2SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
3Preliminary results pertaining to coseismic displacement and preseismic strain accumulation of the Lushan M_S7.0 earthquake,as reflected by GPS surveying显示文摘This paper presents the coseismic displacement and preseismic deformation fields of the Lushan MS7.0 earthquake that occurred on April 20,2013.The results are based on GPS observations along the Longmenshan fault and within its vicinity.The coseismic displacement and preseismic GPS results indicate that in the strain release of this earthquake,the thrust rupture is dominant and the laevorotation movement is secondary.Furthermore,we infer that any possible the rupture does not reach the earth’s surface,and the seismogenic fault is most likely one fault to the east of the Guanxian-Anxian fault.Some detailed results are obtainable.(1)The southern segment of the Longmenshan fault is locked preceding the Lushan earthquake.After the Wenchuan earthquake,the strain accumulation rate in the southeast direction accelerates in the epicenter of the Lushan earthquake,and the angle between the principal compressional strain and the seismogenic fault indicates that a sinistral deformation background in the direction of the seismogenic fault precedes the Lushan earthquake.Therefore,it is evident that the Wenchuan MS8.0 earthquake accelerated the pregnancy of the Lushan earthquake.(2)The coseismic displacements reflected by GPS data are mainly located in a region that is 230 km(NW direction)×100 km(SW direction),and coseismic displacements larger than 10 mm lie predominantly in a100-km region(NW direction).(3)On a large scale,the coseismic displacement shows thrust characteristics,but the associated values are remarkably small in the near field(within 70 km)of the earthquake fault.Meanwhile,the thrust movement in this70-km region does not correspond with the attenuation characteristics of the strain release,indicating that the rupture of this earthquake does not reach the earth’s surface.(4)The laevorotation movements are remarkable in the 50-km region,which is located in the hanging wall that is close to the earthquake fault,and the corresponding values in this case correlate with the attenuation characteristics of the strain release.WU YanQiang JIANG ZaiSen WANG Min CHE Shi LIAO Hua LI Qiang LI Peng YANG YongLin XIANG HePing SHAO ZhiGang WANG WuXing WEI WenXin LIU XiaoXia 2013Chinese Science Bulletin2013,58,28:33
4Rapid afterslip and short-term viscoelastic relaxation following the 2008 M_W7.9 Wenchuan earthquake显示文摘Significant postseismic deformation of the 2008 M W 7.9 Wenchuan earthquake has been observed from GPS data of the first 14 days after the earthquake. The possible mechanisms for the rapid postseismic deformation are assumed to be afterslip on the earthquake rupture plane and viscoelastic relaxation of coseismiclly stress change in the lower crust or upper mantle. We firstly use the constrained least squares method to find an afterslip model which can fit the GPS data best. The afterslip model can explain near-field data very well but shows considerable discrepancies in fitting far-field data. To estimate the effect due to the viscoelastic relaxation in the lower crust, we then ignore the contribution from the afterslip and attempt to invert the viscosity structure beneath the Longmenshan fault where the Wenchuan earthquake occurred from the postseismic deformation data. For this purpose, we use a viscoelastic model with a 2D geometry based on the geological and seismological observations and the coseismic slip distribution derived from the coseismic GPS and InSAR data. By means of a grid search we find that the optimum viscosity is 9×10 18 Pa·s for the middle-lower crust in the Chengdu Basin, 4×10 17 Pa·s for the middle-lower crust in the Chuanxi Plateau and 7×10 17 Pa·s for the low velocity zone in the Chuanxi plateau. The viscoelastic model explains the postseismic deformation observed in the far-field satisfactorily, but it is considerably worse than the afterslip model in fitting the near-fault data. It suggests therefore a hybrid model including both afterslip and relaxation effects. Since the viscoelastic model produces mainly the far-field surface deformation and has fewer degree of freedoms (three viscosity parameters) than the afterslip model with a huge number of source parameters, we fix the viscositiy structure as obtained before but redetermine the afterslip distribution using the residual data from the viscoelastic modeling. The redetermined afterslip distribution becomes physically more reasonable; it is more localized and exhibits a pattern spatially complementary with the coseismic rupture distribution. 