6Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 7Prognostic significance of computed tomography-detected extramural vascular invasion in colon cancer显示文摘AIM: To compare disease-free survival(DFS) between extramural vascular invasion(EMVI)-positive and-negative colon cancer patients evaluated by computed tomography(CT).METHODS: Colon cancer patients(n = 194) undergoing curative surgery between January 2009 and December 2013 were included. Each patient's demographics, cancer characteristics, EMVI status, pathological status and survival outcomes were recorded. All included patients had been routinely monitored until December 2015. EMVI was defined as tumor tissue within adjacent vessels beyond the colon wall as seen on enhanced CT. Disease recurrence was defined as metachronous metastases, local recurrence, or death due to colon cancer. Kaplan-Meier analyses were used to compare DFS between the EMVI-positive and-negative groups. Cox's proportional hazards models were used to measure the impact of confounding variables on survival rates.RESULTS: EMVI was observed on CT(ct EMVI) in 60 patients(30.9%, 60/194). One year after surgery, there was no statistically significant difference regarding the rates of progressive events between EMVI-positive and-negative patients [11.7%(7/60) and 6.7%(9/134), respectively; P = 0.266]. At the study endpoint, the EMVI-positive patients had significantly more progressive events than the EMVI-negative patients [43.3%(26/60) and 14.9%(20/134), respectively; oddsratio = 4.4, P < 0.001]. Based on the Kaplan-Meier method, the cumulative 1-year DFS rates were 86.7%(95%CI: 82.3-91.1) and 92.4%(95%CI: 90.1-94.7) for EMVI-positive and EMVI-negative patients, respectively. The cumulative 3-year DFS rates were 49.5%(95%CI: 42.1-56.9) and 85.8%(95%CI: 82.6-89.0), respectively. Cox proportional hazards regression analysis revealed that ctE MVI was an independent predictor of DFS with a hazard ratio of 2.15(95%CI: 1.12-4.14, P = 0.023). 8Antagonism of Protease-Activated Receptor 4 Protects Against Traumatic Brain Injury by Suppressing Neuroinflammation via Inhibition of Tab2/NF-κB Signaling显示文摘Traumatic brain injury(TBI)triggers the activation of the endogenous coagulation mechanism,and a large amount of thrombin is released to curb uncontrollable bleeding through thrombin receptors,also known as protease-activated receptors(PARs).However,thrombin is one of the most critical factors in secondary brain injury.Thus,the PARs may be effective targets against hemorrhagic brain injury.Since the PAR1 antagonist has an increased bleeding risk in clinical practice,PAR4 blockade has been suggested as a more promising treatment.Here,we explored the expression pattern of PAR4 in the brain of mice after TBI,and explored the effect and possible mechanism of BMS-986120(BMS),a novel selective and reversible PAR4 antagonist on secondary brain injury.Treatment with BMS protected against TBI in mice.mRNA-seq analysis,Western blot,and qRT-PCR verification in vitro showed that BMS significantly inhibited thrombin-induced inflammation in astrocytes,and suggested that the Tab2/ERK/NF-κB signaling pathway plays a key role in this process.Our findings provide reliable evidence that blocking PAR4 is a safe and effective intervention for TBI,and suggest that BMS has a potential clinical application in the management of TBI.Jianing Luo Xun Wu Haixiao Liu Wenxing Cui Wei Guo Kang Guo Hao Guo Kai Tao Fei Li Yingwu Shi Dayun Feng Hao Yan Guodong Gao Yan Qu 2021Neuroscience Bulletin2021,37,2:5

10Acrolein Induces Systemic Coagulopathy via Autophagy-dependent Secretion of von Willebrand Factor in Mice after Traumatic Brain Injury显示文摘Traumatic brain injury(TBI)-induced coagulopathy has increasingly been recognized as a significant risk factor for poor outcomes,but the pathogenesis remains poorly understood.In this study,we aimed to investigate the causal role of acrolein,a typical lipid peroxidation product,in TBI-induced coagulopathy,and further explore the underlying molecular mechanisms.We found that the level of plasma acrolein in TBI patients suffering from coagulopathy was higher than that in those without coagulopathy.Using a controlled cortical impact mouse model,we demonstrated that the acrolein scavenger phenelzine prevented TBI-induced coagulopathy and recombinant ADAMTS-13 prevented acrolein-induced coagulopathy by cleaving von Willebrand factor(VWF).Our results showed that acrolein may contribute to an early hypercoagulable state after TBI by regulating VWF secretion.mRNA sequencing(mRNA-seq)and transcriptome analysis indicated that acrolein over-activated autophagy,and subsequent experiments revealed that acrolein activated autophagy partly by regulating the Akt/mTOR pathway.In addition,we demonstrated that acrolein was produced in the perilesional cortex,affected endothelial cell integrity,and disrupted the blood-brain barrier.In conclusion,in this study we uncovered a novel pro-coagulant effect of acrolein that may contribute to TBI-induced coagulopathy and vascular leakage,providing an alternative therapeutic target.Wenxing Cui Xun Wu Dayun Feng Jianing Luo Yingwu Shi Wei Guo Haixiao Liu Qiang Wang Liang Wang Shunnan Ge Yan Qu 2021Neuroscience Bulletin2021,37,8:3
