| 1 | Mating patterns and pollen dispersal in a Japanese larch(Larix kaempferi) clonal seed orchard: a case study显示文摘Pollination dynamics highly determines the genetic quality of seed orchard crops. However, there is less research about the effect of mating patterns on seed productivity of orchard crops. So far, clonal seed orchards have been producing genetically improved seedlings used for most Japanese larch(Larix kaempferi(Lamb.) Carr.) plantations in China. In the present study, a total of 17 highly variable simple sequence repeat(SSR) markers were used for genotyping a progeny trial population consisting of 647 open-pollinated progenies germinated from seeds which were collected from 63 maternal clones with 140 potential paternal clones in a Japanese larch clonal seed orchard in China. Paternity analysis was used in the present case study in order to evaluate the level of paternal gametic contribution, estimate pollen contamination and selfing rates, and investigate pollination patterns,pollen dispersal patterns and the impact of mating patterns on seed productivity of orchard crops. We observed 93.7% of the success rate of the parental assignment, unequal paternal gametic contribution(0–12.4%) with 6.3% of the progenies derived from pollen contamination or unsampled pollen donors, and absence of evidence for selfing. We also found that pollination rate highly depended on the distance between pollen donors and maternal parents, the majority of the identified crossing(65.7%)occurred between clones within a 150-m radius, and large variations in growth performance existed among the paternal halfsiblings. Progeny growth performance(diameter at breast(DBH) and height(HGT)) was measured at Age-20 in order to investigate the impact of mating patterns on timber production of orchard crops. As either the paternal or maternal, two clones(i.e., clones Z38 and Z62) were identified to have produced progenies with higher average stem volume breeding values than that of all of the progenies. Specifically, the genetic gains for volume were 3.53% for the two clones as paternal parents, and 8.26% as the maternal parents at Age-20. Thus, both elite clones were ideal candidates for the construction of next-generation clonal seed orchards due to their synchronous reproductive phenology with greater crossing rate and higher genetic gain. | 3 | The roles of graphene in advanced Li-ion hybrid supercapacitors显示文摘Lithium-ion hybrid supercapacitors(LIHSs), also called Li-ion capacitors, are electrochemical energy storage devices that combining the advantages of high power density of supercapacitor and high energy density of Li-ion battery. However, high power density and long cycle life are still challenges for the current LIHSs due to the imbalance of charge-storage capacity and electrode kinetics between capacitor-type cathode and battery-type anode. Therefore, great efforts have been made on designing novel cathode materials with high storage capacity and anode material with enhanced kinetic behavior for LIHSs. With unique two-dimensional form and numerous appealing properties, for the past several years, the rational designed graphene and its composites materials exhibit greatly improved electrochemical performance as cathode or anode for LIHSs. Here, we summarized and discussed the latest advances of the stateof-art graphene-based materials for LIHSs applications. The major roles of graphene are highlighted as(1) a superior active material,(2) ultrathin 2D flexible support to remedy the sluggish reaction of the metal compound anode, and(3) good 2D building blocks for constructing macroscopic 3D porous carbon/graphene hybrids. In addition, some high performance aqueous LIHSs using graphene as electrode were also summarized. | 8 | Comparative Secretome Analysis of Magnaporthe oryzae Identified Proteins Involved in Virulence and Cell Wall Integrity显示文摘Plant fungal pathogens secrete numerous proteins into the apoplast at the plant–fungus contact sites to facilitate colonization.However,only a few secretory proteins were functionally characterized in Magnaporthe oryzae,the fungal pathogen causing rice blast disease worldwide.Asparagine-linked glycosylation 3(Alg3)is an a-1,3-mannosyltransferase functioning in the Nglycan synthesis of N-glycosylated secretory proteins.Fungal pathogenicity and cell wall integrity are impaired inΔalg3 mutants,but the secreted proteins affected inΔalg3 mutants are largely unknown.In this study,we compared the secretomes of the wild-type strain and theΔalg3 mutant and identified 51 proteins that require Alg3 for proper secretion.These proteins were predicted to be involved in metabolic processes,interspecies interactions,cell wall organization,and response to chemicals.Nine proteins were selected for further validation.We found that these proteins were localized at the apoplastic region surrounding the fungal infection hyphae.Moreover,the Nglycosylation of these proteins was significantly changed in theΔalg3 mutant,leading to the decreased protein secretion and abnormal protein localization.Furthermore,we tested the biological functions of two genes,INV1(encoding invertase 1,a secreted invertase)and AMCase(encoding acid mammalian chinitase,a secreted chitinase).The fungal virulence was significantly reduced,and the cell wall integrity was altered in theΔinv1 andΔamcase mutant strains.Moreover,the N-glycosylation was essential for the function and secretion of AMCase.Taken together,our study provides new insight into the role of N-glycosylated secretory proteins in fungal virulence and cell wall integrity. | Ning Liu Linlu Qi Manna Huang Deng Chen Changfa Yin Yiying Zhang Xingbin Wang Guixin Yuan Rui-Jin Wang Jun Yang You-Liang Peng Xunli Lu | 2022 | Genomics, Proteomics & Bioinformatics2022,20,4: | 0 |