52D nanoplate assembled nitrogen doped hollow carbon sphere decorated with Fe3O4 as an efficient electrocatalyst for oxygen reduction reaction and Zn-air batteries显示文摘Designing a highly efficient non-precious based oxygen reduction reaction(ORR)electrocatalyst is critical for the commercialization of various sustainable energy storage and conversion devices such as metal-air batteries and fuel cells.Herein,we report a convenient strategy to synthesis Fe3O4 embedded in N doped hollow carbon sphere(NHCS)for ORR.What's interesting is that the carbon microsphere is composed of two-dimensional(2D)nanoplate that could provide more exposed active sites.The usage of solid ZnO nanowires as zinc source is crucial to obtain this structure.The Fe3O4@NHCS-2 exhibits better catalytic activity and durability than the commercial PtC catalyst.Moreover,it further displays high-performance of Zn-air batteries as a cathode electrocatalyst with a high-power density of 133 mW·cm^-2 and high specific capacity of 701 mA·h·g^-1.The special hollow structure composed 2D nanoplate,high surface area,as well as synergistic effect between the high active Fe3O4 nanoparticles and N-doped matrix endows this outstanding catalytic activity.The work presented here can be easily extended to prepare metal compounds decorated carbon nanomaterials with special structure for a broad range of energy storage and conversion devices.Yanqiang Li Huiyong Huang Siru Chen Xin Yu Chao Wang Tingli Ma 2019Nano Research2019,12,11:9
6Manipulation of SlMXl for enhanced carotenoids accumulation and drought resistance in tomato显示文摘Plants are the ultimate source of nutrients in the human diet. To ensure adequate availability of high quality food for an increasing world population, traits including improved tolerance of stresses and nutrient levels need to be selected in crops, both individually and in combination. Here we report the identification of Sl MX1 encoding a MIXTA-like MYB transcription factor in tomato that simultaneously modulates drought resistance and metabolic processes through regulating key structural and regulatory genes of the corresponding pathways. Over-expression of Sl MX1 results in substantially increased drought tolerance and improved fruit quality, while knocking down Sl MX1 resulted in the opposite phenotypes. 7Dietary encapsulated essential oils and organic acids mixture improves gut health in broiler chickens challenged with necrotic enteritis显示文摘Background:The poultry industry is in need of effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE)due to Clostridium perfringens.In the present study,we investigated the effects of dietary supplementation with a blend of encapsulated essential oils and organic acids(BLJ)on growth performance and gut health using a coinfection model of NE in broiler chickens.Methods:Two hundred and eighty-eight one-day-old male Arbor Acres broiler chicks were randomly assigned using a 2×2 factorial design into two groups fed either 0 or 500 mg/kg dietary BLJ and co-challenged(or not challenged for the control)with Eimeria spp./C.perfringens.Results:Infected birds fed the BLJ-supplemented diet exhibited an improved feed conversion ratio throughout the trial(P<0.01),a higher villus height and villus height/crypt depth ratio,and reduced intestinal C.perfringens counts,liver C.perfringens carriage,gut lesion scores and serum fluorescein isothiocyanate dextran(FITC-D)concentrations at 7 d postinfection compared with those of birds without BLJ supplementation(P<0.05).NE-infected birds fed BLJ exhibited significantly upregulated claudin-1 and IGF-2 mRNA levels(P<0.05),increased A20 mRNA expression and significantly downregulated TRAF-6,TNFSF15 and TOLLIP mRNA levels in the jejunum at 7 d post-infection compared with those in birds without BLJ supplementation(P<0.05).Compared with the uninfected and untreated birds,the uninfected birds fed BLJ displayed increased relative abundances of Lactobacillus and Coprococcus but reduced Rikenellaceae levels.Compared with the unsupplemented NE-challenged birds,infected birds fed BLJ showed an increased relative abundance of Unclassified_Lachnospiraceae and a significantly decreased relative abundance of Erysipelotrichaceae.Conclusion:BLJ supplementation improved growth performance and gut health in NE-infected broiler chickens by strengthening the intestinal barrier function,positively modulating the gut microbiota community and differentially regulating intestinal immune responses.Our results also suggested that adding BLJ effectively controlled NE infections after experimental Eimeria and Clostridium perfringens coinfection.Van Hieu Pham Liugang Kan Jinyu Huang Yanqiang Geng Wenrui Zhen Yuming Guo Waseem Abbas Zhong Wang 2020Journal of Animal Science and Biotechnology2020,11,3:8
