Heavy metal accumulation by panicled goldenrain tree(Koelreuteria paniculata) and common elaeocarpus(Elaeocarpus decipens) in abandoned mine soils in southern China显示文摘Phytoremediation can be used as a sustainable technology for mine spoil remediation to remove heavy metals. This study investigated the concentration of 7 heavy metal contamination in soil and plant samples at an abandoned mine site.We found that, after vegetation remediation at the abandoned mine site, the reduction rates for 7 heavy metals were in the range of 4.2%-86%,where reduction rates over 50% were achieved for four heavy metals(Zn,Mn,Cd,Ni).Transfer coeffcients of the panicled goldenrain tree(Koelreuteria paniculata Laxm) and the common elaeocarpus(Elaeocarpus decipens) for Zn,Mn,Ni,and Co were more than 1.Enrichment coeffcients of both trees for Mn were higher than 1.Our results suggest that the panicled goldenrain tree and the common elaeocarpus tree may act as accumulators in remediation. Moreover, the woody vegetation remediation in abandoned mining areas play an important role in improving scenery besides removing heavy metal from contaminated soil. | TIAN Dalun ZHU Fan YAN Wende Fang Xi XIANG Wenhua DENG Xiangwen WANG Guangjun PENG Changhui | 2009 | Journal of Environmental Sciences2009,21,3: | 11
Application of artificial neural networks in global climate change and ecological research:An overview显示文摘Fields that employ artificial neural networks(ANNs)have developed and expanded continuously in recent years with the ongoing development of computer technology and artificial intelligence.ANN has been adopted widely and put into practice by research-ers in light of increasing concerns over ecological issues such as global warming, frequent El Nio-Southern Oscillation(ENSO)events, and atmospheric circulation anomalies.Limitations exist and there is a potential risk for misuse in that ANN model pa-rameters require typically higher overall sensitivity, and the chosen network structure is generally more dependent upon individ-ual experience.ANNs,however, are relatively accurate when used for short-term predictions;despite global climate change re-search favoring the effects of interactions as the basis of study and the preference for long-term experimental research.ANNs remain a better choice than many traditional methods when dealing with nonlinear problems, and possesses great potential for the study of global climate change and ecological issues.ANNs can resolve problems that other methods cannot. This is especially true for situations in which measurements are difficult to conduct or when only incomplete data are available. It is anticipated that ANNs will be widely adopted and then further developed for global climate change and ecological research. | LIU ZeLin PENG ChangHui XIANG WenHua TIAN DaLun DENG XiangWen ZHAO MeiFang | 2010 | Chinese Science Bulletin2010,55,34: | 9
Development and validation of a nomogram for predicting the survival of patients with non-metastatic nasopharyngeal carcinoma after curative treatment显示文摘Background: The TNM staging system is far from perfect in predicting the survival of individual cancer patients because only the gross anatomy is considered. The survival rates of the patients who have the same TNM stage disease vary across a wide spectrum. This study aimed to develop a nomogram that incorporates other clinicopathologic factors for predicting the overall survival(OS) of non-metastatic nasopharyngeal carcinoma(NPC) patients after curative treatments. Methods: We retrospectively collected the clinical data of 1520 NPC patients who were diagnosed histologically between November 2000 and September 2003. The clinical data of a separate cohort of 464 patients who received intensity-modulated radiation therapy(IMRT) between 2001 and 2010 were also retrieved to examine the extensibility of the model. Cox regression analysis was used to identify the prognostic factors for building the nomogram. The predictive accuracy and discriminative ability were measured using the concordance index(c-index).Results: We identiied and incorporated 12 independent clinical factors into the nomogram. The calibration curves showed that the prediction of OS was in good agreement with the actual observation in the internal validation set and IMRT cohort. The c-index of the nomogram was statistically higher than that of the 7th edition TNM staging system for predicting the survival in both the primary cohort(0.69 vs. 0.62) and the IMRT cohort(0.67 vs. 0.63).Conclusion: We developed and validated a novel nomogram that outperformed the TNM staging system in predicting the OS of non-metastatic NPC patients who underwent curative therapy. | Wenhua Liang Guanzhu Shen Yaxiong Zhang Gang Chen Xuan Wu Yang Li Anchuan Li Shiyang Kang Xi Yuan Xue Hou Peiyu Huang Yan Huang Hongyun Zhao Ying Tian Chong Zhao Li Zhang | 2016 | Chinese Journal of Cancer2016,35,12: | 6
