1Regulation of glucose and lipid metabolism in health and disease显示文摘Glucose and fatty acids are the major sources of energy for human body. Cholesterol, the most abundant sterol in mammals, is a key component of cell membranes although it does not generate ATP. The metabolisms of glucose, fatty acids and cholesterol are often intertwined and regulated. For example, glucose can be converted to fatty acids and cholesterol through de novo lipid biosynthesis pathways. Excessive lipids are secreted in lipoproteins or stored in lipid droplets. The metabolites of glucose and lipids are dynamically transported intercellularly and intracellularly, and then converted to other molecules in specific compartments. The disorders of glucose and lipid metabolism result in severe diseases including cardiovascular disease, diabetes and fatty liver. 2Predicting geological hazards during tunnel construction显示文摘The complicated geological conditions and geological hazards are challenging problems during tunnel construction,which will cause great losses of life and property.Therefore,reliable prediction of geological defective features,such as faults,karst caves and groundwater,has important practical significances and theoretical values.In this paper,we presented the criteria for detecting typical geological anomalies using the tunnel seismic prediction(TSP) method.The ground penetrating radar(GPR) signal response to water-bearing structures was used for theoretical derivations.And the 3D tomography of the transient electromagnetic method(TEM) was used to develop an equivalent conductance method.Based on the improvement of a single prediction technique,we developed a technical system for reliable prediction of geological defective features by analyzing the advantages and disadvantages of all prediction methods.The procedure of the application of this system was introduced in detail.For prediction,the selection of prediction methods is an important and challenging work.The analytic hierarchy process(AHP) was developed for prediction optimization.We applied the newly developed prediction system to several important projects in China,including Hurongxi highway,Jinping II hydropower station,and Kiaochow Bay subsea tunnel.The case studies show that the geological defective features can be successfully detected with good precision and efficiency,and the prediction system is proved to be an effective means to minimize the risks of geological hazards during tunnel construction.Shucai Li Shuchen Li Qingsong Zhang Yiguo Xue Bin Liu Maoxin Su Zhechao Wang Shugang Wang 2010Journal of Rock Mechanics and Geotechnical Engineering2010,2,3:25

4Effect of acupuncture at Neiguan (PC 6) on cardiac function using echocardiography in myocardial ischemia rats induced by isoproterenol显示文摘OBJECTIVE: To investigate the effect of acupuncture at Neiguan(PC 6) on cardiac function using echocardiography in rat models of myocardial ischemia(MI) induced by isoproterenol(ISO).METHODS: Twenty-seven Sprague-Dawley rats were randomly assigned to normal, model, and acupuncture groups. The model and acupuncture groups were given injections of ISO(85 mg/kg) to establish the MI model. After model establishment,the acupuncture group was treated with acupuncture at Neiguan(PC 6) for 30 min. Echocardiography was used to monitor diastolic and systolic function for 30 min starting from the time after the acupuncture needles were removed. Changes in the length of left ventricular internal diameter at end-diastole(LVIDd), length of left ventricular internal diameter at end-systole(LVIDs), the ratio of mitral peak velocity at early diastole and atrial contraction(E/A), ejection fraction(EF), fractional shortening(FS), and stroke volume(SV) were measured.RESULTS: Compared with the model group at 0and 15 min after needles were removed, the means of LVIDd and LVIDs were significantly lower(P <0.01) and E/A, EF, FS, and SV significantly higher(P < 0.01) in the acupuncture group. In the acupuncture group, the means of LVIDd and LVIDs15 min after the needles were removed were significantly higher(P < 0.01) than those at 0 min. 5PKM2 coordinates glycolysis with mitochondrial fusion and oxidative phosphorylation显示文摘A change in the metabolic flux of glucose from mitochondrial oxidative phosphorylation (OXPHOS) to aerobic glycolysis is regarded as one hallmark of cancer. However, the mechanisms underlying the metabolic switch between aerobic glycolysis and OXPHOS are unclear. Here we show that the M2 isoform of pyruvate kinase (PKM2), one of the rate-limiting enzymes in glycolysis, interacts with mitofusin 2 (MFN2), a key regulator of mitochondrial fusion, to promote mitochondrial fusion and OXPHOS, and attenuate glycolysis. mTOR increases the PKM2:MFN2 interaction by phosphorylating MFN2 and thereby modulates the effect of PKM2: MFN2 on glycolysis, mitochondrial fusion and OXPHOS. 