1Construction of an integrated map and location of a bruchid resistance gene in mung bean显示文摘Bruchid beetle(Callosobruchus chinensis) poses a serious threat to the production and storage of mung bean(Vigna radiata). Mapping bruchid resistance(Br) will provide an important basis for cloning the responsible gene(s) and elucidating its functional mechanism, and will also facilitate marker-assisted selection in mung bean breeding. Here, we report the construction of the genetic linkage groups of mung bean and mapping of the Br1 locus using an RIL population derived from a cross between Berken, a bruchid-susceptible line, and ACC41, a bruchid-resistant line. A total of 560 markers were mapped onto 11 linkage groups,with 38.0% of the markers showing distorted segregation. The lengths of the linkage groups ranged from 45.2 to 117.0 c M with a total coverage of 732.9 c M and an average interval of1.3 c M between loci. Br1 was located on LG9 between BM202(0.7 c M) and Vr2-627(1.7 c M).Based on 270 shared SSR markers, most of the linkage groups were assigned to specific chromosomes. 2Inferring the Tree of Life of the order Cypriniformes, the earth's most diverse clade of freshwater fishes: Implications of varied taxon and character sampling显示文摘The phylogenetic relationships of species are fundamental to any biological investigation, including all evolutionary studies. Accurate inferences of sister group relationships provide the researcher with an historical framework within which the attributes or geographic origin of species (or supraspecific groups) evolved. Taken out of this phylogenetic context, interpretations of evolutionary processes or origins, geographic distributions, or speciation rates and mechanisms, are subject to nothing less than a biological experiment without controls. Cypriniformes is the most diverse clade of freshwater fishes with estimates of diversity of nearly 3,500 species. These fishes display an amazing array of morphological, ecological, behavioral, and geographic diversity and offer a tremendous opportunity to enhance our understanding of the biotic and abiotic factors associated with diversification and adaptation to environments. Given the nearly global distribution of these fishes, they serve as an important model group for a plethora of biological investigations, including indicator species for future cli-matic changes. The occurrence of the zebrafish, Danio rerio, in this order makes this clade a critical component in understanding and predicting the relationship between mutagenesis and phenotypic expressions in vertebrates, including humans. With the tremendous diversity in Cypriniformes, our understanding of their phylogenetic relationships has not proceeded at an acceptable rate, despite a plethora of morphological and more recent mo-lecular studies. Most studies are pre-Hennigian in origin or include relatively small numbers of taxa. Given that analyses of small numbers of taxa for molecular characters can be compromised by peculiarities of long-branch attraction and nodal-density effect, it is critical that significant progress in our understanding of the relationships of these important fishes occurs with increasing sampling of species to mitigate these potential problems. The recent Cypriniformes Tree of Life initiative is an effort to achieve this goal with morphological and molecular (mitochondrial and nuclear) data. In this early synthesis of our understanding of the phylogenetic relationships of these fishes, all types of data have contributed historically to improving our understanding, but not all analyses are complementary in taxon sampling, thus precluding direct understanding of the impact of taxon sampling on achieving accurate phylogenetic inferences. However, recent molecular studies do provide some insight and in some instances taxon sampling can be implicated as a variable that can influence sister group relationships. Other instances may also exist but without inclusion of more taxa for both mitochondrial and nuclear genes, one cannot distinguish between inferences being dictated by taxon sampling or the origins of the molecular data.Richard L. 3First-principles calculations of electronic structure and optical properties of strained Mg_2Si显示文摘A detailed theoretical study on structural,electronic and optical properties of Mg2Si under the isotropic lattice deformation was performed based on the first-principles pseudopotential method.The results show that the isotropic lattice deformation results in a linear decrease in the energy gap for the directΓ15-Γ1 and indirectΓ15-L1 transitions from 93%to 113%,while the indirect band gapΓ15-X1 increases from 93%to 104%and then reduces over 104%.When the crystal lattice is 93%compressed and 113% stretched,the magnesium silicide is a zero-gap semiconductor.Furthermore,the isotropic lattice deformation makes the dielectric function shift and the static dielectric constant change.CHEN Qian XIE Quan ZHAO FengJuan CUI DongMeng LI XuZhen 2010Chinese Science Bulletin2010,55,21:4
