we resulted 94,593 ADR records reported to liver injury and then created a new database ready for computer mining.The reporting status of L-ADR showed a persistent 1.62-fold change over the past five years.The national population-adjusted reporting numbers of L-ADR manifested an upward trend with age increasing and more evident in men.The annual reporting rate of L-ADR in age group over 80 years old strikingly exceeded the annual DILI incidence rate in general population,despite known underreporting situation in spontaneous ADR reporting system.The percentage of herbal and traditional medicines(H/TM)L-ADR reports in the whole number was 4.5%,while 80.60%of the H/TM reports were new findings.There was great geographical disparity of reported agents,i.e.more cardiovascular and antineoplastic agents were reported in higher socio-demographic index(SDI)regions and more antimicrobials,especially antitubercular agents,were reported in lower SDI regions.In conclusion,this study presented a large-scale,unbiased,unified,and computer-minable L-ADR database for further investigation.Age-,sex-and SDI-related risks of L-ADR incidence warrant to emphasize the precise pharmacovigilance policies within China or other regions in the world. | Jiabo Wang Haibo Song Feilin Ge Peng Xiong Jing Jing Tingting He Yuming Guo Zhuo Shi Chao Zhou Zixin Han Yanzhong Han Ming Niu Zhaofang Bai Guangbin Luo Chuanyong Shen Xiaohe Xiao | 2022 | Acta Pharmaceutica Sinica B2022,12,12: | 12 |
| 5 | Changing the strategy and culture of stroke awareness education in China: implementing Stroke 1-2-0显示文摘This project implemented the Stroke1-2-0 stroke awareness programme across China and investigated its impact over a 2-year period.We initiated the Stroke1-2-0 educational campaign and Stroke1-2-0 special task forces(STF)across the nation.Massive media coverage,community-based educational sessions with videos and other related materials and induction of Stroke1-2-0 STF were the major means of promotion.We delivered a survey at the end of 2016 and 2018 to evaluate the impact of our effort.A total of 3066 participants responded to the first survey in 2016,and 15207 participants responded in 2018 across China.The acceptance rate for Stroke1-2-0 versus FAST(an English-language stroke awareness tool)was 50.2%versus 19.1%in 2016,and changed significantly to 82.2%versus 8.0%in 2018(p<0.001).Stroke1-2-0 was well accepted by all ages and by people with different academic qualifications.Only 6.5%of survey respondents were aware that there was a therapeutic window for thrombolytic therapy in 2016,but this awareness increased significantly to 32.8%in 2018.Only 12.6%of people in 2016 indicated that they would send patients with stroke to the nearest hospital capable of performing thrombolytic therapy,but there was a nearly threefold increase(52.5%)in this number by 2018.More than 1000 major hospitals joined the Stroke1-2-0 STF,and more than 20000‘stroke warriors’have joined our stroke awareness improvement effort so far.Stroke1-2-0 stroke awareness programme is well-implemented and accepted,and is generating profound improvement in stroke awareness in China. | Jing Zhao Xutong Li Xiaochuan Liu Yuming Xu Jihong Xu Anding Xu Yongjun Wang Renyu Liu | 2020 | Stroke & Vascular Neurology2020,5,4: | 6 |
| 6 | Advancements in the preparation of high-performance liquid chromatographic organic polymer monoliths for the separation of small-molecule drugs显示文摘The various advantages of organic polymer monoliths, including relatively simple preparation processes,abundant monomer availability, and a wide application range of pH, have attracted the attention of chromatographers. Organic polymer monoliths prepared by traditional methods only have macropores and mesopores, and micropores of less than 50 nm are not commonly available. These typical monoliths are suitable for the separation of biological macromolecules such as proteins and nucleic acids, but their ability to separate small molecular compounds is poor. In recent years, researchers have successfully modified polymer monoliths to achieve uniform compact pore structures. In particular, microporous materials with pores of 50 nm or less that can provide a