1A Simple DNA Preparation Method for PCR Amplifications in Marker-Assisted Selection of Wheat显示文摘An important, but often limiting step in marker-assisted breeding is the efficient isolation of plant DNA for polymerasechain reaction (PCR) amplification. A simple method using an alkali treatment to extract wheat DNA for marker-assistedselection (MAS) in wheat breeding programs was compared to a commercial kit and cetyltrimethylammonium bromide(CTAB) extraction. DNA concentration from the alkali extraction was higher than the other two methods but purity waslower than CTAB extraction. The alkali extraction method was used on breeding lines to determine its usefulness. 2Computed tomography and magnetic resonance imaging evaluation of pelvic lymph node metastasis in bladder cancer显示文摘Background:Accurate evaluation of lymph node metastasis in bladder cancer(BCa)is important for disease staging,treatment selection,and prognosis prediction.In this study,we aimed to evaluate the diagnostic accuracy of com-puted tomography(CT)and magnetic resonance imaging(MRI)for metastatic lymph nodes in BCa and establish criteria of imaging diagnosis.Methods:We retrospectively assessed the imaging characteristics of 191 BCa patients who underwent radical cys-tectomy.The data regarding size,shape,density,and diffusion of the lymph nodes on CT and/or MRI were obtained and analyzed using Kruskal-Wallis test and χ^(2) test.The optimal cutoff value for the size of metastatic node was deter-mined using the receiver operating characteristic(ROC)curve analysis.Results:A total of 184 out of 3317 resected lymph nodes were diagnosed as metastatic lymph nodes.Among 82 imaging-detectable lymph nodes,51 were confirmed to be positive for metastasis.The detection rate of metastatic nodes increased along with more advanced tumor stage(P<0.001).Once the ratio of short-to long-axis diameter≤0.4 or fatty hilum was observed in lymph nodes on imaging,it indicated non-metastases.Besides,lymph nodes with spiculate or obscure margin or necrosis indicated metastases.Furthermore,the short diameter of 6.8 mm was the optimal threshold to diagnose metastatic lymph node,with the area under ROC curve of 0.815.Conclusions:The probability of metastatic nodes significantly increased with more advanced T stages.Once lymph nodes are detected on imaging,the characteristic signs should be paid attention to.The short diameter>6.8 mm may indicate metastatic lymph nodes in BCa.Yong Li Feiyu Diao Siya Shi Kaiwen Li Wangshu Zhu Shaoxu Wu Tianxin Lin 2018Cancer Communications2018,38,1:7
3Integrated analysis of high-throughput sequencing data shows abscisic acidresponsive genes and miRNAs in strawberry receptacle fruit ripening显示文摘The perception and signal transduction of the plant hormone abscisic acid(ABA)are crucial for strawberry fruit ripening,but the underlying mechanism of how ABA regulates ripening-related genes has not been well understood.By employing high-throughput sequencing technology,we comprehensively analyzed transcriptomic and miRNA expression profiles simultaneously in ABA-and nordihydroguaiaretic acid(NDGA,an ABA biosynthesis blocker)-treated strawberry fruits with temporal resolution.The results revealed that ABA regulated many genes in different pathways,including hormone signal transduction and the biosynthesis of secondary metabolites.Transcription factor genes belonging to WRKY and heat shock factor(HSF)families might play key roles in regulating the expression of ABA inducible genes,whereas the KNOTTED1-like homeobox protein and Squamosa Promoter-Binding-like protein 18 might be responsible for ABA-downregulated genes.Additionally,20 known and six novel differentially expressed miRNAs might be important regulators that assist ABA in regulating target genes that are involved in versatile physiological processes,such as hormone balance regulation,pigments formation and cell wall degradation.Furthermore,degradome analysis showed that one novel miRNA,Fa_novel6,could degrade its target gene HERCULES1,which likely contributed to fruit size determination during strawberry ripening.These results expanded our understanding of how ABA drives the strawberry fruit ripening process as well as the role of miRNAs in this process.Dongdong Li Wangshu Mou Rui Xia Li Li Christopher Zawora Tiejin Ying Linchun Mao Zhongchi Liu Zisheng Luo 2019Horticulture Research2019,6,1:6
4THE ASYMPTOTIC BEHAVIOR OF GLOBAL SMOOTH SOLUTIONS TO THE MACROSCOPIC MODELS FOR SEMICONDUCTORS显示文摘The authors study the asymptotic behavior of the smooth solutions to the Cauchy problems for two macroscopic models (hydrodynamic and drift-diffusion models) for semiconductors and the related relaxation limit problem. First, it is proved that the solutions to these two systems converge to the unique stationary solution time asymptotically without the smallness assump- tion on doping profile.