4Cyprininae phylogeny revealed independent origins of the Tibetan Plateau endemic polyploid cyprinids and their diversifications related to the Neogene uplift of the plateau显示文摘Origin and diversification of the Tibetan polyploid cyprinids(schizothoracins) may help us to explore relationships between diversification of the cyprinids and the Tibetan Plateau uplift. Cyprininae phylogeny was analyzed using mitochondrial and nuclear DNA sequences to trace origins of polyploidy and diversifications of schizothoracins. Ancestral states reconstruction for ploidy levels indicated that the Cyprininae was diploid origin and the schizothoracin clades tetraploid origins. There were two diversification rate shifts along with diversification of the cyprinine fishes in response to the Tibetan uplift. The unusual diversification shifts were located to branches subtending the clades of Tibetan polyploid cyprinids. Our analyses suggested that(i) phylogeny of Cyprininae recovered two independent origins of the Tibetan polyploidy schizothoracins;(ii) diversifications of the schizothoracins were closely related to the Neogene uplift of the Tibetan plateau in the following ways: the relatively ancient Late Oligocene-Middle Miocene adaptive radiation may be associated with the uplift of the southern Tibet and Himalaya; the Middle Miocene-Early Pleistocene lineage-specific diversification broadly coincident with major phase of the Neogene Tibetan uplift; and the most recent Pleistocene diversification shift in Schizothorax closely coincident with the successive Kunlun-Huanghe and Gonghe movements of the Tibetan uplift and the glaciation-induced climate oscillations on the plateau.Xuzhen Wang Xiaoni Gan Junbing Li Yiyu Chen Shunping He 2016Science China(Life Sciences)2016,59,11:4
5Cyprinid phylogeny based on Bayesian and maximum likelihood analyses of partitioned data:implications for Cyprinidae systematics显示文摘Cyprinidae is the biggest family of freshwater fish, but the phylogenetic relationships among its higher-level taxa are not yet fully resolved. In this study, we used the nuclear recombination activating gene 2 and the mitochondrial 16S ribosomal RNA and cytochrome b genes to reconstruct cyprinid phylogeny. Our aims were to (i) demonstrate the effects of partitioned phylogenetic analyses on phylogeny reconstruction of cyprinid fishes; (ii) provide new insights into the phylogeny of cyprinids. Our study indicated that unpartitioned strategy was optimal for our analyses; partitioned analyses did not provide better-resolved or-supported estimates of cyprinid phylogeny. Bayesian analyses support the following relationships among the major monophyletic groups within Cyprinidae: (Cyprininae, Labeoninae), ((Acheilognathinae, ((Leuciscinae, Tincinae), Gobioninae)), Xenocyprininae). The placement of Danioninae was poorly resolved. Estimates of divergence dates within the family showed that radiation of the major cyprinid groups occurred during the Late Oligocene through the Late Miocene. 8Integration of Desulfurization and Lithium-Sulfur Batteries Enabled by Amino-Functionalized Porous Carbon Nanofibers显示文摘Hydrogen sulfide(H_(2)S)is an industrial exhausted gas that is highly toxic to humans and the environment.Combining desulfurization and fabrication of cathode materials for lithium-sulfur batteries(LSBs)can solve this issue with a double benefit.Herein,the amino-functionalized lotus root-like carbon nanofibers(NH_(2)-PLCNFs)are prepared by the amination of electrospinning carbon nanofibers under dielectric barrier discharge plasma.Selective catalytic oxidation of H_(2)S to elemental sulfur(S)is achieved over the metalfree NH_(2)-PLCNFs catalyst,and the obtained composite S@NH_(2)-PLCNFs is further used as cathode in LSBs.NH_(2)-PLCNFs enable efficient desulfurization(removal capacity as high as 3.46 g H_(2)S g^(−1) catalyst)and strongly covalent stabilization of S on modified carbon nanofibers.LSBs equipped with S@NH_(2)-PLCNFs deliver a high specific capacity of 705.8 mA h g^(−1) at 1 C after 1000 cycles based on the spatial confinement and the covalent stabilization of electroactive materials on amino-functionalized porous carbon matrix.It is revealed that S@NH_(2)-PLCNFs obtained by this kind of chemical vapor deposition leads to a more homogeneous S distribution and superior electrochemical performance to the sample S/NH_(2)-PLCNF-M prepared by the traditional molten infusion.This work opens a new avenue for the combination of environment protection and energy storage.Minghui Sun Xuzhen Wang Yong Li Zongbin Zhao Jieshan Qiu 2023Energy & Environmental Materials2023,6,2:1

