1Seroprevalence of SARS-CoV-2 infections among children visiting a hospital显示文摘Importance:In this study,we retrospectively investigated the seroprevalence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)antibodies within serum samples from children in Beijing,China.These findings provide preliminary guidance regarding population susceptibility to SARS-CoV-2,which will aid in establishing policy toward coronavirus disease 2019(COVID-19)prevention and control.Objective:To understand the seropositivity of anti-SARS-CoV-2 IgM/IgG antibodies among children in Beijing,China,evaluate the susceptibility of children in Beijing to SARS-CoV-2,and provide prima facie evidence to guide SARS-CoV-2 prevention and control.Methods:IgM/IgG antibody kits(colloidal gold)were used to conduct preliminary screening of SARS-CoV-2 IgM/IgG antibodies in serum samples of children who presented to Beijing Children’s Hospital,Capital Medical University,having fever or requiring hospitalization,from March 2020 to August 2020.Statistical analysis of anti-SARS-CoV-2 antibody seropositivity was performed according to the children’s general demographic characteristics,timing of admission to hospital,presence of pneumonia,and viral nucleic acid test results.Results:The study included 19797 children with both IgM and IgG antibody results.Twenty-four children had anti-SARS-CoV-2 IgM-positive results(positive rate of 1.2‰),twelve children had anti-SARS-CoV-2 IgG-positive results(positive rate of 0.6‰).Viral nucleic acid test results were negative for the above-mentioned children with positive antibody findings;during the study,two children exhibited positive viral nucleic acid test results,but their anti-SARS-CoV-2 IgM/IgG antibody results were negative.Anti-SARS-CoV-2 IgM antibody seropositivity was higher in the<1-year-old group than in the≥6-year-old group.The rates of anti-SARS-CoV-2 IgM seropositivity was highest in August from March to August;IgG results did not significantly differ over time.The rates of anti-SARS-CoV-2 IgM or IgG seropositivity among children with and without suspected pneumonia did not significantly differ between groups.Interpretation:During the study period,the rates of anti-SARS-CoV-2 IgM/IgG antibody seropositivity were low among children who presented to Beijing Children’s Hospital,Capital Medical University.The findings suggest that children in Beijing are generally susceptible to SARS-CoV-2 infection;COVID-19 prevention and control measures should be strengthened to prevent disease in children.Ran Wang Fang Jin Shuhui Cao Hong Yuan Jingchen Qu Jiaqi Zhang Yuxuan Li Xiangpeng Chen Wenqi Song Zhengde Xie 2020Pediatric Investigation2020,4,4:2
2Interplay between polarization rotation and crack propagation in PMN-PT relaxor single crystals显示文摘Investigations on the interconnection between the polarization rotation and crack propagation are performed for [110]-oriented 74Pb(Mg1/3Nb2/3)O3-26PbTiO3 relaxor ferroelectric single crystal under electric loadings along [001]direction. The crystal is of predominantly monoclinic MA phase with scatter distributed rhombohedral(R) phase under a moderate poling field of 900 V/mm in[001] direction. With magnitude of 800 V/mm, a through thickness crack is initiated near the electrode by electric cycling. Static electric loadings is then imposed to the single crystal. As the applied static electric field increases, domain switching in the monoclinic MA phase and phase transition from MA to R phase occur near the crack. 3Application of ERA5 Reanalysis to the Construction of Initial Conditions for WACCM Simulations显示文摘This study uses ECMWF fifth-generation reanalysis, ERA5, which extends to the mesopause, to construct the Initial Conditions(IC) for WACCM(Whole Atmosphere Community Climate Model) simulations. Because the biases between ERA5 and Sounding of the Atmosphere using Broadband Emission Radiometry(SABER) temperature data are within ±5 K below the lower mesosphere,ERA5 reanalysis is used to construct IC in the lower atmosphere. Four experiments are performed to simulate a Stratospheric Sudden Warming(SSW) event from 5 to 15 February 2016. The simulation using the WACCM default climatic IC cannot represent the sharp meteorological variation during SSW. In contrast, the 0~4 d forecast results driven by ERA5-constructed IC is consistent with ERA5 reanalysis below the middle mesosphere. Comparing with WACCM climatology ICs scheme, the ICs constructing method based on ERA5 reanalysis can obtain 67%, 40%, 22%, 4% and 6% reduction of temperature forecast RMSE at 10 hPa, 1 hPa, 0.1 hPa, 0.01 hPa and 0.001 hPa respectively. 