1Acute kidney injury in acute-on-chronic liver failure is different from in decompensated cirrhosis显示文摘AIM To evaluate the differences in acute kidney injury(AKI) between acute-on-chronic liver failure(ACLF) and decompensated cirrhosis(DC) patients. METHODS During the period from December 2015 to July 2017, 280 patients with hepatitis B virus(HBV)-related ACLF(HBV-ACLF) and 132 patients with HBV-related DC(HBV-DC) who were admitted to our center were recruited consecutively into an observational study. Urine specimens were collected from all subjects and the levels of five urinary tubular injury biomarkers were detected,including neutrophil gelatinase-associated lipocalin(NGAL), interleukin-18(IL-18), liver-type fatty acid binding protein(L-FABP), cystatin C(CysC), and kidney injury molecule-1(KIM-1). Simultaneously, the patient demographics, occurrence and progression of AKI, and response to terlipressin therapy were recorded. All patients were followed up for 3 mo or until death after enrollment. RESULTS AKI occurred in 71 and 28 of HBV-ACLF and HBV-DC patients, respectively(25.4% vs 21.2%, P = 0.358). Among all patients, the levels of four urinary biomarkers(NGAL, CysC, L-FABP, IL-18) were significantly elevated in patients with HBV-ACLF and AKI(ACLF-AKI), compared with that in patients with HBV-DC and AKI(DC-AKI) or those without AKI. There was a higher proportion of patients with AKI progression in ACLF-AKI patients than in DC-AKI patients(49.3% vs 17.9%, P = 0.013). Fortythree patients with ACLF-AKI and 19 patients with DC-AKI were treated with terlipressin. The response rate of ACLFAKI patients was significantly lower than that of patients with DC-AKI(32.6% vs 57.9%, P = 0.018). 2Cystatin C is a biomarker for predicting acute kidney injury in patients with acute-on-chronic liver failure显示文摘AIM:To investigate serum cystatin C level as an early biomarker for predicting acute kidney injury(AKI)in patients with acute-on-chronic liver failure(ACLF).METHODS:Fifty-six consecutive patients with hepatitis B virus-related ACLF who had normal serum creatinine(Cr)level(<1.2 mg/dL in men,or<1.1 mg/dL in women)were enrolled in the Liver Failure Treatment and Research Center of Beijing 302 Hospital between August 2011 and October 2012.Thirty patients with chronic hepatitis B(CHB)and 30 healthy controls in the same study period were also included.Measurement of serum cystatin C(CysC)was performed by a particle-enhanced immunonephelometry assay using the BN Prospec nephelometer system.The ACLF patients were followed during their hospitalization period.RESULTS:In the ACLF group,serum level of CysC was 1.1±0.4 mg/L,which was significantly higher(P<0.01)than those in the healthy controls(0.6±0.3mg/L)and CHB patients(0.7±0.2 mg/L).During the hospitalization period,eight ACLF patients developed AKI.Logistic regression analysis indicated that CysC level was an independent risk factor for AKI development(odds ratio=1.8;95%CI:1.4-2.3,P=0.021).The cutoff value of serum CysC for prediction of AKI in ACLF patients was 1.21 mg/L.The baseline CysC-based estimated glomerular filtration rate(eGFR CysC)was significantly lower than the creatinine-based eGFR(eGFR CG and eGFR MDRD)in ACLF patients with AKI,suggesting that baseline eGFR CysC represented early renal function in ACLF patients while the Cr levels were still within the normal ranges.CONCLUSION:Serum CysC provides early prediction of renal dysfunction in ACLF patients with a normal serum Cr level.Zhi-Hong Wan Jian-Jun Wang Shao-Li You Hong-Ling Liu Bing Zhu Hong Zang Chen Li Jing Chen Shao-Jie Xin 2013World Journal of Gastroenterology2013,19,48:20
3Formaldehyde up-regulates TRPV1 through MAPK and PI3K signaling pathways in a rat model of bone cancer pain显示文摘Objective Our previous study showed that tumor tissue-derived formaldehyde at low concentrations plays an important role in bone cancer pain through activating transient receptor potential vanilloid subfamily member 1 (TRPV1). The present study further explored whether this tumor tissue-derived endogenous formaldehyde regulates TRPV1 expression in a rat model of bone cancer pain, and if so, what the possible signal pathways are during the development of this type of pain. Methods A rat model of bone cancer pain was established by injecting living MRMT-1 tumor cells into the tibia. The formaldehyde levels were determined by high performance liquid chromatography, and the expression of TRPV1 was examined with Western blot and RT-PCR. In primary cultured dorsal root ganglion (DRG) neurons, the expression