1Antimicrobial activity of silver loaded MnO_2 nanomaterials with different crystal phases against Escherichia coli显示文摘Silver-loaded MnO_2 nanomaterials(Ag/MnO_2),including Ag/α-MnO_2,Ag/β-MnO_2,Ag/γ-MnO_2and Ag/δ-MnO_2 nanorods,were prepared with hydrothermal and impregnation methods.The bactericidal activities of four types of Ag/MnO_2 nanomaterials against Escherichia coli were investigated and an inactivation mechanism involving Ag^+ and reactive oxygen species(ROS)was also proposed.The bactericidal activities of Ag/MnO_2 depended on the MnO_2 crystal phase.Among these nanomaterials,Ag/β^-MnO_2 showed the highest bactericidal activity.There was a 6-log decrease in E.coli survival number after treatment with Ag/β^-MnO_2 for120 min.The results of 5,5-dimethyl-l-pyrroline-N-oxide spin-trapping measurements by electron spin resonance indicate OH and O_2^- formation with addition of Ag/β-MnO_2,Ag/γ-MnO_2 or Ag/δ-MnO_2.The strongest peak of OH appeared for Ag/β-MnO_2,while no OH or ·O_2^-signal was found over Ag/α-MnO_2.Through analysis of electron spin resonance(ESR) and Ag+elution results,it could be deduced that the toxicity of Ag^+ eluted from Ag/MnO_2 nanomaterials and ROS played the main roles during the bactericidal process.Silver showed the highest dispersion on the surface of β-MnO_2,which promoted ROS formation and the increase of bactericidal activity.Experimental results also indicated that Ag/MnO_2 induced the production of intracellular ROS and disruption of the cell wall and cell membrane.Lian Wang Hong He Changbin Zhang Li Sun Sijin Liu Shaoxin Wang 2016Journal of Environmental Sciences2016,28,3:6
2Superatomic Aub clusters ligated by different N-heterocyclic carbenes and their ligand-dependent catalysis, photoluminescence, and proton sensitivity显示文摘We report herein a class of superatomic AU13 clusters stabilized by different N-heterocyclic carbenes(NHCs).The clusters show diverse metal surface structures,properties and functions as exemplified by:(1)the first anionic AU13 cluster[Au1g(NHC-1)sBr6],which has bulky NHC-1 ligands that lead to a rather open metal surface contributing to its high catalytic activity;(2)the tricationic cluster[Au13(NHC-2)sBr2]+which has bidentate,benzyl-rich NHC-2 ligands that make it ultra-stable and highly-luminescent,suitable for bio-imaging;and(3)by bearing two pyridyl groups on NHC-3,the dicationic cluster[Au:a(NHC-3)gClg]f+exhibits reversible and stable visible absorption and solubility responses to protonation/deprotonation cycles,making it a potential pH sensor(NHC-1=1,3-disopropylbenzimidazolin-ylidene;NHC-2=1,3-bis(1-benzyl-1H-benzimidazol-1-ium-yl)propane;NHC-3=1,3-bis(icolyI)benzimidazolin-ylidene).The study nicely demonstrates the importance of ligands in designing metal nanoclusters with desired functionalities.Hui Shen Sijin Xiang Zhen Xu Chen Liu Xihua Li Cunfa Sun Shuichao Lin Boon K.Teo Nanfeng Zheng 2020Nano Research2020,13,7:6
3Primary cilia in hard tissue development and diseases显示文摘Bone and teeth are hard tissues.Hard tissue diseases have a serious effect on human survival and quality of life.Primary cilia are protrusions on the surfaces of cells.As antennas,they are distributed on the membrane surfaces of almost all mammalian cell types and participate in the development of organs and the maintenance of homeostasis.Mutations in cilium-related genes result in a variety of developmental and even lethal diseases.Patients with multiple ciliary gene mutations present overt changes in the skeletal system,suggesting that primary cilia are involved in hard tissue development and reconstruction.Furthermore,primary cilia act as sensors of external stimuli and regulate bone homeostasis.Specifically,substances are trafficked through primary cilia by intraflagellar transport,which affects key signaling pathways during hard tissue development.In this review,we summarize the roles of primary cilia in long bone development and remodeling from two perspectives:primary cilia signaling and sensory mechanisms.In addition,the cilium-related diseases of hard tissue and the manifestations of mutant cilia in the skeleton and teeth are described.We believe that all the findings will help with the intervention and treatment of related hard tissue genetic diseases.Sijin Li Han Zhang Yao Sun 2021Frontiers of Medicine2021,15,5:4
