| 2 | Acrolein Aggravates Secondary Brain Injury After Intracerebral Hemorrhage Through Drp1-Mediated Mitochondrial Oxidative Damage in Mice显示文摘Clinical advances in the treatment of intracranial hemorrhage(ICH)are restricted by the incomplete understanding of the molecular mechanisms contributing to secondary brain injury.Acrolein is a highly active unsaturated aldehyde which has been implicated in many nervous system diseases.Our results indicated a significant increase in the level of acrolein after ICH in mouse brain.In primary neurons,acrolein induced an increase in mitochondrial fragmentation,loss of mitochondrial membrane potential,generation of reactive oxidative species,and release of mitochondrial cytochrome c.Mechanistically,acrolein facilitated the translocation of dynaminrelated protein 1(Drpl)from the cytoplasm onto the mitochondrial membrane and led to excessive mitochondrial fission.Further studies found that treatment with hydralazine(an acrolein scavenger)significantly reversed Drpl translocation and the morphological damage of mitochondria after ICH.In parallel,the neural apoptosis,brain edema,and neurological functional deficits induced by ICH were also remarkably alleviated.In conclusion,our results identify acrolein as an important contributor to the secondary brain injury following ICH.Meanwhile,we uncovered a novel mechanism by which Drpl-mediated mitochondrial oxidative damage is involved in acroleininduced brain injury. | Xun Wu Wenxing Cui Wei Guo Haixiao Liu Jianing Luo Lei Zhao Hao Guo Longlong Zheng Hao Bai Dayun Feng Yan Qu | 2020 | Neuroscience Bulletin2020,36,10: | 3 |
| 3 | Reappraise role of lymph node status in patterns of recurrence following curative resection of gastric adenocarcinoma显示文摘Objective: To examine the association between lymph node status and recurrence patterns in completely resected gastric adenocarcinoma.Methods: We retrospectively assessed 1,694 patients who underwent curative gastrectomy from January 2010 to August 2014. Patients stratified according to lymph node status and recurrence patterns among different subgroups were compared.Results: Of all, 517(30.5%) patients developed recurrent disease, and complete data of recurrence could be obtained in 493(95.4%) patients. For p^(N0) patients, the patterns of recurrence were different according to p T stage: locoregional recurrence was most common in patients with p T1-2 disease(57.1%), distant recurrence was most common in patients with p T3 disease(57.1%), and peritoneal recurrence was most common in patients with p T4 a disease(66.7%). For p^(N+) patients, distant metastasis was most common pattern irrespective of p T stage. The site-specific trend of recurrence showed that locoregional recurrence increased within 5 years in patients with p^(N0)-2 disease but plateaued 3 years after surgery in patients with p N3 disease. Time to recurrence was significantly longer for the p^(N0) patients compared with the p^(N+) patients(median: 25 vs. 16 months, P=0.001).Moreover, post-recurrence survival was significantly better for the p^(N0) patients than for the p^(N+) patients(median:12 vs. 6 months, P<0.001), especially in patients with non-peritoneal recurrence, late recurrence, single recurrence,and receipt of potential curative treatment.Conclusions: Among clinicopathologic factors, lymph node status is the most important factor associated with recurrence patterns after curative gastrectomy. | 6 | Inconsistent effect of dynamic load waveform on macro-and micro-scale responses of ballast bed characterized in individual cycle:a numerical study显示文摘Cyclic load is widely adopted in laboratory to simulate the effect of train load on ballast bed.The effectiveness of such load equivalence is usually testified by having similar results of key concerns of ballast bed,such as deformation or stiffness,while the consistency of particle scale characteristics under two loading patterns is rarely examined,which is insufficient to well-understand and use the load simplification.In this study,a previous laboratory model test of ballast bed under cyclic load is rebuilt using 3D discrete element method(DEM),which is validated by dynamic responses monitored by high-resolution sensors.Then,train load having the same magnitude and amplitude as the cyclic load is applied in the numerical model to obtain the statistical characteristics of inter-particle contact force and particle movements in ballast bed.The results show that particle scale responses under two loading patterns could have quite deviation,even when macro-scale responses of ballast bed under two loading patterns are very close.This inconsistency indicates that the application scale of the DEM model should not exceed the validation scale.Moreover,it is important to examine multiscale responses to validate the effectiveness of load simplification. | Longlong Fu Yuexiao Zheng Yongjia Qiu Shunhua Zhou | 2023 | Railway Engineering Science2023,31,4: | 0 |