11Genetic diversity and a population structure analysis of accessions in the Chinese cowpea [Vigna unguiculata(L.)Walp.]germplasm collection显示文摘Cowpea(Vigna imguicuiata) is an important legume crop with diverse uses. The species is presently a minor crop, and evaluation of its genetic diversity has been very limited. In this study, a total of 200 genic and 100 genomic simple sequence repeat(SSR) markers were developed from cowpea unigene and genome sequences, respectively. Among them, 27 genic and 27 genomic SSR markers were polymorphic and were used for assessment of genetic diversity and population structure in 105 selected cowpea accessions. A total of 155 alleles and 2.9 alleles per marker were identified, and the average polymorphic information content(PIC) value was 0.3615. The average PIC of genomic SSRs(0.3996) was higher than that of genic SSRs(0.3235), and most of the polymorphic genomic SSRs were composed of di-and trinucleotide repeats(51.9% and 37.0% of all loci, respectively). The low level of detected genetic diversity may be attributed to a severe genetic bottleneck that occurred during the cowpea domestication process. The accessions were classified by structure and cluster analysis into four subgroups that correlated well with their geographic origins or collection sites. The classification results were also consistent with the results from principal coordinate analysis and can be used as a guide during future germplasm collection and selection of accessions as breeding materials for cultivar improvement. 16Molecular evolution of connective tissue growth factor in Cyprinidae (Teleostei:Cypriniformes)显示文摘Connective tissue growth factor (CTGF) plays an important role in regulation of cell growth,differentiation,apoptosis and individual development in animals. The study of sequences variation and molecular evolution of CTGF gene across various species of the cyprinid could be helpful for understanding of speciation and gene divergence in this kind of fish. In this study,19 novel sequences of CTGF gene were obtained from the representative species of the family Cyprinidae using PCR amplification,cloning and sequencing. Phylogenetic relationships of Cyprinidae were reconstructed by neighbor-joining (NJ),maximum parsimony (MP),maximum likelihood (ML) and Bayesian method. Oryzias latipes from the family Cyprinodontidae was assigned to be the outgroup taxon. Leuciscini and Barbini were clustered into the monophyletic lineages,respectively,with the high nodal supports. The estimation of the ratio of non-synonymous to synonymous substitution (dN/dS) for the various branches indicated that there stood the different evolution rates between the Leuciscini and the Barbini. With the ratio of dN/dS of the Leuciscini being lower than that of the Barbini,species within the Barbini were demonstrated to be subjected to the relatively less selection pressure and under the relaxable evolution background. A 6 bp indel (insertion/deletion) was found at the 50 end of CTGF gene of Cyprinidae,and this 6 bp deletion only appeared in the Leuciscini,which is a typical characteristic of the Leuciscini and provides evidence for the monophylogeny of the Leuciscini. 17EZH2 identifies the precursors of human natural killer cells with trained immunity显示文摘Objective:Trained immunity of natural killer(NK)cells has shown great potential in the treatment of cancers by eliciting enhanced effector responses to restimulation by cytokines or cancer cells for long time periods after preactivation.However,the human NK cells responsible for the generation and maintenance of trained immunity are largely unknown.We hypothesized that heterogeneous human NK cells would respond differentially to stimulation with a combination of IL-12,IL-15,and IL-18,and that an NK cell subset might exist that is mainly responsible for the induction of trained immunity.On the basis