4A non-metabolic function of hexokinase 2 in small cell lung cancer:promotes cancer cell stemness by increasing USP11-mediated CD133 stability显示文摘Background:Maintenance of cancer stem-like cell(CSC)stemness supported by aberrantly regulated cancer cell metabolism is critical for CSC self-renewal and tumor progression.As a key glycolytic enzyme,hexokinase 2(HK2)plays an instrumental role in aerobic glycolysis and tumor progression.However,whether HK2 directly contribute to CSC stemness maintenance in small cell lung cancer(SCLC)is largely unclear.In this study,we aimed to investgate whether HK2 independent of its glycolytic activity is directly involved in stemness maintenance of CSC in SCLC.Methods:Immunoblotting analyses were conducted to determine the expression of HK2 in SCLC CSCs and their differentiated counterparts.CSC-like properties and tumorigenesis of SCLC cells with or without HK2 depletion or overexpression were examined by sphere formation assay and xenograft mouse model.Immunoprecipitation and mass spectrometry analyses were performed to identify the binding proteins of CD133.The expression levels of CD133-associated and CSC-relevant proteins were evaluated by immunoblotting,immunoprecipitation,immunofluorescence,and immunohistochemistry assay.RNA expression levels of Nanog,POU5F1,Lin28,HK2,Prominin-1 were analyzed through quantitative reverse transcription PCR.Polyubiquitination of CD133 was examined by in vitro or in vivo ubiquitination assay.CD133+cells were sorted by flow cytometry using an anti-CD133 antibody.Results:We demonstrated that HK2 expression was much higher in CSCs of SCLC than in their differentiated counterparts.HK2 depletion inhibited CSC stemness and promoted CSC differentiation.Mechanistically,nonmitochondrial HK2 directly interacted with CD133 and enhanced CD133 expression without affecting CD133 mRNA levels.The interaction of HK2 and CD133 promoted the binding of the deubiquitinase ubiquitin-specific protease 11(USP11)to CD133,thereby inhibiting CD133 polyubiquitylation and degradation.HK2-mediated upregulation of CD133 expression enhanced the expression of cell renewal regulators,SCLC cell stemness,and tumor growth in mice.In addition,HK2 expression was positively correlated with CD133 expression in human SCLC specimens,and their expression levels were associated with poor prognosis of SCLC patients.Conclusions:These results revealed a critical non-metabolic function of HK2 in promotion of cancer cell stemness.Our findings provided new insights into the multifaceted roles of HK2 in tumor development.Juhong Wang Fei Shao Yannan Yang Wei Wang Xueying Yang Renda Li Hong Cheng Sijin Sun Xiaoli Feng Yibo Gao Jie He Zhimin Lu 2022Cancer Communications2022,42,10:2
5Application of bacteria-plant association in bio-degradation of diesel oil pollutants in soil显示文摘The bacteria and plants were associated to remove diesel oil pollutants from soil.Three efficiently degrading bacteria(named strains Q10,Q14 and Q18,respectively) were isolated.Two plants(alfalfa and Indian mustard) were selected to form the association.Biodegradation of diesel oil pollutants in soil was accelerated by bacteria-plants association.The main results are summarized as follows.The plants-bacteria association was more effective in biodegradation of diesel oil pollutants in soil than in respective experiments carried out with plants or bacteria alone.Strain Q18-Indian mustard association resulted in the maximum diesel oil reduction(69.18%).The activities of catalase and polyphenol oxidase in soil were enhanced and microbial populations in soil,especially in rhizosphere,were also stimulated in the treatment of bacteria-plant association.Overall,the soil conditions might be improved by alfalfa or Indian mustard to benefit the growth of bacteria,which resulted in degradation of diesel oil pollutants more effective by the bacteria-plant association.The bacteria-plants association may be a better approach to the removal of diesel oil pollutants from soil.Lu, Sijin Teng, Yanguo Sun, Zongjian Wang, Jinsheng 2011Chinese Journal Of Geochemistry2011,30,2:2

