1MiR-181b suppresses proliferation of and reduces chemoresistance to temozolomide in U87 glioma stem cells显示文摘MicroRNAs regulate self renewal and differentiation of cancer stem cells.There,we sought to identify the expression of miR-181b in glioma stem cells and investigate the biological effect of miR-181b on glioma stem cells in this study.MiR-181b expression was measured by real-time PCR in glioma stem cells isolated from U87 cells by FACS sorting.After miR-181b was overexpressed in U87 glioma stem cells by miR-181b lentiviral expression vector and/or treatment of temozolomide,secondary neurosphere assay,soft agar colony assay and MTT assay were performed.Compared with U87 cells,the expression of miR-181b was significantly decreased in U87 glioma stem cells.Overexpression of miR-181b decreased neurosphere formation by U87 glioma stem cells in vitro and suppressed colony formation in soft agar,and the cell growth inhibition rates increased in a time-dependent manner in U87 glioma stem cells infected with miR-181b lentivirus.Furthermore,miR-181b had a synergistic effect on temozolomide-induced inhibition of secondary neurosphere and soft agar colony,and on cell growth inhibition rates.MiR-181b functions as a tumor suppressor that suppresses proliferation and reduces chemoresistance to temozolomide in glioma stem cells.Ping Li Xiaoming Lu Yingyi Wang Lihua Sun Chunfa Qian Wei Yan Ning Liu Yongping You Zhen Fu 2010The Journal of Biomedical Research2010,24,6:14

4Evaluation of human epidermal growth factor receptor 2 status of breast cancer using preoperative multidetector computed tomography with deep learning and handcrafted radiomics features显示文摘Objective:To evaluate the human epidermal growth factor receptor 2(HER2)status in patients with breast cancer using multidetector computed tomography(MDCT)-based handcrafted and deep radiomics features.Methods:This retrospective study enrolled 339 female patients(primary cohort,n=177;validation cohort,n=162)with pathologically confirmed invasive breast cancer.Handcrafted and deep radiomics features were extracted from the MDCT images during the arterial phase.After the feature selection procedures,handcrafted and deep radiomics signatures and the combined model were built using multivariate logistic regression analysis.Performance was assessed by measures of discrimination,calibration,and clinical usefulness in the primary cohort and validated in the validation cohort.Results:The handcrafted radiomics signature had a discriminative ability with a C-index of 0.739[95%confidence interval(95%CI):0.661-0.818]in the primary cohort and 0.695(95%CI:0.609-0.781)in the validation cohort.The deep radiomics signature also had a discriminative ability with a C-index of 0.760(95%CI:0.690-0.831)in the primary cohort and 0.777(95%CI:0.696-0.857)in the validation cohort.The combined model,which incorporated both the handcrafted and deep radiomics signatures,showed good discriminative ability with a C-index of 0.829(95%CI:0.767-0.890)in the primary cohort and 0.809(95%CI:0.740-0.879)in the validation cohort.Conclusions:Handcrafted and deep radiomics features from MDCT images were associated with HER2 status in patients with breast cancer.Thus,these features could provide complementary aid for the radiological evaluation of HER2 status in breast cancer.Xiaojun Yang Lei Wu Ke Zhao Weitao Ye Weixiao Liu Yingyi Wang Jiao Li Hanxiao Li Xiaomei Huang Wen Zhang Yanqi Huang Xin Chen Su Yao Zaiyi Liu Changhong Liang 2020Chinese Journal of Cancer Research2020,32,2:4

8Biological receptor-inspired flexible artificial synapse based on ionic dynamics显示文摘The memristor has been regarded as a promising candidate for constructing a neuromorphic computing platform that is capable of confronting the bottleneck of the traditional von Neumann architecture.Here,inspired by the working mechanism of the G-protein-linked receptor of biological cells,a novel double-layer memristive device with reduced graphene oxide(rGO)nanosheets covered by chitosan(an ionic conductive polymer)as the channel material is constructed.The protons in chitosan and the functional groups in rGO nanosheets imitate the functions of the ligands and receptors of biological cells,respectively.Smooth changes in the response current depending on the historical applied voltages are observed,offering a promising pathway toward biorealistic synaptic emulation.The memristive behavior is mainly a result of the interaction between protons provided by chitosan and the defects and functional groups in the rGO nanosheets.The channel current is due to the hopping of protons through functional groups and is limited by the traps in the rGO nanosheets.The transition from short-term to long-term potentiation is achieved,and learning-forgetting behaviors of the memristor mimicking those of the human brain are demonstrated.Overall,the bioinspired memristor-type artificial synaptic device shows great potential in neuromorphic networks.Qifeng Lu Fuqin Sun Lin Liu Lianhui Li Yingyi Wang Mingming Hao Zihao Wang Shuqi Wang Ting Zhang 2020Microsystems & Nanoengineering2020,6,1:2

