1Ribozyme probe based on molecular beacon for real time monitoring of enzymatic cleavage process显示文摘Ribozyme probe based on molecular beacon (MBR) for monitoring enzymatic cleavage process in real time is designed and studied. The approach relies on ri-bozyme substrates modified at the two arms, with a fluores-cent moiety attached to the end of one arm and a non-fluorescent quenching moiety attached to the end of the other arm. MBR is employed to directly convert the cleavage information into fluorescence signal in real time. Compared with traditional approach, this method provides a no-radiolabeling, sensitive and effective way to research on the ribozyme activity, enzymatic dynamic process and ri-bozyme function during gene therapy. 2Novel protein detection method based on proximity- dependent polymerase reaction and aptamers显示文摘In recent years, specific detection of proteins is one of the hot issues about aptamers in proteomics. Here we reported a simple, sensitive and specific proximity-dependent protein assay with dual DNA aptamers. Thrombin was used as the model protein, and two aptamer probes with complementary se- quence at 3′-end were designed for the two distinct epitopes of the protein. Association of the two ap- tamers with thrombin resulted in stable hybrids due to the proximity of 3′-end, then polymerase reac- tion was induced. The amount of obtained dsDNA was indicated using the fluorescence dye Sybr Green I. The results showed that the initial velocity of polymerase reaction had a positive correlation with concentration of thrombin. The advantages of this dual-aptamer-based approach included simple and flexible design of aptamer probes, high selectivity and high sensitivity. 3Ammonia promotes the proliferation of bone marrow-derived mesenchymal stem cells by regulating the Akt/mTOR/S6k pathway显示文摘Ammonia plays an important role in cellular metabolism.However,ammonia is considered a toxic product.In bone marrow-derived mesenchymal stem cells,multipotent stem cells with high expression of glutamine synthetase(GS)in bone marrow,ammonia and glutamate can be converted to glutamine via glutamine synthetase activity to support the proliferation of MSCs.As a major nutritional amino acid for biosynthesis,glutamine can activate the Akt/mTOR/S6k pathway to stimulate cell proliferation.The activation of mTOR can promote cell entry into S phase,thereby enhancing DNA synthesis and cell proliferation.Our studies demonstrated that mesenchymal stem cells can convert the toxic waste product ammonia into nutritional glutamine via GS activity.Then,the Akt/mTOR/S6k pathway is activated to promote bone marrow-derived mesenchymal stem cell proliferation.These results suggest a new therapeutic strategy and potential target for the treatment of diseases involving hyperammonemia.Yu Liu Xiangxian Zhang Wei Wang Ting Liu Jun Ren Siyuan Chen Tianqi Lu Yan Tie Xia Yuan Fei Mo Jingyun Yang Yuquan Wei Xiawei Wei 2022Bone Research2022,10,4:1
4Plasmonic Refractive Index Sensors Based on One-and Two-Dimensional Gold Grating on a Gold Film显示文摘In this paper,we propose two kinds of composite structures based on the one-and two-dimensional(1D&2D)gold grating on a gold film for plasmonic refractive index sensing.The resonance modes and sensing characteristics of the composite structures are numerically simulated by the finite-difference time-domain method.The composite structure of the 1D gold semi-cylinder grating and gold film is analyzed first,and the optimized parameters of the grating period are obtained.The sensitivity and figure of merit(FOM)can reach 660 RIU/nm and 169 RIU-1,respectively.Then,we replace the 1D grating with the 2D gold semi-sphere particles array and find that the 2D grating composite structure can excite strong surface plasmon resonance intensity in a wider period range.The sensitivity and FOM of the improved composite structure can reach 985 RIU/nm and 298 RIU-1,respectively.At last,the comparison results of the sensing performance of the two structures are discussed.The proposed structures can be used for bio-chemical refractive index sensing.Jiankai ZHU Xiangxian WANG Yuan WU Yingwen SU Tianxu JIA Hua YANG Liping ZHANG Yunping QI Xiaolei WEN 2020Photonic Sensors2020,10,4:1

8Iron Speciation of Mud Breccia from the Dushanzi Mud Volcano in the Xinjiang Uygur Autonomous Region,NW China显示文摘Organic-inorganic interactions occurring in petroleum-related mud volcanoes can help predict the chemical processes that are responsible for methane emissions to the atmosphere. Seven samples of mud breccia directly ejected from one crater were collected in the Dushanzi mud volcano, along with one argillite sample of the original reddish host rocks distal from the crater, for comparison purposes. The mineral and chemical compositions as well as iron species of all samples were determined using XRD, XRF and M?ssbauer spectroscopy, respectively. The results indicate that a series of marked reactions occurred in the mud volcano systems, more specifically in the mud breccia when compared to the original rocks. Changes mainly included:(1) some conversion of clay minerals from smectite into chlorite and illite, and the precipitation of secondary carbonate minerals such as calcite and siderite;(2) silicon depletion and significant elemental enrichment of iron, manganese, magnesium, calcium and phosphorus; and(3) transformation of iron from ferric species in hematite and smectite into ferrous species in siderite, chlorite and illite. These geochemical reactions likely induced the color changes of the original reddish Neogene argillite to the gray or black mud breccia, as a result of reduction of elements and/or alteration of minerals associated with the oxidation of hydrocarbons. 