of TRPV1 was assessed after treatment with 100 μmol/L formaldehyde with or without pre-addition of PD98059 [an inhibitor for extracellular signal-regulated kinase], SB203580 (a p38 inhibitor), SP600125 [an inhibitor for c-Jun N-terminal kinase], BIM [a protein kinase C (PKC) inhibitor] or LY294002 [a phosphatidylinositol 3-kinase (PI3K) inhibitor]. Results In the rat model of bone cancer pain, formaldehyde concentration increased in blood plasma, bone marrow and the spinal cord. TRPV1 protein expression was also increased in the DRG. In primary cultured DRG neurons, 100 μmol/L formaldehyde significantly increased the TRPV1 expression level. Pre-incubation with PD98059, SB203580, SP600125 or LY294002, but not BIM, inhibited the formaldehyde-induced increase of TRPV1 expression. Conclusion Formaldehyde at a very low concentration up-regulates TRPV1 expression through mitogen-activated protein kinase and PI3K, but not PKC, signaling pathways. 4Situation Awareness of Active Distribution Network:Roadmap,Technologies,and Bottlenecks显示文摘With the rapid development of local generation and demand response,the active distribution network(ADN),which aggregates and manages miscellaneous distributed resources,has moved from theory to practice.Secure and optimal operations now require an advanced situation awareness(SA)system so that operators are aware of operation states and potential risks.Current solutions in distribution supervisory control and data acquisition(DSCADA)as well as the distribution automation system(DAS)generally are not able to meet the technology requirements of SA.In this paper,the authors’participation in the project of developing an SA system as the basic component of a practical active distribution management system(ADMS)deployed in Beijing,China,is presented.This paper reviews the ADN’s development roadmap by illustrating the changes that are made in elements,topology,structure,and control scheme.Taking into consideration these hardware changes,a systematic framework is proposed for the main components and the functional hierarchy of an SA system for the ADN.The SA system’s implementation bottlenecks are also presented,including,but not limited to issues in big data platform,distribution forecasting,and security evaluation.Potential technology solutions are also provided.Jin Lin Can Wan Yonghua Song Renle Huang Xiaoshuang Chen Wanfang Guo Yi Zong You Shi 2016CSEE Journal of Power and Energy Systems2016,2,3:13
5Clinical characteristics and corticosteroid therapy in patients with autoimmune-hepatitis-induced liver failure显示文摘AIM:To investigate the clinical features,response to corticosteroids,and prognosis of autoimmune hepatitis(AIH)-induced liver failure in China.METHODS:A total of 22 patients(19 female and 3male;average age 51±15 years)with AIH-induced liver failure treated in our hospital from 2004 to 2012were retrospectively analyzed.Clinical,biochemical and pathological characteristics of the 22 patients and responses to corticosteroid treatment in seven patients were examined retrospectively.The patients were divided into survivor and non-survivor groups,and the clinical characteristics and prognosis were compared between the two groups.The t test was used for data analysis of all categorical variables,and overall survival was calculated by the Kaplan-Meier method.RESULTS:At the time of diagnosis,mean IgG was2473±983 mg/dL,with three(18.8%)patients showing normal levels.All of the patients had elevated serum levels of antinuclear antibody(≥1:640).Liver histology from one patient showed diagnostic pathological changes,including massive necrosis and plasma cell infiltration.Four patients survived(18.2%)and 18died(81.8%)without liver transplantation.The results showed that patients with low admission Model for End-Stage Liver Disease(MELD)scores(21.50±2.08vs 30.61±6.70,P<0.05)and corticosteroid therapy(100%vs 16.7%,P<0.05)had better prognosis.A total of seven patients received corticosteroid therapy,of whom,four responded and survived,and the other three died.Survivors showed young age,shorter duration from diagnosis to corticosteroid therapy,low MELD score,and absence of hepatic encephalopathy at the time of corticosteroid administration.Six patients who were administered corticosteroids acquired fungal infections but recovered after antifungal therapy.CONCLUSION:Early diagnosis and corticosteroid therapy are essential for improving the prognosis of patients with AIH-induced liver failure without liver transplantation.Bing Zhu Shao-Li You Zhi-Hong Wan Hong-Ling Liu Yi-Hui Rong Hong Zang Shao-Jie Xin 2014World Journal of Gastroenterology2014,20,23:10