4Variability of Atlantic Meridional Overturning Circulation in FGOALS-g2显示文摘The variability of Atlantic Meridional Overturning Circulation(AMOC) in the pre-industrial control experiment of the Flexible Global Ocean–Atmosphere–Land System model, Grid-point Version 2(FGOALS-g2) was investigated using the model outputs with the most stable state in a 512-yr time window from the total 1500-yr period of the experiment. The period of AMOC in FGOALS-g2 is double peaked at 20 and 32 years according to the power spectrum, and 22 years according to an auto-correlation analysis, which shows very obvious decadal variability. Like many other coupled climate models, the decadal variability of AMOC in FGOALS-g2 is closely related to the convection that occurs in the Labrador Sea region. Deep convection in the Labrador Sea in FGOALS-g2 leads the AMOC maximum by 3–4 years. The contributions of thermal and haline effects to the variability of the convection in three different regions [the Labrador, Irminger and Greenland–Iceland– Norwegian(GIN) Seas] were analyzed for FGOALS-g2. The variability of convection in the Labrador and Irminger Seas is thermally dominant, while that in the colder GIN Seas can be mainly attributed to salinity changes due to the lower thermal expansion. 6Engineering single-molecule fluorescence withasymmetric nano-antennas显示文摘As a powerful tool for studying molecular dynamics in bioscience,single-molecule fluorescence detection providesdynamical information buried in ensemble experiments.Fluorescence in the near-infrared(NIR)is particularly usefulbecause it offers higher signal-to-noise ratio and increased penetration depth in tissue compared with visiblefluorescence.The low quantum yield of most NIR fluorophores,however,makes the detection of single-moleculefluorescence difcult.Here,we use asymmetric plasmonic nano-antenna to enhance the fluorescence intensity ofAlEE1000,a typical NIR dye,by a factor up to 405.The asymmetric nano-antenna achieve such an enhancement mainlyby increasing the quantum yield(to~80%)rather than the local field,which degrades the molecules'photostability.Our coupled-mode-theory analysis reveals that the enhancements stem from resonance-matching between antennaand molecule and,more importantly,from optimizing the coupling between the near-and far-ield modes withdesigner asymmetric structures.Our work provides a universal scheme for engineering single-molecule fluorescence inthe near-infrared regime.Wenqi Zhao Xiaochaoran Tian Zhening Fang Shiyi Xiao Meng Qiu Qiong He Wei Feng Fuyou Li Yuanbo Zhang Lei Zhou Yan-Wen Tan 2021Light(Science & Applications)2021,10,5:1

10Function of resveratrol derived from transgenic plant expressing resveratrol syn-thase gene显示文摘Two genes from grapevine coding for resvera-trol synthase, named RS1 and RS2, were cloned by RT-PCR. An Escherichia coli expression vector was constructed by insertion of RS1 into pBV221. A specific protein with the same molecular weight (42 ku) as the resveratrol synthase was expressed and used to prepare the rabbit antiserum. A plant expression vector was constructed by inserting the RS1 gene into pBin438 downstream of the doubled CaMV 35S promoter and TMV-Ω fragment. PCR-positive transgenic tobacco plants were obtained after transformation with Agrobacterium tumefaclens LBA4404 harboring the plant expression vector. Southern blot analysis demonstrated that the foreign gene was integrated into the tobacco genome. The results of RT-PCR and Western blot indicated that the RS1 gene was transcribed and expressed. Formation of resveratrol in transgenic tobacco was further determined by thin-layer chromatography of silica gel and HPLC. 12Detection of abnormal hydroponic lettuce leaves based on image processing and machine learning显示文摘Accurate and fast detection of abnormal hydroponic lettuce leaves is primary technology for robotic sorting.Yellow and rotten leaves are main types of abnormal leaves in hydroponic lettuce.This study aims to demonstrate a feasibility of detecting yellow and rotten leaves of hydroponic lettuce by machine learning models,i.e.Multiple Linear Regression(MLR),K-Nearest Neighbor(KNN),and Support Vector Machine(SVM).One-way analysis of variance was applied to reduce RGB,HSV,and L*a*b*features number of hydroponic lettuce images.Image binarization,image mask,and image filling methods were employed to segment hydroponic lettuce from an image for models testing.Results