11Acrolein Aggravates Secondary Brain Injury After Intracerebral Hemorrhage Through Drp1-Mediated Mitochondrial Oxidative Damage in Mice显示文摘Clinical advances in the treatment of intracranial hemorrhage(ICH)are restricted by the incomplete understanding of the molecular mechanisms contributing to secondary brain injury.Acrolein is a highly active unsaturated aldehyde which has been implicated in many nervous system diseases.Our results indicated a significant increase in the level of acrolein after ICH in mouse brain.In primary neurons,acrolein induced an increase in mitochondrial fragmentation,loss of mitochondrial membrane potential,generation of reactive oxidative species,and release of mitochondrial cytochrome c.Mechanistically,acrolein facilitated the translocation of dynaminrelated protein 1(Drpl)from the cytoplasm onto the mitochondrial membrane and led to excessive mitochondrial fission.Further studies found that treatment with hydralazine(an acrolein scavenger)significantly reversed Drpl translocation and the morphological damage of mitochondria after ICH.In parallel,the neural apoptosis,brain edema,and neurological functional deficits induced by ICH were also remarkably alleviated.In conclusion,our results identify acrolein as an important contributor to the secondary brain injury following ICH.Meanwhile,we uncovered a novel mechanism by which Drpl-mediated mitochondrial oxidative damage is involved in acroleininduced brain injury.Xun Wu Wenxing Cui Wei Guo Haixiao Liu Jianing Luo Lei Zhao Hao Guo Longlong Zheng Hao Bai Dayun Feng Yan Qu 2020Neuroscience Bulletin2020,36,10:3
12The impacts of infectious complications on outcomes in acute pancreatitis: a retrospective study显示文摘Background: The occurrence of infectious complications characterizes the more severe forms of acute pancreatitis(AP) and is associated with high mortality. We investigated the effects of infection at different sites in patients with AP, including those with necrotizing pancreatitis(NP).Methods: We conducted a retrospective analysis of 285 patients who met the inclusion criteria for AP and were admitted to Tianjin Nankai Hospital between January 2016 and September 2019. According to the source of the culture positivity during hospitalization, patients were divided into four groups: sterile group(n=148), pancreatic infection group(n=65), extrapancreatic infection group(n=22) and combined infection group(n=50). The source of infection, microbiology, biochemical parameters and prognostic indicators were analyzed.Results: In terms of baseline characteristics, the four groups were similar in age, sex, aetiology, previous pancreatitis and diabetes. Compared with the severity of the disease in the other groups, the APACHE II scores(9.91±4.65, 9.46±5.05, respectively) and organ failure rate(40.9% and 50%, respectively)were higher in the extrapancreatic infection group and the combined infection group(P<0.05). The frequency of surgical intervention and hospitalization time in patients with NP complicated with extrapancreatic infection was greatly increased(P<0.05). Regarding the primary outcome, patients in the combined infection group had longer hospital stays(68.28±51.80 vs. 55.58±36.24, P<0.05) and higher mortality(24.0% vs. 9.2%, P<0.05) than patients in the pancreatic infection group. In addition, patients in the extrapancreatic infection group also showed high intensive care utilization(59.1%) and mortality rates(18.2%). Among the 137 AP patients with infection complications, 89 patients exhibited multidrug-resistant(MDR) microorganisms, and the mortality rate of patients with MDR bacterial infection was higher than that of patients with non-MDR bacterial infection(24.7% vs. 3.6%, P=0.001).Conclusions: Clinicians should be aware that extrapancreatic infection(EPI) significantly aggravates the main outcome in pancreatic infection patients. 