3Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. 4Function and regulation of cholecystokinin-octapeptide, β-endorphin and gastrin in anorexic infantile rats treated with ErBao Granules显示文摘AIM To study the role of cholecystokinin octapeptide ( CCK-8), β-endorphin ( β-EP), and gastrin in an anorexic infantile rat model and no subsequent regulation of nose peptides by the Yunpi complex prescription ErBao Granule.METHODS We fed infantile rats with special prepared forage. A liquid extract of ErBao Granule was administered to the rats daily for 3weeks, CCK-8, β-EP, and gastrin concentrations in hypothalamus, gastric antrum, and plasma of the rats were measured by radioimmunoassay,and were compared with controls.RESULTS Treatment of rats with ErBao Granule inhibited CCK-8 secretion and increased β-EP and gastrin secretion. CCK-8 concentration in hypothalamus and plasma of model control group increased significantly and correlated negatively with food intake of models.respectively. β-EP concentration in gastric antrum and plasma of model control group decreased significantly and showed a positive correlation with food intake of models,respectively. Hypothalamus concentration of β-EP was similar in models and controls. Gastrin concentration in gastric antrum of models was lower than in the blank control group, and correlated positively to food intake of models.Finally, CCK-8 concentrations in plasma of rats showed a positive correlation with plasma β-EP(r- 0.68, P<0.05).CONCLUSION The increased plasma and hypothalamus concentration of CCK-8,decreased gastric antrum and plasma level of β-EP. and decreased gastric antrum concentration of gastrin are associated significantly with the anorexia of infantile anorexic rat models produced by special forage. 6Insight into fouling behavior of poly(vinylidene fluoride)(PVDF)hollow fiber membranes caused by dextran with different pore size distributions显示文摘Membrane fouling is the key problem that occurs in membrane process for water treatment. However, how membrane microstructure influences the fouling behavior is still not clear. In this study, fouling behavior caused by dextran was deeply and systematically investigated by employing four poly(vinylidene fluoride)(PVDF)membranes with different pore sizes, ranging from 24 to 94 nm. The extent of fouling by dextran was accurately characterized by pore reduction, flux decline, and the change of critical flux. The result shows that membrane with the smallest pore size of 24 nm experienced the smallest fouling rate and the lowest fouling extent. As the membrane pore size increased, the critical flux ranges were 105-114, 63-73, 38-44 and 34-43 L·m^(-2)·h^(-1),respectively. The critical flux and fouling resistances indicated that the fouling propensity increases with the increase of membrane pore size. Two pilot membrane modules with mean pore size of 25 nm and 60 nm were applied in membrane filtration of surface water treatment. 12A Difference Method of the Gravity Center with Double Pilots for the MC CDMA System显示文摘A carrier recovery method of the power spectrum center difference adapting to the mobile channel of the MC CDMA system with serious Doppler shift using double pilots is presented. In the transmitter, two pilots of equal frequency distance to the carrier with one putting on the left position and the other on the right position of the carrier frequency are used. Even if a continuous sine wave is transmitted, the power spectrum is expanded to a Doppler band in the receiver owing to the multi path transmission and the Doppler shift. The pilot spectrum is made to pass through two narrow band filters which are transformed to the base band with local carrier wave in the receiver. 16Comparative chemical characters of Sparganii Rhizoma from different regions in China显示文摘Objective: To compare the chemical characters of Sparganii Rhizoma from different areas via chromatographic analysis and to establish a sensitive LC/MS method for quality assessment of Sparganii Rhizoma.Methods: Under the optimised HPLC-PDA chromatographic conditions,twenty batches of Sparganii Rhizoma were analyzed by HPLC fingerprints.Principal component analysis(PCA),orthogonal projections to latent structures discriminant analysis(OPLS-DA)and hierarchical cluster analysis(HCA)were performed based on all peak areas of Sparganii Rhizoma fingerprints.Meanwhile,part of common peaks were subsequently quantified by UFLC-QTRAP-MS.Results: The similarity values of HPLC fingerprints fluctuated in a wide range of 0.511–0.973,which showed variable differences of chemical characters among Sparganii Rhizoma from twenty habitats.PCA,OPLS-DA and HCA indicated that samples could be divided into five groups with different chemical characters,which generally corresponded with their geographical distributions.A total of 31 peaks in HPLC fingerprints were marked,and eight of them were identified and quantified.The quantitative result was generally in agreement with the classifications based on HPLC fingerprints,which indicated that Sparganii Rhizoma samples from eastern China mostly contained more contents including phenolic acids and flavonoids.Conclusion: This study not only proved that there were relationships between geographic distributions and internal chemical compositions of plants,which could provide evidence to the traditional Chinese medicine concept 'geo-authentic',but also supplied a sensitive and rapid simultaneous quantitive method for the quality estimation of Sparganii Rhizoma.Meng-ru Sang Xiao-long Zhang Qi-nan Liu Chan-chan Liu Yi-ming Xu Ling-ling Zhao Qi-nan Wu Wei Yue Chuan Chai 2018Chinese Herbal Medicines2018,10,1:2
17Chromatin accessibility and translational Iandscapes of tea plants under chilling stress显示文摘Plants have evolved regulatory mechanisms at multiple levels to regulate gene expression in order to improve their cold adaptability.However,limited information is available regarding the stress response at the chromatin and translational levels.Here,we characterize the chromatin accessibility,transcriptional,and translational landscapes of tea plants in vivo under chilling stress for the first time.Chilling stress signi ficantly affected both the transcription and translation levels as well as the translation efficiency of tea plants.A total of 3010 genes that underwent rapid and independent translation under chilling stress were observed,and they were signi ficantly enriched in the photosynthesis-antenna protein and phenylpropanoid biosynthesis pathways.A set of genes that were signi ficantly responsive to cold at the transcription and translation levels,including four(+)-neomenthol dehydrogenases(MNDs)and two(E)-nerolidol synthases(NESs)arranged in tandem on the chromosomes,were also found.We detected potential upstream open reading frames(uORFs)on 3082 genes and found that tea plants may inhibit the overall expression of genes by enhancing the translation of uORFs under chilling stress.In addition,we identi fied distal transposase hypersensitive sites(THSs)and proximal THSs and constructed a transcriptional regulatory network for tea plants under chilling stress.We also identi fied 13 high-con fidence transcription factors(TFs)that may play a crucial role in cold regulation.These results provide valuable information regarding the potential transcriptional regulatory network in plants and help to clarify how plants exhibit flexible responses to chilling stress.Pengjie Wang Shan Jin Xuejin Chen Liangyu Wu Yucheng Zheng Chuan Yue Yongchun Guo Xingtan Zhang Jiangfan Yang Naixing Ye 2021Horticulture Research2021,8,1:2
18DmpIRFs and DmpST:DAMPE instrument response functions and science tools for gamma-ray data analysis显示文摘Observing GeV gamma-rays is an important goal of the DArk Matter Particle Explorer(DAMPE)for indirect dark matter searching and high energy astrophysics. In this work, we present a set of accurate instrument response functions for DAMPE(DmpIRFs) including the effective area, point-spread function and energy dispersion, which are crucial for gamma-ray data analysis based on statistics from simulation data. A dedicated software named DmpST is developed to facilitate the scientific analyses of DAMPE gamma-ray data. Considering the limited number of photons and angular resolution of DAMPE, the maximum likelihood method is adopted in DmpST to better disentangle different source components.