Soil respiration dynamics in Cinnamomum camphora forest and a nearby Liquidambar formosana forest in Subtropical China显示文摘Climate change can significantly affect carbon cycling of forest ecosystems. The diurnal and seasonal dynamics of soil respiration (Rs) in Cinnamomum camphora and Liquidambar formosana forests were investigated by using infrared gas exchange analyzer of Li-Cor 6400-09 each month in 2006.Soil temperature and moisture were also measured.Diurnal variations in Rs varied with daily soil temperature in the two forests.Across the growing season, soil respiration peaked on July 28 due to higher soil temperature and moisture conditions.Seasonal Rs variations were predominantly influenced by soil temperature.Seasonal soil respirations in C.camphora and L.formosana forests were strongly related to soil temperature (x) at 5-cm depth (Rs=0.1598e 0.1377x (R 2=0.9289,P=0.001) and Rs=0.2177e 0.0962x (R 2=0.927,P=0.000)),while annual mean Rs rate was 2.614 and 1.397 μmol m-2 s-1 and Q 10 values were 3.96 and 2.62 for the two forests, respectively. However,Rs was not influenced by seasonal changes in soil moisture (w) in the two forests in 2006.We developed two equations (Rs=-0.020w 2 +0.497w+0.562 (R 2=0.109,P>0.05),Rs=-0.001w 2 +0.072w +0.731(R 2=0.053,P>0.05)) to describe the relationship between Rs and soil moisture.A positive relationship between Rs and soil moisture were observed when soil moisture was below 12.43% and 18.00%,but when soil moisture exceeded threshold values of 12.43% and 18.00%,soil moisture became the restraint factor for Rs.Long-term observations of soil respiration and moisture are required to understand the temporal dynamics of soil respiration. | TIAN DaLun WANG GuangJun YAN WenDe XIANG WenHua PENG ChangHui | 2010 | Chinese Science Bulletin2010,55,8: | 2
Metabolic reprogramming and redox adaptation in sorafenib-resistant leukemia cells:detected by untargeted metabolomics and stable isotope tracing analysis显示文摘Background:Internal tandem duplications(ITD)within the juxtamembrane domain of FMS-like tyrosine kinase 3(FLT3)represent a poor prognostic indicator in acute myeloid leukemia(AML).Therapeutic benefits of tyrosine kinase inhibitors, such as sorafenib, are limited due to the emergence of drug resistance. While investigations have been con-ducted to improve the understanding of the molecular mechanisms underlying the resistance to this FLT3 inhibitor,a profile of cell functioning at the metabolite level and crosstalk between metabolic pathways has yet to be created. This study aimed to elucidate the alteration of metabolomic profile of leukemia cells resistant to the FLT3 inhibitor. Methods:We established two sorafenib-resistant cell lines carrying FLT3/ITD mutations, namely the murine BaF3/ITD-R and the human MV4-11-R cell lines. We performed a global untargeted metabolomics and stable isotope-labe-ling mass spectrometry analysis to identify the metabolic alterations relevant to the therapeutic resistance. Results:The resistant cells displayed fundamentally rewired metabolic profiles, characterized by a higher demand for glucose, accompanied by a reduction in glucose flux into the pentose phosphate pathway(PPP);and by an increase in oxidative stress, accompanied by an enhanced glutathione synthesis. We demonstrated that the highest scoring network of altered metabolites in resistant cells was related to nucleotide degradation.A stable isotope tracing experiment was performed and the results indicated a decrease in the quantity of glucose entering the PPP in resistant cells.Further experiment suggested that the inhibition of major enzymes in the PPP consist of glucose-6-phosphate dehydrogenase deficiency(G6PD)in the