6Multigene editing via CRISPR/Cas9 guided by a single-sgRNA seed in Arabidopsis显示文摘We report that a solo single-guide RNA(sg RNA) seed is capable of guiding Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPRàassociated 9(CRISRP/Cas_9) to simultaneously edit multiple genes At RPL_(10)A, At RPL_(10)B and At RPL_(10)C in Arabidopsis. Our results also demonstrate that it is possible to use CRISPR/Cas_9 technology to create At RPL_(10) triple mutants which otherwise cannot be generated by conventional genetic crossing. Compared to other conventional multiplex CRISPR/Cas systems, a single sg RNA seed has the advantage of reducing off-target gene-editing. 8Effect of electronic stimulation at Neiguan(PC 6) acupoint on gene expression of adenosine triphosphate-sensitive potassium channel and protein kinases in rats with myocardial ischemia显示文摘OBJECTIVE:To investigate the effects of electronic stimulation at acupoints Neiguan(PC 6) and Lieque(LU 7) on the gene expression of the adenosine triphosphate(ATP)-Sensitive potassium channel(KATP:Kir6.1,Kir6.2,SUR2 A,and SUR2B) and protein kinases(PKA,PKG,and PKCβ2) in myocardial cells of rats with myocardial ischemia(Ml) induced by isoproterenol(ISO).METHODS:Rats were randomly divided into a control,model,Neiguan(PC 6),Lieque(LU 7),and non-acupoint groups.The Ml model was established by injecting rats with ISO.Electro-acupuncture treatment was given to the acupuncture groups,once a day for 7 days.Gene expression was analyzed with real-time PCR.RESULTS:The gene expression of KATP and protein kinases in the model group was higher than those in the control group(P < 0.05).After acupuncture treatment,the KATP and protein kinase expression levels were significantly lower in the Neiguan(PC 6)and Lieque(LU 7) groups compared with the model group[P < 0.05).The Neiguan(PC 6) group lowered these levels significantly more than that of the Lieque(LU 7) group(P < 0.05).No significant differences were observed between the model and non-acupoint groups(P > 0.05).CONCLUSION:Our findings suggest that electronic needling of Neiguan(PC 6) can both reduce the gene expression of KATP and protein kinases in rats with ISO-induced Ml.Wang Wei Li Jiquan Meng Xiangyu Chen Yiguo 2015Journal of Traditional Chinese Medicine2015,35,5:4

13A new evaluation method for site selection of large underground water-sealed petroleum storage depots显示文摘Scientific site selection is the first step in constructing underground water-sealed petroleum storage depots, but no uniform standard and code for such activity has been established. Therefore, the main objective of this study is to propose an evaluation method for the site selection of an underground water-sealed petroleum storage depot. The first large underground water-sealed petroleum storage depot being built in China served as the background of this study. The following 12 indexes were used as evaluation factors based on comprehensive evaluation criteria and specifications for key project site selection: geographic structure development feature, topographic feature, lithostratic formation feature, crustal stress, strength of rock mass, joint development feature, hydrogeological conditions, long-term water sealing conditions, environmental/ecological vulnerability, regional stability, technical and economic conditions, and meteorological and hydrological conditions. The weight back analysis and power coefficient methods were also used to evaluate the site selected for the first underground water-sealed petroleum storage depot project. Petroleum site classification models based on the two aforementioned methods were established and used to verify the feasibility of the evaluation criteria and methods, and the evaluation results show the grade of the site selected for the underground water-sealed petroleum storage depot in Huangdao, China is good. 14Protein and RNA expression of acid-sensing ion channels 2 and 3 in myocardial ischemia rats induced by isoproterenol显示文摘OBJECTIVE:To investigate the impact of electro-acupuncture at the Neiguan(PC 6) acupoint on protein and RNA expression of acid-sensing ion channel 2(ASIC2) and ASIC3 in myocardial ischemia rats.METHODS:Fifty male Sprague-Dawley rats were used,weighing(230 ± 50) g.The rats were randomized into a normal group A,model group B,Neiguan(PC 6) group C,Lieque(LU 7) group D,and A-shi points group E.There were 10 rats in each group.Rats were continuously administered 85 mg/kg intravenous isoproterenol daily to establish the model.Successfully modeled rats in groups C,D,and E were given electro-acupuncture treatment.Each group of rats was sacrificed with chloral hydrate(1 mL/100 g) intraperitoneal injection.The left ventricular myocardium was extracted and placed at- 70 ℃ until use.Western blot analysis and real-time PCR were performed to assay protein and RNA expressions of ASIC2 and