10Real time monitoring of nucleic acids ligation based on molecular beacon显示文摘A novel method has been developed to monitor the nucleic acids ligation process. Molecular beacon was employed here to convert the ligation information into fluo-rescence signal quickly and quantitatively. This method pro-vides effective and original approach to researching the dy-namic ligation process and the interactions between nucleic acids and ligase. An analytical method for T4 DNA ligase based on this way has been built up with a linear detection range from 2.3×10-4 U/mL to 0.23 U/mL. 11Gender-based violence and hegemonic masculinity in China:an analysis based on the quantitative research显示文摘Based on a survey implemented in a county in central China,the researchers found it is common for women to experience gender-based violence,especially violence at the hands of intimate partners.About half of men surveyed reported inflicting physical or sexual violence on their female partners.One in five men reported having raped a partner or non-partner woman.The physical,mental and reproductive health of the female and male respondents were found to be significantly associated with women’s victimization and men’s perpetration of intimate partner violence.Gender-based violence,including intimate partner violence,is a construction of the social-ecological system.Four elements that are key to hegemonic masculinity are identified:male decision-making,male reputation,violence and heterosexuality.By positing the four elements as standards that define a“real man”,the domination of men over women is naturalized and legitimized.It is necessary to foster other non-violent and more equitable masculinities.Xiangxian Wang Gang Fang Hongtao Li 2019China Population and Development Studies2019,3,3:1
12Ultrasensitive monitoring of ribozyme cleavage product using molecular-beacon-ligation system显示文摘This paper reports a new approach to detect ribozyme cleavage product based on the molecular-beacon-ligation system. The molecular beacon, designed in such a way that one-half of its loop is complementary to ribozyme cleavage product, is used to monitor ligation process of RNA/DNA com-plex in a homogeneous solution and to convert directly cleavage product information into fluorescence signal. The method need not label ribozyme and ribozyme substrate, which is fast, simple and ultra-sensitive for detection of cleavage product. Detection limit of the assay is 0.05 nmol/L. The cleavage product of hammerhead ribozyme against hepatitis C virus RNA (HCV-RNA) was detected perfectly based on this assay. 13Virtual fire drill system supporting co-located collaboration显示文摘Background Due to the restriction of display mode,in most of the virtual reality systems with multiple people in the same physical space,the program renders the scene based on the position and perspective of the one user,so that other users just see the same scene,resulting in vision disorder.Methods To improve experience of multi-user co-located collaboration,in this study,we propose a fire drill system supporting co-located collaboration,in which three co-located users can collaborate to complete the virtual firefighting mission.Firstly,with multi-view stereoscopic projective display technology and ultra wideband(UWB)technology,co-located users can roam independently and watch virtual scenes through the correct perspective view based on their own position by wearing dedicated shutter glasses,thus carrying out different virtual tasks,which improves the flexibility of co-located collaboration.Secondly,we design simulated firefighting water-gun using the micro-electromechanical system sensor,through which users can interact with virtual environment,and thus provide a better interactive experience.Finally,we develop a workbench including a holographic display module and multi touch operation module for virtual scene assembly and virtual environment control.Results The controller can use the workbench to adjust the virtual layout in real time,and control the virtual task process to increase the flexibility and playability of system.Conclusions Our work can be employed in a wide range of related virtual reality applications.Yuan WEI Dongdong GUAN Qiuchen WANG Xiangxian LI Yulong BIAN Pu QIN Yanning XU Chenglei YANG 2019Virtual Reality & Intelligent Hardware2019,1,3:1

15Nation from Village: The Evolving Landscape for Rural Women in the First Half of the Twentieth Century显示文摘In the first half of the 20th century, during China's rapid transition from tradition to modernity, three major historical events occurred successively in Ding County, Hebei Province. These events illustrate the gradual broadening of the horizons of some rural women: shifting from the Confucian expectation that 'women's rightful place is in the home' to a gradual extension into the public domain. The Zhaicheng village system took the first small step in breaking the Confucian gender norms by promoting female education in public spaces, yet it did not truly challenge the fundamental gender order of males outside the home and females within it. The Mass Education Movement saw the emergence of the first group of professional women in rural Ding County. However, they faced social exclusion as a consequence of entering the public domain. The Zhaicheng system and the gender-based teaching practices during the Mass Education Movement sowed the seeds of the impending tension and conflict experienced by women juggling dual roles within and outside the family. Amidst the bloodshed of the Chinese People's War of Resistance against Japanese Aggression and the War of Liberation(wars of Resistance and Liberation), female warriors, through sacrifice and devotion, ventured into the newly expanded public sphere of war, yet traditional gender norms continued to confine their broadened horizons to a transient phase in their lives.