7CCGD-ESCC: A Comprehensive Database for Genetic Variants Associated with Esophageal Squamous Cell Carcinoma in Chinese Population显示文摘Esophageal squamous-cell carcinoma(ESCC) is one of the most lethal malignancies in the world and occurs at particularly higher frequency in China. While several genome-wide association studies(GWAS) of germline variants and whole-genome or whole-exome sequencing studies of somatic mutations in ESCC have been published, there is no comprehensive database publically available for this cancer. Here, we developed the Chinese Cancer Genomic Database-Esophageal Squamous Cell Carcinoma(CCGD-ESCC) database, which contains the associations of 69,593 single nucleotide polymorphisms(SNPs) with ESCC risk in 2022 cases and 2039 controls, survival time of 1006 ESCC patients(survival GWAS) and gene expression(expression quantitative trait loci,eQTL) in 94 ESCC patients. Moreover, this database also provides the associations between8833 somatic mutations and survival time in 675 ESCC patients. Our user-friendly database is a resource useful for biologists and oncologists not only in identifying the associations of genetic variants or somatic mutations with the development and progression of ESCC but also in studying the underlying mechanisms for tumorigenesis of the cancer. 8Facile discovery of red blood cell deformation and compromised membrane/skeleton assembly in Prader—Willi syndrome显示文摘Prader—Willi syndrome(PWS)is a rare congenital disease with genetic alterations in chromosome 15.Although genetic disorders and DNA methylation abnormalities involved in PWS have been investigated to a significant degree,other anomalies such as those in erythrocytes may occur and these have not been clearly elucidated.In the present study,we uncovered slight anemia in children with PWS that was associated with increased red blood cell(RBC)distribution width(RDW)and contrarily reduced hematocrit(HCT)values.Intriguingly,the increased ratio in RDW to HCT allowed sufficient differentiation between the PWS patients from the healthy controls and,importantly,with individuals exhibiting conventional obesity.Further morphologic examinations revealed a significant deformity in erythrocytes and mild hemolysis in PWS patients.Comprehensive mechanistic investigations unveiled compromised membrane skeletal assembly and membrane lipid composition,and revealed a reduced F-actin/G-actin ratio in PWS patients.We ascribed these phenotypic changes in erythrocytes to the observed genetic defects,including DNA methylation abnormalities.Our collective data allowed us to uncover RBC deformation in children with PWS,and this may constitute an auxiliary indicator of PWS in early childhood.Yashuang Yang Guimei Li Yanzhou Wang Yan Sun Chao Xu Zhen Wei Shuping Zhang Ling Gao Sijin Liu Jiajun Zhao 2022Frontiers of Medicine2022,16,6:0
9Scalable synthesis of lipid nanoparticles for nucleic acid drug delivery using an isometric channel-size enlarging strategy显示文摘Lipid nanoparticles(LNPs)have emerged as highly effective delivery systems for nucleic acid-based therapeutics.However,the broad clinical translation of LNP-based drugs is hampered by the lack of robust and scalable synthesis techniques that can consistently produce formulations from early development to clinical application.In this work,we proposed a method to achieve scalable synthesis of LNPs by scaling inertial microfluidic mixers isometrically in three dimensions.Moreover,a theoretical predictive method,which controls the mixing time to be equal across different chips,is developed to ensure consistent particle size and size distribution of the synthesized LNPs.LNPs loaded with small interfering RNA(siRNA)were synthesized at different flow rates,exhibiting consistent physical properties,including particle size,size distribution and encapsulation efficiency.This work provides a practical approach for scalable synthesis of LNPs consistently,offering the potential to accelerate the transition of nucleic acid drug development into clinical application.Zesen Ma Haiyang Tong Sijin Lin Li Zhou Demeng Sun Baoqing Li Changlin Tian Jiaru Chu 2024Nano Research2024,17,4:0