of our hypothesis,we aimed to identify the subset from which cytokine-trained human NK cells originate and to explore possible regulatory targets for drug intervention.Methods:Flow cytometry assays were performed to analyze the functions of cytokine-trained NK cells and examine cell division and protein expression in NK cell subsets.Single-cell RNA sequencing(scRNA-seq)plus TotalSeq™technology was used to track the heterogeneity of NK cells during the induction of trained immunity.Results:Traditional developmental markers for peripheral NK cells were unable to identify the precursors of human NK cells with trained immunity.Therefore,we used scRNA-seq plus TotalSeq™technology to track the heterogeneity of NK cells during the induction of trained immunity and identified a unique cluster of CD57−NKG2A+EZH2+IFNG+MKI67+IL12R+IL15R+IL18R+NK cells.Enrichment and pseudotime trajectory analyses suggested that this cluster of NK cells contained the precursor of trained NK cells.We then used flow cytometry to further investigate the role of EZH2 in trained NK precursors and found that CD57−NKG2A+EZH2+NK cells had faster cell cycles and an enhanced trained phenotype,and EZH2 inhibition significantly impaired the induction of trained immunity in NK cells.These results suggested that EZH2 is a unique epigenetic marker of precursors of human NK cells with trained immunity.Conclusions:Our work revealed human NK heterogeneity in the induction of trained immunity,identified the precursor subset for trained NK cells,and demonstrated the critical role of EZH2 in the induction of trained immunity in human NK cells.Chen Zhang Jie Yin Jian Zheng Jun Xiao Jiajian Hu Yudong Su Kaichen Zhou Yingchi Zhang Xuzhen Zhang Hong Zhang Qian Sun Yang Wang Wenwen Yu Feng Wei Qiang Zhao Long Li Xiubao Ren 2021Cancer Biology & Medicine2021,18,4:0

19Robust THP Design with the Presence of Errors in CSIT Acquisition显示文摘In this paper, a robust THP is proposed for broadcast channel in multi-user multi-input multi-output (MU-MIMO) system, in which the channel uncertainty caused by feedback delay and mobile speed is taken into account. In order to acquire the imperfect channel state information at transmitter (CSIT), a dynamic CSIT model is introduced where the channel temporal and spatial correlations are considered. By utilizing the unchanged channel statistics and accurate outdated feedback channel information jointly, the imperfect CSIT is obtained as the channel estimate and error covariance. Based on the obtained imperfect CSIT, a Minimum Mean-Square Error (MMSE) Tomlinsion-Harashima precoding (THP) is designed and the closed-form solution is derived by the Lagrangian multiplier approach under total power constraint. Simulation results are provided to indicate the preferable performances of the robust THP. 20Significant expansion and red-shifting of fluorescent protein chromophore determined through computational design and genetic code expansion显示文摘Fluorescent proteins (FPs)with emission wavelengths in the far-red and infrared regions of the spectrum provide powerful tools for deep-tissue and super-resolution imaging.The development of red-shifted FPs has evoked widespread interest and continuous engineering efforts.In this article, based on a computational design and genetic code expansion,we report a rational approach to significantly expand and red-shift the chromophore of green fluorescent protein (GFP).We applied computational calculations to predict the excitation and emission wavelengths of a FP chromophore harboring unnatural amino acids (UAA)and identify in silico an appropriate UAA,2-amino-3-(6- hydroxynaphthalen-2-yl)propanoic acid (naphthol-Ala).Our methodology allowed us to formulate a GFP variant (cpsfGFP-66-Naphthol-Ala)with red-shifted absorbance and emission spectral maxima exceeding 60 and 130 nm,respectively,compared to those of GFP.The GFP chromophore is formed through autocatalytic post-translational modification to generate a planar 4-(p-hydroxybenzylidene)-5- imidazolinone chromophore.We solved the crystal structure of cpsfGFP-66-naphthol-Ala at 1.3 ■ resolution and demonstrated the formation of a much larger conjugated n-system when the phenol group is replaced by naphthol.These results explain the significant red-shifting of the excitation and emission spectra of cpsfGFP-66-naphthol-Ala.Li Wang Xian Chen Xuzhen GUO liasong Li Qi Liu Fuying Kang Xudong Wang Cheng HU Haiping Liu Weimin Gong Wei Zhuang Xiaohong Liu Jiangyun Wang 2018Biophysics Reports2018,4,5:0
