2Contaminants of emerging concern in landfill leachate in China:A review显示文摘This review paper summarizes the occurrence,removal and ecological risk of contaminants of emerging concern(CEC)reported in landfill leachate in China since 1996(43 studies in 10 regions).Results show that many more studies are conducted in developed southeastern China than in developing western and northeastern regions in China.Phthalate esters(PAEs,with 15 studies)and pharmaceuticals and personal care products(PPCPs,with 13 studies)are the two most frequently studied CEC classes.Concentrations of nine CECs classes were in a wide range from 0.03(organochlorine pesticides)to approximately 4500 mg/L(alkylphenol polyethoxylates/bisphenol analog).Meanwhile,concentrations of CEC compounds range from below detection limit(e.g.doxycycline)to approximately 4500 mg/L(bisphenol A).Several PAEs(diethyl phthalate,di-n-butyl phthalate,and di(2-ethylhexyl)phthalate)and PPCPs(diclofenac and gemfibrizol)have significant variation between sampling sites.Typically,advanced treatment processes can achieve higher removal efficiencies of CEC compounds from landfill leachate compared with conventional treatment processes.Furthermore,environmental risk assessments of CEC compounds in treated landfill leachate using a risk quotient method show that 2(substituted)polycyclic aromatic hydrocarbons(sPAHs)(benzo(a)anthracene and benzo(b)fluoranthene),2 PPCPs(bezafibrate and sulfapyridine),g-hexachlorocyclohexane,and bisphenol A pose high risk.The importance of monitoring and potential risks of CECs in the leachate to vicinity aquatic environment cross China is addressed.Chengdu Qi Jun Huang Bin Wang Shubo Deng Yujue Wang Gang Yu 2018Emerging Contaminants2018,4,1:10

6Weighted Correlation Network Analysis(WGCNA) of Japanese Flounder(Paralichthys olivaceus) Embryo Transcriptome Provides Crucial Gene Sets for Understanding Haploid Syndrome and Rescue by Diploidization显示文摘Artificial gynogenesis is of great research value in fish genetics and breeding technology. However, existing studies did not explain the mechanism of some interesting phenomena. Severe developmental defects in gynogenetic haploids can lead to death during hatching. After diploidization of chromosomes, gynogenetic diploids may dispense from the remarkable malformation and restore the viability, although the development time is longer and the survival rate is lower compared with normal diploids. The aim of this study was to reveal key mechanism in haploid syndrome of Japanese flounder, a commercially important marine teleost in East Asia. We measured genome-scale gene expression of flounder haploid, gynogenetic diploid and normal diploid embryos using RNA-Seq, constructed a module-centric co-expression network based on weighted correlation network analysis(WGCNA) and analyzed the biological functions of correlated modules. Module gene content analysis revealed that the formation of gynogenetic haploids was closely related to the abnormality of plasma proteins, and the up-regulation of p53 signaling pathway might rescue gynogenetic embryos from haploid syndrome via regulating cell cycle arrest, apoptosis and DNA repair. Moreover, normal diploid has more robust nervous system. 9Occurrence and discharge of pharmaceuticals and personal care products in dewatered sludge from WWTPs in Beijing and Shenzhen显示文摘This study mainly focused on the occurrence of pharmaceutical and personal care products(PPCPs)in sewage dewatered sludge and their discharges through sludge disposal from wastewater treatment plants(WWTPs).The data were obtained and calculated from seven PPCPs in dewatered sludge collected from 12WWTPs in two typical cities,Beijing and Shenzhen in China.Four of seven PPCPs,diclofenac acid,carbamazepine,mefenamic acid and N,N-diethyl-m-toluamide were detectable in dewatered sludge from Beijing and Shenzhen with concentration up to 4240,11,060,92 and 219μg/kg respectively.While,the other three compounds,trimethoprim,chloramphenicol and bezafibrate were not detected in collected samples from these two cities.The highest discharge of diclofenac acid and carbamazepine were 1023 g/d and 494 g/d respectively.In addition,the total discharge of these four detected PPCPs from each plant ranged from 5 to 1092 g/d in Beijing and 4e497 g/d in Shenzhen.Thus,PPCP discharge through sludge disposal cannot be neglected,and further research on transfer of PPCPs during sludge disposal onto agriculture land and influence of sludge application is required and essential.Rui Dong Gang Yu Yuntao Guan Bin Wang Jun Huang Shubo Deng Yujue Wang 2016Emerging Contaminants2016,2,1:2
