1Genetic basis of high aroma and stress toleranee in the oolong tea cultivar genome显示文摘Tea plants(Camellia sinensis)are commercially cultivated in>60 countries,and their fresh leaves are processed into tea,which is the most widely consumed beverage in the world.Although several chromosome-level tea plant genomes have been published,they collapsed the two haplotypes and ignored a large number of allelic variations that may underlie important biological functions in this species.Here,we present a phased chromosome-scale assembly for an elite oolong tea cultivar,'Huangdan',that is well known for its high levels of aroma.Based on the two sets of haplotype genome data,we identi fi ed numerous genetic variations and a substantial proportion of allelic imbalance related to important traits,including aroma-and stress-related alleles.Comparative genomics revealed extensive structural variations as well as expansion of some gene families,such as terpene synthases(TPSs),that likely contribute to the high-aroma characteristics of the backbone parent,underlying the molecular basis for the biosynthesis of aroma-related chemicals in oolong tea.Our results uncovered the genetic basis of special features of this oolong tea cultivar,providing fundamental genomic resources to study evolution and domestication for the economically important tea crop.Pengjie Wang Jiaxin Yu Shan Jin Shuai Chen Chuan Yue Wenling Wang Shuilian Gao Hongli Cao Yucheng Zheng Mengya Gu Xuejin Chen Yun Sun Yuqiong Guo Jiangfan Yang Xingtan Zhang Naixing Ye 2021Horticulture Research2021,8,1:4
2Identification of PAL genes related to anthocyanin synthesis in tea plants and its correlation with anthocyanin content显示文摘The phenylalanine ammonia-lyase(PAL)gene family in tea plants(Camellia sinensis L.)encodes the enzyme that catalyzes the first reaction of the phenylpropane metabolic pathway.The present study aimed to characterize the PAL genes in tea plants,and get better insights on the CsPALs in anthocyanins accumulation.Seven CsPAL genes were identified and characterized in tea plants by bioinformatics analysis.Systematic analysis of CsPALs was conducted for its phylogenetic relationship,gene structure,chromosomal location,and protein conserved motifs based on tea plant genome.The cis-elements of CsPALs were responsive to light,abiotic stress,hormone,and MYB-binding site.Furthermore,tissuespecific expression analysis showed that CsPAL4 was expressed preferentially in young leaves and buds.Correlation analysis was performed in purple-leaf tea with anthocyanin components,and it was suggested that CsPAL4 was closely related with different anthocyanin accumulated,especially with cyanidin 3-O-galactoside,cyanidin 3-O-glucoside,and delphinidin 3-O-glucoside.Additionally,the putative upstream regulation factors CsMYBs(CsMYB59,CsARR1,CsSRM1,CsMYB101,and CsMYB52)and CsbHLHs(CsbHLH104,CsbHLH3,CsbIM1,CsTCP14,and CsPIF4)could bind to the promoter of CsPALs,thereby activating its transcription.This study provides a theoretical basis for further research to elucidate the functions of the CsPAL genes.Xuejin Chen Pengjie Wang Mengya Gu Binghao Hou Churan Zhang Yucheng Zheng Yun Sun Shan Jin Naixing Ye 2022Horticultural Plant Journal2022,8,3:4

4Research on controllable alkaline soil degradation of 5-substituted chlorsulfuron显示文摘Sulfonylurea(SU) has become one class of the most important herbicides worldwide due to their ultralow application rate and mammalian toxicity. Recently application licenses of 3 classical SU herbicides chlorsulfuron, metsulfuron-methyl and ethametsulfuron were suspended due to their undesirable long persistence which is incompatible with the particular rotation planting system in China. Our previous study has discovered that electron-donating groups, especially dimethylamino and diethylamino substituents, on the 5 th position of the benzene ring in chlorsulfuron, greatly accelerated its degradation rate in an acidic soil(p H 5.41). Owing to the natural slower degradation of SUs in alkaline soil,dimethylamino and diethylamino substituted chlorsulfuron Ia and Ib were further studied in an alkaline soil(p H 8.46) with chlorsulfuron as a control. The experimental data indicated that the half-life of degradation(DT_(50)) of Ia was 3.36 days while Ib was 6.25 days which amounted to 30 and 15 folds faster than chlorsulfuron(DT_(50)84.53 days), respectively. The research confirmed that our newly-designed structures Ia and Ib can hasten their degradation rate in alkaline soil as well as in acidic soil. 8Food resources of the Khog Gzung site on the Tibetan Plateau revealed by sedimentary ancient DNA显示文摘Traditional zooarchaeological and archaeobotanical methods based on morphological identification of the excavated faunal and floral remains have been broadly used in reconstructing ancient subsistence economies. However, the accuracy and reliability of these methods rely heavily on the preservation state of the remains. By sequencing the ancient DNA of plants,animals, and microorganisms preserved in sediment, sedimentary ancient DNA(sedaDNA) now