8N_2O Emission from Paddy Field under Different Rice Planting Modes显示文摘Measurements of N2O emissions from conventional rice cultivation (CRC), CRC with straw mulching, system of rice intensification (SRI) and SRI with plastic film mulching were conducted through static chamber/gas-chromatography techniques. The results show that daily fluctuation of N2O emissions in jointing stage are much higher than in others. A type peak of N2O seasonal emission presented between jointing and bearing stages companying with high daily average temperature and low precipitation. Biomass and leaf stomatal conductance were observed. Total quantities of N2O emission were budgeted. The results showed that after jointing stage the average N2O emission flux of SRI with plastic film mulching increased significantly than CRC with straw mulching and SRI, the leaf stomatal conductance of those showed the same trend (p< 0.05 ). Yield and total quantity of N2O emission in CRC with straw mulching enhanced 13.7% and 10.7% compared with those of CRC, respectively. 9Zircon U-Pb Age and Geochemistry of the Ore-hosting Ultramafic Complex of Zhou'an PGE-Cu-Ni Deposit,Henan Province,Central China显示文摘The Zhou'an PGE-Cu-Ni deposit was recently discovered in the Qinling orogenic belt bound by the Yangtze and the North China Cratons. It is a blind deposit thoroughly covered by the Cenozoic alluvial sediments in the Nanyang Basin. As the first large PGE-Cu-Ni deposit discovered in the Qinling-Dabie-Sulu orogenic belt, its geological and geochemical characteristic, isotope age, genesis and tectonic setting are of wide concern in both scientific studies and ore exploration. In this contribution, we report the results obtained from a pioneering study. The Zhou'an ultramafic complex is ferruginous, with m/f=4.79-5.08, and shows the nature of tholeiite series. It is rich in light rare earth elements, Rb, Th, U, La, Sm, Zr and Hf, and poor in heavy rare earth elements, Nd and Ta, suggesting an intraplate setting. It has high 87Sr/86Sr and low 143Nd/144Nd ratios. The ratios of Zr/Nb, La/Nb, Ba/Nb, Rb/Nb, Th/Nb, Th/La and Ba/La, suggest the magma originated from lithosphere mantle. The Fo values of olivine and Pd/Ir-Ni/Cu diagram suggest primary magma was High Mg basalt. 11HrcQ is necessary for Xanthomonas oryzae pv. oryzae HR-induction in non-host tobacco and pathogenicity in host rice显示文摘Bacterial blight, caused by Xanthomonas oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice(Oryza sativa L.) worldwide. The type III secretion system(T3SS) of Xoo, encoded by the hrp(hypersensitive response and pathogenicity) genes, plays critical roles in conferring pathogenicity in host rice and triggering a hypersensitive response(HR) in non-host plants. To investigate the major genes conferring the pathogenicity and avirulence of Xoo, we previously constructed a random Tn5-insertion mutant library of Xoo strain PXO99A. We report here the isolation and characterization of a Tn5-insertion mutant PXM69. Tn5-insertion mutants were screened on indica rice JG30, which is highly susceptible to PXO99A, by leaf-cutting inoculation.Four mutants with reduced virulence were obtained after two rounds of screening. Among them, the mutant PXM69 had completely lost virulence to the rice host and ability to elicit HR in non-host tobacco. Southern blotting analysis showed a single copy of a Tn5-insertion in the genome of PXM69. PCR walking and sequencing analysis revealed that the Tn5 transposon was inserted at nucleotide position 70,192–70,201 in the genome of PXO99A, disrupting the type III hrc(hrp-conserved) gene hrcQ, the first gene in the D operon of the hrp cluster in Xoo. To confirm the relationship between the Tn5-insertion and the avirulence phenotype of PXM69, we used the marker exchange mutagenesis to create a PXO99Amutant, ΔhrcQ::KAN, in which the hrcQ was disrupted by a kanamycin-encoding gene cassette at the same site as that of the Tn5-insertion. ΔhrcQ::KAN showed the same phenotype as mutant PXM69. Reintroduction of the wild-type hrcQ gene partially complemented the pathogenic function of PXM69. RT-PCR and cellulase secretion assays showed that the Tn5-disruption of hrcQ did not affect transcription of downstream genes in the D operon and function of the type II secretion system. 12The influence of intimacy on the ’iterative reactions’ during OX-ZEO process for aromatic production显示文摘The oxide-zeolite process provides a promising way for one-step production of aromatics from syngas,whereas the reasons for the dramatic effect of intimacy between oxide and zeolite in the composite catalyst on the product selectivity are still unclear. In order to explore the optimal intimacy and the essential influence factors, ZnCrOxcombined with ZSM-5 are employed to prepare the composite catalysts with different distances between the two components by changing the mixing methods. An aromatic selectivity of 74%(with CO conversion to be 16%) is achieved by the composite catalyst when the intimacy is in the range of nanometer. A so-called ‘iterative reactions’ mechanism of intermediates over oxides is then proposed and studied: the intermediate chemical can undergo a hydrogenation reaction on oxide.So the shorter the intermediates stay on oxide, the more of chance for C-C coupling takes place on zeolite to form aromatics. Moreover, the aero-environments of reaction is found to impact on the extent of iterative reaction as well. Therefore, when the intimacy between the two components changes, the extent of iterative reactions vary, resulting in alteration of product distribution. 