large enough surface area are the key to the separation of small molecules. In this review, preparation methods of polymer monoliths for high-performance liquid chromatography, including ultra-high cross-linking technology, post-surface modification, and the addition of nanomaterials, are discussed. | 7 | Methionine deficiency and its hydroxy analogue influence chicken intestinal 3-dimensional organoid development显示文摘Methionine and its hydroxy analogue(MHA)have been shown to benefit mouse intestinal regeneration.The intestinal organoid is a good model that directly reflects the impact of certain nutrients or chemicals on intestinal development.Here,we aimed to establish a chicken intestinal organoid culture method first and then use the model to explore the influence of methionine deficiency and MHA on intestinal organoid development.The results showed that 125-mm cell strainer exhibited the highest efficiency for chicken embryo crypt harvesting.We found that transforming growth factor-b inhibitor(A8301)supplementation promoted enterocyte differentiation at the expense of the proliferation of intestinal stem cells(ISC).The mitogen-activated protein kinase p38 inhibitor(SB202190)promoted intestinal organoid formation and enterocyte differentiation but suppressed the differentiation of enteroendocrine cells,goblet cells and Paneth cells.However,the suppression of enteroendocrine cell and Paneth cell differentiation by SB202190 was alleviated at the presence of A8301.The glycogen synthase kinase 3 inhibitor(CHIR99021),valproic acid(VPA)alone and their combination promoted chicken intestinal organoid formation and enterocyte differentiation at the expense of the expression of Paneth cells and goblet cells.Chicken serum significantly improved organoid formation,especially in the presence of A8301,SB202190,CHIR99021,and VPA,but inhibited the differentiation of Paneth cells and enteroendocrine cells.Chicken serum at a concentration of 0.25%meets the requirement of chicken intestinal organoid development,and the beneficial effect of chicken serum on chicken intestinal organoid culture could not be replaced by fetal bovine serum and insulin-like growth factor-1.Moreover,commercial mouse organoid culture medium supplemented with A8301,SB202190,CHIR99021,VPA,and chicken serum promotes chicken organoid budding.Based on the chicken intestinal organoid model,we found that methionine deficiency mimicked by cycloleucine suppressed organoid formation and organoid size,and this effect was reinforced with increased cycloleucine concentrations.Methionine hydroxy analogue promoted regeneration of ISC but decreased cell differentiation compared with the results obtained with L-methionine.In conclusion,our results provide a potentially excellent guideline for chicken intestinal organoid culture and insights into methionine function in crypt development. | Youli Wang Qihang Hou Yuqin Wu Yanwei Xu Yan Liu Jing Chen Lingling Xu Yuming Guo Shuai Gao Jianmin Yuan | 2022 | Animal Nutrition2022,,1: | 4 |

| 9 | 细胞因子诱导杀伤细胞治疗肾细胞癌的疗效观察(英文)显示文摘Objective: To observe the effects of cytokine-induced killer (CIK) cells on the treatment of renal cell carcinoma. Methods: Twenty-eight postoperative cases with stage I or stage II renal cell carcinoma were admitted in our hospital from January 2002 to June 2006, all cases were pathologically confirmed, and were divided into group A (18 cases) and group B (10 cases). Group A was administrated 3–12 periods of CIK cells treatment combined with 5–7 cycles of IL-2 and INFα-2b, together with 4 cycles of chemotherapy (5-Fu + CF). Group B was given 4 cycles of chemotherapy (5-Fu + CF) and 5–7 cycles of IL-2 and INFα-2b. Results: Three cases in group A had metastatic masses in two lungs within 1 year and died within 2 years postoperatively. The other 15 cases are still alive and in good health. Six cases in group B had metastatic masses in two lungs or/and in abdominal cavity within 1 year, and 4 of them died within 2 years. All the 6 cases died within 3 years. The other 4 cases are still alive and in good health. | 10 | Three-Dimensional In Situ Stress-Field Simulations of Shale Gas Formations:A Case Study of the 5th Member of the Xujiahe