10Evaluation of the technoeconomic feasibility of electrochemical hydrogen peroxide production for decentralized water treatment显示文摘This study evaluated the feasibility of electrochemical hydrogen peroxide(H2O2)production with gas diffusion electrode(GDE)for decentralized water treatment.Carbon black-polytetrafluoroethylene GDEs were prepared and tested in a continuous flow electrochemical cell for H2O2 production from oxygen reduction.Results showed that because of the effective oxygen transfer in GDEs,the electrode maintained high apparent current efficiencies(ACEs,>80%)for H_(2)O_(2) production over a wide current density range of 5–400 mA/cm^(2),and H_(2)O_(2) production rates as high as~202 mg/h/cm^(2) could be obtained.Long-term stability test showed that the GDE maintained high ACEs(>85%)and low energy consumption(<10 kWh/kg H2O2)for H_(2)O_(2) production for 42 d(~1000 h).However,the ACEs then decreased to~70%in the following 4 days because water flooding of GDE pores considerably impeded oxygen transport at the late stage of the trial.Based on an electrode lifetime of 46 days,the overall cost for H2O2 production was estimated to be~0.88$/kg H_(2)O_(2),including an electricity cost of 0.61$/kg and an electrode capital cost of 0.27$/kg.With a 9 cm^(2) GDE and 40 mA/cm^(2) current density,~2–4 mg/L of H2O2 could be produced on site for the electro-peroxone treatment of a 1.2 m^(3)/d groundwater flow,which considerably enhanced ibuprofen abatement compared with ozonation alone(~43%–59%vs.7%).These findings suggest that electrochemical H_(2)O_(2) production with GDEs holds great promise for the development of compact treatment technologies for decentralized water treatment at a household and community level.Yang Li Yixin Zhang Guangshen Xia Juhong Zhan Gang Yu Yujue Wang 2021Frontiers of Environmental Science & Engineering2021,15,1:2
11Direct conversion of mouse astrocytes into neural progenitor cells and specific lineages of neurons显示文摘Background:Cell replacement therapy has been envisioned as a promising treatment for neurodegenerative diseases.Due to the ethical concerns of ESCs-derived neural progenitor cells(NPCs)and tumorigenic potential of iPSCs,reprogramming of somatic cells directly into multipotent NPCs has emerged as a preferred approach for cell transplantation.Methods:Mouse astrocytes were reprogrammed into NPCs by the overexpression of transcription factors(TFs)Foxg1,Sox2,and Brn2.The generation of subtypes of neurons was directed by the force expression of cell-type specific TFs Lhx8 or Foxa2/Lmx1a.Results:Astrocyte-derived induced NPCs(AiNPCs)share high similarities,including the expression of NPC-specific genes,DNA methylation patterns,the ability to proliferate and differentiate,with the wild type NPCs.The AiNPCs are committed to the forebrain identity and predominantly differentiated into glutamatergic and GABAergic neuronal subtypes.Interestingly,additional overexpression of TFs Lhx8 and Foxa2/Lmx1a in AiNPCs promoted cholinergic and dopaminergic neuronal differentiation,respectively.Conclusions:Our studies suggest that astrocytes can be converted into AiNPCs and lineage-committed AiNPCs can acquire differentiation potential of other lineages through forced expression of specific TFs.Understanding the impact of the TF sets on the reprogramming and differentiation into specific lineages of neurons will provide valuable strategies for astrocyte-based cell therapy in neurodegenerative diseases.Kangmu Ma Xiaobei Deng Xiaohuan Xia Zhaohuan Fan Xinrui Qi Yongxiang Wang Yuju Li Yizhao Ma Qiang Chen Hui Peng Jianqing Ding Chunhong Li Yunlong Huang Changhai Tian Jialin C.Zheng 2018Translational Neurodegeneration2018,7,1:2

16Large-Scale Screening of Growth-Related Variants in Chinese Tongue Sole(Cynoglossus semilaevis)显示文摘Growth is the most valuable economic trait for improving aquaculture fish species,since fast growth can reduce labor cost and make more economic benefits.However,the knowledge about how many and which genes are related to the growth of Chi-nese tongue sole is limited.High-throughput sequencing screening of variants is a fast,economical and accurate assay to identify genes related to growth in crops,livestock and other aquaculture products.In the present study,genome-wide resequencing of 30 female Chinese tongue sole individuals from large and small groups to screen growth trait-related variations(SNPs,InDels)was carried out.In total,6545735 SNPs and 1016745 InDels were detected,while 31 genes related to growth traits were identified.Their functions were mainly involved in muscle architecture,post-embryonic development,neurosensory development and hormone regulation.Fur-thermore,18 of the 31 genes related to growth trait are located on W chromosome,indicating that W chromosome plays an important role in regulating the body size of female tongue soles.The markers and genes identified in our study can be applied to selective breeding of tongue sole and thus will promote the aquaculture industry and increase economic efficiency.SONG Weihao ZHU He WANG Yujue ZHANG Kai ZHANG Quanqi HE Yan 2021Journal of Ocean University of China2021,20,3:1