oxidative arm and transketolase(TKT)in the non-oxidative arm.In addition,we observed that chronic treatment with sorafenib resulted in an increased oxidative stress in FLT3/ITD-positive leu-kemia cells, which was accompanied by decreased cell proliferation and an enhanced antioxidant response. Conclusions:Our data regarding comparative metabolomics characterized a distinct metabolic and redox adaptation that may contribute to sorafenib resistance in FLT3/ITD-mutated leukemia cells. | Xin You Weiye Jiang Wenhua Lu Hui Zhang Tiantian Yu Jingyu Tian Shijun Wen Guillermo Garcia-Manero Peng Huang Yumin Hu | 2019 | Cancer Communications2019,39,1: | 1

Novel neutrophil extracellular trap-related mechanisms in diabetic wounds inspire a promising treatment strategy with hypoxia-challenged small extracellular vesicles显示文摘Neutrophil extracellular traps(NETs)have been considered a significant unfavorable factor for wound healing in diabetes, but the mechanisms remain unclear. The therapeutic application of small extracellular vesicles(sEVs)derived from mesenchymal stem cells(MSCs)has received considerable attention for their properties.Hypoxic preconditioning is reported to enhance the therapeutic potential of MSC-derived sEVs in regenerative medicine. Therefore, the aim of this study is to illustrate the detailed mechanism of NETs in impairment of diabetic wound healing and develop a promising NET-targeting treatment based on hypoxic pretreated MSC-derived sEVs(Hypo-sEVs).Excessive NETs were found in diabetic wounds and in high glucose(HG)-induced neutrophils.Further research showed that high concentration of NETs impaired the function of fibroblasts through activating endoplasmic reticulum(ER)stress.Hypo-sEVs efficiently promoted diabetic wound healing and reduced the excessive NET formation by transferring miR-17-5p.Bioinformatic analysis and RNA interference experiment revealed that miR-17-5p in Hypo-sEVs obstructed the NET formation by targeting TLR4/ROS/MAPK pathway.Additionally,miR-17-5p overexpression decreased NET formation and overcame NET-induced impairment in fibroblasts, similar to the effects of Hypo-sEVs.Overall,we identify a previously unrecognized NET-related mechanism in diabetic wounds and provide a promising NET-targeting strategy for wound treatment. | Ziqiang Chu Qilin Huang Kui Ma Xi Liu Wenhua Zhang Shengnan Cui Qian Wei Huanhuan Gao Wenzhi Hu Zihao Wang Sheng Meng Lige Tian Haihong Li Xiaobing Fu Cuiping Zhang | 2023 | Bioactive Materials2023,,9: | 1
Comparative analyses of aging-related genes in long-lived mammals provide insights into natural longevity显示文摘Extreme longevity has evolved multiple times during the evolution of mammals, yet its underlying molecular mechanisms remain largely underexplored. Here,we compared the evolution of 115 aging-related genes in 11 long-lived species and 25 mammals with non-increased lifespan(control group)in the hopes of better understanding the common molecular mechanisms behind longevity. We identified 16 unique positively selected genes and 23 rapidly evolving genes in long-lived species, which included nine genes involved in regulating lifespan through the insulin/IGF-1 signaling(IIS)pathway and 11 genes highly enriched in immune-response-related pathways, suggesting that the IIS pathway and immune response play a particularly important role in exceptional mammalian longevity.Interestingly,11 genes related to cancer progression, including four positively selected genes and seven genes with convergent amino acid changes, were shared by two or more long-lived lineages, indicating that long-lived mammals might have evolved convergent or similar mechanisms of cancer resistance that extended their lifespan. This suggestion was further corroborated by our identifi-cation of 12 robust candidates for longevity-related genes closely related to cancer. | Zhenpeng Yu Inge Seim Mengxin Yin Ran Tian Di Sun Wenhua Ren Guang Yang Shixia Xu | 2021 | The Innovation2021,2,2: | 1