showed that G,H,and a*were selected from RGB,HSV,and L*a*b*for training models.It took about 20.25 s to detect an image with 30244032 pixels by KNN,which was much longer than MLR(0.61 s)and SVM(1.98 s).MLR got detection accuracies of 89.48%and 99.29%for yellow and rotten leaves,respectively,while SVM reached 98.33%and 97.91%,respectively.SVM was more robust than MLR in detecting yellow and rotten leaves of hydroponic.Thus,it was possible for abnormal hydroponic lettuce leaves detection by machine learning methods.Ruizhe Yang Zhenchao Wu Wentai Fang Hongliang Zhang Wenqi Wang Longsheng Fu Yaqoob Majeed Rui Li Yongjie Cui 2023Information Processing in Agriculture2023,10,1:0
13Pseudo-chromosome–length genome assembly for a deep-sea eel Ilyophis brunneus sheds light on the deep-sea adaptation显示文摘High hydrostatic pressure,low temperature,and scarce food supply are the major factors that limit the survival of vertebrates in extreme deep-sea environments.Here,we constructed a high-quality genome of the deep-sea Muddy arrowtooth eel(MAE,Ilyophis brunneus,captured below a depth of 3,500 m)by using Illumina,Pac Bio,and Hi-C sequencing.We compare it against those of shallow-water eel and other outgroups to explore the genetic basis that underlies the adaptive evolution to deep-sea biomes.The MAE genome was estimated to be 1.47 Gb and assembled into 14 pseudo-chromosomes.Phylogenetic analyses indicated that MAE diverged from its closely related shallow-sea species,European eel,~111.9 Mya and experienced a rapid evolution.The genome evolutionary analyses primarily revealed the following:(i)under high hydrostatic pressure,the positively selected gene TUBGCP3 and the expanded family MLC1 may improve the cytoskeleton stability;ACOX1 may enhance the fluidity of cell membrane and maintain transport activity;the expansion of ABCC12 gene family may enhance the integrity of DNA;(ii)positively selected HARS likely maintain the transcription ability at low temperatures;and(iii)energy metabolism under a food-limited environment may be increased by expanded and positively selected genes in AMPK and m TOR signaling pathways.Jie Chen Honghui Zeng Wenqi Lv Ning Sun Cheng Wang Wenjie Xu Mingliang Hu Xiaoni Gan Lisheng He Shunping He Chengchi Fang 2023Science China(Life Sciences)2023,66,6:0

15Sympatric or micro-allopatric speciation in a glacial lake?Genomic islands support neither显示文摘Apparent cases of sympatric speciation may actually be due to micro-allopatric or micro-parapatric speciation.One way to distinguish between these models is to examine the existence and nature of genomic islands of divergence,wherein divergent DNA segments are interspersed with low-divergence segments.Such islands should be rare or absent under micro-allopatric speciation but common in cases of speciation with gene flow.Sympatric divergence of endemic fishes is known from isolated saline,crater,postglacial,and ancient lakes.Two morphologically distinct cyprinid fishes,Gymnocypris eckloni scoliostomus(GS)and G.eckloni eckloni(GE),in a small glacial lake on the Qinghai-Tibet Plateau,Lake Sunmcuo,match the biogeographic criteria of sympatric speciation.In this study,we examined genome-wide variation in 46individuals from these two groups.The divergence time between the GS and GE lineages was estimated to be 20-60 Kya.We identified 54 large genomic islands(≥100 kb)of speciation,which accounted for 89.4%of the total length of all genomic islands.These islands harboured divergent genes related to olfactory receptors and olfaction signals that may play important roles in food selection and assortative mating in fishes.Although the genomic islands clearly indicated speciation with gene flow and rejected micro-allopatric speciation,they were too large to support the hypothesis of sympatric speciation.Theoretical and recent empirical studies suggested that continual gene flow in sympatry should give rise to many small genomic islands(as small as a few kilobases in size).Thus,the observed pattern is consistent with the extensive evidence on parapatric speciation,in which adjacent habitats facilitate divergent selection but also permit gene flow during speciation.We suggest that many,if not most,of the reported cases of sympatric speciation are likely to be micro-parapatric speciation.Ning Sun Liandong Yang Fei Tian Honghui Zeng Ziwen He Kai Zhao Cheng Wang Minghui Meng Chenguang Feng Chengchi Fang Wenqi Lv Jing Bo Yongtao Tang Xiaoni Gan Zuogang Peng Yiyu Chen Shunping He 2022National Science Review2022,9,12:0