17Serial deletion reveals structural basis and stability for the core enzyme activity of human glutaminase 1 isoforms:relevance to excitotoxic neurodegeneration显示文摘Background:Glutaminase 1 is a phosphate-activated metabolic enzyme that catalyzes the first step of glutaminolysis,which converts glutamine into glutamate.Glutamate is the major neurotransmitter of excitatory synapses,executing important physiological functions in the central nervous system.There are two isoforms of glutaminase 1,KGA and GAC,both of which are generated through alternative splicing from the same gene.KGA and GAC both transcribe 1–14 exons in the N-terminal,but each has its unique C-terminal in the coding sequence.We have previously identified that KGA and GAC are differentially regulated during inflammatory stimulation and HIV infection.Furthermore,glutaminase 1 has been linked to brain diseases such as amyotrophic lateral sclerosis,Alzheimer’s disease,and hepatic encephalopathy.Core enzyme structure of KGA and GAC has been published recently.However,how other coding sequences affect their functional enzyme activity remains unclear.Methods:We cloned and performed serial deletions of human full-length KGA and GAC from the N-terminal and the C-terminal at an interval of approximately 100 amino acids(AAs).Prokaryotic expressions of the mutant glutaminase 1 protein and a glutaminase enzyme activity assay were used to determine if KGA and GAC have similar efficiency and efficacy to convert glutamine into glutamate.Results:When 110 AAs or 218 AAs were deleted from the N-terminal or when the unique portions of KGA and GAC that are beyond the 550 AA were deleted from the C-terminal,KGA and GAC retained enzyme activity comparable to the full length proteins.In contrast,deletion of 310 AAs or more from N-terminal or deletion of 450 AAs or more from C-terminal resulted in complete loss of enzyme activity for KGA/GAC.Consistently,when both Nand C-terminal of the KGA and GAC were removed,creating a truncated protein that expressed the central 219 AA-550 AA,the protein retained enzyme activity.Furthermore,expression of the core 219 AA-550 AA coding sequence in cells increased extracellular glutamate concentrations to levels comparable to those of full-length KGA and GAC expressions,suggesting that the core enzyme activity of the protein lies within the central 219 AA-550 AA.Full-length KGA and GAC retained enzyme activities when kept at 4°C.In contrast,219 AA-550 AA truncated protein lost glutaminase activities more readily compared with full-length KGA and GAC,suggesting that the Nterminal and C-terminal coding regions are required for the stability KGA and GAC.Conclusions:Glutaminase isoforms KGA and GAC have similar efficacy to catalyze the conversion of glutamine to glutamate.The core enzyme activity of glutaminase 1 protein is within the central 219 AA-550 AA.The N-terminal and C-terminal coding regions of KGA and GAC help maintain the long-term activities of the enzymes.Yuju Li Justin Peer Runze Zhao Yinghua Xu Beiqing Wu Yi Wang Changhai Tian Yunlong Huang Jialin Zheng 2017Translational Neurodegeneration2017,6,1:1

20Discharge inventory of pharmaceuticals and personal care products in Beijing,China显示文摘Pharmaceuticals and personal care products(PPCPs)are emerging environmental contaminants,whose potential risk for the ecological environment has caused wide attention in recent years.In China,quite a large amount of PPCPs were annually emitted into the environment.Their existence in different matrix has been reported frequently,including river water,sediment and soil.However,the contribution from different sources was seldom reported and still unclear in China.Wastewater treatment plant(WWTP)was usually considered to be the main source to the urban river,but livestock and aquaculture farms were also reported as significant pollution sources of PPCPs due to poor environmental management in China.This study summarized environmental discharges of different PPCPs from various sources and obtained the discharge data through different environment media in Beijing,the statistical source of PPCPs was analyzed in detail.The sources comprised WWTPs,excess sludge,hospital wastewater,municipal untreated wastewater,aquaculture wastewater and landfill leachate.This article helps understand the general situation and the potential risk of PPCPs in Beijing.Zhixia Zhang Bin Wang Honglin Yuan Ruixue Ma Jun Huang Shubo Deng Yujue Wang Gang Yu 2016Emerging Contaminants2016,2,3:1