ASIC3,respectively.Fold changes in RNA expression were quantified with the 2~^(-△△Ct) method.Blood samples were drawn from the aorta abdominalis and tested for creatine kinase-MB(CK-MB) and lactate dehydrogenase(LDH) levels using enzyme-linked immunosorbent assay.RESULTS:Myocardial ischemia rats given electro-acupuncture at the Neiguan(PC 6) acupoint had significantly lower protein and RNA expression of ASIC2 and ASIC3,and CK-MB and LDH levels,compared with model rats(P < 0.01).CONCLUSION:Electro-acupuncture at the Neiguan(PC 6) acupoint can not only decrease the protein and RNA expression of ASIC2 and ASIC3,but also inhibit the opening of ASICs and reduce the cardiomyocyte damage in myocardial ischemia rats.Wang Shudong Zhang Lide Dong Baoqiang Zhang Wenshun Ho Chin Ee Guo Na Chen Yiguo 2015Journal of Traditional Chinese Medicine2015,35,2:2
15Overexpression of amplified in breast cancer 1 (AIB1) gene promotes lung adenocarcinoma aggressiveness in vitro and in vivo by upregulating C-X-C motif chemokine receptor 4显示文摘Background:We previously found that overexpression of the gene known as amplified in breast cancer 1(AIB1)was associated with lymph node metastasis and poor prognosis in patients with lung adenocarcinoma.However,the role of AIB1 in that malignancy remains unknown.The present study aimed to investigate the function of AIB1 in the process of lung adenocarcinoma cell metastasis.Methods:A series of in vivo and in vitro assays were performed to elucidate the function of AIB1,while real-time PCR and Western blotting were utilized to identify the potential downstream targets of AIB1 in the process of lung adenocarcinoma metastasis.Rescue experiments and in vitro assays were performed to investigate whether the invasive-ness of AIB1-induced lung adenocarcinoma was mediated by C-X-C motif chemokine receptor 4(CXCR4).Results:The ectopic overexpression of AIB1 in lung adenocarcinoma cells substantially enhanced cell migration and invasive abilities in vitro and tumor metastasis in vivo,whereas the depletion of AIB1 expression substantially inhibited lung adenocarcinoma cell migration and invasion.CXCR4 was identified as a potential downstream target of AIB1 in lung adenocarcinoma.The knockdown of AIB1 greatly reduced CXCR4 gene expression at both the transcription and protein levels,whereas the knockdown of CXCR4 in cells with AIB1 ectopic overexpression diminished AIB1-induced migration and invasion in vitro and tumor metastasis in vivo.Furthermore,we found a significant positive association between the expression of AIB1 and CXCR4 in lung adenocarcinoma patients(183 cases),and the co-overexpression of AIB1 and CXCR4 predicted the poorest prognosis.Conclusions:These findings suggest that AIB1 promotes the aggressiveness of lung adenocarcinoma in vitro and in vivo by upregulating CXCR4 and that it might be usable as a novel prognostic marker and/or therapeutic target for this disease.Liru He Haixia Deng Shiliang Liu Jiewei Chen Binkui Li Chenyuan Wang Xin Wang Yiguo Jiang Ningfang Ma Mengzhong Liu Dan Xie 2018Cancer Communications2018,38,1:2

17The miR157-SPL-CNR module acts upstream of bHLH101 to negatively regulate iron deficiency responses in tomato显示文摘Iron(Fe)homeostasis is critical for plant growth,development,and stress responses.Fe levels are tightly controlled by intricate regulatory networks in which transcription factors(TFs)play a central role.A series of basic helix-loop-helix(b HLH)TFs have been shown to contribute to Fe homeostasis,but the regulatory layers beyond b HLH TFs remain largely unclear.Here,we demonstrate that the SQUAMOSA PROMOTER-BINDING PROTEIN-LIKE(SPL)TF Sl SPL-CNR negatively regulates Fe-deficiency responses in tomato(Solanum lycopersicum)roots.Fe deficiency rapidly repressed the expression of Sl SPL-CNR,and Fe deficiency responses were intensified in two clustered regularly interspaced palindromic repeats(CRISPR)/CRISPRassociated protein 9-generated Sl SPL-CNR knockout lines compared to the wild-type.Comparative transcriptome analysis identified 47 Fe deficiencyresponsive genes the expression of which is negatively regulated by Sl SPL-CNR,one of which,Slb HLH101,helps regulate Fe uptake genes.Sl SPLCNR localizes the nucleus and interacts with the GTAC and BOX 4(ATTAAT)motifs in the Slb HLH101 promoter to repress its expression.Inhibition of Sl SPL-CNR expression in response to Fe deficiency was well correlated with the expression of the micro RNA Slymi R157.Slymi R157-overexpressing tomato lines displayed enhanced Fe deficiency responses,as did Sl SPL-CNR loss-of-function mutants.We propose that the Slymi R157-Sl SPL-CNR module represents a novel pathway that acts upstream of Slb HLH101 to regulate Fe homeostasis in tomato roots.Huihui Zhu Jiayi Wang Dan Jiang Yiguo Hong Jiming Xu Shaojian Zheng Jianli Yang Weiwei Chen 2022Journal of Integrative Plant Biology2022,64,5:1