13Application of an antibody chip for screening differentially expressed proteins during peach ripening and identification of a metabolon in the SAM cycle to generate a peach ethylene biosynthesis model显示文摘Peach(Prunus persica)is a typical climacteric fruit that produces ethylene rapidly during ripening,and its fruit softens quickly.Stony hard peach cultivars,however,do not produce large amounts of ethylene,and the fruit remains firm until fully ripe,thus differing from melting flesh peach cultivars.To identify the key proteins involved in peach fruit ripening,an antibody-based proteomic analysis was conducted.A mega-monoclonal antibody(mAb)library was generated and arrayed on a chip(mAbArray)at a high density,covering~4950 different proteins of peach.Through the screening of peach fruit proteins with the mAbArray chip,differentially expressed proteins recognized by 1587 mAbs were identified,and 33 corresponding antigens were ultimately identified by immunoprecipitation and mass spectrometry.These proteins included not only important enzymes involved in ethylene biosynthesis,such as ACO1,SAHH,SAMS,and MetE,but also novel factors such as NUDT2.Furthermore,protein–protein interaction analysis identified a metabolon containing SAHH and MetE.By combining the antibody-based proteomic data with the transcriptomic and metabolic data,a mathematical model of ethylene biosynthesis in peach was constructed.Simulation results showed that MetE is an important regulator during peach ripening,partially through interaction with SAHH.Wenfang Zeng Liang Niu Zhaohui Wang Xiaobei Wang Yan Wang Lei Pan Zhenhua Lu Guochao Cui Weining Weng Mingqiao Wang Xun Meng Zhiqiang Wang 2020Horticulture Research2020,7,1:2

19Estrogen-dependent depressor response of melatonin via baroreflex afferent function and intensification of PKC-mediated Nav1.9 activation显示文摘Recent studies suggest that melatonin(Mel)plays an important role in the regulation of blood pressure(BP)via the aortic baroreflex pathway.In this study,we investigated the interaction between the baroreflex afferent pathway and Mel-mediated BP regulation in rats under physiological and hypertensive conditions.Mel(0.1,0.3,and 1.0 mg/mL)was microinjected into the nodose ganglia(NG)of rats.We showed that Mel-induced reduction of mean arterial pressure in female rats was significantly greater than that in male and in ovariectomized rats under physiological condition.Consistently,the expression of Mel receptors(MTNRs)in the NG of female rats was significantly higher than that of males.In L-NAME-induced hypertensive and spontaneously hypertensive rat models,MTNRs were upregulated in males but downregulated in female models.Interestingly,Mel-induced BP reduction was found in male hypertensive models.In whole-cell recording from identified baroreceptor neurons(BRNs)in female rats,we found that Mel(0.1μM)significantly increased the excitability of a female-specific subpopulation of Ah-type BRNs by increasing the Nav1.9 current density via a PKC-mediated pathway.Similar results were observed in baroreceptive neurons of the nucleus tractus solitarius,showing the facilitation of spontaneous and evoked excitatory post-synaptic currents in Ah-type neurons.Collectively,this study reveals the estrogen-dependent effect of Mel/MTNRs under physiological and hypertensive conditions is mainly mediated by Ah-type BRNs,which may provide new theoretical basis and strategies for the gender-specific anti-hypertensive treatment in clinical practice.Di Wu Dan Zhao Di Huang Xun Sun Ke-xin Li Yan Feng Qiu-xin Yan Xin-yu Li Chang-peng Cui Hu-die Li Bai-yan Li 2022Acta Pharmacologica Sinica2022,43,9:1
20Development of low-coherence high-power laser drivers for inertial confinement fusion显示文摘The use of low-coherence light is expected to be one of the effective ways to suppress or even eliminate the laser–plasma instabilities that arise in attempts to achieve inertial confinement fusion.In this paper,a review of low-coherence high-power laser drivers and related key techniques is first presented.Work at typical low-coherence laser facilities,including Gekko XII,PHEBUS,Pharos III,and Kanal-2 is described.The many key techniques that are used in the research and development of low-coherence laser drivers are described and analyzed,including low-coherence source generation,amplification,harmonic conversion,and beam smoothing of low-coherence light.Then,recent progress achieved by our group in research on a broadband low-coherence laser driver is presented.During the development of our low-coherence high-power laser facility,we have proposed and implemented many key techniques for working with low-coherence light,including source generation,efficient amplification and propagation,harmonic conversion,beam smoothing,and precise beam control.Based on a series of technological breakthroughs,a kilojoule low-coherence laser driver named Kunwu with a coherence time of only 300 fs has been built,and the first round of physical experiments has been completed.This high-power laser facility provides not only a demonstration and verification platform for key techniques and system integration of a low-coherence laser driver,but also a new type of experimental platform for research into,for example,high-energy-density physics and,in particular,laser–plasma interactions.Yanqi Gao Yong Cui Lailin Ji Daxing Rao Xiaohui Zhao Fujian Li Dong Liu Wei Feng Lan Xia Jiani Liu Haitao Shi Pengyuan Du Jia Liu Xiaoli Li Tao Wang Tianxiong Zhang Chong Shan Yilin Hua Weixin Ma Xun Sun Xianfeng Chen Xiuguang Huang Jian Zhu Wenbing Pei Zhan Sui Sizu Fu 2020Matter and Radiation at Extremes2020,5,6:1