6An experimental study of salt expansion in sodium saline soils under transient conditions显示文摘Salt expansion in sulfate saline soils that are widely distributed in northwestern China causes serious infrastructural damages under low-temperature conditions. However, the mechanism of salt expansion under low temperatures is not clear. In this study, we conducted a series of cooling experiments combined with salt crystallization to study this mechanism, and employed an ionic model to calculate the supersaturation ratio of the solution. During the experiments, the strength and the process of salt expansion were examined under different cooling rates and various crystal morphologies. The relationship between temperature and supersaturation ratio under transient conditions was also considered. Results indicate that the initial supersaturation ratio of a sodium sulfate solution is closely related to environmental conditions, and that this ratio decreases with slowing the cooling rates and stabilizing the crystal forms. Higher initial supersaturation ratios lead to an increased non-steady-state zone, resulting in less salt expansion. On the other hand, chloride ion content has a distinct influence on the crystallization supersaturation ratio of the sodium sulfate solution, and higher chloride ion content can inhibit salt expansion in sodium saline soils. 10A novel deletion mutation, c.1296delT in the BCOR gene, is associated with oculo-facio-cardio-dental syndrome显示文摘The purpose of the present study was to analyze the clinical phenotypes of a girl with oculo-facio-cardio-dental(OFCD)syndrome and to identify the potential pathogenic mutation responsible for her disease. The patient underwent detailed clinical examinations and phenotype data were collected over a follow-up period of 9 years. Mutation analysis of the candidate gene BCOR was performed with polymerase chain reaction and Sanger sequencing. BCOR of 60 unrelated normal individuals were also sequenced as a control group. Clinical phenotyping and follow-up study results indicate that this patient had multiple system anomalies including ocular, facial, cardiac, dental, and limb malformations. In addition, papilloma of the choroid plexus was identified, which represents the first report of this phenotype in an OFCD patient. A novel deletion mutation, c.1296 delT in exon4 of the BCOR gene, was identified in this patient and was not found in her parents or in 60 normal unrelated individuals. This deletion was a frameshift mutation and is proposed to encode a premature stop codon, thus producing a truncated protein. Our patient fitted the diagnostic criteria for OFCD syndrome and we report the first papilloma of the choroid plexus in an OFCD patient, expanding the recognized phenotypic spectrum of this disease. 11Elevated Neurosteroids in the Lateral Thalamus Relieve Neuropathic Pain in Rats with Spared Nerve Injury显示文摘Neurosteroids are synthesized in the nervous system from cholesterol or steroidal precursors imported from peripheral sources. These compounds are important allosteric modulators of c-aminobutyric acid A receptors(GABA_ARs), which play a vital role in pain modulation in the lateral thalamus, a main gate where somatosensory information enters the cerebral cortex. Using high-performance liquid chromatography/tandem mass spectrometry,we found increased levels of neurosteroids(pregnenolone,progesterone, deoxycorticosterone, allopregnanolone, and tetrahydrodeoxycorticosterone) in the chronic stage of neuropathic pain(28 days after spared nerve injury) in rats.The expression of the translocator protein TSPO, the upstream steroidogenesis rate-limiting enzyme, increased at the same time. In vivo stereotaxic microinjection of neurosteroids or the TSPO activator AC-5216 into the lateral thalamus(AP-3.0 mm, ML ±3.0 mm, DV 6.0 mm)alleviated the mechanical allodynia in neuropathic pain,while the TSPO inhibitor PK 11195 exacerbated it. The analgesic effects of AC-5216 and neurosteroids were significantly attenuated by the GABA_AR antagonist bicuculline. 12A gate size estimation algorithm for data association filters显示文摘The problem of forming validation regions or gates for new sensor measurements obtained when tracking targets in clutter is considered. Since the gate size is an integral part of the data association filter, this paper is intended to describe a way of estimating the gate size via the performance of the data association filter. That is, the gate size can be estimated by looking for the optimal performance of the data association filter. 