| 9 | Apogossypolone对人前列腺癌LNCaP移植瘤的生长抑制作用(英文)显示文摘Objective:The aim of this study was to investigate the inhibitory effect of apogossypolone (ApoG2) on subcutaneous implants of human LNCaP prostatic carcinoma cells, and explore its mechanism. Methods:To establish human LNCaP prostatic carcinoma cell line subcutaneous xenograft models and observe the inhibitory effect of ApoG2 on the tumor model. Immunohistochemistry was employed to observe the expression of Bcl-2, PCNA, CD31, caspase-3 and-8 in tumor tissues. The microvessel density was calculated. Results:ApoG2 could obviously inhibit the growth of subcutaneous prostatic carcinoma implant. ApoG2 decreased the expression of PCNA and CD31, and increased the expression of caspases-3,-8 in tumor tissues. | 11 | A novel coumarin derivative DBH2 inhibits proliferation and induces apoptosis of chronic myeloid leukemia cells显示文摘With the development of tyrosine kinase inhibitor(TKI)resistance,finding the novel effective chemotherapeutic agent is of seminal importance for chronic myelogenous leukemia(CML)treatment.This study aims to find the effective anti-leukemic candidates and investigate the possible underlying mechanism.We synthesized the novel coumarin derivatives and evaluated their anti-leukemic activity.Cell viability assay revealed that compound DBH2 exhibited the potent inhibitory activity on the proliferation of CML K562 cells and TKI resistant K562 cells.Morphological observation and flow cytometry confirmed that DBH2 could selectively induce cell apoptosis and cell cycle arrest at G2/M phase of the K562 cells,which was further confirmed on the bone marrow cells from CML transgenic model mice and CD34+bone marrow leukemic cells from CML patients.Treatments of DBH2 in combination with imatinib could prolong the survival rate of SCL-tTA-BCR/ABL transgenic model mice significantly.Quantitative RT-PCR revealed that DBH2 inhibited the expression of STAT3 and STAT5 in K562 cells,and caspase-3 knockout alleviated the DBH2 induced apoptosis.Furthermore,DBH2 could induce the expression of PARP1 and ROCK1 in K562 cells,which may play the important role in caspase-dependent apoptosis.Our results concluded that coumarin derivative DBH2 serves as a promising candidate for the CML treatment,especially in the combination with imatinib for the TKI resistant CML,and STAT/caspase-3 pathway was involved in the molecular mechanism of anti-leukemic activity of DBH2. | Jiajia Xin Huijie Zhang Dandan Yin Ning An Yaozhen Chen Jinmei Xu Jing Zhang Zhixin Liu Yongsheng Liu Wen Yin Mingkai Li Xingbin Hu | 2023 | Genes & Diseases2023,10,2: | 0 |
| 12 | The size distribution of airborne bacteria and human pathogenic bacteria in a commercial composting plant显示文摘Composting plants are regarded as one of the important sources of environmental bioaerosols.However,limitations in the size distribution of airborne bacteria have prevented our comprehensive understanding of their risk to human health and their dispersal behavior.In this study,different sizes of airborne bacteria were collected using an eight-stage impactor from a full-scale composting facility.Size-related abundance and communities of airborne bacteria as well as human pathogenic bacteria(HPB)were investigated using 16S rRNA gene sequencing coupled with droplet digital PCR.Our results indicate that the bacterial concentrations from the eight stages were approximately 10^(4)-105copies/m^(3).Although no statistical correlation was detected between the particle size and the Shannon index,the influence of size on bacterial lineages was observed in both composting and packaging areas.For airborne bacteria from different stages,the dominant phyla were Firmicutes,Proteobacteria,and Actinobacteria,and the dominant genera was Bacillus.Seven out of eight HPB with a small geometric mean aerodynamic diameter had a high concentration in composting areas.Based on diameters of 2.42 to 5.09μm,most HPB in the composting areas were expected to be deposited on the bronchus and secondary bronchus.However,in the packaging areas,the deposition of HPB(diameters 3.70 to 8.96μm)occurred in the upper part of the respiratory tract.Our results on the size distribution,abundance,and diversity of these bacteria offer important information for the systematic evaluation of bacterial pathogenicity and the potential health impacts on workers in composting plants and the surrounding residents. | Min Gao Ziye Yang Yajie Guo Mo Chen Tianlei Qiu Xingbin Sun Xuming Wang | 2021 | Frontiers of Environmental Science & Engineering2021,15,3: | 0 |