11A hybrid intelligent soft computing method for ammonia nitrogen prediction in aquaculture显示文摘Ammonia nitrogen is one of the key parameters in determining the aquaculture water quality condition in pond.The high level of ammonia nitrogen is likely to cause deterioration of water quality and mass death of cultured subjects.Therefore,accurate detection of the cultured water ammonia nitrogen content is crucially important for aquaculture management.While,at present,the accuracy of equipment for measuring the ammonia nitrogen content of aquaculture water in real time cannot meet the requirements for aquaculture.In this paper,the soft computing method is firstly proposed to predict the ammonia nitrogen content in aquaculture water in real time.This method includes empirical mode decomposition(EMD),improved particle swarm optimization(IPSO)and extreme learning machine(ELM).To evaluate the performance of the soft computing techniques,three different statistic indicators were used,including root mean square error(RMSE),the mean absolute error(MAE),and the mean absolute percentage error(MAPE)to compare three artificial soft computing methods.Results showed that the EMD-IPSO-ELM soft computing method showed the best performance among other studied methods in the ammonia nitrogen real time prediction.The EMD-IPSO-ELM model provides moderately and roughly accurately real time prediction value of ammonia nitrogen in aquaculture water.Huihui Yu Ling Yang Daoliang Li Yingyi Chen 2021Information Processing in Agriculture2021,8,1:1

14“Top-down” and “bottom-up” strategies for wafer-scaled miniaturized gas sensors design and fabrication显示文摘Manufacture of large-scale patterned nanomaterials via top-down techniques,such as printing and slurry coating,have been used for fabrication of miniaturized gas sensors.However,the reproducibility and uniformity of the sensors in wafer-scale fabrication are still a challenge.In this work,a“top-down”and“bottom-up”combined strategy was proposed to manufacture wafer-scaled miniaturized gas sensors with high-throughput by in-situ growth of Ni(OH)2 nanowalls at specific locations.First,the micro-hotplate based sensor chips were fabricated on a two-inch(2″)silicon wafer by micro-electro-mechanical-system(MEMS)fabrication techniques(“top-down”strategy).Then a template-guided controllable de-wetting method was used to assemble a porous thermoplastic elastomer(TPE)thin film with uniform micro-sized holes(relative standard deviation(RSD)of the size of micro-holes<3.5%,n>300),which serves as the patterned mask for in-situ growing Ni(OH)2 nanowalls at the micro-hole areas(“bottom-up”strategy).The obtained gas microsensors based on this strategy showed great reproducibility of electric properties(RSD<0.8%,n=8)and sensing response toward real-time H _(2)S detection(RSD<3.5%,n=8).Lin Liu Yingyi Wang Fuqin Sun Yanbing Dai Shuqi Wang Yuanyuan Bai Lianhui Li Tie Li Ting Zhang Sujie Qin 2020Microsystems & Nanoengineering2020,6,1:1