13Intrastriatal glial cell line-derived neurotrophic factors for protecting dopaminergic neurons in the substantia nigra of mice with Parkinson disease显示文摘BACKGROUND:Substantia nigra is deep in position and limited in range,the glial cell line-derived neurotrophic factor(GDNF)injection directly into substantia nigra has relatively greater damages with higher difficulty.GDNF injection into striatum,the target area of dopaminergic neuron,may protect the dopaminergic neurons in the compact part of substantia nigra through retrograde transport.OBJECTIVE:To investigate the protective effect of intrastriatal GDNF on dopaminergic neurons in the substantia nigra of mice with Parkinson disease(PD),and analyze the action pathway.DESIGN:A controlled observation.SETTING:Neurobiological Laboratory of Xuzhou Medical College.MATERIALS:Twenty-four male Kunming mice of 7-8 weeks old were used.GDNF,1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)were purchased from Sigma Company(USA);LEICAQWin image processing and analytical system.METHODS:The experiments were carried out in the Neurobiological Laboratory of Xuzhou Medical College from September 2005 to October 2006.The PD models were established in adult KunMing mice by intraperitoneal injection of MPTP.The model mice were were randomly divided into four groups with 6 mice in each group:GDNF 4-day group,phosphate buffer solution(PSB)4-day group,GDNF 6-day group and PSB 6-day group.Mice in the GDNF 4 and 6-day groups were administrated with 1 μL GDNF solution(20 μg/L,dispensed with 0.01 mol/L PBS)injected into right striatum at 4 and 6 days after model establishment.Mice in the PSB 4 and 6-day groups were administrated with 0.01 mol/L PBS of the same volume to the same injection at corresponding time points.② On the 12th day after model establishment,the midbrain tissue section of each mice was divided into 3 areas from rostral to caudal sides.The positive neurons of tyroxine hydroxylase(TH)and calcium binding protein(CB)with obvious nucleolus and clear outline were randomly selected for the measurement,and the number of positive neurons in unit area was counted.MAIN OUTCOME MEASURES:Number of positive neurons of TH and CB in midbrain substantia nigra of mice in each group.RESULTS:All the 24 mice were involved in the analysis of results.The numbers of TH+ and CB+ neurons in the GDNF 4-day group(54.33±6.92,46.33±5.54)were obviously more than those in the PBS 4-day group(27.67±5.01,21.50±5.96,P < 0.01).The numbers of TH+ and CB+ neurons in the GDNF 6-day group(75.67±5.39,69.67±8.69)were obviously more than those in the PBS 6-day group(27.17±4.50,21.33±5.72,P < 0.01)and those in the GDNF 4-day group(P < 0.01).CONCLUSION:Intrastriatal GDNF can protect dopaminergic neurons in substantia nigra of PD mice,and it may be related to the increase of CB expression.Chenghua Xiao Yanqiang Wang Hongmei Liu Hongjun Wang Junping Cao Dianshuai Gao 2007Neural Regeneration Research2007,2,4:4
14Identification and gene mapping of a narrow and upper-albino leaf mutant in rice(Oryza sativa L.)显示文摘The leaf blade consists of color and shape traits.Studies of leaf-blade development are important for improvement of rice yield and quality because it is an essential organ for photosynthesis.A narrow and upper-albino leaf mutant(nul1) was identified from among progeny of the indica restorer line Jinhui10 raised from seeds treated with ethyl methane sulfonate.Under field conditions,the mutant displayed narrow and upper-albino leaf blades with significantly decreased photosynthetic pigment contents throughout their development.The narrow-leaf trait is caused by a decreased number of small veins.In contrast to the wild type,the growth period was extended by approximately 8 d and agronomic traits,such as effective panicle number,percentage seed set and 1000-grain weight,declined significantly in the nul1 mutant.Genetic analysis suggested that the narrow and upper-albino leaf characteristics showed coseparation and were controlled by one recessive gene.The Nul1 gene was mapped onto chromosome 7 between the Indel marker Ind07-1 and the Simple Sequence Repeat marker RM21637.The physical distance between the markers was 75 kb and eight genes were annotated in this region based on the rice Nipponbare genome sequence.These results provide a foundation for cloning and function analysis of Nul1.WANG Feng TANG YanQiang MIAO RunLong XU FangFang LIN TingTing HE GuangHua SANG XianChun 2012Chinese Science Bulletin2012,57,28:4