Construction of Tobacco Bax Inhibitor-1 ihpRNA Gene Silencing Vector and Transformation into Agrobacterium tumefaciens EHA105显示文摘The plasmid pGSA1285 was first modified by substituting its GUS sequence with the Chalcone synthase intron fragment from vector pFGC5941 to get the plant silencing expression vector that contained Kanamycin resistance site and was named as pGSA2285. Using PCR-based amplification, two different restriction sites at both ends of tobacco Bax inhibitor-1 (NtBI-1) gene were created, respectively, which made the construction of ihpRNA gene silencing vector more efficiently. Then, NtBI-1 genes were inserted into Multiple Cloning Site (MCS) of pGSA2285 respectively to form Bax inhibitor-1 ihpRNA gene silencing vector, named as pGSA4285, containing sense and anti-sense BI-1 sequence which was spliced by chalcone synthase intron. Combined PCR identification and enzyme restriction analyses, the results showed that Bax inhibitor-1 ihpRNA gene silencing vector had been constructed and transferred into Agrobacterium tumefaciens EHA105 successfully, which laid a foundation for the further study on the function of BI-1 in plant PCD regulation. | ZHU Wenhua XIE Meng LIN Yuheng YANG Mingyu MA Qiumin TIAN Huiqin QU Guiqin | 2011 | Journal of Northeast Agricultural University(English Edition)2011,18,2: | 0

Broadening and enhancing 2.7 μm emission spectra in Er/Ho co-doped oxyfluoride germanosilicate glass ceramics by imparting multiple local structures to rare earth ions显示文摘Er/Ho co-doped oxyfluoride germanosilicate glass and glass ceramics are prepared and compared. The results indicate that the glass consists of SiO_4 and GeO_4 structural units, while the network of the glass ceramics consists of SiO_4, GeO_4, and GeO_6 units together with NaYF_4 nanocrystals. The presence of multiple local structures in glass ceramics creates a range of dipole environments, which is beneficial to the broadening of 2.7 μm emission. Two other reasons are attributed to the broadening of 2.7 μm emission in glass ceramics: the energy-level splitting of Er^(3+) and the enhancement of the Ho^(3+):~5I_6→~5I_7 transition in NaYF_4 nanocrystals. | QUNHUO LIU YING TIAN WENHUA TANG FEIFEI HUANG XUFENG JING JUNJIE ZHANG SHIQING XU | 2018 | Photonics Research2018,6,4: | 0
FAM89A and IFI44L for distinguishing between viral and bacterial infections in children with febrile illness显示文摘Importance: The current lack of reliable rapid tests for distinguishing between bacterial and viral infections has contributed to antibiotic misuse. Objective: This study aimed to develop a novel biomarker assay that integratesFAM89A andIFI44L measurements to assist in differentiating between bacterial and viral infections. Methods: This prospective study recruited children with febrile illness from two hospitals between July 1, 2018, and June 30, 2019. A panel of three experienced pediatricians performed reference standard diagnoses of all patients (i.e., bacterial or viral infection) using available clinical and laboratory data, including a 28-day follow-up assessment. Assay operators were blinded to the reference standard diagnoses. The expression levels ofFAM89A andIFI44L were determined by quantitative real-time polymerase chain reaction assessment. Results: Of 133 potentially eligible patients with suspected bacterial or viral infection, 35 were excluded after the application of exclusion criteria. The resulting cohort included 98 patients: 59 with viral diagnoses and 39 with bacterial diagnoses. The areas under the curve (AUCs) of diagnoses usingFAM89A andIFI44L were 0.694 [95% confidence interval (CI): 0.583-0.804] and 0.751 (95%CI: 0.651-0.851), respectively. The disease risk score (DRS) [log2(FAM89A expression) - log2(IFI44L expression)] signature achieved an improved area under the receiver operating characteristic curve (AUC, 0.825;95%CI: 0.735-0.915), compared with the AUC generated from individual host RNA. A combination of the DRS and the C-reactive protein (CRP) level achieved an AUC of 0.896 (95%CI: 0.825-0.966). Optimal cutoffs for the DRS and CRP level were -3.18 and 19.80 mg/L, respectively.Interpretation: The DRS was significantly more accurate than the CRP level in distinguishing between bacterial and viral infections;the combination of these two parameters exhibited greater sensitivity and specificity. This study provides information that could be useful for the clinical application ofFAM89A andIFI44L in terms of distinguishing between viral and bacterial infections. | Shufeng Tian Jikui Deng Wenhua Huang Linlin Liu Yunsheng Chen Yongqiang Jiang Gang Liu | 2021 | Pediatric Investigation2021,5,3: | 0