17Clinical Study on the Anti-Aging Effect of a Cream Containing Ginseng Root Extract and L-Carnosine显示文摘The anti-aging effects of a cream containing L-carnosine and ginseng root extract to provide a reference for the evaluation and development of anti-aging cosmetics were tested. Sixty healthy females, aged 35-60,were grouped into test sample and placebo groups. Skin elasticity, gloss, and wrinkles of the participants were analyzed before use and after use for 8 weeks. Instrument measurement results showed that after 8 weeks,skin elasticity, gloss, and wrinkles in the members of the test sample group obviously improved relative to those of the placebo group. Clinical effect assessment results showed that subjects did not experience adverse skin reactions, and their skin gloss showed obvious improvement. Self-assessment results showed that 87%subjects are satisfied with the test sample. 18Influencing factors of colostrum exposure to low-level lead and their relationship with early neural development of infants显示文摘BACKGROUND: Exposure to low-level lead has a toxic effect on the development of neonates, which has attracted wide attention. Colostrum lead level can be used as the indication of lead exposure. OBJECTIVE: To observe the relationship of colostrum lead level and the neurobehavioral development of infants. DESIGN: A prospective control observation. SETTING: Center for Maternal and Child Health, Shanxi Provincial Children's Hospital. PARTICIPANTS: Totally 128 neonates of full-term normal delivery, 76 male and 52 female, from Shanxi Provincial Maternal and Child Health Center and Jiexiu Maternal and Child Health Center were involved in this study. All the involved neonates had no peripartal ischemic/hypoxic history or fetus intrauterine developmental lag. Pregnant women had no various acute and chronic diseases in pregnancy, family history of neurological disease or occupational lead exposure. 128 portions of colostrum sample of full-term normal delivery were collected. Informed consents of detected items were obtained from the puerperants and their relatives. METHODS: ① Experimental grouping: Lead level in the colostrum was determined by atomic absorption spectrometry. According to lead level in the colostrum, the neonates were classified into two exposure groups of greater than or equal to 0.24 μmol/L in a high-level lead group and less than 0.24 μ mol/L in a low-level lead group. ② Experimental evaluation: Mental developmental index (MDI) and psychological developmental index (PDI) of 3-month-old infants were evaluated with Bayley Scales of Infant Development (BSID). The relationships of MDI, PDI and colostrum lead level were performed correlation regression analysis; The relationship of colostrum lead level and development was performed multi-factor analysis with family environment and health questionnaires. MAIN OUTCOME MEASURES: ① Evaluation results of MDI and PDI. ② Multi-factor analysis results. RESULTS: Totally 128 neonates were involved in the study. Ten and eleven neonates were lost due to emigration in the high-level lead group and low-level lead group respectively, and the other 107 neonates participated in the final analysis. ① MDI and PDI in the high-level lead group were significantly lower than those in the low-level lead group, respectively (P < 0.01); Regression analysis results showed that two developmental indexes were statistically negatively correlated with colostrum lead level (regression equation y = 1.9+0.01x1,-0.04x2,+0.04x3,+0.03x4). ② Four variables of the factors included by family environment and health questionnaires were taken into equation. Large maternal age, irrational dietary pattern in pregnancy and pollution degree of habitation environment in pregnancy were the risk factors of colostrum lead level (partial regression coefficien t =0.598 4,0.426 8,0.306 7,P < 0.05-0.01), and calcium supplementation in pregnancy was a protective factor (partial regression coefficien t =-0.455 8, P < 0.01). 20Single-cell-resolution transcriptome map revealed novel genes involved in testicular germ cell progression and somatic cells specification in Chinese tongue sole with sex reversal显示文摘Female-to-male sex reversals(pseudomales)are common in lower vertebrates and have been found in natural populations,which is a concern under rapid changes in environmental conditions.Pseudomales can exhibit altered spermatogenesis.However,the regulatory mechanisms underlying pseudomale spermatogenesis remain unclear.Here,we characterized spermatogenesis in Chinese tongue sole(Cynoglossus semilaevis),a species with genetic and environmental sex determination,based on a highresolution single-cell RNA-seq atlas of cells derived from the testes of genotypic males and pseudomales.We identified five germ cell types and six somatic cell types and obtained a single-cell atlas of dynamic changes in gene expression during spermatogenesis in Chinese tongue sole,including alterations in pseudomales.We detected decreased levels of Ca^(2+)signaling pathway-related genes in spermatogonia,insufficient meiotic initiation in spermatocytes,and a malfunction of somatic niche cells in pseudomales.However,a cluster of CaSR genes and MAPK signaling factors were upregulated in undifferentiated spermatogonia of pseudomales.Additionally,we revealed that Z chromosome-specific genes,such as piwil2,dhx37,and ehmt1,were important for spermatogenesis.These results improve our understanding of reproduction after female-to-male sex-reversal and provide new insights into the adaptability of reproductive strategies in lower vertebrates.Hong-Yan Wang Xiang Liu Jian-Yang Chen Yingyi Huang Yifang Lu Fujian Tan Qun Liu Mingming Yang Shuo Li Xianghui Zhang Yating Qin Wenxiu Ma Yingming Yang Liang Meng Kaiqiang Liu Qian Wang Guangyi Fan Rafael H.Nóbrega Shanshan Liu Francesc Piferrer Changwei Shao 2023Science China(Life Sciences)2023,66,5:1