15Experimental Investigation on the Hydrodynamic Characteristics of Fluidized Bed Particle Solar Receiver with Gas-Solid Countercurrent Flow Pattern显示文摘To design a particle solar receiver(PSR),a vital energy conversion system,is still a bottleneck for researchers.This study presents a novel PSR based on countercurrent fluidized bed(CCFB)technology,named CCFB receiver.In this design,downward-moving particles are subjected to the action of an up-flow gas to reduce the falling speed and enhance the radial disturbance,and hence increase the residence time of particles and improve the heat transfer.A cold-mold visual experimental setup is established.The influence factors are investigated experimentally,including the superficial gas velocity,solid flux,aeration gas,particle size and transport tube diameter.The results indicate that the maximum solid holdup can exceed 9%or so with fine particles of diameter d_(p)=113.5 μm and a tube diameter of 40 mm.It is proved that the CCFB can operate stably and adjust the solid flux rapidly.The results of this study provide a new structure for PSRs in the concentrated solar power field and could fill the research insufficiency in the gas-solid counterflow field.JIANG Kaijun WANG Fengli KONG Yanqiang XU Chao DU Xiaoze 2021Journal of Thermal Science2021,30,6:4

17Nitrogen-doped hierarchically porous carbon spheres for low concentration CO_(2) capture显示文摘Synthesis of spherical carbon beads with effective CO_2 capture capability is highly desirable for large scale application of CO2 sorption, but remains challenging. Herein, a facile and efficient strategy to prepare nitrogen-doped hierarchically porous carbon spheres was developed via co-pyrolyzation of poly(vinylidene chloride) and melamine in alginate gel beads. In this approach, melamine not only serves as the nitrogen precursor, but also acts as a template for the macropores structures. The nitrogen contents in the hierarchically porous carbon spheres reach a high level, ranging from 11.8 wt% to 14.7 wt%, as the melamine amount increases. 18Spatiotemporal Changes of Hazard Intensity-Adjusted Population Exposure to Multiple Hazards in Tibet During 1982–2015显示文摘The dynamic changes of population exposure to hazards in high-altitude areas are an important factor in the scientific evaluation of environmental risks. In this study,the hazards of hypoxia, earthquakes, and snowstorms in Tibet were respectively described by the percentage of oxygen at sea level, earthquake intensity, and mean annual maximum snow depth. The rates of population affected by hypoxia, earthquakes, and snowstorms were calculated by chronic mountain sickness and historical disaster data.Based on these, the study examined the change in population exposure to the three hazards and their combinations by hazard intensity level at the 1 km 9 1 km grid scale in1982–2015. The results show that population exposures to hypoxia, earthquakes, and snowstorms were about 745 thousand, 97 thousand, and 168 thousand in 2015,respectively, among a total population in Tibet of 3.24 million. These exposures were mainly concentrated in the3400–5000 m above sea level zone. The population exposed to hypoxia and earthquakes showed a rising trend from 1982 to 2015, while the population exposed tosnowstorms decreased after 2000 due to reduced snowstorm intensity. Hypoxia-earthquake and hypoxia-snowstorm are the main multiple hazard combinations that people in Tibet suffered from and their personátime exposures were estimated at around 842 thousand and 913 thousand in 2015, respectively, with an average annual increase of 1.7% and 1.3%. Hypoxia is the most important health risk in Tibet. 20Efficient fluorescence detection of a single neutral atom with low background in a microscopic optical dipole trap显示文摘A single cesium atom is trapped in a far-off-resonance optical dipole trap(FORT) from the magneto-optical trap(MOT) and directly imaged by using a charge-coupled device(CCD) camera.The binary single-atom steps and photon anti-bunching are observed by a photon-counting-based HBT system using fluorescence light.The average atom dwelling time in the FORT is about 9 s.To reduce the background noise in the detection procedure we employ a weak probe laser tuned to the D 1 line to illuminate the single atom from the direction perpendicular to the large-numerical-aperture collimation system.The second order degree of coherence g(2)()=0.12±0.02 is obtained directly from the fluorescence light of the single atom without deducting the background.The background light has been suppressed to 10 counts per 50 ms,which is much lower compared with the reported results.The measured g(2)() is in good agreement with theoretical analysis.The system provides a simple and efficient method to manipulate and measure single neutral atoms,and opens a way to create an efficient controlled single-photon source.GUO YanQiang LI Gang ZHANG YanFeng ZHANG PengFei WANG JunMin ZHANG TianCai 2012Science China(Physics,Mechanics & Astronomy)2012,55,9:3