offers a novel approach for reconstructing the taxa composition dated back to hundreds of thousands of years. Yet, its application in open-air archaeological sites is rarely reported. In this study, we attempted to apply sedaDNA shotgun metagenomics on the archaeological deposits of the Khog Gzung site(an open-air site dated to 3160–2954 cal yr BP) on the Tibetan Plateau, and then compared the reconstructed taxonomic composition to the unearthed remains. Results showed that most of the crops and domestic animals identified by the two approaches, such as barley(Hordeum vulgare) and sheep(Ovis aries), are in general consistent. Some species, such as foxtail millet(Setaria italica), however, were only detected by sedaDNA. In addition, a variety of microorganisms were also detected by the sedaDNA. The two approaches combined revealed diversified food resources at the Khog Gzung site, which included crops such as millet, barley and wheat, domestic animals such as sheep and cattle, and likely also wild animals from fishing and hunting. 9Defect-rich and highly porous carbon nanosheets derived from Ti_(3)AlC_(2) MAX with good lithium storage properties显示文摘Defect-rich,highly porous two-dimensional carbon nanosheets(CNS) have attracted tremendous research interests in catalysis and environmental purification and other fields,because of their unique micromorphology,chemical stability and high specific surface area.Herein,in this work,we report a new solution to synthesize an ultrathin two-dimensional CNS with rich defects and abundant pores via two-step etching the Ti_(3)AlC_(2)with the help of I2and NaOH.The CNS thickness,specific surface area and pore volume could be all tunable by adding the amount of I2.And the highest specific surface area and pore volume of the synthesized 2D CNS can be achieved 1134.4 m^(2)/g and 0.80 cm^(3)/g,with a thickness of only 0.64 nm and a yield of 35.9%.When employed as the anodes for lithium-ion batteries,the synthesized CNS anodes exhibit good cycling and rate capabilities.This work provides a novel and facile strategy for synthesizing highly porous and defective 2D carbon materials with good lithium storage properties.Zhihao Zhu Peng Liu Yucheng Liu Xingxing Gu Zhi Su Zhong Su Chao Lai 2023Chinese Chemical Letters2023,34,2:0

11N-acetyldopamine dimer inhibits neuroinflammation through the TLR4/NF-κB and NLRP3/Caspase-1 pathways显示文摘Neuroinflammation mediated by microglia is an important pathophysiological mechanism in neurodegenerative diseases.However,there is a lack of effective drugs to treat neuroinflammation.N-acetyldopamine dimer(NADD)is a natural compound from the traditional Chinese medicine Isaria cicada.In our previous study,we found that NADD can attenuate DSS-induced ulcerative colitis by suppressing the NF-κB and MAPK pathways.Does NADD inhibit neuroinflammation,and what is the target of NADD?To answer this question,lipopolysaccharide(LPS)-stimulated BV-2 microglia was used as a cell model to investigate the effect of NADD on neuroinflammation.Nitric oxide(NO)detection,reactive oxygen species(ROS)detection and enzyme-linked immunosorbent assay(ELISA)results show that NADD attenuates inflammatory signals and proinflammatory cytokines in LPS-stimulated BV-2 microglia,including NO,ROS,tumor necrosis factor(TNF)-α,interleukin(IL)-1βand interleukin-6(IL-6).Western blot analysis show that NADD inhibits the protein levels of Toll-like receptor 4(TLR4),nuclear factor kappa-B(NF-κB),NOD-like receptor thermal protein domain associated protein 3(NLRP3),ASC and cysteinyl aspartate specific proteinase(Caspase)-1,indicating that NADD may inhibit neuroinflammation through the TLR4/NF-κB and NLRP3/Caspase-1 signaling pathways.In addition,surface plasmon resonance assays and molecular docking demonstrate that NADD binds with TLR4 directly.Our study reveals a new role of NADD in inhibiting the TLR4/NF-κB and NLRP3/Caspase-1 pathways,and shows that TLR4-MD2 is the direct target of NADD,which may provide a potential therapeutic candidate for the treatment of neuroinflammation.Lijun Huang Leiqiang Gong Xueyan Huo Lirong Lei Qi Zhang Yunjie Hu Qixuan Kuang Yu Gui Yifei Dai Yucheng Gu Yun Deng Dong Wang Dale Guo 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
12The Quantification of Micro-structural Damage of Weak Muddy Intercalation in Dry-wet Cycles Combining in-situ SEM and DIP显示文摘In order to reflect truly the damage evolution mechanism of weak muddy intercalation in dry-wet cycles, two typical weak muddy intercalations were selected for dry-wet cycles. The mineral changes of specimens were analyzed via X-ray diffraction after dry-wet cycles. By combining in-situ SEM and digital image processing(DIP), the damage evolution process and damage characteristic parameters of each stage were obtained. The experimental results indicate that the hydration and dissolution of minerals can not be a determinant factor in structure damage. The micro-structural damage is due to disintegration of mineral aggregates, leading to changes in the number and size of cracks and pores.
