| 1 | Ablation of gut microbiota alleviates obesity-induced hepatic steatosis and glucose intolerance by modulating bile acid metabolism in hamsters显示文摘Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesityrelated metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor(FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However,the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet(HFD) were administeredvehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue(sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1(CYP7 B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro-β-muricholic acid(TβMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. | 2 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 3 | Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression. | Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu | 2020 | Cancer Biology & Medicine2020,17,1: | 19 |

This paper aims to establish the finite-time adaptive consensus criterion for a class of MASs with nonlinear dynamics. Traditionally, the finite-time consensus criterion is often established based on the prior information on Lipschitz constants and the eigenvalues of Laplacian matrix. However, it is difficult to acquire the above prior information for most real-world engineering systems. To overcome the above difficulty, this paper develops the finite-time consensus criteria for a class of MASs with nonlinear dynamics via adaptive technique. In detail, we design the finite-time distributed node-based and edge-based adaptive consensus protocols for a class of MASs with fixed and switching topologies. | 6 | Molecular Characterization of a Leaf Senescence-Related Transcription Factor BrWRKY75 of Chinese Flowering Cabbage显示文摘WRKY is a plant-specific transcription factor(TF) involved in the regulation of many biological processes; however, its role in leaf senescence of leafy vegetables remains unknown. In the present work, a WRKY TF, termed Br WRKY75 was isolated from Chinese flowering cabbage [Brassica rapa L. ssp. chinensis(L.) Mokino var. utilis Tsen et Lee]. Analysis of deduced amino acid sequence and the phylogenetic tree showed that Br WRKY75 has high homology with WRKY75 from Brassica oleracea and Arabidopsis thaliana, and belongs to the II c sub-group. Sub-cellular localization and transcriptional activity analysis revealed that Br WRKY75 is a nuclear protein with transcriptional repression activity, and was up-regulated during leaf senescence. Electrophoretic mobility shift assay confirmed that Br WRKY75 directly bound to the W-box(TTGAC) cis-element. | 12 | Reduced macular inner retinal thickness and microvascular density in the early stage of patients with dysthyroid optic neuropathy显示文摘Background:The goal was to investigate changes of the inner intra-retinal layer thicknesses and retinal capillary density(RCD)around the macula in thyroid-associated ophthalmopathy(TAO)patients with or without dysthyroid optic neuropathy(DON).Methods:Forty-four TAO patients including 23 non-DON and 21 DON patients,and 38 healthy participants were enrolled.Spectral domain optical coherence tomography equipped with Angiovue was used to obtain threedimensional retinal thickness maps and microvascular images of the superficial and deep retinal capillary layers(SRCL and DRCL,respectively)around the macula.Quantitative analyses were performed using a custom automated algorithm.Results:The thicknesses of the nerve fiber layer,ganglion cell layer+inner plexiform layer,and ganglion cell complex(GCC)as well as the RCDs in the SRCL and DRCL in both TAO groups were significantly decreased compared to the controls.In addition,the RCDs in DRCL of the DON group were further decreased compared to the non-DON group.GCC thickness in both TAO groups was positively correlated with the RCDs of the SRCL in the total annular zone and in the temporal,inferior,and nasal sectors.The areas under the receiver operating characteristic curves for the GCC thickness combined with the RCD were generally larger than those of each single indicator.Conclusions:Thinned inner intra-retinal layers and decreased RCDs in the TAO patients without DON revealed that morphological changes might precede visual dysfunction.The composite index of the retinal structure and the microvascular density might be valuable in the diagnosing,monitoring,and intervention for early DON. | Yufei Wu Yunhai Tu Chaoming Wu Lulu Bao Jianhua Wang Fan Lu Meixiao Shen Qi Chen | 2020 | Eye and Vision2020,7,1: | 3 |

