| 2 | Exotic Plant Alnus trabeculosa Alters the Composition and Diversity of Native Rhizosphere Bacterial Communities of Phragmites australis显示文摘Alnus trabeculosa, a rhizobia-nodulating tree, was introduced into the Chongxi tidal wetland in the Yangtze River estuary of China to increase the biodiversity of plants and restore tidal wetland functions. However, the effect of the introduced plant on soil bacterial communities and restoration outcomes remains unknown. In this study, the rhizosphere bacterial community structure and diversity were compared between Phragmites australis monospecific community and A. trabeculosa-P. australis mixed communities,aiming to assess whether A. trabeculosa influenced the rhizosphere bacterial communities of P. australis and to investigate whether different taxonomic groups within a soil community may respond similarly to the presence of an introduced exotic plant. Among the14 phylogenetic phyla detected, Proteobacteria and Acidobacteria were the dominant bacterial taxa in the rhizosphere. Phylogenetic analysis of the predominant Proteobacteria showed that the clones from the rhizosphere soils of A. trabeculosa and P. australis in A. trabeculosa-P. australis mixed communities were more diverse than those in the rhizosphere soil of P. australis in P. australis monospecific community. The rhizosphere community in the wetland potentially included active microbial community related to carbon, nitrogen, and sulfur cycling in the Yangtze River estuary. The rhizosphere soil of P. australis in A. trabeculosa-P. australis mixed communities exhibited the highest Shannon diversity index(H′) and Simpson diversity index(1/D)(H = 4.52, 1/D =253). | 3 | Deletion of large-conductance calcium-activated potassium channels promotes vascular remodelling through the CTRP7-mediated PI3K/Akt signaling pathway显示文摘The large-conductance calcium-activated potassium(BK)channel is a critical regulator and potential therapeutic target of vascular tone and architecture,and abnormal expression or dysfunction of this channel is linked to many vascular diseases.Vascular remodelling is the early pathological basis of severe vascular diseases.Delaying the progression of vascular remodelling can reduce cardiovascular events,but the pathogenesis remains unclear.To clarify the role of BK channels in vascular remodelling,we use rats with BK channelαsubunit knockout(BKα‒/‒).The results show that BKα‒/‒rats have smaller inner and outer diameters,thickened aortic walls,increased fibrosis,and disordered elastic fibers of the aortas compared with WT rats.When the expression and function of BKαare inhibited in human umbilical arterial smooth muscle cells(HUASMCs),the expressions of matrix metalloproteinase 2(MMP2),MMP9,and interleukin-6 are enhanced,while the expressions of smooth muscle cell contractile phenotype proteins are reduced.RNA sequencing,bioinformatics analysis and qPCR verification show that C1q/tumor necrosis factor-related protein 7(CTRP7)is the downstream target gene.Furthermore,except for that of MMPs,a similar pattern of IL-6,smooth muscle cell contractile phenotype proteins expression trend is observed after CTRP7 knockdown.Moreover,knockdown of both BKαand CTRP7 in HUASMCs activates PI3K/Akt signaling.Additionally,CTRP7 is expressed in vascular smooth muscle cells(VSMCs),and BKαdeficiency activates the PI3K/Akt pathway by reducing CTRP7 level.Therefore,we first show that BK channel deficiency leads to vascular remodelling.The BK channel and CTRP7 may serve as potential targets for the treatment of cardiovascular diseases. | Jing Bi Yanru Duan Meili Wang Chunyu He Xiaoyue Li Xi Zhang Yan Tao Yunhui Du Huirong Liu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,12: | 0 |
| 4 | Study on dynamic sorption characteristics of modified biochars for ammonium in biogas slurry显示文摘The objective of this study was to investigate the relationship between ammonium(NH^(+)_(4)-N)dynamic sorption capacity and physicochemical properties of modified biochars.Biochars,producing from three species of agricultural wastes of cornstalk(A),corncob(B)and sawdust(C)at 550°C,600°C and 650°C,were modified by four methods of NaOH plus microwave(NaM),KOH(K),FeCl 3(Fe)and HNO 3(H).The static and dynamic adsorption experiments were conducted to investigate sorption characteristics of modified biochars on NH^(+)_(4)-N in slurry of piggery manure anaerobic digestate.Four modified biochars with better NH^(+)_(4)-N adsorption rates were selected through static kinetics adsorption tests,which showed that chemical reactions might have occurred during the NH^(+)_(4)-N sorption process and the maximum NH^(+)_(4)-N removal rates of B-550-Fe,A-550-NaM,A-550-K and C-600-NaM were 66.64%,57.50%,52.39%and 45.20%,respectively.The dynamic adsorption column experiment was conducted in a three-stage adsorption column packed with the selected three modified biochars.NH^(+)_(4)-N dynamic adsorption depended on the slurry inflow flow rate,the type of biochar and the depth of packed biochar.The optimal adsorption process was elected by the method of orthogonal experiment.The data showed that using deeper packed biochar and applying lower flow rates could be a better strategy to increase NH^(+)_(4)-N adsorption.The maximum NH^(+)_(4)-N removal rate in the slurry could reach 85.60%in the three-stage adsorption process.It concluded that NH^(+)_(4)-N adsorption in three-stage adsorption process could be an effective method to recover nitrogen from piggery manure anaerobic digestate. | Jingtao Ding Yujun Shen Yanru Ma Haibo Meng Hongsheng Cheng Xi Zhang Jian Wang Pengyue Zhang | 2020 | International Journal of Agricultural and Biological Engineering2020,13,1: | 0 |
