| 2 | Drug resistance mutation profiles of the drug-nave and first-line regimen-treated HIV-1-infected population of Suzhou, China显示文摘Little is known about the prevalence of drug-resistant mutations in HIV-1-positive individuals in Suzhou, China. To elucidate the transmitted drug resistance(TDR) and acquired drug resistance mutation(ADR) profiles, we collected blood specimens from 127 drug-naive and 117 first-line drugtreated HIV-1-infected individuals sampled from 2014 to 2016 in Suzhou. We successfully amplified po/fragments from 100 drug-naive and 20 drug-treated samples. We then determined the drugresistant mutations to protease(PR) and reverse-transcriptase(RT) inhibitors according to the Stanford drug resistance database. Overall, 11 and 13 individuals had transmitted(drug-naive group) and acquired(treated group) resistance mutations, respectively. Six transmitted drugresistant mutations were found, including two mutations(L33F and L76V) in the protease region and four(K70N/E and V179D/E) in the RT region. Only L76 V was a major mutation, and K70N/E and V179D/E are known to cause low-level resistance to RT inhibitors. All 13 treated participants who had major drug resistance mutations demonstrated intermediate to high resistance to efavirenz and nevirapine, and six had a treatment duration of less than three months. No major mutations to RT inhibitors were found, implying that the epidemic of transmitted resistance mutations was not significant in this area. Our results suggest that more frequent virus load and drug resistance mutation tests should be conducted for individuals receiving antiretroviral treatment, especially for newly treated patients. | 8 | Impact of China's SO_2 Emissions on SO_2 Concentration and Sulfur Deposition in Japan显示文摘Based on the monitored data of references,EANET and the CMAQ( 4. 7. 1) model scenario simulations,the annual average concentration of SO2 in China in the recent 10 years and the contribution of China's SO2 emissions to SO2 concentration and sulfur deposition at various sites in Japan downwind were analyzed. The results showed that from 2001 to 2009 annual average concentration of SO2 was 0- 150 μg / m3 and showed a decreasing trend on the whole in China. The correlation coefficient of annual average concentration of SO2 at different sites in Japan and China's SO2 emissions was lower than that of annual average concentration and emission of SO2 in China's cities on the whole,showing that SO2 discharged in China had small impact on SO2 concentration in Japan. | 10 | The E3 ubiquitin ligase CSIT1 regulates critical sterility-inducing temperature by ribosome-associated quality control to safeguard two-line hybrid breeding in rice显示文摘Two-line hybrid breeding can fully utilize heterosis in crops.In thermo-sensitive genic male sterile(TGMS)lines,low critical sterility-inducing temperature(CSIT)is vital to safeguard the production of two-line hybrid seeds in rice(Oryza sativa),but the molecular mechanism determining CSIT is unclear.Here,we report the cloning of CSIT1,which encodes an E3 ubiquitin ligase,and show that CSIT1 modulates the CSIT of thermo-sensitive genic male sterility 5(tms5)-based TGMS lines through ribosome-associated quality control(RQC).Biochemical assays demonstrated that CSIT1 binds to the 80S ribosomes and ubiquitinates abnormal nascent polypeptides for degradation in the RQC process.Loss of CSIT1 function inhibits the possible damage of tms5 to the ubiquitination system and protein translation,resulting in enhanced accumulation of anther-related proteins such as catalase to suppress abnormal accumulation of reactive oxygen species and premature programmed cell death in the tapetum,thereby leading to a much higher CSIT in the tms5-based TGMS lines.Taken together,our findings reveal a regulatory mechanism of CSIT,providing new insights into RQC and potential targets for future two-line hybrid breeding. | Guoqing Peng Minglong Liu Liya Zhu Wenlong Luo Qinghua Wang Mumei Wang Huiqiong Chen Ziliang Luo Yueping Xiao Yongjie Zhang Haona Hong Zhenlan Liu Lingyan Zhou Guoqiang Guo Yingxiang Wang Chuxiong Zhuang Hai Zhou | 2023 | Molecular Plant2023,16,10: | 0 |