15A Context-Based Analgesia Model in Rats: Involvement of Prefrontal Cortex显示文摘Cognition and pain share common neural substrates and interact reciprocally: chronic pain compromises cognitive performance, whereas cognitive processes modulate pain perception. In the present study, we established a non-drug-dependent rat model of context-based analgesia,where two different contexts(dark and bright) were matched with a high(52°C) or low(48°C) temperature in the hot-plate test during training. Before and after training,we set the temperature to the high level in both contexts.Rats showed longer paw licking latencies in trials with the context originally matched to a low temperature than those to a high temperature, indicating successful establishment of a context-based analgesic effect in rats. This effect was blocked by intraperitoneal injection of naloxone(an opioid receptor antagonist) before the probe. The context-based analgesic effect also disappeared after optogenetic activation or inhibition of the bilateral infralimbic or prelimbic sub-region of the prefrontal cortex. In brief, we established a context-based, non-drug dependent, placebo-like analgesia model in the rat. 17Nitric Oxide Inducing Function and Intracellular Movement of Chicken Interleukin-18 in Cultured Cells显示文摘To evaluate the characteristics of chicken interleukin-18 (ChIL-18) in different forms in vitro,the ChIL-18 full-length gene (ChIL-18-F) and the ChIL-18 presumed mature protein gene (ChIL-18-M) werecloned and inserted into the eukaryotic expression vector pCI,to construct recombinant pCI-ChIL-18-Fand pCI-ChIL-18-M.The recombinant plasmids were then transferred into chicken splenic lymphocytes(CSLs).Western blot showed that ChIL-18-F,with a molecular weight of 23.0 kDa,was produced in CSLstransfected by pCI-ChlL-18-F;ChIL-18-M,with a molecular weight of 19.5 kDa,was produced in CSLstransfected by pCI-ChIL-18-M.The nitric oxide (NO) level in the transfected CSLs and the culture mediumat different time points was further examined under confocal microscopy using 4.5-diaminofluoresceinstaining.The results showed that both pCI-ChIL-18-F and pCI-ChIL-18-M groups showed significantincrease in intracellular and extracellular NO production compared with pCI transfected control cells.Theseresults suggest that both ChIL-18-F and ChIL-18-M could stimulate NO secretion in CSLs.To characterizethe intracellular distribution of ChIL-18,ChIL-18-F and ChIL-18-M were each fused to the enhanced greenfluorescent protein gene,and expressed in Vero cells.The results showed that the ChIL-18-F tended to themembranous region in Vero cells,while ChIL-18-M did not.This indicates that the N-terminal 27 amino acidpeptide helped ChIL-18 target to Veto cell membranes.Jian XU Tong-Le DENG Long LI Zhen-Qiang YOU Wang-Jun WAN Lian YU 2005Acta Biochimica et Biophysica Sinica2005,37,10:3
18OsPKpa_1 encodes a plastidic pyruvate kinase that affects starch biosynthesis in the rice endosperm显示文摘Pyruvate kinase(PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice(Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpa_1, which encodes a plastidic isoform of PK(PKp). OsPKpa_1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpa_1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, atthe early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein–protein interaction analysis. 19Calibration procedures for the CHASE/HIS science data显示文摘The Hα line is an important optical line in solar observations containing the information from the photosphere to the chromosphere. To study the mechanisms of solar eruptions and the plasma dynamics in the lower atmosphere, the Chinese Hα Solar Explorer(CHASE) was launched into a Sun-synchronous orbit on October 14, 2021. The scientific payload of the CHASE satellite is the Hα Imaging Spectrograph(HIS). The CHASE/HIS acquires, for the first time, seeing-free Hα spectroscopic observations with high spectral and temporal resolutions. It consists of two observational modes. The raster scanning mode provides full-Sun or region-of-interest spectra at Hα(6559.7-6565.9 ?) and Fe I(6567.8-6570.6 ?) wavebands. The continuum imaging mode obtains full-Sun photospheric images at around 6689 ?. In this paper, we present detailed calibration procedures for the CHASE/HIS science data, including the dark-field and flat-field correction, slit image curvature correction, wavelength and intensity calibration, and coordinate transformation.