| 17 | Hazards of extreme events in China under different global warming targets显示文摘Changes in the frequency and intensity of extreme events affect the risk pattern of risk-bearing bodies and the effectiveness of response measures.Based on future climate scenario data,in this study,we identify global warming periods of 1.5℃ and 2℃ above pre-industrial levels,and the temporal and spatial patterns of temperature and precipitation in China during the same period,propose a methodology for risk assessment and integrated analysis of extreme events,build a database of extreme events and their hazards in China in the context of climate change,and assess the hazards caused by extreme events(i.e.,heatwaves,floods,and droughts)in China under different warming periods.Under the RCP4.5 and RCP8.5 scenarios,global temperatures increase by 1.5℃ and 2℃ above preindustrial levels by 2029 and 2026 and by 2050 and 2040,respectively.The warming would be fast in the north and slow in the south,the warming of the Qinghai-Tibet Plateau would accelerate,and the precipitation would increase in the north and decrease in the south,but the trend of the precipitation reduction in the south would be more significant under the RCP8.5 scenario.For 2℃ of warming,the hazard index of severe heatwaves in the North China Plain,parts of South China,and the western part of Inner Mongolia would double that of 1.5℃ warming.Regional differences in extreme precipitation would increase,especially under the RCP8.5 scenario,the hazard index of severe floods in parts of East China would be about twice that of 1.5℃ warming.Under the joint influence of rising temperature and changing precipitation,the drought situation would be alleviated under the RCP4.5 scenario,but it is intensified under the RCP8.5 scenario.For 2℃ of warming,the hazard index of severe droughts in some areas would reach three times that of 1.5℃ warming.For the integrated hazard of the three types of extreme events,the areas of high and medium hazards would expand,while the areas of low hazard would decrease.For 2℃ of warming,the areas of high and medium hazards would be more than half the total area. | Jiangbo Gao Lulu Liu Shaohong Wu | 2020 | Big Earth Data2020,4,2: | 2 |

| 20 | Cardioprotective effect of Yiqi Huoxue granule through regulation of mitophagy after myocardial infarction in rats显示文摘Objective:To investigate the cardioprotective effect of Yiqi Huoxue granule(YQHXG)in the regulation of autophagy in rats induced with myocardial infarction(MI).Methods:An acute MI animal model was established by ligation of the left anterior descending branch of the coronary artery in Sprague-Dawley rats.Besides,20 rats received sham operation were classified into a control group.The remaining 59 rats were randomly divided into MI model group(n=19),YQHXG group(n=20),and perindopril group(n=20).Relevant indicators on days 7 and 28 were observed in each group.Left ventricular function was determined by echocardiography.The structure and morphology of mitochondria,and the number of autophagic vesicles,were observed by transmission electron microscopy.The mRNA and protein expression levels of LC3,FUNDC1,Beclin-1,and BNIP3 were examined in the tissue of the MI marginal area.Results:Compared with the MI model group,YQHXG showed obvious improvements in cardiac functions.Observing the microscopic morphology of the heart tissue,myocardial tissue damage attenuated,autophagic signs of autophagosomes and autolysosomes reduced,vacuolization in mitochondria mitigated,and mitochondria arranged in order.YQHXG could reduce the degree of tissue lesion after MI and regulate the expression of autophagy-related molecules at different stages.On Day 7,YQHXG significantly downregulated the expression of Fundc1,Becn1,Bnip3 mRNA and reduced the levels of FUNDC1,Beclin-1,BNIP3,and LC3 B proteins expression(all P<.001).On Day 28,YQHXG could upregulate the expression of Becn1,Fundc1 and Bnip3 mRNA and increased the levels of the corresponding proteins expression(all P<.001).Besides,it also increased LC3 B protein expression level(P=.0344).Conclusion:YQHXG regulated the expression of mitochondrial autophagy-related factors in myocardial tissue and mitochondrial autophagic activity at different stages to protect the heart following MI. | Xi Chen Jiangong Wu Shuwen Guo Hanrong Mo Tianwei Zhan Fanghe Li Hui Wang Pengfei Feng Lulu Wei | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,2: | 1 |