Formation in the Xinchang Gas Field,West Sichuan Depression显示文摘This work established a geological model for the 5^(th) member of the Xujiahe Formation(X5 member) in the Xinchang gas field of the West Sichuan Depression based on the lithological, structural and depositional properties, as well as logging and well completion data and drill-core observations. Rock mechanical parameters were calculated according to rock mechanic experiments and rock mechanic interpretations from logging data. We also calculated the magnitudes and orientations of the in situ stresses based on acoustic emission tests, differential strain tests, fracturing behaviour and logging interpretations as well as anisotropy logging tests, borehole-breakout measurements and well-log data. Additionally, the present stress field of the X5 member was simulated using finite element numerical(FEM) simulation methods. The numerical simulation results indicate that the distributions of lithology and fractures are key factors that influence the present stress field. The stress field in the study area is discontinuous as a result of fractures and faults in the central and eastern areas. Stress is concentrated at the end sections and bends of faults, but dissipates with distance away from both sides of the faults. A longitudinal profile clearly demonstrates the zonality and continuity of the stress field and an increase with depth. | 14 | ATP生物荧光肿瘤药敏在复发卵巢癌并腹水化疗中的应用价值(英文)显示文摘Objective: To investigate the clinical value and application of ATP based bioluminescence tumor chemo-sensitivity assay (ATP-TCA) in the chemotherapy for ascites caused by recurrent ovarian cancer. Methods: More than 10 kinds of chemotherapeutic drugs or combinations were applied and 35 ascites specimens from recurrent ovarian cancer were analyzed by ATP-TCA. Sensitivity of chemotherapeutic drugs was assessed. After 2-4 chemotherapeutic cycles, clinical outcomes were analyzed, which were compared with those of 40 cases by empirical regimens. Results: 32 of 35 specimens were evaluated with an overall evaluation rate of 91%. The assay results suggested that chemo-naive patients responded to chemotherapeutic drugs with individualized profiles. The sensitivity rates of GEM, EPI, OXA, DDP, CBP, ADM, VP-16, CTX, NVB, 5-FU, PTX and TXT were 40%, 30%, 33%, 29%, 33%, 38%, 25%, 33%, 38%, 33%, 25% and 20%, respectively. While the sensitivity rates of combinations GEM + EPI, GEM + CBP, GEM + DDP, NVB + DDP, CTX + ADM + DDP, CTX + ADM, DDP + VP-16, OXA + 5-FU, VP-16 + IFO, PTX + DDP, TXT + CBP, VCR + CTX + MTX, DDP + CPT-11, OXA + CPT-11, and DTIC + CTX were 47%, 50%, 36%, 44%, 30%, 33%, 27%, 33%, 40%, 27%, 23%, 14%, 28%, 30% and 17%, respectively. In vitro results correlated well with clinical outcomes. Objective response rate (RR) in chemo-sensitivity-guided group was of significance compared with that in empirical-regimen-guided group. Conclusion: ATP-TCA is a choice for the screening of chemotherapeutic drugs against ascites caused by recurrent ovarian cancer with excellent sensitivity and reliability. | 16 | 肿瘤药敏指导下的化疗对非小细胞肺癌并恶性胸水治疗的有效性(英文)显示文摘Objective:The aim of the study was to investigate the clinical value and application of ATP based bioluminescence tumor chemosensitivity assay (ATP-TCA) in the chemotherapy for hydrothorax caused by non-small cell lung cancer (NSCLC). Methods:Hydrothorax specimens from 120 NSCLC patients were analyzed by ATP-TCA and the most sensitive chemotherapeutic drugs were used in NSCLC patients (treatment group). At the same time, 56 NSCLC patients with hydrothorax were admitted in our Hospital (Department of Oncology, The No. 2 People's Hospital of Yibin, China) and given chemotherapy without guidance of the ATP-TCA (control group). Before the third chemotherapeutic cycle, clinical outcomes were analyzed in the two groups. Results:Effective rate of hydrothorax in treatment group was 67%, while 46% in control group (P < 0.05). In refractory hydrothorax patients, they were 69% and 40% (P < 0.05), respectively. In vitro results correlated well with clinical outcomes (P < 0.01). Conclusion:Effective rate of chemotherapy for hydrothorax in NSCLC is higher in treatment group than that in control group.
